| 1 | Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility显示文摘Premature ovarian failure(POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study,umbilical cord mesenchymal stem cells on a collagen scaffold(collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3 a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follicles. Successful clinical pregnancy was achieved in women with POF after transplantation of collagen/UC-MSCs or UC-MSCs. | 2 | Transplantation of collagen scaffold with autologous bone marrow mononuclear cells promotes functional endometrium reconstruction via downregulating ΔNp63 expression in Asherman's syndrome显示文摘Asherman's syndrome(AS) is a common disease that presents endometrial regeneration disorder. However, little is known about its molecular features of this aregenerative endometrium in AS and how to reconstruct the functioning endometrium for the patients with AS. Here, we report that ΔNp63 is significantly upregulated in residual epithelial cells of the impaired endometrium in AS; the upregulated-ΔNp63 induces endometrial quiescence and alteration of stemness. Importantly, we demonstrate that engrafting high density of autologous bone marrow mononuclear cells(BMNCs) loaded in collagen scaffold onto the uterine lining of patients with AS downregulates ΔNp63 expression, reverses ΔNp63-induced pathological changes, normalizes the stemness alterations and restores endometrial regeneration. Finally, five patients achieved successful pregnancies and live births. Therefore, we conclude that ΔNp63 is a crucial therapeutic target for AS. | 4 | Re-Os and U-Pb Geochronology of the Erlihe Pb-Zn Deposit,Qinling Orogenic Belt,Central China,and Constraints on Its Deposit Genesis显示文摘The Erlihe Pb-Zn deposit is an important mine of the Pb-Zn metallogenic zone in the South Qinling Orogen.It has been considered a sedimentary exhalative deposit in previous investigations because the ore body occurs concordantly at the transitional location of an upright fold.Re and Os isotopic analyses for paragenetic pyrites with sphalerite and galena from the ore body have been used to determine the timing of mineralization and to trace the source of metallogenic materials.The Re-Os isotopic data of four pyrite samples construct an isochron,yielding a weighted average age of 226±17 Ma(mean square weighted deviation=1.7),which is considered the main mineralization age.A dioritic porphyrite vein sample,showing weaker mineralization,was also dated using the SHRIMP zircon UPb isotopic method to constrain the youngest metallogenic age of the ore deposit,because it distributes along a group of tensional joints cutting not only the upright fold in the deposit field,but also the main ore bodies.The dioritic porphyrite sample yields a weighted mean ^(206)Pb/^(238)U age of 221±3 Ma,which is slightly younger than the Re-Os isotopic isochron age of the pyrites,considered as the upper age limit of the mineralization,namely the ending age of the mineralization.The Os isotopic compositions of sulfide minerals distribute within a range between Os isotopic compositions of the crust and the mantle, indicating that the ore deposit can be derived from magma-related fluid,and the metallogenic materials are most likely derived from the mixing source of the crust and the mantle.The Erlihe Pb-Zn deposit and associated dioritic porphyrite vein,important records of Qinling tectonic-magmatism-mineralization activities,were formed during the Triassic collisional orogeny processes. | ZHANG Fan LIU Shuwen LI Qiugen SUN Yali WANG Zongqi YAN Quanren YAN Zhen | 2011 | Acta Geologica Sinica(English Edition)2011,85,3: | 30 |
