1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29

3Chemical characterization of size-resolved aerosols in four seasons and hazy days in the megacity Beijing of China显示文摘Size-resolved aerosol samples were collected by MOUDI in four seasons in 2007 in Beijing. The PM10 and PM1.8mass concentrations were 166.0 ± 120.5 and 91.6 ± 69.7 μg/m^3, respectively,throughout the measurement, with seasonal variation: nearly two times higher in autumn than in summer and spring. Serious fine particle pollution occurred in winter with the PM1.8/PM10 ratio of 0.63, which was higher than other seasons. The size distribution of PM showed obvious seasonal and diurnal variation, with a smaller fine mode peak in spring and in the daytime. OM(organic matter = 1.6 × OC(organic carbon)) and SIA(secondary inorganic aerosol) were major components of fine particles, while OM, SIA and Ca_2+were major components in coarse particles. Moreover, secondary components, mainly SOA(secondary organic aerosol) and SIA,accounted for 46%–96% of each size bin in fine particles, which meant that secondary pollution existed all year. Sulfates and nitrates, primarily in the form of(NH_4)_2SO_4, NH_4NO_3, Ca SO_4, Na_2SO_4 and K_2SO_4, calculated by the model ISORROPIA II, were major components of the solid phase in fine particles. The PM concentration and size distribution were similar in the four seasons on non-haze days, while large differences occurred on haze days, which indicated seasonal variation of PM concentration and size distribution were dominated by haze days. 4A fishy odor episode in a north China reservoir: Occurrence, origin, and possible odor causing compounds显示文摘A significant outbreak of fishy odor occurred in a reservoir located in Inner Mongolia, China, in the winter of 2011, and the odor rating, algal density and concentrations of some potential odorous compounds were monitored over a period of two months. The peak odor rating of the fishy odor was 7 according to flavor profile analysis. Among the dominant algal species(two diatom and one chrysophyte species) observed during the survey, the chrysophyte Dinobryon sp. was the most abundant species, with the peak density recorded at 88,520 cells/mL. Seven potential algal metabolites including heptanal, 2,4-heptadienal, 2,4-decadienal, nonanal, 2-octenal, 2,6- nonadienal and hexanal were detected. The principal component analysis result showed that n-hexanal, n-heptanal and 2,4-decadienal, possibly the metabolites of diatoms, and 2,4-heptadienal, possibly the metabolite of Dinobryon sp., might have contributed to the fishy odor episode. 5Single-cell transcriptomic analysis defines the interplay between tumor cells,viral infection,and the microenvironment in nasopharyngeal carcinoma显示文摘Nasopharyngeal carcinoma(NPC)is an Epstein-Barr virus(EBV)-associated malignancy with a complex tumor ecosystem.How the interplay between tumor cells,EBV,and the microenvironment contributes to NPC progression and immune evasion remains unclear.Here we performed single-cell RNA sequencing on^104,000 cells from 19 EBV^+NPCs and 7 nonmalignant nasopharyngeal biopsies,simultaneously profiling the transcriptomes of malignant cells,EBV,stromal and immune cells.Overall,we identified global upregulation of interferon responses in the multicellular ecosystem of NPC.Notably,an epithelial–immune dual feature of malignant cells was discovered and associated with poor prognosis.Functional experiments revealed that tumor cells with this dual feature exhibited a higher capacity for tumorigenesis.Further characterization of the cellular components of the tumor microenvironment(TME)and their interactions with tumor cells revealed that the dual feature of tumor cells was positively correlated with the expression of co-inhibitory receptors on CD8^+tumor-infiltrating T cells.In addition,tumor cells with the dual feature were found to repress IFN-γproduction by T cells,demonstrating their capacity for immune suppression.Our results provide new insights into the multicellular ecosystem of NPC and offer important clinical implications.Shanzhao Jin Ruoyan Li Ming-Yuan Chen Chao Yu Lin-Quan Tang Yan-Min Liu Jiang-Ping Li Yi-Na Liu Yi-Ling Luo Yifan Zhao Yu Zhang Tian-Liang Xia Shang-Xin Liu Qi Liu Guan-Nan Wang Rui You Jing-Yun Peng Jiang Li Feng Han Jianwei Wang Qiu-Yan Chen Li Zhang Hai-Qiang Mai Benjamin E.Gewurz Bo Zhao Lawrence S.Young Qian Zhong Fan Bai Mu-Sheng Zeng 2020Cell Research2020,30,11:16
