| 1 | Rapid species identification of pathogenic bacteria from a minute quantity exploiting three-dimensional quantitative phase imaging and artificial neural network显示文摘The healthcare industry is in dire need of rapid microbial identification techniques for treating microbial infections.Microbial infections are a major healthcare issue worldwide,as these widespread diseases often develop into deadly symptoms.While studies have shown that an early appropriate antibiotic treatment significantly reduces the mortality of an infection,this effective treatment is difficult to practice.The main obstacle to early appropriate antibiotic treatments is the long turnaround time of the routine microbial identification,which includes time-consuming sample growth.Here,we propose a microscopy-based framework that identifies the pathogen from single to few cells.Our framework obtains and exploits the morphology of the limited sample by incorporating three-dimensional quantitative phase imaging and an artificial neural network.We demonstrate the identification of 19 bacterial species that cause bloodstream infections,achieving an accuracy of 82.5%from an individual bacterial cell or cluster.This performance,comparable to that of the gold standard mass spectroscopy under a sufficient amount of sample,underpins the effectiveness of our framework in clinical applications.Furthermore,our accuracy increases with multiple measurements,reaching 99.9%with seven different measurements of cells or clusters.We believe that our framework can serve as a beneficial advisory tool for clinicians during the initial treatment of infections. | Geon Kim Daewoong Ahn Minhee Kang Jinho Park DongHun Ryu YoungJu Jo Jinyeop Song Jea Sung Ryu Gunho Choi Hyun Jung Chung Kyuseok Kim Doo Ryeon Chung In Young Yoo Hee Jae Huh Hyun-seok Min Nam Yong Lee YongKeun Park | 2022 | Light(Science & Applications)2022,11,7: | 2 |

| 7 | Two Types of Diurnal Variations in Heavy Rainfall during July over Korea显示文摘This study examined the characteristics of the diurnal variations of heavy rainfall(≥110 mm in 12 hours)in Korea and the related atmospheric circulation for July from 1980−2020.During the analysis period,two dominant pattens of diurnal variation of the heavy rainfall emerged:all-day heavy rainfall(AD)and morning only heavy rainfall(MO)types.For the AD-type,the heavy rainfall is caused by abundant moisture content in conjunction with active convection in the morning(0000−1200,LST;LST=UTC+9)and the afternoon hours(1200−2400 LST).These systems are related to the enhanced moisture inflow and upward motion induced by the strengthening of the western North Pacific subtropical high and upper-tropospheric jet.For the MO-type,heavy rainfall occurs mostly in the morning hours;the associated atmospheric patterns are similar to the climatology.We find that the atmospheric pattern related to severe heavy rainfalls in 2020 corresponds to a typical AD-type and resembles the 1991 heavy-rainfall system in its overall synoptic/mesoscale circulations.The present results imply that extremely heavy rainfall episodes in Korea during the 2020 summer may occur again in the future associated with the recurring atmospheric phenomenon related to the heavy rainfall. | Chang-Kyun PARK Minhee CHANG Chang-Hoi HO Kyung-Ja HA Jinwon KIM Byung-Ju SOHN | 2021 | Advances in Atmospheric Sciences2021,38,12: | 1 |

| 11 | Accumulation of poly(adenosine diphosphate-ribose)by sustained supply of calcium inducing mitochondrial stress in pancreatic cancer cells显示文摘BACKGROUND The biochemical phenomenon defined as poly adenosine diphosphate(ADP)-ribosylation(PARylation)is essential for the progression of pancreatic cancer.However,the excessive accumulation of poly ADP-ribose(PAR)induces apoptosis-inducing factor(AIF)release from mitochondria and energy deprivation resulting in the caspase-independent death of cancer cells.AIM To investigate whether sustained calcium supply could induce an anticancer effect on pancreatic cancer by PAR accumulation.METHODS Two pancreatic cancer cell lines,AsPC-1 and CFPAC-1 were used for the study.Calcium influx and mitochondrial reactive oxygen species(ROS)were observed by fluorescence staining.Changes in enzyme levels,as well as PAR accumulation and energy metabolism,were measured using assay kits.AIF-dependent cell death was investigated followed by confirming in vivo anticancer effects by sustained calcium administration.RESULTS Mitochondrial ROS levels were elevated with increasing calcium influx into pancreatic cancer cells.Then,excess PAR accumulation,decreased PAR glycohydrolase and ADP-ribosyl hydrolase 3 levels,and energy deprivation were observed.In vitro and in vivo antitumor effects were confirmed to accompany elevated AIF levels.CONCLUSION This study visualized the potential anticancer effects of excessive PAR accumulation by sustained calcium supply on pancreatic cancer,however elucidating a clear mode of action remains a challenge,and it should be accompanied by further studies to assess its potential for clinical application. | Keun-Yeong Jeong Jae Jun Sim Minhee Park Hwan Mook Kim | 2022 | World Journal of Gastroenterology2022,28,27: | 0 |