| 5 | Indosinian isotope ages of plutons and deposits in southwestern Miaoershan-Yuechengling,northeastern Guangxi and implications on Indosinian mineralization in South China显示文摘The Miaoershan-Yuechengling complex pluton is the largest granitoid complex in the western Nanling metallogenic belt with a surface exposure of >3000 km2.The complex pluton is composed of an early stage granitoid batholith and late stage small intrusions.The early stage batholith contains mainly medium-grained porphyritic mica granite and porphyritic monzonite granite,whereas the late stage intrusions are composed of muscovite granite porphyry and muscovite monzonitic granite.There are many W-Sn-Mo-Pb-Zn-Cu ores in the contact zone between the batholith and strata,forming an ore-rich belt around the batholith.Based on zircon LA-ICP-MS U-Pb ages,the southwestern part of the early stage batholith formed at 228.7 ± 4.1 Ma(MSWD = 2.49),with slightly earlier magmatic activity at 243.0 ± 5.8 Ma(MSWD = 2.62).The Yuntoujie muscovite granite was associated with W-Mo mineralization and has a zircon LA-ICP-MS U-Pb age of 216.8 ± 4.9 Ma(MSWD = 1.44).The Re-Os isochron age of molybdenite from the Yuntoujie W-Mo ore was 216.8 ± 7.5 Ma(MSWD = 11.3).Our new isotope data suggest that the late stage intrusive stocks from the southwestern Miaoershan-Yuechengling batholith were closely associated with W-Mo mineralization from the Indosinian period.These new results together with previous isotope data,suggest that South China underwent not only the well-known Yanshanian mineralization event,but also a widespread Indosinian metallogenic event during the Mesozoic period.Therefore,South China has a greater potential for Indosinian mineralization than previously thought such that more attention should be given to the Indosinian ore prospecting in South China. | WU Jing LIANG HuaYing HUANG WenTing WANG ChunLong SUN WeiDong SUN YaLi LI Jing MO JiHai WANG XiuZhang | 2012 | Chinese Science Bulletin2012,57,9: | 28 |
| 6 | Efficacy and safety of Xinfeng capsule in patients with rheumatoid arthritis:a multi-center parallel-group double-blind randomized controlled trial显示文摘OBJECTIVE:To evaluate the efficacy and safety of Xinfeng capsule in patients suffering rheumatoid arthritis(RA).METHODS:A multi-center parallel-group designed,double-blind,randomized,controlled trial was conducted.Totally 304 RA patients were assigned to two groups:one group was administered Xinfeng capsule(XFC) plus the placebo of leflunomide and the other given leflunomide(LEF) plus the placebo of XFC for twelve weeks.The clinical and laboratory parameters were compared at baseline and fourth,eighth,and twelfth weeks.RESULTS:After twelve-week treatment,patients in two groups all showed some trend of effectiveness when compared in terms of American Rheumatism Association(ACR) recommended 20%,50%,70%improvement criteria,but it was insignificant.The validity in ameliorate modified disease activity score(DAS28) and laboratory indexes as erythrocyte sedimentation rate(ESR),C-reactive protein(CRP),rheumatoid factor(RF) were also found no difference.The score of health assessment questionnaire(HAQ),self-rating anxiety scale(SAS),self-rating depression scale(SDS) and quality of life questionnaire with rheumatoid arthritis(RAQOL)both lower than the first week and the changes showed no difference.However,the score of SDS dropped more in XFC group than in the other.A total of 147 adverse reaction cases were reported,which shows no difference between the two groups.The most common adverse reactions were hepatic impairment,anemia,leukocytopenia,epigastric discomfort and phalacrosis.CONCLUSION:XFC demonstrated better improvement in the scores of SDS and compared with those of LEF group. | Liu Jian Wang Yuan Huang Chuanbing Xu Jianhua Li Zhijun Xu Liang He Liyun Sun Yue Wang Yali Xu Shengqian Zhao Ping Mao Tongjun Tan Bin Zhu Fubing Zhang Pingheng Fang Li | 2015 | Journal of Traditional Chinese Medicine2015,35,5: | 27 |
| 7 | Heat and power load dispatching considering energy storage of district heating system and electric boilers显示文摘As one of promising clean and low-emission energy, wind power is being rapidly developed in China.However, it faces serious problem of wind curtailment,particularly in northeast China, where combined heat and power(CHP) units cover a large proportion of the district heat supply. Due to the inherent strong coupling between the power and the heat load, the operational flexibility of CHP units is severely restricted in winter to meet the heat supply demand, which imparts considerable stress on the wind power connection to the grid. To promote the integration of wind power and enhance the flexibility of CHP units, this paper presented a method of heat and power