2Multi-omics analyses of 398 foxtail millet accessions reveal genomic regions associated with domestication,metabolite traits,and antiinflammatory effects显示文摘Foxtail millet(Setaria italica),which was domesticatedfromthewild speciesgreenfoxtail(Setaria viridis),isa richsource of phytonutrientsfor humans.To evaluate how breeding changed themetabolome offoxtail millet grains,we generated and analyzed the datasets encompassing the genomes,transcriptomes,metabolomes,and anti-inflammatory indices from 398 foxtail millet accessions.We identified hundreds of common variants that influence numerous secondary metabolites.We observed tremendous differences in natural variations of the metabolites and their underlying genetic architectures between distinct sub-groups of foxtail millet.Furthermore,we found that the selection of the gene alleles associated with yellow grains led to altered profiles of metabolites such as carotenoids and endogenous phytohormones.Using CRiSPR-mediated genome editing wevalidated the function of PHYTOENE SYNTHASE1(PSY1)gene in affecting milletgrain colorand quality.Interestingly,our in vitro cell inflammation assays showed that 83 metabolites in millet grains have anti-inflammatory effects.Taken together,ourmulti-omics study illustrates how the breeding history of foxtail millet has shaped its metabolite profile.The datasets we generated in this study also provide important resources for further understanding how millet grain quality is affected by different metabolites,laying the foundations for future millet genetic research and metabolome-assisted improvement.Xukai Li Jianhua Gao Jingyi Song Kai Guo Siyu Hou Xingchun Wang Qiang He Yanyan Zhang Yakun Zhang Yulu Yang Jiaoyan Tang Hailang Wang Staffan Persson Mingquan Huang Lishuai Xu Linlin Zhong Dongqin Li Yongming Liu Hua Wu Xianmin Diao Peng Chen Xiaowen Wang Yuanhuai Han 2022Molecular Plant2022,15,8:8
3Magnetic polyamidoamine dendrimers for magnetic separation and sensitive determination of organochlorine pesticides from water samples by high-performance liquid chromatography显示文摘Organochlorine pesticides(OCPs)have received much attention due to their toxicity.Reliable methods to monitor their residues in the environment are needed.Here,magnetic polyamidoamine dendrimers were prepared by co-precipitation,Michael addition,and amidation.The magnetic polyamidoamine dendrimers demonstrated good adsorption ability for OCPs—this feature was utilized to construct a sensitive tool for monitoring OCPs in water samples.The proposed method provided remarkable linearity from 0.1 to 500μg/L and satisfactory limits of detection from 0.012 to 0.029μg/L.The spiked recoveries of the four target analytes were 91.8%-103.5%with relative standard deviations less than 4.5%.The magnetic materials had good reusability.The results indicated that the resulting method was an efficient,easy,rapid,economical,and eco-friendly tool for monitoring OCPs in aqueous samples.Qingxiang Zhou Yalin Wu Yi Sun Xueying Sheng Yayan Tong Jinghan Guo Boyao Zhou Jingyi Zhao 2021Journal of Environmental Sciences2021,33,4:5
4Urinary donor-derived cell-free DNA as a non-invasive biomarker for BK polyomavirus-associated nephropathy显示文摘BK polyomavirus-associated nephropathy(BKPyVAN)is a common cause of allograft failure.However,differentiation between BKPyVAN and type I T cell-mediated rejection(TCMR)is challenging when simian virus 40(SV40)staining is negative,because of the similarities in histopathology.This study investigated whether donor-derived cell-free DNA(ddcfDNA)can be used to differentiate BKPyVAN.Target region capture sequencing was applied to detect the ddcfDNAs of 12 recipients with stable graft function,22 with type I TCMR,21 with proven BKPy VAN,and 5 with possible Py VAN.We found that urinary ddcfDNA levels were upregulated in recipients with graft injury,whereas plasma ddcfDNA levels were comparable for all groups.The median urinary concentrations and fractions of ddcfDNA in proven BKPyVAN recipients were significantly higher than those in type I TCMR recipients(10.4 vs.6.1 ng/mL,P<0.001 and 68.4%vs.55.3%,P=0.013,respectively).Urinary ddcfDNA fractions(not concentrations)were higher in the BKPyVAN-pure subgroup than in the BKPyVAN-rejection-like subgroup(81.30%vs.56.64%,P=0.025).With a cut-off value of 7.81 ng/m L,urinary ddcf DNA concentrations distinguished proven BKPyVAN from type I TCMR(area under the curve(AUC)=0.848,95%confidence interval(95%CI):0.734 to 0.963).These findings suggest that urinary ddcf DNA is a non-invasive biomarker which can reliably differentiate BKPy VAN from type I TCMR.Jia SHEN Luying GUO Wenhua LEI Shuaihui LIU Pengpeng YAN Haitao LIU Jingyi ZHOU Qin ZHOU Feng LIU Tingya JIANG Huiping WANG Jianyong WU Jianghua CHEN Rending WANG 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,11:4
