1CRISPR/Cas9-mediated PINK1 deletion leads to neurodegeneration in rhesus monkeys显示文摘Dear Editor,PINK1 mutations cause autosomal recessive and early-onset Parkinson’s disease(PD)with selective neurodegeneration.Unfortunately,current PINK1 knockout(KO)mouse1,2 and pig models3,4 are unable to recapitulate the selective and overt neurodegeneration seen in PD patients.Furthermore,endogenous Pink1 in the mouse brain is expressed at very low levels and can only be detected via immunoprecipitation,5 meaning that PINK1’s function in the mammalian brain needs to be assessed using larger animals that are closer to humans.We previously used CRISPR/Cas9 to target the monkey gene in one-cell stage embryos.6 Using the same approach,we designed two gRNAs to target exon 2(T1)and exon 4(T2),which encode a kinase domain in the PINK1 gene of rhesus monkeys(Fig.1a).CRISPR/Cas9 and PINK1 gRNAs were injected into one-cell stage rhesus monkey embryos.A T7E1 assay and sequencing of PCR products from the injected embryos showed high efficiency(61.5%)in targeting PINK1(Fig.1b and Supplementary information,Fig.S1).Transfer of 87 embryos to 28 surrogate rhesus monkeys resulted in 11 pregnancies(39.2%)(Fig.1b).Eleven fetuses developed to term and were born naturally.Of these live monkeys,eight carried PINK1 mutations(M),and three were wild type(WT).However,three mutant monkeys(M1,M3 and M4)were newborn triplets that struggled to survive and died 3-4 days after birth.One WT newborn monkey also died after a difficult labor.Another mutant monkey(M2)died 7 days after birth without noticeable warning signs or symptoms.The other three mutant monkeys(M6,M7 and M8)have lived for three years;M5,however,reduced its food intake and showed weakness at the age of 1.5 years,and died 30 days after anesthesia for MRI examination.Weili Yang Yunbo Liu Zhuchi Tu Chong Xiao Sen Yan Xishan Ma Xiangyu Guo Xiusheng Chen Peng Yin Zhengyi Yang Su Yang Tianzi Jiang Shihua Li Chuan Qin Xiao-Jiang Li 2019Cell Research2019,29,4:12

5Evolution of the microstructure and solute distribution of Sn-10wt% Bi alloys during electromagnetic field-assisted directional solidification显示文摘The effects of forced flows at different velocities on microstructure and solute distribution during the directional solidification of Sn-10 wt% Bi alloys under a simultaneous imposition of a transverse static magnetic field(TSMF) and an external direct current(DC) have been investigated experimentally and numerically. The experimental results show that the solid-liquid interface will gradually become sloping with the increase of the forced flow velocity when the thermoelectric magnetic convection(TEMC)dominates the forced flow at solidification front. However, the interface will gradually become planar as the flow velocity further increases when the electromagnetic convection(EMC) dominates the forced flow. Moreover, when the flow velocity gradually increases, the primary dendrite spacing decreases from384 to 105 μm accordingly. The simulation results show that the solute distribution at the two sides of the sample can be significantly changed by the forced flow at solidification front. The rejected solute will be unidirectionally transported to one side of the sample along the TEMC(a low-velocity forced flow),thereby causing the formation of a sloping interface. However, the rejected solute will be returned back along the EMC(a higher-velocity force flow), which results in a planar interface. Furthermore, the solute content at the two sides of the sample under the forced flows at different velocities was measured. 66.5 wt%Si high silicon steel sheets prepared by composite electrodeposition in magnetic field显示文摘A new preparation method of near-net-shape 6.5 wt% Si high silicon steel sheets was proposed by combining composite electrodeposition(CED) and diffusion annealing under magnetic field. The obtained sheets were characterized by scanning electron microscopy, energy dispersive spectrometry, analytical balance and a silicon steel material measurement system. The results show that the surface morphology,the elemental distribution, the cathode current efficiency and the silicon content of coatings were obviously influenced by the micro and macro magnetohydrodynamics(MHD) flows under magnetic field.With the effect of magnetic field, the silicon particles content of coatings showed an increasing trend and the diffusion process showed that an approximately uniform 6.5 wt% silicon steel sheet has been successfully obtained. The magnetism measurement showed that the high silicon steel sheet has the lower iron loss, and the iron loss further decreased under magnetic field. 