load dispatching by exploring the energy storage ability of electric heating boilers and district heating systems. The optimization results indicate that the proposed method can integrate additional wind power into the grid and reduce the coal consumption of CHP units over the optimized period. | 8 | A Review of Seasonal Climate Prediction Research in China显示文摘The ultimate goal of climate research is to produce climate predictions on various time scales. In China, efforts to predict the climate started in the 1930 s. Experimental operational climate forecasts have been performed since the late 1950 s,based on historical analog circulation patterns. However, due to the inherent complexity of climate variability, the forecasts produced at that time were fairly inaccurate. Only from the late 1980 s has seasonal climate prediction experienced substantial progress, when the Tropical Ocean and Global Atmosphere project of the World Climate Research program(WCRP) was launched. This paper, following a brief description of the history of seasonal climate prediction research, provides an overview of these studies in China. Processes and factors associated with the climate variability and predictability are discussed based on the literature published by Chinese scientists. These studies in China mirror aspects of the climate research effort made in other parts of the world over the past several decades, and are particularly associated with monsoon research in East Asia. As the climate warms, climate extremes, their frequency, and intensity are projected to change, with a large possibility that they will increase. | 9 | Assimilating Doppler radar observations with an ensemble Kalman filter for convection-permitting prediction of convective development in a heavy rainfall event during the pre-summer rainy season of South China显示文摘This study examines the effectiveness of an ensemble Kalman filter based on the weather research and forecasting model to assimilate Doppler-radar radial-velocity observations for convection-permitting prediction of convection evolution in a high-impact heavy-rainfall event over coastal areas of South China during the pre-summer rainy season. An ensemble of 40 deterministic forecast experiments(40 DADF) with data assimilation(DA) is conducted, in which the DA starts at the same time but lasts for different time spans(up to 2 h) and with different time intervals of 6, 12, 24, and 30 min. The reference experiment is conducted without DA(NODA).To show more clearly the impact of radar DA on mesoscale convective system(MCS)forecasts, two sets of 60-member ensemble experiments(NODA EF and exp37 EF) are performed using the same 60-member perturbed-ensemble initial fields but with the radar DA being conducted every 6 min in the exp37 EF experiments from 0200 to0400 BST. It is found that the DA experiments generally improve the convection prediction. The 40 DADF experiments can forecast a heavy-rain-producing MCS over land and an MCS over the ocean with high probability, despite slight displacement errors. The exp37 EF improves the probability forecast of inland and offshore MCSs more than does NODA EF. Compared with the experiments using the longer DA time intervals, assimilating the radial-velocity observations at 6-min intervals tends to produce better forecasts. The experiment with the longest DA time span and shortest time interval shows the best performance.However, a shorter DA time interval(e.g., 12 min) or a longer DA time span does not always help. The experiment with the shortest DA time interval and maximum DA window shows the best performance, as it corrects errors in the simulated convection evolution over both the inland and offshore areas. | 12 | DDIT4 promotes gastric cancer proliferation and tumorigenesis through the p53 and MAPK pathways显示文摘Background:Gastric cancer(GC)is one of the most common malignancies worldwide,particularly in China.DNA damage-inducible transcript 4(DDIT4)is a mammalian target of rapamycin inhibitor and is induced by various cellular stresses;however,its critical role in GC remains poorly understood.The present study aimed to investigate the poten-tial relationship and the underlying mechanism