5Combined amendment reduces soil Cd availability and rice Cd accumulation in three consecutive rice planting seasons显示文摘The scientific application of stabilized materials has been considered an effective method for the in situ remediation of Cd-contaminated soil.This study aimed to investigate the persistence of the effect of a combined amendment of limestone and sepiolite(LS)on soil Cd availability and accumulation in rice grown in a mildly Cd-contaminated paddy field(0.45 mg/kg of Cd)over three consecutive rice seasons.1125–4500 kg/ha of LS was applied to the soil before the first rice planting season and 562.5–2250 kg/ha of LS was supplemented before the third rice planting season.The application of LS(1125–4500 kg/ha)increased the soil p H by 0.44–1.09,0.18–0.53,and 0.42–0.68 in the first,second,and third season,respectively,and decreased the soil acid-extractable Cd content by 18.2–36.4%,17.7–33.5%,and9.6–17.6%.LS application significantly decreased the Cd contents in the rice tissues.The application of 4500 kg/ha of LS decreased the Cd content in brown rice to below the National Food Limit Standard of 0.2 mg/kg(GB 2762-2017)in the three consecutive rice seasons.However,the effect of LS on the soil-rice system was significantly weakened in the third season.The supplementary application of 562.5–2250 kg/ha of LS further decreased the Cd content in brown rice by 26.1–56.5%and decreased the health risk index by 23.7–43.8%.Therefore,it was recommended to apply 4500 kg/ha of LS in the first season and to supplement2250 kg/ha of LS in the third season to effectively guarantee the clean production of rice in three consecutive rice seasons.Wenjun Yang Shilong Wang Hang Zhou Min Zeng Jingyi Zhang Fang Huang Shiping Shan Zhaohui Guo Hongwei Yi Zhiguang Sun Jiaofeng Gu Bohan Liao 2022Journal of Environmental Sciences2022,34,1:4
6Silicone oil-based solar-thermal fluids dispersed with PDMS-modified Fe3O4@graphene hybrid nanoparticles显示文摘One of the most challenging problems that limit the practical application of carbon-based photothermal nanofluids is their poor dispersion stability and tendency to form aggregation. Herein, by using Fe_3O_4@graphene hybrid nanoparticles as a model system, we proposed a new method to prepare stably dispersed silicone oilbased solar-thermal nanofluids that can operate at high temperatures than water-based fluids. The introduction of Fe_3O_4 nanoparticles between graphene nanosheets not only physically increases the inter-plane distance of the graphene nanosheet but also provides numerous anchoring points for surface modification. Phosphate-terminated polydimethylsiloxane chains, which have high compatibility with the silicone oil base fluids and hightemperature stability, were synthesized and utilized to modify the Fe_3 O_4 nanoparticle surfaces. The attached chains create steric hindrance and effectively screen the strong inter-plane van der Waals attraction between graphene sheets. Dispersion stability of the nanofluids with different concentrations of surface-modified hybrid nanoparticles and heated under different temperatures was investigated. We have demonstrated that such fluids could maintain stable dispersion under a heating temperature up to 150 °C depending on the concentration of the hybrid nanoparticles. 