6Determination of N-nitrosodimethylamine in drinking water by UPLC-MS/MS显示文摘The method for detecting N-nitrosodimethylamine(NDMA) in drinking water using ultra performance liquid chromatography(UPLC) coupled with tandem mass spectrometry(MS/MS) was improved by optimizing the clean-up procedure to remove the matrix interference in pretreatment process,and was then applied to a survey of NDMA in both raw and finished water samples from five water treatment plants in South China.The NDMA concentrations ranged from 4.7 to 15.1 ng/L in raw water samples,and from 4.68 to 46.9 ng/L in finished water.The NDMA concentration in raw water was found to be related with nitrite concentration,and during the treatment,the NDMA concentration increased following ozonation but decreased after subsequent activated carbon treatment.Wanfeng Wang Jianying Hu Jianwei Yu Min Yang 2010Journal of Environmental Sciences2010,22,10:16
7Effects of continuously regenerating diesel particulate filters on regulated emissions and number-size distribution of particles emitted from a diesel engine显示文摘The effects of continuously regenerating diesel particulate filter (CRDPF) systems on regulated gaseous emissions, and number-size distribution and mass of particles emanated from a diesel engine have been investigated in this study. Two CRDPF units (CRDPF-1 and CRDPF-2) with different specifications were separately retrofitted to the engine running with European steady-state cycle (ESC). An electrical low pressure impactor (ELPI) was used for particle number-size distribution measurement and mass estimation. The conversion/reduction rate (R CR ) of hydrocarbons (HC) and carbon monoxide (CO) across CRDPF-1 was 83% and 96.3%, respectively. Similarly, the R CR of HC and CO and across CRDPF-2 was 91.8% and 99.1%, respectively. The number concentration of particles and their concentration peaks; nuclei mode, accumulation mode and total particles; and particle mass were highly reduced with the CRDPF units. The nuclei mode particles at downstream of CRDPF-1 and CRDPF-2 decreased by 99.9% to 100% and 97.8% to 99.8% respectively; and the particle mass reduced by 73% to 92.2% and 35.3% to 72.4%, respectively, depending on the engine conditions. 9Addition of hydrogen peroxide for the simultaneous control of bromate and odor during advanced drinking water treatment using ozone显示文摘Complete removal of the characteristic septic/swampy odor from Huangpu River source water could only be achieved under an ozone dose as high as 4.0 mg/L in an ozone-biological activated carbon(O3-BAC) process, which would lead to the production of high concentrations of carcinogenic bromate due to the high bromide content. This study investigated the possibility of simultaneous control of bromate and the septic/swampy odor by adding H2O2 prior to the O3-BAC process for the treatment of Huangpu River water. H2O2 addition could reduce the bromate concentration effectively at an H2O2/O3(g/g) ratio of 0.5 or higher. At the same time, the septic/swampy odor removal was enhanced by the addition of H2O2, although optimization of the H2O2/O3 ratio was required for each ozone dose. At an ozone dose of 2.0 mg/L, the odor was removed completely at an H2O2/O3 ratio of 0.5. 11Characterization of disinfection byproduct formation potential in 13 source waters in China显示文摘The formation potential of four trihalomethanes (THMFP) and seven haloacetic acids (HAA 7 FP) in 13 source waters taken from four major water basin areas in China was evaluated using the simulated distribution system (SDS) chlorination method.The specific ultraviolet absorbance (SUVA 254:the ratio of UV 254 to dissolved organic carbon (DOC)),which ranged between 0.9 and 5.0 L/(mg·m),showed that the organic compounds in different source waters exhibited different reactivities with chlorine.The HAA 7 FP of source waters ranged from 20 to 448 μg/L and the THMFP ranged from 29 to 259 μg/L.The HAA 7 FP concentrations were higher than the THMFP concentrations in all but one of the samples.Therefore,the risks of haloacetic acids (HAAs) should be of concern in some source waters.TCM (chloroform) and BDCM (bromodichloromethane) were the major THM constituents,while TCAA (trichloroacetic acid) and DCAA (dichloroacetic acid) were the major HAA species.Br-THM (brominated THM species) were much higher than BrHAA (brominated HAA species),and the formation of Br-DBP (Br-THM and Br-HAA) should be of concern when the bromide concentration is over 100 μg/L.Junzhi Zhang Jianwei Yu Wei An Juan Liu Yongjing Wang Youjun Chen Jia Tai Min Yang 2011Journal of Environmental Sciences2011,23,2:13

