1Efficient hepatic delivery and protein expression enabled by optimized mRNA and ionizable lipid nanoparticle显示文摘mRNA is a novel class of therapeutic modality that holds great promise in vaccination,protein replacement therapy,cancer immunotherapy,immune cell engineering etc.However,optimization of mRNA molecules and efficient in vivo delivery are quite important but challenging for its broad application.Here we present an ionizable lipid nanoparticle(iLNP)based on iBL0713 lipid for in vitro and in vivo expression of desired proteins using codon-optimized mRNAs.mRNAs encoding luciferase or erythropoietin(EPO)were prepared by in vitro transcription and formulated with proposed iLNP,to form iLP171/mRNA formulations.It was revealed that both luciferase and EPO proteins were successfully expressed by human hepatocellular carcinoma cells and hepatocytes.The maximum amount of protein expression was found at 6 h post-administration.The expression efficiency of EPO with codon-optimized mRNA was significantly higher than that of unoptimized mRNA.Moreover,no toxicity or immunogenicity was observed for these mRNA formulations.Therefore,our study provides a useful and promising platform for mRNA therapeutic development.Tongren Yang Chunhui Li Xiaoxia Wang Deyao Zhao Mengjie Zhang Huiqing Cao Zicai Liang Haihua Xiao Xing-Jie Liang Yuhua Weng Yuanyu Huang 2020Bioactive Materials2020,5,4:7
2Effect of Banxia Xiexin decoction on Helicobacter pylori-related peptic ulcers and its possible mechanism via the TGF-β/Smad signaling pathway显示文摘OBJECTIVE: To investigate the effect of Banxia Xiexin decoction(BXD) on Helicobacter pylori(Hp)-related peptic ulcers(PUs) and the possible mecha-nism underlying BXD actions via the transforming growth factor-β/small mothers against decapentaplegic(TGF-β/Smad) signaling pathway.METHODS: PU patients with cold-heat complex syndrome were randomly assigned to groups that received Chinese or Western medicines with 20 patients in each group. Serum was collected after 7 d of treatment. The healthy group included 20 individuals. Gastric mucosal epithelial cell line GES-1 was cultured in vitro and randomly divided into the following seven groups: control, model, healthy,Western Medicine, prior treatment, low dosage,and high dosage. After 72 h of treatment with the corresponding serum, the m RNA and protein expression levels of TGF-β1, Smad3, and Smad7 were measured by reverse transcription quantitative polymerase chain reaction and western blotting, respectively.RESULTS: The m RNA expression levels of TGF-β1 and Smad3 in GES-1 cells were increased after Hp introduction, and these increased levels were reduced by the BXD-containing serum. The protein levels of p-Smad3, but not TGF-β1 or Smad3, were significantly increased in Hp-treated GES-1 cells,and treatment with the BXD-containing serum markedly decreased the protein levels. 3Optimization of dosage ratio of chlorogenic acid and gardenia glycosides in the treatment of rats with fatty liver disease induced by high-fat feed显示文摘OBJECTIVE: To investigate the optimal dosage ratio of chlorogenic acid and gardenia glycosides in treating the rates with fatty liver disease induced by high-fat feed.METHODS: A rat model of non-alcoholic fatty liver disease(NAFLD) was established by using a high-fat diet. According to mathematical model 'uniform design', varying doses of chlorogenic acid and gardenia glycosides have been combined to form 6 medications for the treatment of NAFLD.Samples were then taken to observe pathological changes of the liver tissue(HE staining); changes in the fat metabolism pathway e.g. triglyceride(TG)and free fatty acid(FFA) content; alterations in liver function, i.e. serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST) activity; and differences in Malondialdehyde(MDA) and superoxide dismutase(SOD) content in the liver tissue. Multiple regression analysis was conducted to test the optimal dosage ratio of chlorogenic acid and gardenia glycosides.RESULTS: Fatty degeneration and vacuole-like changes of different degrees occurred in hepatic cells of the model group. Markers for fat metabolism, serum ALT and AST activities, and expression of MDA in liver tissue significantly increased, while SOD decreased. 4AQR is a novel type 2 diabetes-associated gene that regulates signaling pathways critical for glucose metabolism显示文摘Type 2 diabetes mellitus(T2 DM) is a common metabolic disease influenced by both genetic and environmental factors. In this study, we performed an in-house genotyping and meta-analysis study using three independent GWAS datasets of T2 DM and found that rs3743121, located 1 kb downstream of AQR,was a novel susceptibility SNP associated with T2 DM. The risk allele C of rs3743121 was correlated with the increased expression of AQR in white blood cells, similar to that observed in T2 DM models. The knockdown of AQR in HepG2 facilitated the glucose uptake, decreased the expression level of PCK2,increased the phosphorylation of GSK-3β, and restored the insulin sensitivity. Furthermore, the suppression of AQR inhibited the mTOR pathway and the protein ubiquitination process. 