| 2 | Arabidopsis calcium-dependent protein kinase CPK28 is potentially involved in the response to osmotic stress显示文摘As calcium sensors, plant calcium-dependent protein kinases(CDPKs) play important roles in plants' responses to various abiotic stresses. Here, we report the functional characterization of CPK28, a member of the CDPK family in Arabidopsis, in response to osmotic stress.The cpk28 mutant, a loss-of-function mutant, exhibited an NaCl- and mannitol-sensitive phenotype in green cotyledons, while CPK28-overexpressing plants displayed stronger tolerance to NaCl and mannitol stresses than wildtype plants. Reverse transcriptase-polymerase chain reaction and beta-glucuronidase staining assays showed that NaCl and mannitol stresses induced CPK28. CPK28-overexpressing lines accumulated significantly more proline relative to wild-type plants and mutant plants under NaCl and mannitol stresses. Transient expression of CPK28-GFP in mesophyll cell protoplasts, as well as stable transgenic lines expressing CPK28-GFP, showed that CPK28 was localized in the plasma membrane. Expression levels of known stress-responsive genes were not significantly altered in the null mutant and overexpression lines,suggesting that CPK28 possibly mediated the stress response via the regulation of target proteins rather than via regulation at the level of transcription. Meanwhile, CPK28could autophosphorylate. | 4 | Characterization of a Novel Anti-DR5 Monoclonal Antibody WD1 with the Potential to Induce Tumor Cell Apoptosis显示文摘TNF-related apoptosis-inducing ligand (TRAIL) is a TNF family member capable of inducing apoptosis. Death receptor 5 (DR 5) is a key receptor of TRAIL and plays an important role in TRAIL-induced apoptosis. To prepare monoclonal antibodies (mAbs) against DR5,cDNA encoding soluble DR5 (sDR5) was firstly amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) with specific primers,and then inserted into a prokaryotic expression vector pET-30a. The recombinant plasmid was expressed in Escherichia coli strain BL21 (DE3),and sDR5 was purified by nickel affinity chromatography. As an antigen,sDR5 was used to immunize mice. Hybridomas secreting antibodies against sDR5 were identified. One positive clone was selected to produce antibody,WD1. ELISA and immunofluorescence demonstrated that WD1 could bind recombinant sDR5 and membrane-bound DR5 (mDR5) on Jurkat and Molt-4 cells. ATPLite assays showed that Jurkat and Molt-4 cells were sensitive to the antibody in a dose dependent manner. The Annexin V/PI assays and Giemsa's staining both showed that WD1 could induce Jurkat cell apoptosis efficiently. Transient transfection of 293T cells and indirect immunofluorescence assay demonstrated that mAb (WD1) couldn't cross-react with DR4. Our findings indicated that the novel antibody,WD1 could act as a direct agonist,bind DR5 characteristically,and initiate efficient apoptotic signaling and tumor regression. | 5 | All-optical computing based on convolutional neural networks显示文摘The rapid development of information technology has fueled an ever-increasing demand for ultrafast and ultralow-en-ergy-consumption computing.Existing computing instruments are pre-dominantly electronic processors,which use elec-trons as information carriers and possess von Neumann architecture featured by physical separation of storage and pro-cessing.The scaling of computing speed is limited not only by data transfer between memory and processing units,but also by RC delay associated with integrated circuits.Moreover,excessive heating due to Ohmic losses is becoming a severe bottleneck for both speed and power consumption scaling.Using photons as information carriers is a promising alternative.Owing to the weak third-order optical nonlinearity of conventional materials,building integrated photonic com-puting chips under traditional von Neumann architecture has been a challenge.Here,we report a new all-optical comput-ing framework to realize ultrafast and ultralow-energy-consumption all-optical computing based on convolutional neural networks.The device is constructed from cascaded silicon Y-shaped waveguides with side-coupled silicon waveguide segments which we termed“weight modulators”to enable complete phase and amplitude control in each waveguide branch.The generic device concept can be used for equation