7Reservoir accumulation conditions and key exploration&development technologies for Keshen gas field in Tarim Basin显示文摘The Keshen gas field is located in the central part of Kuqa foreland thrust belt in Tarim Basin,and is another large gas field discovered in Kuqa depression after Kela 2 gas field.Since the breakthrough in 2008,a number of large and medium scale gas reservoirs including Keshen 2,Keshen 5 and Keshen 8 have been discovered,that are characterized by ultra depth,ultra-high pressure,ultra-low porosity,ultra-low permeability,high temperature and high pressure.With natural gas geological reserves of nearly trillion cubic meters and production capacity of nearly 5.5 billion cubic meters,the Keshen gas field is the main natural gas producing area in Tarim Oilfield.The Keshen gas field is located in a series of thrusting imbrication structures in the Kelasu structural belt of Kuqa foreland thrust belt.The salt roof structure,plastic rheology of salt beds and pre-salt faulted anticlinal structure constitute the large wedge-shaped thrust body.The thick delta sandstone of the Cretaceous Bashijike Formation is widely distributed,and it forms the superior reservoir-caprock combination with overlying Paleogene thick gypsum-salt bed.The deep Jurassic-Triassic oil and gas migrate vertically along fault system formed in Late Himalaya,break through the thick Cretaceous mudstone and move laterally along the fracture system of the pre-salt reservoirs,to form anticline and fault anticline high pressure reservoir groups.Through near ten years of studies,the three-dimensional seismic acquisition and processing technology for complex mountainous areas,extrusion salt-related structural modeling technology and fractured low-porosity sandstone reservoir evaluation technology have been established,which lay a foundation for realization of oil and gas exploration objectives.Logging acquisition and evaluation technology for high temperature,high pressure,ultra-deep and low-porosity sandstone gas reservoirs,and efficient development technology for fractured tight sandstone gas reservoirs have been developed,which provide a technical support for efficient exploration&development and rapid production of the Keshen gas field.Haijun Yang Yong Li Yangang Tang Ganglin Lei Xiongwei Sun Peng Zhou Lu Zhou Anming Xu Jingjie Tang Wenhui Zhu Jiangwei Shang Weili Chen Mei Li 2019Petroleum Research2019,4,4:3

9Diketopyrrolopyrrole-Au(I) as singlet oxygen generator for enhanced tumor photodynamic and photothermal therapy显示文摘Diketopyrrolopyrrole(DPP)derivatived photosensitizers(PSs)with near infrared(NIR)absorption and good photophysical properties have drawn tremendous attention in cancer phototherapy.However,current DPP derivatives present unsatisfactory quantum yield of singlet oxygen(1O2)due to the large energy gap between the excited singlet and triplet states.To tackle this challenge,herein the DPP core is functionalized with triphenylphosphine-Au(I)group(Th DPP-Au),leading to a high1O2 quantum yield of 0.65.Theoretical calculation attributes the enhancement to spin-orbit coupling and population of the triplet excition upon photoexcitation.The hydrophilic Th DPP-Au nanoparticals(NPs)are prepared via nano-reprecipitation,which displays homogeneous size and excellent light absorption ability(ε=4.382×104 M-1cm-1).And the Th DPP-Au NPs exhibit low dark toxicity and high phototoxicity,which can effectively kill tumor cells via 1O2 induced mitochondrial apoptotic pathway upon irradiation.Furthermore,in vivo experiments demonstrate that Th DPP-Au NPs can selective accumulation in tumor and present excellent synergistic photodynamic/photothermal therapy guided by fluorescence and photothermal dual imaging.Xiaoyu Huang Rui Gu Jiewei Li Nan Yang Zijin Chen Weili Si Peng Chen Wei Huang Xiaochen Dong 2020Science China Chemistry2020,63,1:3
