| 1 | Isolation and characterization of atrazine-degrading Arthrobacter sp. AD26 and use of this strain in bioremediation of contaminated soil显示文摘A bacterial strain (AD26) capable of utilizing atrazine as a sole nitrogen source for growth was isolated from an industrial wastewater sample by enrichment culture. The 16S rRNA gene sequencing identified AD26 as an Arthrobacter sp. PCR assays indicated that AD26 contained atrazine-degrading genes trzN and atzBC. The trzN gene of AD26 only differs from the trzN of Arthrobacter aurescens TC1 by one base (A→T at 907) and one amino acid (Met→Leu at 303). The specific activity of trzN of AD26 in crude cell extract was 0.28 U/mg, which was 1.2 times that of TC1. This strain has shown faster growth and atrazine-degradation rates in atrazine-containing minimal media than two well characterized atrazine-degrading bacteria, Pseudomonas sp. ADP and Arthrobacter aurescens TC1. After incubating for 48 h at 30°C, the OD600 of AD26 reached 2.6 compared with 1.33 of ADP. AD26 was capable of degrading 500 mg/L of atrazine in minimal medium at 95% in 72 h, while the degradative rates by TC1 and ADP were only 90% and 86%, respectively. | 3 | Effects of “Bioactive” amino acids leucine, glutamate, arginine and tryptophan on feed intake and mRNA expression of relative neuropeptides in broiler chicks显示文摘Feed intake control is vital to ensuring optimal nutrition and achieving full potential for growth and development in poultry. The aim of the present study was to investigate the effects of L-leucine, L-glutamate, L-tryptophan and L-arginine on feed intake and the mRNA expression levels of hypothalamic Neuropeptide involved in feed intake regulation in broiler chicks. Leucine, glutamate, tryptophan or arginine was intra-cerebroventricularly (ICV) administrated to 4d-old broiler chicks respectively and the feed intake were recorded at various time points. Quantitative PCR was performed to determine the hypothalamic mRNA expression levels of Neuropeptide Y (NPY), agouti related protein (AgRP), pro-opiomelanocortin (POMC), melanocortin receptor 4 (MC4R) and corticotrophin releasing factor (CRF). Our results showed that ICV administration of L-leucine (0.15 or 1.5 μmol) significantly (P<0.05) increased feed intake up to 2 h post-administration period and elevated both hypothalamic NPY and AgRP mRNA expression levels. In contrast, ICV administration of L-glutamate (1.6μmol) significantly (P<0.05) decreased feed intake 0.25, 0.5 and 2 h post-injection, and increased hypothalamic CRF and MC4R mRNA expression levels. Meanwhile, both L-tryptophan (10 or 100μg) and L-arginine (20 or 200μg) had no significant effect on feed intake. | 4 | Effect of diacylglycerol acyltransferase 2overexpression in 3T3-L1 is associated to an increase in mono-unsaturated fatty acid accumulation显示文摘Background:Fatty acid(FA) composition is the most important parameter affecting the flavor and nutritional value of the meat.The final and the only committed step in the biosynthesis of triglycerides is catalyzed by diacylglycerol acyltransferase 2(DGAT2).The role of DGAT2 in lipid accumulation has been demonstrated in adipocytes,However,little is known about the effect of DGAT2 on the FA composition of these cells.Methods:To investigate the role of DGAT2 in regulating lipid accumulation,FA composition and the expression of adipogenic genes,we cloned the open reading frame of the porcine DGAT2 gene and established 3T3-L1 cells that overexpressed DGAT2.Cells were then cultured in differentiation medium(DM) without FA,with a mixture of FAs(FA-DM),or containing a ^(13)C stable isotope-labeled FA mixture(IFA-DM).The FA composition of adipocytes was analyzed by gas chromatography-mass spectrometry and gas chromatography-isotope ratio mass spectrometry.Quantitative PCR and western blotting were employed to detect expression of adipogenic genes in 3T3-L1 adipocytes cultured with FA-DM for 12 d.Results:The triacylglyceride(TAG) content was significantly higher in 3T3-L1 adipocytes overexpressing DGAT2 than in control cells.When cultured in DM or FA-DM for 12 d,cells overexpressing DGAT2 showed a higher proportion of unsaturated FAs(C16:1 and C18:1).However,when cells overexpressing DGAT2 were cultured with FA-DM for30 min,the FA composition was almost identical to that of controls.Further,the proportion of stable isotope-labeled FAs were similar in 3T3-L1 adipocytes overexpressing DGAT2 and control cells cultured in IFA-DM for 12 d.These results collectively indicate that the higher proportion of mono-unsaturated FAs,C16:1 and C18:1,may originate from de novo FA synthesis but not from the uptake of specific FAs from the medium.This hypothesis is further supported by evidence that both mRNA and protein expression of genes involved in FA synthesis(ACACA,FASN,SCD1,and A-FABP)were significantly higher in cells overexpressing DGAT2 than in control cells.Conclusions:In conclusion,our study revealed that TAG accumulation,the proportion of MUFAs,and the expression of adipogenic genes were higher in 3T3-L1 cells overexpressing DGAT2 than in control cells. | Zhiqi Zhang Gang Shu Xiaotong Zhu Junming Guo Han Cai Songbo Wang Lina Wang Ping Gao Qianyun Xi Yongliang Zhang Li Yuan Qingyan Jiang | 2014 | Journal of Animal Science and Biotechnology2014,5,4: | 3 |