13Lattice-Based Double-Authentication-Preventing Ring Signature for Security and Privacy in Vehicular Ad-Hoc Networks显示文摘Amidst the rapid development of the Internet of Things (loT), Vehicular Ad-Hoc NETwork (VANET), a typical loT application, are bringing an ever-larger number of intelligent and convenient services to the daily lives of individuals. However, there remain challenges for VANETs in preserving privacy and security. In this paper, we propose the first lattice-based Double-Authentication-Preventing Ring Signature (DAPRS) and adopt it to propose a novel privacy-preserving authentication scheme for VANETs, offering the potential for security against quantum computers. The new construction is proven secure against chosen message attacks. Our scheme is more efficient than other ring signature in terms of the time cost of the message signing phase and verification phase, and also in terms of signature length. 14Spatial and temporal variations of two cyanobacteria in the mesotrophic Miyun reservoir, China显示文摘Spatial variations in phytoplankton community within a large mesotrophic reservoir(Miyun reservoir, North China) were investigated in relation to variations in physico-chemical properties, nutrient concentrations, temperature and light conditions over a 5 month period in 2009. The dynamics of phytoplankton community was represented by the dominance of cyanobacteria through summer and fall, following with a short term dominance of chlorophyta in late fall, and a relatively high abundance of diatom in October; on the other hand, maximum phytoplankton biomass was recorded in the north shallow region of Miyun reservoir with a higher nutrients level. Particular attention was paid to the impacts of environmental conditions on the growth of two cyanobacteria genera, the toxin-producing Microcystis and the taste & odor-producing Oscillatoria. Microcystis biomass was in general greatly affected by water temperature and mixing depth/local water depth ratio in this reservoir, while the Oscillatoria biomass in the surface and middle layers was greatly affected by total dissolved phosphorus, and that in the bottom layer was related with the Secchi depth/local water depth ratio. 15Advanced oxidation of bromide-containing drinking water: A balance between bromate and trihalomethane formation control显示文摘Addition of H2O2 has been employed to repress bromate formation during ozonation of bromide-containing source water.However,the addition of H2O2 will change the oxidation pathways of organic compounds due to the generation of abundant hydroxyl radicals,which could afect the removal efcacy of trihalomethane precursors via the combination of ozone and biological activated carbon(O3-BAC).In this study,we evaluated the efects of H2O2 addition on bromate formation and trihalomethane formation potential(THMFP)reduction during treatment of bromide-containing(97.6–129.1μg/L)source water by the O3-BAC process.At an ozone dose of 4.2 mg/L,an H2O2/O3(g/g)ratio of over 1.0 was required to maintain the bromate concentration below 10.0μg/L,while a much lower H2O2/O3ratio was sufcient for a lower ozone dose.An H2O2/O3(g/g)ratio below 0.3 should be avoided since the bromate concentration will increase with increasing H2O2 dose below this ratio.However,the addition of H2O2 at an ozone dose of 3.2 mg/L and an H2O2/O3 ratio of 1.0 resulted in a 43% decrease in THMFP removal when compared with the O3-BAC process.The optimum H2O2/O3(g/g) ratio for balancing bromate and trihalomethane control was about 0.7–1.0.Fractionation of organic materials showed that the addition of H2O2 decreased the removal efcacy of the hydrophilic matter fraction of DOC by ozonation and increased the reactivity of the hydrophobic fractions during formation of trihalomethane,which may be the two main reasons responsible for the decrease in THMFP reduction efcacy.Overall,this study clearly demonstrated that it is necessary to balance bromate reduction and THMFP control when adopting an H2O2 addition strategy.Yongjing Wang Jianwei Yu Po Han Jing Sha Tao Li Wei An Juan Liu Min Yang 2013Journal of Environmental Sciences2013,25,11:9