10New pathogenic insights from large animal models of neurodegenerative diseases显示文摘Animal models are essential for investigating the pathogenesis and developing the treatment of human diseases.Identification of genetic mutations responsible for neurodegenerative diseases has enabled the creation of a large number of small animal models that mimic genetic defects found in the affected individuals.Of the current animal models,rodents with genetic modifications are the most commonly used animal models and provided important insights into pathogenesis.However,most of genetically modified rodent models lack overt neurodegeneration,imposing challenges and obstacles in utilizing them to rigorously test the therapeutic effects on neurodegeneration.Recent studies that used CRISPR/Cas9-targeted large animal(pigs and monkeys)have uncovered important pathological events that resemble neurodegeneration in the patient’s brain but could not be produced in small animal models.Here we highlight the unique nature of large animals to model neurodegenerative diseases as well as the limitations and challenges in establishing large animal models of neurodegenerative diseases,with focus on Huntington disease,Amyotrophic lateral sclerosis,and Parkinson diseases.We also discuss how to use the important pathogenic insights from large animal models to make rodent models more capable of recapitulating important pathological features of neurodegenerative diseases.Peng Yin Shihua Li Xiao-Jiang Li Weili Yang 2022Protein & Cell2022,13,10:2
11Nondestructive effect of the cusp magnetic field on the dendritic microstructure during the directional solidification of Nickel-based single crystal superalloy显示文摘The mechanical-property improvement of directionally-solidified Nickel-based single crystal(SC)superalloy with the single-direction magnetic fields is limited by their destructiveness on the dendritic microstructure.Here,the work present breaks through the bottleneck.It shows that the application of the cusp magnetic field(CMF)ensures that the dendrites are not destroyed.This feature embodies that the primary dendrite trunks arrange regularly and orderly,as well the secondary dendrite arms grow symmetrically.By contrast,both the unidirectional transverse and longitudinal magnetic field destroy the dendrite morphology,and there are a number of stray grains near the totally-re melted interface.The nondestructive effect is achieved mainly by the combined action of the thermoelectromagnetic force on the dendrites and thermoelectromagnetic convection in the melt during directional solidification.The investigation should contribute a new route for dramatically and effectively improving the crystal quality and mechanical properties of the directionally-solidified alloys.Xiaotan Yuan Tao Zhou Weili Ren Jianchao Peng Tianxiang Zheng Long Hou Jianbo Yu Zhongming Ren Peter K.Liaw Yunbo Zhong 2021Journal of Materials Science & Technology2021,,3:2

13Fracture mechanism of inversed trapezoidal shaped tunnel excavated in 45° inclined rock strata显示文摘In order to understand failure mechanisms of the tunnel excavated in the stratified rock masses in deep mine, the physical modeling experiment by using the large-scale model was carried out. The field case simulated in the experiment is a main connection tunnel located at depth of 1000 m in Qishan coal mine,Xuzhou mining district. Tunnel deformation was monitored by using strain gauges and a video camera simultaneously. Crack initiation and propagation process during the test were analyzed based on image analysis of the captured video photographs. At the same time, deformation process of the key monitoring points around the tunnel section is given by the monitored strain plots. 