5OPTIMIZATION OF Ni BIOACCUMULATION BY SYNECHCOCCUS显示文摘Influencing factors on bioaccumulation of Ni by Synechcoccus were studied in this paper. The equilibration time of Ni bioaccumulation was about 80 min in aqueous solution. Bioaccumulation quantity reached maximum when mass ratio of Ni to dry weight concentration of Synechcoccus was 16?18%. Bioaccumulation quantity increased with in- creasing pH. The optimum pH was 9?10 and higher pH led to precipitation of Ni(OH)2. Bioaccumulation quantity was also influenced by temperature and light intensity reaching their optima at 35oC and 3 000 lx respectively. 6Overexpression of IbSINA5 Increases Cold Tolerance through a CBF SINA-COR Mediated Module in Sweet Potato显示文摘Seven in absentia(SINA)family proteins play a central role in plant growth,development and resistance to abiotic stress.However,their biological function in plant response to cold stress is still largely unknown.In this work,a seven in absentia gene IbSINA5 was isolated from sweet potato.Quantitative real-time polymerase chain reaction(qRT-PCR)analyses demonstrated that IbSINA5 was ubiquitously expressed in various tissues and organs of sweet potato,with a predominant expression in fibrous roots,and was remarkably induced by cold,drought and salt stresses.Subcellular localization assays revealed that IbSINA5-GFP fusion protein was mainly localized in cytoplasm and nucleus.Overexpression of IbSINA5 in sweet potato led to dramatically improved resistance to cold stress in transgenic plants,which was associated with the up-regulated expression of IbCOR(cold-regulated)genes,increased proline production,and decreased malondialdehyde(MDA)and H2O2 accumulation in the leaves of transgenic plants.Furthermore,transient expression of IbCBF3,a C-repeat binding factor(CBF)gene,in the leaf protoplasts of wild type sweet potato plants up-regulated the expression of both IbSINA5 and IbCOR genes.Our results suggest that IbSINA5 could function as a positive regulator in the cold signaling pathway through a CBF-SINA-COR mediated module in sweet potato,and have a great potential to be used as a candidate gene for the future breeding of new plant species with improved cold resistance.Shiyang Li Xue-Ao Liu Lizi Zhao Huiqing Huang Bei Li Zhizhong Song Meixia Liang Hongxia Zhang Limin Wang Shenglin Zhou 2021Phyton-International Journal of Experimental Botany2021,90,3:1

9Artificial Noise Aided Polar Codes for Physical LayerSecurity显示文摘The secrecy rates of the existing practical secrecy coding methods are relative low to satisfy the security requirement of 5 G communications.We propose an artificial noise(AN) aided polar coding algorithm to improve the secrecy rate.Firstly,a secrecy coding model based on AN is presented,where the confidential bits of last transmission code block are adopted as AN to inject into the current codeword.In this way,the AN can only be eliminated from the jammed codeword by the legitimate users.Since the AN is shorter than the codeword,we then develop a suboptimal jamming positions selecting algorithm with the goal of maximizing the bit error rate of the eavesdropper.Theoretical and simulation results demonstrate that the proposed algorithm outperforms the random selection method and the method without AN.Huiqing Bai Liang Jin Ming Yi 2017China Communications2017,14,12:1

11Multi-valent mRNA vaccines against monkeypox enveloped or mature viron surface antigens demonstrate robust immune response and neutralizing activity显示文摘Monkeypox was declared a global health emergency by the World Health Organization,and as of March 2023,86,000 confirmed cases and 111 deaths across 110 countries have been reported.Its causal agent,monkeypox virus(MPV)belongs to a large family of double-stranded DNA viruses,Orthopoxviridae,that also includes vaccinia virus(VACV)and others.MPV produces two distinct forms of viral particles during its replication cycles:the enveloped viron(EV)that is released via exocytosis,and the mature viron(MV)that is discharged through lysis of host cells.This study was designed to develop multi-valent m RNA vaccines against monkeypox EV and MV surface proteins,and examine their efficacy and mechanism of action.Four m RNA vaccines were produced with different combinations of surface proteins from EV(A35R and B6R),MV(A29L,E8L,H3L and M1R),or EV and MV,and were administered in Balb/c mice to assess their immunogenicity potentials.A dynamic immune response was observed as soon as seven days after initial immunization,while a strong Ig G response to all immunogens was detected with ELISA after two vaccinations.The higher number of immunogens contributed to a more robust total Ig G response and correlating neutralizing activity against VACV,indicating the additive potential of each immunogen in generating immune response and nullifying VACV infection.Further,the m RNA vaccines elicited an antigen-specific CD4^(+)T cell response that is biased towards Th1.The m RNA vaccines with different combinations of EVand MV surface antigens protected a mouse model from a lethal dose VACV challenge,with the EV and MV antigens-combined vaccine offering the strongest protection.These findings provide insight into the protective mechanism of multi-valent m RNAvaccines against MPV,and also the foundation for further development of effective and safe m RNA vaccines for enhanced protection against monkeypox virus outbreak.Niubing Zhang Xiang Cheng Yilong Zhu Ouyang Mo Huiqing Yu Liqi Zhu Juan Zhang Linlin Kuang Ying Gao Ruiyuan Cao Xiaozhen Liang Haikun Wang Honglin Li Song Li Wu Zhong Xuan Li Xiao Li Pei Hao 2023Science China(Life Sciences)2023,66,10:0