solving,multifunctional logic operations as well as many other mathematical operations.Multiple computing functions including transcendental equation solvers,multifarious logic gate operators,and half-adders were experimentally demonstrated to validate the all-optical computing performances.The time-of-flight of light through the network structure corresponds to an ultrafast computing time of the order of several picoseconds with an ultralow energy consumption of dozens of femtojoules per bit.Our approach can be further expan-ded to fulfill other complex computing tasks based on non-von Neumann architectures and thus paves a new way for on-chip all-optical computing. | Kun Liao Ye Chen Zhongcheng Yu Xiaoyong Hu Xingyuan Wang Cuicui Lu Hongtao Lin Qingyang Du Juejun Hu Qihuang Gong | 2021 | Opto-Electronic Advances2021,4,11: | 3 |
| 6 | Activation of medial septum cholinergic neurons restores cognitive function in temporal lobe epilepsy显示文摘Cognitive impairment is the most common complication in patients with temporal lobe epilepsy with hippocampal scle rosis.There is no effective treatment for cognitive impairment.Medial septum cholinergic neurons have been reported to be a potential target for controlling epileptic seizures in tempo ral lobe epile psy.However,their role in the cognitive impairment of temporal lobe epilepsy remains unclear.In this study,we found that patients with temporal lobe epile psy with hippocampal sclerosis had a low memory quotient and severe impairment in verbal memory,but had no impairment in nonverbal memory.The cognitive impairment was slightly correlated with reduced medial septum volume and medial septum-hippocampus tra cts measured by diffusion tensor imaging.In a mouse model of chronic temporal lobe epilepsy induced by kainic acid,the number of medial septum choline rgic neurons was reduced and acetylcholine release was reduced in the hippocampus.Furthermore,selective apoptosis of medial septum cholinergic neurons mimicked the cognitive deficits in epileptic mice,and activation of medial septum cholinergic neurons enhanced hippocampal acetylcholine release and restored cognitive function in both kainic acid-and kindling-induced epile psy models.These res ults suggest that activation of medial septum cholinergic neurons reduces cognitive deficits in temporal lobe epilepsy by increasing acetylcholine release via projections to the hippocampus. | Junzi Chen Yu Wang Cong Chen Qingyang Zhang Shuang Wang Yi Wang Jiajia Fang Ying Wang | 2023 | Neural Regeneration Research2023,18,11: | 1 |

| 13 | A chromosome-level genome assembly provides insights into Cornus wilsoniana evolution, oil biosynthesis, and floral bud development显示文摘Cornus wilsoniana W.is a woody oil plant with high oil content and strong hypolipidemic effects,making it a valuable species for medicinal,landscaping,and ecological purposes in China.To advance genetic research on this species,we employed PacBio together with Hi-C data to create a draft genome assembly for C.wilsoniana.Based on an 11-chromosome anchored chromosome-level assembly,the estimated genome size was determined to be 843.51 Mb.The N50 contig size and N50 scaffold size were calculated to be 4.49 and 78.00 Mb,respectively.Furthermore,30474 protein-coding genes were annotated.Comparative genomics analysis revealed that C.wilsoniana diverged from its closest species∼12.46 million years ago(Mya).Furthermore,the divergence between Cornaceae and Nyssaceae occurred>62.22 Mya.We also found evidence of whole-genome duplication events and whole-genome triplicationγ,occurring at∼44.90 and 115.86 Mya.We further inferred the origins of chromosomes,which sheds light on the complex evolutionary history of the karyotype of C.wilsoniana.Through transcriptional and metabolic analysis,we identified two FAD2 homologous genes that may play a crucial role in controlling the oleic to linoleic acid ratio.We further investigated the correlation between metabolites and genes and identified 33 MADS-TF homologous genes that may affect f lower morphology in C.wilsoniana.Overall,this study lays the groundwork for future research aimed at identifying the genetic basis of crucial traits in C.wilsoniana. | Zhenxiang He Haoyu Chao Xinkai Zhou Qingyang Ni Yueming Hu Ranran Yu Minghuai Wang Changzhu Li Jingzhen Chen Yunzhu Chen Yong Chen Chunyi Cui Liangbo Zhang Ming Chen Dijun Chen | 2023 | Horticulture Research2023,10,11: | 0 |