7Uranium sorption on oxyhydroxide minerals by surface complexation and precipitation显示文摘During the chemical weathering of the uranium mill tailings,released uranium could be immobilized by the newly formed secondary minerals such as oxyhydroxides.A deeper understanding of the interaction between uranium and common oxyhydroxides under environmental conditions is necessary.In this work,uranium sorption behaviors on Al-,Mn-and Fe-oxyhydroxide minerals(boehmite,manganite,goethite,and lepidocrocite)were investigated by batch experiments.Results showed that the uranium sorption on Al-oxyhydroxide behaved significantly differently from the other three minerals.The sorption edge of the Mn-and Fe-oxyhydroxides located around pH 5,while the sorption edge of boehmite shifted about 1.5 pH unit to near neutral.The sorption isotherms of uranium on manganite,goethite and lepidocrocite at pH 5.0 could be well fitted by the Langmuir model.Instead of surface complexation,sorption on boehmite happened mainly by uranium-bearing carbonates and hydroxides precipitation as illustrated by the characterization results.Both carbonate and phosphate strongly affected the uranium sorption behavior.The removal efficiency of uranium by boehmite exceeded 98%after three sorption-desorption cycles,indicating it may be a potential material for uranium removal and recovery.Jingyi Wang Wanqiang Zhou Yanlin Shi Yao Li Dongfan Xian Ning Guo Chunli Liu 2022Chinese Chemical Letters2022,33,7:3
8The role of watermelon caffeic acid O-methyltransferase (ClCOMT1) in melatonin biosynthesis and abiotic stress tolerance显示文摘Melatonin is a pleiotropic signaling molecule that regulates plant growth and responses to various abiotic stresses.The last step of melatonin synthesis in plants can be catalyzed by caffeic acid O-methyltransferase(COMT),a multifunctional enzyme reported to have N-acetylserotonin O-methyltransferase(ASMT)activity;however,the ASMT activity of COMT has not yet been characterized in nonmodel plants such as watermelon(Citrullus lanatus).Here,a total of 16 putative O-methyltransferase(ClOMT)genes were identified in watermelon.Among them,ClOMT03(Cla97C07G144540)was considered a potential COMT gene(renamed ClCOMT1)based on its high identities(60.00–74.93%)to known COMT genes involved in melatonin biosynthesis,expression in almost all tissues,and upregulation under abiotic stresses.The ClCOMT1 protein was localized in the cytoplasm.Overexpression of ClCOMT1 significantly increased melatonin contents,while ClCOMT1 knockout using the CRISPR/Cas-9 system decreased melatonin contents in watermelon calli.These results suggest that ClCOMT1 plays an essential role in melatonin biosynthesis in watermelon.In addition,ClCOMT1 expression in watermelon was upregulated by cold,drought,and salt stress,accompanied by increases in melatonin contents.Overexpression of ClCOMT1 enhanced transgenic Arabidopsis tolerance against such abiotic stresses,indicating that ClCOMT1 is a positive regulator of plant tolerance to abiotic stresses.Jingjing Chang Yanliang Guo Jingyi Yan Zixing Zhang Li Yuan Chunhua Wei Yong Zhang Jianxiang Ma Jianqiang Yang Xian Zhang Hao Li 2021Horticulture Research2021,8,1:2

11Integrated microdevice with a windmill-like hole array for the clog-free,efficient,and self-mixing enrichment of circulating tumor cells显示文摘Circulating tumor cells(CTCs)have tremendous potential to indicate disease progression and monitor therapeutic response using minimally invasive approaches.Considering the limitations of affinity strategies based on their cost,effectiveness,and simplicity,size-based enrichment methods that involve low-cost,label-free,and relatively simple protocols have been further promoted.Nevertheless,the key challenges of these methods are clogging issues and cell aggregation,which reduce the recovery rates and purity.Inspired by the natural phenomenon that the airflow around a windmill is disturbed,in this study,a windmill-like hole array on the SU-8 membrane was designed to perturb the fluid such that cells in a fluid would be able to self-mix and that the pressure acting on cells or the membrane would be dispersed to allow a greater velocity.In addition,based on the advantages of fluid coatings,a lipid coating was used to modify the membrane surface to prevent cell aggregation and clogging of the holes.Under the optimal conditions,recovery rates of 93%and 90%were found for A549 and HeLa cells in a clinical simulation test of our platform with a CTC concentration of 20-100 cells per milliliter of blood.The white blood cell(WBC)depletion rate was 98.7%(n=15),and the CTC detection limit