| 6 | Tauroursodeoxycholic acid(TUDCA)improves intestinal barrier function associated with TGR5-MLCK pathway and the alteration of serum metabolites and gut bacteria in weaned piglets显示文摘Background:Tauroursodeoxycholic acid(TUDCA),a hydrophilic bile acid,is the main medicinal component of bear bile and is commonly used to treat a variety of hepatobiliary diseases.Meanwhile,TUDCA has been shown to modulate the intestinal barrier function and alleviate DSS-induced colitis in mice.However,the effect of TUDCA on the intestinal barrier of weaned piglets remains largely unclear.Methods:The weaned piglets and porcine IPEC-J2 intestinal epithelial cells were used to investigate the effects of TUDCA on intestinal barrier function in weaned piglets and explore the possible underlying mechanisms.In vivo,72 healthy weaned piglets were randomly allocated into 2 groups according to their gender and body weight,and piglets were fed the basal diet with 0(control,CON)and 200 mg/kg TUDCA for 30 d,respectively.Three female and three male piglets reflecting the average bodyweight were slaughtered in each group and samples were collected.In vitro,IPEC-J2 cells were subjected to 100μmol/L TUDCA to explore the possible underlying mechanisms.Results:Our results demonstrated that dietary TUDCA supplementation significantly reduced the diarrhea incidence of weaned piglets,possibly attributing to the TUDCA-enhanced intestinal barrier function and immunity.In addition,TUDCA supplementation altered serum metabolites and the relative abundance of certain gut bacteria,which might contribute to the improved intestinal barrier function.Furthermore,the in-vitro results showed that TUDCA improved the E.coli-induced epithelial barrier impairment of IPEC-J2 cells and increased Takeda G-coupled protein receptor 5(TGR5)protein expression.However,knockdown of TGR5 and inhibition of myosin light chain kinase(MLCK)pathway abolished the TUDCA-improved epithelial barrier impairment in E.coli-treated IPEC-J2 cells,indicating the involvement of TGR5-MLCK in this process.Conclusions:These findings showed that TUDCA improved intestinal barrier function associated with TGR5-MLCK pathway and the alteration of serum metabolites and gut bacteria in weaned piglets,suggesting the potential application of TUDCA in improving gut health in piglet production. | Min Song Fenglin Zhang Yiming Fu Xin Yi Shengchun Feng Zhichang Liu Dun Deng Qiang Yang Miao Yu Canjun Zhu Xiaotong Zhu Lina Wang Ping Gao Gang Shu Xianyong Ma Qingyan Jiang Songbo Wang | 2022 | Journal of Animal Science and Biotechnology2022,13,6: | 1 |

| 13 | Enhancing milk quality and modulating rectal microbiota of dairy goats in starch‑rich diet:the role of bile acid supplementation显示文摘Background Diets rich in starch have been shown to increase a risk of reducing milk fat content in dairy goats.While bile acids(BAs)have been used as a lipid emulsifier in monogastric and aquatic animals,their effect on ruminants is not well understood.This study aimed to investigate the impact of BAs supplementation on various aspects of dairy goat physiology,including milk composition,rumen fermentation,gut microbiota,and BA metabolism.Results We randomly divided eighteen healthy primiparity lactating dairy goats(days in milk=100±6 d)into two groups and supplemented them with 0 or 4 g/d of BAs undergoing 5 weeks of feeding on a starch-rich diet.The results showed that BAs supplementation positively influenced milk yield and improved the quality of fatty acids in goat milk.BAs supplementation led to a reduction in saturated fatty acids(C16:0)and an increase in monounsaturated fatty acids(cis-9 C18:1),resulting in a healthier milk fatty acid profile.We observed a significant increase in plasma total bile acid concentration while the proportion of rumen short-chain fatty acids was not affected.Furthermore,BAs supplementation induced significant changes in the composition of the gut microbiota,favoring the enrichment of specific bacterial groups and altering the balance of microbial populations.Correlation analysis revealed associations between specific bacterial groups(Bacillus and Christensenellaceae R-7 group)and BA types,suggesting a role for the gut microbiota in BA metabolism.Functional prediction analysis revealed notable changes in pathways associated with lipid metabolism,suggesting that BAs supplementation has the potential to modulate lipid-related processes.Conclusion These findings highlight the potential benefits of BAs supplementation in enhancing milk production,improving milk quality,and influencing metabolic pathways in dairy goats.Further research is warranted to elucidate the underlying mechanisms and explore the broader implications of these findings. | Qingyan Yin Junjian Yu Jiaxiao Li Tianci Zhang Tianyu Wang Yufei Zhu Jun Zhang Junhu Yao | 2024 | Journal of Animal Science and Biotechnology2024,15,2: | 0 |

