1Topographic species-habitat associations of tree species in a heterogeneous tropical karst seasonal rain forest, China显示文摘Aims Tropical and subtropical karst forests of south China are under increas-ing pressure from over-exploitation causing widespread habitat degra-dation and biodiversity loss.Previous research has demonstrated that topography,as a proxy for resource availability,plays an important role in shaping tree species distributions in tropical forests.However,the association between growth stages and habitats types has not been considered in this analysis.Our aim was to examine the differences among different habitat types to determine whether tree species show similar species-habitat associations at young and mature life stages.Methods We used multivariate regression tree analysis to examined species-habitat associations among eight topographically defined habitats.The results were tested with a torus-translation test and canonical correspondence analysis(CCA)for 74 species in a 15 ha karst tropi-cal seasonal rain forest at the Nonggang National Natural Reserve in south China.We considered two life stages(young and mature)of trees species that showed a positive association with topography.Important Findings We found marked differences in community characteristics and number of associations among the eight habitats.Of the 74 species subjected to torus-translation test,63 had significant positive and 70 had significant negative associations with one or more of the eight habitats.Positive associations were more frequent in higher elevation habitats and negative associations were more frequent in lower eleva-tion habitats.This suggests that edaphic and hydrological variation related to topography play important roles in habitat partitioning in heterogeneous karst forests.For the 63 tree species with significant positive associations to at least one habitat,40 of them had the same positive association at young and mature life stages.The CCA revealed that the six topographic variables considered had consistent relation-ships with species distribution among all individuals and their two life stages.This indicates that most of the karst forest tree species show consistent associations with a single habitat throughout their life.We conclude that niche differentiation plays an important role in main-taining the diversity of this heterogeneous species-rich karst forest.Yili Guo Bin Wang Azim U.Mallik Fuzhao Huang Wusheng Xiang Tao Ding Shujun Wen Shuhua Lu Dongxing Li Yunlin He Xiankun Li 2017Journal of Plant Ecology2017,10,3:6
2An optimized protocol for stepwise optimization of real-time RT-PCR analysis显示文摘Computational tool-assisted primer design for real-time reverse transcription(RT)PCR(qPCR)analysis largely ignores the sequence similarities between sequences of homologous genes in a plant genome.It can lead to false confidence in the quality of the designed primers,which sometimes results in skipping the optimization steps for qPCR.However,the optimization of qPCR parameters plays an essential role in the efficiency,specificity,and sensitivity of each gene’s primers.Here,we proposed an optimized approach to sequentially optimizing primer sequences,annealing temperatures,primer concentrations,and cDNA concentration range for each reference(and target)gene.Our approach started with a sequence-specific primer design that should be based on the single-nucleotide polymorphisms(SNPs)present in all the homologous sequences for each of the reference(and target)genes under study.By combining the efficiency calibrated and standard curve methods with the 2−ΔΔCt method,the standard cDNA concentration curve with a logarithmic scale was obtained for each primer pair for each gene.As a result,an R 2≥0.9999 and the efficiency(E)=100±5% should be achieved for the best primer pair of each gene,which serve as the prerequisite for using the 2^(−ΔΔCt) method for data analysis.We applied our newly developed approach to identify the best reference genes in different tissues and at various inflorescence developmental stages of Tripidium ravennae,an ornamental and biomass grass,and validated their utility under varying abiotic stress conditions.We also applied this approach to test the expression stability of six reference genes in soybean under biotic stress treatment with Xanthomonas axonopodis pv.glycines(Xag).Thus,these case studies demonstrated the effectiveness of our optimized protocol for qPCR analysis.Fangzhou Zhao Nathan A.Maren Pawel Z.Kosentka Ying-Yu Liao Hongyan Lu James R.Duduit Debao Huang Hamid Ashrafi Tuanjie Zhao Alejandra I.Huerta Thomas G.Ranney Wusheng Liu 2021Horticulture Research2021,8,1:2

20Engineered Cleistogamy in Camelina sativa for bioconfinement显示文摘Camelina sativa is a self-pollinating and facultative outcrossing oilseed crop.Genetic engineering has been used to improve camelina yield potential for altered fatty acid composition,modified protein profiles,improved seed and oil yield,and enhanced drought resistance.The deployment of transgenic camelina in the field posits high risks related to the introgression of transgenes into nontransgenic camelina and wild relatives.Thus,effective bioconfinement strategies need to be developed to prevent pollen-mediated gene f low(PMGF)from transgenic camelina.In the present study,we overexpressed the cleistogamy(i.e.f loral petal non-openness)-inducing PpJAZ1 gene from peach in transgenic camelina.Transgenic camelina overexpressing PpJAZ1 showed three levels of cleistogamy,affected pollen germination rates after anthesis but not during anthesis,and caused a minor silicle abortion only on the main branches.We also conducted field trials to examine the effects of the overexpressed PpJAZ1 on PMGF in the field,and found that the overexpressed PpJAZ1 dramatically inhibited PMGF from transgenic camelina to non-transgenic camelina under the field conditions.Thus,the engineered cleistogamy using the overexpressed PpJAZ1 is a highly effective bioconfinement strategy to limit PMGF from transgenic camelina,and could be used for bioconfinement in other dicot species.Debao Huang Liwei Gao Jeremy McAdams Fangzhou Zhao Hongyan Lu Yonghui Wu Jeremy Martin Sherif MSherif Jayasankar Subramanian Hui Duan Wusheng Liu 2023Horticulture Research2023,10,2:0