18A precipitate-free AlCoFeNi eutectic high-entropy alloy with strong strain hardening显示文摘Over recent years,eutectic high-entropy alloys(EHEAs)have intrigued substantial research enthusiasms due to their good castability as well as balanced strength-ductility synergy.In this study,a bulk cast Al_(19.25)Co_(18.86)Fe_(18.36)Ni_(43.53)EHEA is developed with fine in-situ lamellar eutectics.The eutectics comprise alternating ordered face-centered-cubic(L1_(2))and ordered body-centered-cubic(B2)phases with semicoherent interfaces.The resulting microstructure resembles that of most reported as-cast EHEAs,but the B2 lamellae are devoid of nano-precipitates because of the Cr-element removal in current tailored eutectic composition.Surprisingly,the B2 lamellae still feature much higher deformation resistance than the L1_(2) lamellae,so that less lattice defects are detected in the B2 lamellae until the fracture.More interestingly,in the L1_(2) lamellae we identify a dynamic microstructure refinement that correlates to extraordinary strain hardening in tension.The precipitate-free EHEA consequently shows excellent tensile ductility of~10%and high ultimate strength up to~956 MPa.Peijian Shi Yi Li Yuebo Wen Yiqi Li Yan Wang Weili Ren Tianxiang Zheng Yifeng Guo Long Hou Zhe Shen Ying Jiang Jianchao Peng Pengfei Hu Ningning Liang Qingdong Liu Peter K.Liaw Yunbo Zhong 2021Journal of Materials Science & Technology2021,,30:1
19An overview of the thermography-based experimental studies on roadway excavation in stratified rock masses at CUMTB显示文摘This paper presents an overview of experimental investigations conducted at China University of Mining and Technology Beijing(CUMTB) on roadway excavation using large-scale geomechanical model tests.The simulated sedimentary rocks are composed by alternating layers of sandstone, mudstone and coal seam inclined at varied angles with respect to the horizontal including 0°, 45°, 60°, and 90°. During the excavation, infrared thermography was employed to detect the thermal response of the surrounding rocks under excavation. The obtained raw thermograms were processed using denoising algorithm, data reduction procedure and Fourier analysis. The infrared temperature(IRT) characterizes the overall rock response; the processed thermal images represent the structural behavior, and the Fourier spectrum describes damage development in the frequency domain. 20Genomic analysis,trajectory tracking,and field surveys reveal sources and long-distance dispersal routes of wheat stripe rust pathogen in China显示文摘Identifying sources of phytopathogen inoculum and determining their contributions to disease outbreaks are essential for predicting disease development and establishing control strategies.Puccinia striiformis f.sp.tritici(Pst),the causal agent of wheat stripe rust,is an airborne fungal pathogen with rapid virulence variation that threatens wheat production through its long-distance migration.Because of wide variation in geographic features,climatic conditions,and wheat production systems,Pst sources and related dispersal routes in China are largely unclear.In the present study,we performed genomic analyses of 154 Pst isolates from all major wheat-growing regions in China to determine Pst population structure and diversity.Through trajectory tracking,historical migration studies,genetic introgression analyses,and field surveys,we investigated Pst sources and their contributions to wheat stripe rust epidemics.We identified Longnan,the Himalayan region,and the Guizhou Plateau,which contain the highest population genetic diversities,as the Pst sources in China.Pst from Longnan disseminates mainly to eastern Liupan Mountain,the Sichuan Basin,and eastern Qinghai;that from the Himalayan region spreads mainly to the Sichuan Basin and eastern Qinghai;and that from the Guizhou Plateau migrates mainly to the Sichuan Basin and the Central Plain.These findings improve our current understanding of wheat stripe rust epidemics in China and emphasize the need for managing stripe rust on a national scale.Yuxiang Li Jichen Dai Taixue Zhang Baotong Wang Siyue Zhang Conghao Wang Jiguang Zhang Qiang Yao Mingju Li Chengyun Li Yuelin Peng Shiqin Cao Gangming Zhan Fei Tao Haifeng Gao Weili Huang Xiaojun Feng Yingwen Bai Zhuoma Qucuo Hongsheng Shang Chong Huang Wancai Liu Jiasui Zhan Xiangming Xu Xianming Chen Zhensheng Kang Xiaoping Hu 2023Plant Communications2023,4,4:1
