| 1 | Lymphocyte subsets predictive value and possible involvement of human papilloma virus infection on breast cancer molecular subtypes显示文摘AIM To detect human papilloma virus(HPV) presence and to characterize cellular immune response in breast cancer patients. METHODS A total of 74 women were included, of which 48 samples were from patients diagnosed with breast cancer and 26 patients with benign pathology of the breast. Molecular subtype classification was performed based on the immunohistochemical reports of the tumor piece. HPV genome detection and genotyping from fresh breast biopsies was performed using the INNO-LIPA HPV Genotyping Extra test(Innogenetics, Ghent, Belgium). CD3+, CD4+, CD8+ and natural killer(NK)+ cells levels from peripheral blood samples from patients with breast cancer and benign pathology were measured by flow cytometry. RESULTS Luminal A was the most frequent breast cancer molecular subtype(33.33%). HPV was detected in 25% of the breast cancer patients, and genotype 18 was the most frequent in the studied population. The mean of CD3+, CD4+ and CD8+ subpopulations were decreased in patients with breast cancer, in relation to those with benign pathology, with a statistically significant difference in CD8+ values(P = 0.048). The mean of NK+ cells was increased in the benign pathology group. The average level of CD3+, CD4+, CD8+ and NK+ cells decreased as the disease progressed. HER2+ and Luminal B HER2+ tumors had the lowest counts of cell subsets. HPV breast cancer patients had elevated counts of cellular subsets. | 3 | 胃肠的道上的职业应力的效果显示文摘 The aim of this review is to provide a general overview of the relationship between occupational stress and gastrointestinal alterations. The International Labour Organization suggests occupational health includes psychological aspects to achieve mental well-being. However, the definition of health risks for an occupation includes biological, chemical, physical and ergonomic factors but does not address psychological stress or other affective disorders. Nevertheless, multiple investigations have studied occupational stress and its physiological consequences, focusing on specific risk groups and occupations considered stressful. Among the physiological effects of stress, gastrointestinal tract(GIT) alterations are highly prevalent. The relationship between occupational stress and GIT diseases is evident in everyday clinical practice; however, the usual strategy is to attack the effects but not the root of the problem. That is, in clinics, occupational stress is recognized as a source of GIT problems, but employers do not ascribe it enough importance as a risk factor, in general, and for gastrointestinal health, in particular. | 4 | Betulinic acid hydroxamate prevents colonic inflammation and fibrosis in murine models of inflammatory bowel disease显示文摘Intestinal fibrosis is a common complication of inflammatory bowel disease (IBD) and is defined as an excessive accumulation of scar tissue in the intestinal wall. Intestinal fibrosis occurs in both forms of IBD: ulcerative colitis and Crohn’s disease. Small-molecule inhibitors targeting hypoxia-inducing factor (HIF) prolyl-hydroxylases are promising for the development of novel antifibrotic therapies in IBD. Herein, we evaluated the therapeutic efficacy of hydroxamate of betulinic acid (BHA), a hypoxia mimetic derivative of betulinic acid, against IBD in vitro and in vivo. We showed that BAH (5–20 μM) dose-dependently enhanced collagen gel contraction and activated the HIF pathway in NIH-3T3 fibroblasts;BAH treatment also prevented the loss of trans-epithelial electrical resistance induced by proinflammatory cytokines in Caco-2 cells. In two different murine models (TNBS- and DSS-induced IBD) that cause colon fibrosis, oral administration of BAH (20, 50 mg/kg·d, for 17 days) prevented colon inflammation and fibrosis, as detected using immunohistochemistry and qPCR assays. BAH-treated animals showed a significant reduction of fibrotic markers (Tnc, Col1a2, Col3a1, Timp-1, α-SMA) and inflammatory markers (F4/80+, CD3+, Il-1β, Ccl3) in colon tissue, as well as an improvement in epithelial barrier integrity and wound healing. BHA displayed promising oral bioavailability, no significant activity against a panel of 68 potential pharmacological targets and was devoid of genotoxicity and cardiotoxicity. | 7 | Species of Lasiodiplodia associated with mango in Brazil显示文摘Mango(Mangifera indica)is a major tropical fruit species cultivated in Brazil.The objective of this study was to identify species of Lasiodiplodia associated with dieback and stem-end rot of mango in the semi-arid region of Northeastern Brazil,and compare the species in relation to mycelial growth,pathogenicity and virulence.A total of 120 isolates of Lasiodiplodia were used and identifications were made using a combination of morphology and phylogenetic analysis based on partial translation elongation factor 1-αsequence(EF1-α)and internal transcribed spacers(ITS).The following species were identified:Lasiodiplodia crassispora,L.egyptiacae,L.hormozganensis,L.iraniensis,L.pseudotheobromae,L.theobromae and Lasiodiplodia sp..Lasiodiplodia theobromae was the most frequently isolated species,which represented 41%of all the isolates.Only this species had been previously reported on mango in Brazil,while the other species represent the first report associated with mango tree diseases in this country.Lasiodiplodia crassispora is reported for the first time associated with mango diseases worldwide.There were significant differences in mycelial growth rates among the Lasiodiplodia species and also in the optimum temperature for growth.All species of Lasiodiplodia were pathogenic on mango fruit.There were significant differences in virulence among the species,wherein L.hormozganensis and Lasiodiplodia sp.were the most virulent,while the least virulent were L.iraniensis,L.pseudotheobromae,L.crassispora and L.egyptiacae. | Marília W.Marques Nelson B.Lima Marcos Antônio de Morais Jr Maria Angélica G.Barbosa Breno O.Souza Sami J.Michereff Alan J.L.Phillips Marcos P.S.Câmara | 2013 | Fungal Diversity2013,,4: | 2 |