between DDIT4 and GC development.Methods:We used western blotting,real-time polymerase chain reaction,and immunohistochemical or immunoflu-orescence to determine DDIT4 expression in GC cells and tissues.High-content screening,cell counting kit-8 assays,colony formation,and in vivo tumorigenesis assays were performed to evaluate cell proliferation.Flow cytometry was used to investigate cell apoptosis and cell cycle distribution.Results:DDIT4 was upregulated in GC cells and tissue.Furthermore,downregulating DDIT4 in GC cells inhibited proliferation both in vitro and in vivo and increased 5-fluorouracil-induced apoptosis and cell cycle arrest.In contrast,ectopic expression of DDIT4 in normal gastric epithelial cells promoted proliferation and attenuated chemosensitivity.Further analysis indicated that the mitogen-activated protein kinase and p53 signaling pathways were involved in the suppression of proliferation,and increased chemosensitivity upon DDIT4 downregulation.Conclusion:DDIT4 promotes GC proliferation and tumorigenesis,providing new insights into the role of DDIT4 in the tumorigenesis of human GC. | Feng Du Lina Sun Yi Chu Tingyu Li Chao Lei Xin Wang Mingzuo Jiang Yali Min Yuanyuan Lu Xiaodi Zhao Yongzhan Nie Daiming Fan | 2018 | Cancer Communications2018,38,1: | 10 |

| 15 | Effect of Xinfeng capsule in the treatment of active rheumatoid arthritis: a randomized controlled trial显示文摘OBJECTIVE: To explore the use of Xinfeng capsule(XFC) in the treatment of active rheumatoid arthritis(RA) and its effect on immunoglobulin titer, B cell-activating factor(BAFF) and its receptor(BAFF-R).METHODS: A multi-center randomized, doubleblind, parallel-controlled study was conducted. 45 RA patients were assigned to two groups: one was treated with XFC plus the placebo for leflunomide(LEF) and the second group was treated with LEF plus XFC placebo, for 12 weeks. The clinical and laboratory parameters were collected at baseline and at 12 weeks.RESULTS: After 12 weeks of treatment, patients in the two groups all showed an therapeutic effect when ACR20, ACR50 and ACR70 were compared,but the differences between two groups were not significant(P < 0.05). The serum levels of Ig G1,BAFF and BAFF-R in the XFC group were lower than those in the LEF group(P < 0.05). | 16 | A Study of the Rhenium-Osmium Geochronometry of Molybdenites显示文摘This study deals with the first application of the isotope dilution - inductively coupled plasma mass spectrometry in the rhenium-osmium geochronometry in China. The chemical separation procedure included decomposition of samples by means of alkali fusion, extraction of rhenium by acetone and extraction of osmium by distillation. The recovery of both elements in the entire process was more than 90% . The blanks of analyses were 0.07 ng for rhenium and 0.01 ng for 187Os. The mineralization ages of molybdenites from four molybdenum deposits in China were determined by using this method, with a precision (2σ)of about ±3% . These results agree satisfactorily with their geological backgrounds. The Re-Os method can directly determine ages of metal deposits and reflect the true ages more reliably than other dating methods that only determine the ages of country rocks from which mineralization ages are inferred. | 17 | Preparation of 5-fluorouracil-loaded chitosan nanoparticles and study of the sustained release in vitro and in vivo显示文摘The sustained-release properties of the biodegradable nano-drug delivery systems were used to improve the residence time of the chemotherapeutic agent in the body. These drug delivery systems were widely used to deliver chemotherapeutic drugs. The 5-fluorouracil loaded chitosan nanoparticles prepared in this paper have the above advantage. Here, we found that when the mass ratio of 5-fluorouracil and chitosan was 1:1, the maximum drug loading of nanoparticles was 20.13 ± 0.007%, the encapsulation efficiency was 44.28 ± 1.69%, the particle size was 283.9 ± 5.25 nm and the zeta potential was 45.3 ± 3.23 mV. The prepared nanoparticles had both burst-release and sustained-release phases in vitro release studies.In addition, the inhibitory effect of the prepared nanoparticles on gastric cancer SGC-7901 cells was