| 16 | MEIC-global-CO_(2):A new global CO_(2)emission inventory with highly-resolved source category and sub-country information显示文摘CO_(2)emission inventory provides fundamental data for climate research and emission mitigation.Currently,most global CO_(2)emission inventories were developed with energy statistics from International Energy Agency(IEA)and were available at country level with limited source categories.Here,as the first step toward a high-resolution and dynamic updated global CO_(2)emission database,we developed a data-driven approach to construct seamless and highly-resolved energy consumption data cubes for 208 countries/territories,797 sub-country administrative divisions in 29 countries,42 fuel types,and 52 sectors,with the fusion of activity data from 24 international statistics and 65 regional/local statistics.Global CO_(2)emissions from fossil fuel combustion and cement production in 1970–2021 were then estimated with highly-resolved source category(1,484 of total)and sub-country information(797 of total).Specifically,73%of global CO_(2)emissions in 2021 were estimated with sub-country information,providing considerably improved spatial resolution for global CO_(2)emission accounting.With the support of detailed information,the dynamics of global CO_(2)emissions across sectors and fuel types were presented,representing the evolution of global economy and progress of climate mitigation.Remarkable differences of sectoral contribution were found across sub-country administrative divisions within a given country,revealing the uneven distribution of energy and economic structure among different regions.Our estimates were generally consistent with existing databases at aggregated level for global total or large emitters,while large discrepancies were observed for middle and small emitters.Our database,named the Multiresolution Emission Inventory model for Climate and air pollution research(MEIC)is publicly available through http://gffzz6c575fb1d0a54d36hcuqfn09pfvpn65vw.ffgz.tsg.suse.edu.cn with highly-resolved information and timely update,which provides an independent carbon emission accounting data source for climate research. | Ruochong XU Dan TONG Qingyang XIAO Xinying QIN Cuihong CHEN Liu YAN Jing CHENG Can CUI Hanwen HU Wenyu LIU Xizhe YAN Huaxuan WANG Xiaodong LIU Guannan GENG Yu LEI Dabo GUAN Kebin HE Qiang ZHANG | 2024 | Science China Earth Sciences2024,67,2: | 0 |

| 18 | Influence and characteristic of shading on photovoltaic performance of bifacial modules and method for estimating bifacial gain显示文摘Bifacial PV modules have unique advantages in low-carbon building applications such as BIPV systems but often suffer from the shading problem resulting from higher surrounding objects or building facades.Point-blank quantitative studies of PV performance of bifacial modules operating in actual environments as affected by shading on PV cells are lacking due to the difficulties of analysis caused by the existing multiple variable factors.By constructing an experimental comparison system on a flat roof of a building,we experimentally tested and analyzed the comparative variation characteristics of PV performance of bifacial and mono-facial modules under different shading area fractions.The results show that from the viewpoint of photoelectric efficiency,the PV performance of both bifacial and mono-facial PV modules clearly varied with the shading fraction of PV cell in some linear rules,though it is difficult to find regularity from the perspective of output power which was also affected by dynamic solar radiation intensity.An abnormal phenomenon emerged that the photoelectric efficiencies of the bifacial modules with small shading fraction were higher compared to the case without shading.Based on the findings of the experimental results,a regression approximation method based on shading test results(RAST Method)is further proposed to analyze and calculate the bifacial gain of bifacial modules.In the case of the existing roof installation,the mean bifacial gains of the two bifacial modules with different inclination angles were 8.86%and 11.30%,respectively. | Chenglong Luo Yuandan Wu Xiaoxiao Su Wu Zou Yanshun Yu Qingyang Jiang Lijie Xu | 2023 | Building Simulation2023,16,10: | 0 |