| 15 | Expression of Inducible Nitric Oxide Synthase, p53 and Bcl-2 in Gastric Precancerous and Cancerous Lesions: Correlation with Clinical Features显示文摘OBJECTIVE To explore the expression of inducible nitric oxide synthase (iNOS), p53 and bcl -2 in gastric precancerous and cancerous lesions and to examine the expression of these proteins in relation to clinical features. METHODS The expressions of iNOS, p53 and bcl-2 proteins in gastric precancerous and cancerous lesions and their correlations with the clinical features were determined using immunohistochemical assays (Power VisionTM two -step method) on 84 gastric carcinomas and 54 gastric atypical hyperplastic tissues. Apoptotic cells were evaluated by terminal deoxynucleotidyl transferase- mediated dUTP-biotin nick-end labeling (TUNEL). RESULTS Expression of iNOS, p53 and bcl-2 was significantly higher in gastric carcinoma (GC) tissues than in gastric atypical hyperplastic tissues. Among the 84 carcinomas, the expression of p53 was observed in 50 (59.52%), bcl-2 in 43 (51.19%), and iNOS in 65 (77.58%). Overex-pression of iNOS and bcl-2 in gastric carcinoma was related to tumor size and iNOS was related to the presence of lymph node metastasis (P< 0.05). The expression of proteins did not correlate with age, sex, stage of disease, or differentiation. Expression of iNOS in gastric carcinoma tissues was positively correlated with bcl-2 expression (X2=8.926, P=0.003), and also with p53 expression (X2= 5.2430, P= 0.022). The mean apoptotic indexes (Al) were 1.29%±0.50 in low-grade atypical hyperplasia (LG), 0.96%±0.36 in high-grade atypical hyperplasia (HG) and 0.70%±0.43 in GC, with the difference being significant between LG, HG and GC (P< 0.05). There was a significant positive correlation between iNOS expression and the Al in GC (t=3.0815, P=0.0028). Along with the development of computer hardware and software, various kinds of platforms have been provided to different simulation methods that can be carried out with distinct working flows. The jet flow element is the core part of the hydrokinetic hammer. We can build the ideal simulation model of it by means of CFD (computational fluid dynamics) technology. On the other hand, to set up the digital model of piston and hammer, the best way is to build the virtual prototype using automatic dynamic analysis of mechanical system. | 19 | Vertical distributions of wintertime atmospheric nitrogenous compounds and the corresponding OH radicals production in Leshan, southwest China显示文摘Ground-based multi-axis differential optical absorption spectroscopy(MAX-DOAS)observations were operated from 02 to 21 December 2018 in Leshan,southwest China,to measure HONO,NO_(2) and aerosol extinction vertical distributions,and these were the first MAX-DOAS measurement results in Sichuan Basin.During the measurement period,characteristic ranges for surface concentration were found to be 0.26-4.58 km^(−1) and averaged at 0.93 km^(−1) for aerosol extinction,0.49 to 35.2 ppb and averaged at 4.57 ppb for NO_(2) and 0.03 to 7.38 ppb and averaged at 1.05 ppb for HONO.Moreover,vertical profiles of aerosol,NO_(2) and HONO were retrieved from MAX-DOAS measurements using the Heidelberg Profile(HEIPRO)algorithm.By analysing the vertical gradients of pollutants and meteorological information,we found that aerosol and HONO are strongly localised,while NO_(2) is mainly transmitted from the north direction(city center direction).Nitrogen oxides such as HONO and NO_(2) are important for the production of hydroxyl radical(OH)and oxidative capacity in the troposphere.In this study,the averaged value of OH production rate from HONO is about 0.63 ppb/hr and maximum value of ratio between OH production from HONO and from(HONO+O_(3))is>93%before12:00 in Leshan.In addition,combustion emission contributes to 26%for the source of HONO in Leshan,and we found that more NO_(2) being converted to HONO under the conditions with high aerosol extinction coefficient and high relative humidity is also a dominant factor for the secondary produce of HONO. | Chengzhi Xing Cheng Liu Qihou Hu Qingyan Fu Shanshan Wang Hua Lin Yizhi Zhu Shuntian Wang Weiwei Wang Zeeshan Javed Xiangguang Ji Jianguo Liu | 2021 | Journal of Environmental Sciences2021,33,7: | 0 |