similar to that of 5-fluorouracil injection, and the blank vector had no obvious inhibitory effect on SGC-7901 cells. | 19 | Viral and Antibody Kinetics of COVID-19 Patients with Different Disease Severities in Acute and Convalescent Phases:A 6-Month Follow-Up Study显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),has spread rapidly around the world,posing a major threat to human health and the economy.Currently,long-term data on viral shedding and the serum antibody responses in COVID-19 patients are still limited.Herein,we report the clinical features,viral RNA loads,and serum antibody levels in a cohort of 112 COVID-19 patients admitted to the Honghu People’s Hospital,Hubei Province,China.Overall,5.36%(6/112)of patients showed persistent viral RNA shedding(>45 days).The peak viral load was higher in the severe disease group than in the mild group(median cycle threshold value,36.4 versus 31.5;P=0.002).For most patients the disappearance of IgM antibodies occurred approximately 4–6 weeks after symptoms onset,while IgG persisted for over 194 days after the onset of symptoms,although patients showed a 46%reduction in antibodies titres against SARS-CoV-2 nucleocapsid protein compared with the acute phase.We also studied18 asymptomatic individuals with RT-qPCR confirmed SARS-CoV-2 infection together with 17 symptomatic patients,and the asymptomatic individuals were the close contacts of these symptomatic cases.Delayed IgG seroconversion and lower IgM seropositive rates were observed in asymptomatic individuals.These data indicate that higher viral loads and stronger antibody responses are related to more severe disease status in patients with SARS-CoV-2 infection,and the antibodies persisted in the recovered patient for more than 6 months so that the vaccine may provide protection against SARS-CoV-2 infection. | Xiaoyong Zhang Suwen Lu Hui Li Yi Wang Zhen Lu Zhihong Liu Qingtao Lai Yali Ji Xuan Huang Yongyin Li Jian Sun Yingsong Wu Xiaoning Xu Jinlin Hou | 2020 | Virologica Sinica2020,35,6: | 4 |
| 20 | Mechanism studies of Xinfeng capsule on improving cardiovascular function though toll-like receptor 4/nuclear factor kappa B pathway in a rat model of adjuvant arthritis显示文摘OBJECTIVE: To observe the impact of Xinfeng capsule(XFC) on cardiovascular function in adjuvant arthritis(AA) model rats and investigate the mechanism though toll-like receptor 4(TLR4)/nuclear factor kappa B(NF-κB) signaling pathway.METHODS: Seventy rats were randomly divided into seven groups: normal control(NC), model control(MC), tripterygium glycosides tablet(TPT),methotrexate(MTX), high, moderate and low dose XFC group. The administration began from day 19 after modeling for 30 day. Paw swelling, arthritic in-dex(AI), cardiac function indexes and myocardial pathological pattern were detected. The expression of TLR4, myeloid differentiation factor(My D) 88, interleukin-1 receptor-associated kinase(IRAK) 1, tumor necrosis factor receptor associated factor(TRAF) 6, NF-κB, tumor necrosis factor-alpha(TNF-α) proteins in myocardial tissue were determined by western blot method.RESULTS: Paw swelling and AI in MC group increased in MC group(P < 0.01), and decreased in high and moderate dose XFC groups(P < 0.01 or P > 0.05). Left ventricular systolic pressure(LVSP),left ventricular end-diastolic pressure(LVEDP),heart rate(HR) were elevated in MC group(P <0.01), and ± dp/dtmax and CI were reduced(P <0.01); while LVSP, LVEDP and HR declined and ±dp/dtmax, CI improved in high dose XFC group(P <0.05 or P < 0.01). LVSP in high dose XFC group were reduced more than other treatment groups(P <0.05 or P < 0.01). The improvements on LVEDP, dp/dt-max were superior to MTX and low dose XFC group, and the improvement on CI was better than low dose XFC group(P < 0.05 or P < 0.01). Myocardial fibers arranged irregular in MC group with intracellular edema and mitochondria damage. The modifications on myocardial structural were shown in each treatment group, but more prominent in TPT, high and moderate dose XFC group. The proteins of TLR4, My D88, IRAK1, TRAF6, NF-κB, TNF-αwere highly expressed in MC group, and those proteins declined in high and moderate dose XFC group(P < 0.05 or P < 0.01).
