| 1 | Fluid–solid coupling analysis of rock pillar stability for concealed karst cave ahead of a roadway based on catastrophic theory显示文摘In order to study the mechanism of water inrush from a concealed, confined karst cave, we established a fluid–solid coupling model of water inrush from a concealed karst cave ahead of a roadway and a strength reduction method in a rock pillar for preventing water inrush based on catastrophic theory. Fluid–solid coupling effects and safety margins in a rock pillar were studied. Analysis shows that rock pillar instability, exerted by disturbance stress and seepage stress, is the process of rock pillar catastrophic destabilization induced by nonlinear extension of plastic zones in the rock pillar. Seepage flow emerges in the rock pillar for preventing water inrush, accompanied by mechanical instability of the rock pillar. Taking the accident of a confined karst cave water-inrush of Qiyi Mine as an example, by studying the safety factor of the rock pillar and the relationship between karst cave water pressure and thickness of the rock pillar,it is proposed that rock pillar thickness with a safety factor equal to 1.5 is regarded as the calculated safety thickness of the rock pillar, which should be equal to the sum of the blasthole depth, blasting disturbance depth and the calculated safety thickness of the rock pillar. The cause of the karst water inrush at Qiyi Mine is that the rock pillar was so small that it did not possess a safety margin. | 2 | Pancreatic head cancer with gastrointestinal bleeding as the initial manifestation:a case report and literature review显示文摘Gastrointestinal bleeding(GIB)as the initial symptom is rare in patients with pancreatic cancer,which is prone to misdiagnosis and should be paid attention to by clinicians.Here,we present a patient with pancreatic head cancer whose first symptom was only severe upper gastrointestinal bleeding,which initially led to misdiagnosis.However,subsequent imaging revealed a mass in the pancreatic head which was considered a malignant tumor.After surgical resection,the pathology confirmed that the mass in the pancreatic head was pancreatic ductal adenocarcinoma,and the duodenal bulb’s full thickness was infiltrated.A literature review found that pancreatic cancer can present gastrointestinal bleeding by invading the digestive tract and blood vessels,left-sided portal hypertension(LSPH),digestive tract metastasis,and canceration of ectopic pancreatic tissue in the digestive tract.For these cases,clinicians should utilize multidisciplinary diagnosis and treatment based on digestive endoscopy,imaging,interventional therapy,and surgery to control bleeding,identify etiology,and remove tumors. | Minghui Zheng Zhiping Fu Ting Niu Anan Liu Danlei Chen Junfeng Peng Judong Li Zhiyuan Hao Qingyang Yu Zechao Lyu Chenghao Shao | 2023 | Journal of Pancreatology2023,6,3: | 0 |

| 4 | LncRNA LOC103694972 promotes fibrosis of NRK-49F cells by regulating STAT3-dependent Smad/CTGF pathway via targeting miR-29c-3p显示文摘Background:Renalfibrosis is an important process in the development of chronic kidney disease.Understanding the pathogenesis andfinding effective treatments for renalfibrosis is crucial.This study aims to investigate whether a newly discovered long non-coding RNA(lncRNA)called LOC103694972 could be a potential target for treatingfibrosis of NRK-49F cells.Methods:LncRNA Chip was used to identify differentially expressed lncRNAs between TGF-β1-induced NRK-49F cells and normal cells.The dual-luciferase assay confirmed the binding between miR-29c-3p and signal transducer and activator of transcription(STAT3),as well as between miR-29c-3p and lncRNA LOC103694972.Si-LOC103694972 and miR-29c-3p mimic were then transfected into TGF-β1-induced NRK-49F cells.Results:The study found that LOC103694972 was highly expressed in TGF-β1-induced NRK-49F cells.These cells exhibited increased cell length and activity compared to the control group.The expression levels of Collagen I,α-Smooth muscle actin(α-SMA),and tissue inhibitor of metalloproteinase(TIMP-1)were increased,while matrix Metalloproteinase 2(MMP2)and matrix Metalloproteinase 9(MMP9)expression was decreased.However,transfection with si-LOC103694972 and miR-29c-3p mimics restored cell morphology and reduced cell viability.This led to a decrease in the levels of Collagen I,α-SMA,and TIMP-1,as well as an increase in MMP2 and MMP9 expression.Additionally,TGF-β1-induced NRK-49F cells transfected with miR-29c-3p mimics activated the STAT3-Smad3/CTGF pathway.Conclusion:Based on thesefindings,lncRNA LOC103694972 shows promise as a target for treating renalfibrosis.It negatively regulates miR-29c-3p and activates the STAT3-Smad3/CTGF pathway. | YAN LI HUZHI CAI XIAOLING PENG YOUHUI LIU QINGYANG CHEN XIANGDONG LIN XINYU CHEN | 2024 | BIOCELL2024,48,3: | 0 |

| 7 | Definition and verification of novel metastasis and recurrence related signatures of ccRCC: A multicohort study显示文摘Background:Cancer metastasis and recurrence remain major challenges in renal carcinoma patient management.There are limited biomarkers to predict the metastatic probability of renal cancer,especially in the early-stage subgroup.Here,our study applied robust machine-learning algorithms to identify metastatic and recurrence-related signatures across multiple renal cancer cohorts,which reached high accuracy in both training and testing cohorts.Methods:Clear cell renal cell carcinoma(ccRCC)patients with primary or metastatic site sequencing information from eight cohorts,including one outhouse cohort,were enrolled in this study.Three robust machine-learning algorithms were applied to identify metastatic signatures.Then,two distinct metastatic-related subtypes were identified and verified;matrix remodeling associated 5(MXRA5),as a promising diagnostic and therapeutic target,was investigated in vivo and in vitro.Results:We identified five stable metastasis-related signatures(renin,integrin subunit beta-like 1,MXRA5,mesenchyme homeobox 2,and anoctamin 3)from multicenter cohorts.Additionally,we verified the specificity and sensibility of these signatures in external and out-house cohorts,which displayed a satisfactory consistency.According to these metastatic signatures,patients were grouped into two distinct and heterogeneous ccRCC subtypes named metastatic cancer subtype 1(MTCS1)and type 2(MTCS2).MTCS2 exhibited poorer clinical outcomes and metastatic tendencies than MTCS1.In addition,MTCS2 showed higher immune cell infiltration and immune signature expression but a lower response rate to immune blockade therapy than MTCS1.The MTCS2 subgroup was more sensitive to saracatinib,sunitinib,and several molecular targeted drugs.In addition,MTCS2 displayed a higher genome mutation burden and instability.Furthermore,we constructed a prognosis model based on subtype biomarkers,which performed well in training and validation cohorts.Finally,MXRA5,as a promising biomarker,significantly suppressed malignant ability,including the cell migration and proliferation of ccRCC cell lines in vitro and in vivo.Conclusions:This study identified five robust metastatic signatures and proposed two metastatic probability clusters with stratified prognoses,multiomics landscapes,and treatment options.The current work not only provided new insight into the heterogeneity of renal cancer but also shed light on optimizing decision‐making in immunotherapy and chemotherapy. | Aimin Jiang Qingyang Pang Xinxin Gan Anbang Wang Zhenjie Wu Bing Liu Peng Luo Le Qu Linhui Wang | 2022 | Cancer Innovation2022,1,2: | 0 |