16Chemical characteristics of size-resolved aerosols in winter in Beijing显示文摘Size-resolved aerosols were continuously collected by a Nano Sampler for 13 days at an urban site in Beijing during winter 2012 to measure the chemical composition of ambient aerosol particles. Data collected by the Nano Sampler and an ACSM(Aerodyne Aerosol Chemical Speciation Monitor) were compared. Between the data sets,similar trends and strong correlations were observed,demonstrating the validity of the Nano Sampler. PM10 and PM2.5concentrations during the measurement were 150.5 ± 96.0 μg/m3(mean ± standard variation)and 106.9 ± 71.6 μg/m3,respectively. The PM2.5/PM10 ratio was 0.70 ± 0.10,indicating that PM2.5dominated PM10. The aerosol size distributions showed that three size bins of 0.5–1,1–2.5 and 2.5–10 μm contributed 21.8%,23.3% and 26.0% to the total mass concentration(TMC),respectively. OM(organic matter) and SIA(secondary ionic aerosol,mainly SO42-,NO3-and NH4+) were major components of PM2.5. Secondary compounds(SIA and secondary organic carbon) accounted for half of TMC(about 49.8%) in PM2.5,and suggested that secondary aerosols significantly contributed to the serious particulate matter pollution observed in winter. Coal burning,biomass combustion,vehicle emissions and SIA were found to be the main sources of PM2.5. 18Experimental study on the goaf flow field of the ‘‘U+I” type ventilation system for a comprehensive mechanized mining face显示文摘‘‘U' and ‘‘U+I' type ventilation experiments were performed on a three-dimensional fully mechanized caving face simulation experimental platform. The distribution laws of the pressure field and gas field in the mine goaf were obtained. Results show that the flow field in the goaf is generally asymmetric;the location of the gas accumulation area changes with ventilation parameters and can be used as an evaluation indicator to study the air leakage extent in the goaf. Hence, drainage pipes buried in the goaf to intensively extract gas can be designed in such gas areas, which can give considerations in both improving gas drainage efficiency and reducing air leakage. By comparing the gas extraction effect of model experiments with that of on-site underground practices, the basic laws are commonly consistent according to comparative analysis. 19Risk assessment of Giardia in rivers of southern China based on continuous monitoring显示文摘The occurrence and risks of Giardia in China have been unclear to date,which has made it difficult to properly manage source water as well as to create reasonable drinking water standards.The levels of Giardia in river networks of several cities in Zhejiang Province,China were found to be in the range of 0-5 oocysts/10 L in the rainy season in 2008.The mortality due to Giardia infection for people in this region was calculated to be from 0 to 1.95 × 10?8 persons using a conditional probability equation.Based on multiple unboiled water intake routes,the disability-adjusted life years(DALYs) due to Giardia infection for people who consumed conventionally treated water was 0.625(95% CI:0.137-2.05) per 105 persons,with the symptom of hospitalization making the highest contribution to total DALYs(0.56 per 105 persons;95% CI:0.122-1.84).The DALYs decreased to 0.425(95% CI:0.137-2.05) per 105 persons per year for those consuming water treated with advanced technology.These values were lower than the acceptable risk(1.97 × 10?5 DALYs per year).This study revealed the risk of Giardia infection to the people in river networks of Zhejiang Province for the first time,and provides a method to evaluate the risk of Giardia infection.The results are useful for the modification of drinking water quality standards based on cost-benefit analysis.Wei An Dongqing Zhang Shumin Xiao Jianwei Yu Min Yang 2012Journal of Environmental Sciences2012,24,2:8
20Shaking table experimental study of recycled concrete frame-shear wall structures显示文摘In this study, four 1/5 scaled shaking table tests were conducted to investigate the seismic performance of recycled concrete frame-shear wall structures with different recycled aggregates replacement rates and concealed bracing detail. The four tested structures included one normal concrete model, one recycled coarse aggregate concrete model, and two recycled coarse and fi ne aggregate concrete models with or without concealed bracings inside the shear walls. The dynamic characteristics, dynamic response and failure mode of each model were compared and analyzed. Finite element models were also developed and nonlinear time-history response analysis was conducted. The test and analysis results show that the seismic performance of the recycled coarse aggregate concrete frame-shear wall structure is slightly worse than the normal concrete structure. The seismic resistance capacity of the recycled concrete frame-shear wall structure can be greatly improved by setting up concealed bracings inside the walls.
