| 4 | Exposure to endocrine disrupting chemicals perturbs lipid metabolism and circadian rhythms显示文摘A growing body of evidence indicates that exposure to environmental chemicals can contribute to the etiology of obesity by inappropriately stimulating adipogenesis as well as perturbing lipid metabolism and energy balance. One potential mechanism by which chemical exposure can influence lipid metabolism is through disturbance of circadian rhythms, endogenously-driven cycles of roughly 24 hr in length that coordinate biochemical, physiological, and behavioral processes in all organisms. Here we show for the first time that exposure to endocrine disrupting compounds(EDCs), including the pesticide tributyltin, two commercial flame retardants, and a UV-filter chemical found in sunscreens,can perturb both circadian clocks and lipid metabolism in vertebrates. Exposure of developing zebrafish to EDCs affects core clock activity and leads to a remarkable increase in lipid accumulation that is reminiscent of the effects observed for longdaysin, a known disruptor of circadian rhythms. Our data reveal a novel obesogenic mechanism of action for environmental chemicals, an observation which warrants further research. | 5 | Characterization of two rat models of cystic fibrosis—KO and F508del CFTR—Generated by Crispr-Cas9显示文摘Background: Genetically engineered animals are essential for gaining a proper understanding of the disease mechanisms of cystic fibrosis(CF). The rat is a relevant laboratory model for CF because of its zootechnical capacity, size, and airway characteristics, including the presence of submucosal glands.Methods: We describe the generation of a CF rat model(F508 del) homozygous for the p.Phe508 del mutation in the transmembrane conductance regulator(Cftr) gene. This model was compared to new Cftr-/-rats(CFTR KO). Target organs in CF were examined by histological staining of tissue sections and tooth enamel was quantified by micro-computed tomography. The activity of CFTR was evaluated by nasal potential difference(NPD) and short-circuit current measurements. The effect of VX-809 and VX-770 was analyzed on nasal epithelial primary cell cultures from F508 del rats.Results: Both newborn F508 del and Knock out(KO) animals developed intestinal obstruction that could be partly compensated by special diet combined with an osmotic laxative. The two rat models exhibited CF phenotypic anomalies such as vas deferens agenesis and tooth enamel defects. Histology of the intestine, pancreas, liver, and lungs was normal. Absence of CFTR function in KO rats was confirmed ex vivo by short-circuit current measurements on colon mucosae and in vivo by NPD, whereas residual CFTR activity was observed in F508 del rats. | 6 | Endoscopy-based management decreases the risk of postoperative recurrences in Crohn's disease显示文摘AIM: To investigate whether an endoscopy-based management could prevent the long-term risk of postoperative recurrence.METHODS: From the pathology department database, we retrospectively retrieved the data of all the patients operated on for Crohn's disease(CD) in our center(1986-2015). Endoscopy-based management was defined as systematic postoperative colonoscopy(median time after surgery = 9.5 mo) in patients with no clinical postoperative recurrence at the time of endoscopy. RESULTS: From 205 patients who underwent surgery, 161 patients(follow-up > 6 mo) were included. Endoscopic postoperative recurrence occurred in 67.6%, 79.7%, and 95.5% of the patients, respectively 5, 10 and 20 years after surgery. The rate of clinical postoperative recurrence was 61.4%, 75.9%, and 92.5% at 5, 10 and 20 years, respectively. The rate of surgical postoperative recurrence was 19.0%, 38.9% and 64.7%, respectively, 5, 10 and 20 years after surgery. In multivariate analysis, previous intestinal resection, prior exposure to anti-TNF therapy before surgery, and fistulizing phenotype(B3) were postoperative risk factors. Previous perianal abscess/fistula(other perianal lesions excluded), were predictive of only symptomatic recurrence. | 8 | Ultrafast response of harmonic modelocked THz lasers显示文摘The use of fundamental modelocking to generate short terahertz(THz)pulses and THz frequency combs from semiconductor lasers has become a routine affair,using quantum cascade lasers(QCLs)as a gain medium.However,unlike classic laser diodes,no demonstrations of harmonic modelocking,active or passive,have been shown in THz QCLs,where multiple pulses per round trip are generated when the laser is modulated at the harmonics of the cavity’s fundamental round-trip frequency.Here,using time-resolved THz techniques,we show for the first time harmonic injection and mode-locking in which THz QCLs are modulated at the harmonics of the round-trip frequency.We demonstrate the generation of the harmonic electrical beatnote within a QCL,its injection locking to an active modulation and its direct translation to harmonic pulse generation using the unique ultrafast nature of our approach.Finally,we show indications of self-starting harmonic emission,i.e.,without external modulation,where the QCL operates exclusively on a harmonic(up to its 15th harmonic)of the round-trip frequency.This behaviour is supported by time-resolved simulations of induced gain and loss in the system and shows the importance of the electronic,as well as photonic,nature of QCLs.These results open up the prospect of passive harmonic modelocking and THz pulse generation,as well as the generation of low-noise microwave generation in the hundreds of GHz region. | Feihu Wang Valentino Pistore Michael Riesch Hanond Nong Pierre-Baptiste Vigneron Raffaele Colombelli Olivier Parillaud Juliette Mangeney Jerome Tignon Christian Jirauschek Sukhdeep S.Dhillon | 2020 | Light(Science & Applications)2020,9,1: | 3 |