was less than 10 cells per milliliter of blood(n=6).Moreover,compared with conventional membrane filtration,the advantages of the proposed device for the rapid(2 mL/min)and efficient enrichment of CTCs without clogging were shown both experimentally and theoretically.Due to its advantages in the efficient,rapid,uniform,and clog-free enrichment of CTCs,our platform offers great potential for metastatic detection and therapy analyses.Hao Li Jinze Li Zhiqi Zhang Zhen Guo Changsong Zhang Zixu Wang Qiuquan Guo Chao Li Chuanyu Li Jia Yao Anran Zheng Jingyi Xu Qingxue Gao Wei Zhang Lianqun Zhou 2022Microsystems & Nanoengineering2022,8,1:1

17Plant G proteins interact with endoplasmic reticulum luminal protein receptors to regulate endoplasmic reticulum retrieval显示文摘Maintaining endoplasmic reticulum(ER) homeostasis is essential for the production of biomolecules.ER retrieval, i.e., the retrograde transport of compounds from the Golgi to the ER, is one of the pathways that ensures ER homeostasis. However, the mechanisms underlying the regulation of ER retrieval in plants remain largely unknown. Plant ERD2-like proteins(ERD2 s) were recently suggested to function as ER luminal protein receptors that mediate ER retrieval. Here, we demonstrate that heterotrimeric G protein signaling is involved in ERD2-mediated ER retrieval. We show that ERD2 s interact with the heterotrimeric G protein Ga and Ggsubunits at the Golgi. Silencing of Ga, Gb, or Gg increased the retention of ER luminal proteins. Furthermore,overexpression of Ga, Gb, or Gg caused ER luminal proteins to escape from the ER, as did the co-silencing of ERD2 a and ERD2 b. 18Key-Recovery Attacks on LED-Like Block Ciphers显示文摘Asymmetric cryptographic schemes, represe nted by RSA, have bee n show n to be in secure un der quantum computing conditions. Correspondingly, there is a need to study whether the symmetric cryptosystem can still guarantee high security with the advent of quantum computers. In this paper, based on the basic principles of classical slide attacks and Simon's algorithm, we take LED-like lightweight block ciphers as research objects to present a security analysis under both classical and quantum attacks, fully considering the influence on the security of the ciphers of adding the round constants. By analyzing the information leakage of round constants, we can introduce the differential of the round constants to propose a classical slide attack on full-round LED-64 with a probability of 1. The analysis result shows that LED-64 is unable to resist this kind of classical slide attack, but that attack method is not applicable to LED-128. As for quantum attacks, by improving on existing quantum attack methods we dem on strate a qua ntum single-key slide attack on LED-64 and a quantum related-key attack on LED- 128, and indicators of the two attack algorithms are analyzed in detail. The attack results show that adding round consta nts does not completely improve the security of the ciphers, and quantum attacks can provide an exp on ential speed-up over the same attacks in the classical model. 20Pad-scale control improves SAGD performance显示文摘Steam Assisted Gravity Drainage(SAGD)is widely used in the Athabasca oil sands deposit to recover bitumen.Since the viscosity of bitumen is high at original reservoir conditions,heat is required to lower its viscosity to the point it becomes mobile enough to be recovered under gravity drainage.To heat the reservoir,steam is injected into the formation and thus SAGD is energy intense.Given that the fuel used to generate steam is the largest operating cost,the steam-to-oil ratio is one of the key parameter for evaluating the economics of any SAGD project.Here,the use of dynamic distributed steam injection within a pad of SAGD wellpairs is explored.The results demonstrate that feedback control leads to improvements of the SOR over that of constant pressure.The results show that the controller is able to detect the“sweet spots”(oil zones with better geological properties)in the reservoir and dynamically deliver more steam to that region.Meanwhile,it reduces the steam injection towards relatively worse quality zones to lower the local SOR.Tao Guo Jingyi Wang Ian D.Gates 2018Petroleum2018,4,3:1
