1Osthole inhibits proliferation of human breast cancer cells by inducing cell cycle arrest and apoptosis显示文摘Recent studies have revealed that osthole,an active constituent isolated from the fruit of Cnidium monnieri(L.) Cusson,a traditional Chinese medicine,possesses anticancer activity.However,its effect on breast cancer cells so far has not been elucidated clearly.In the present study,we evaluated the effects of osthole on the proliferation,cell cycle and apoptosis of human breast cancer cells MDA-MB 435.We demonstrated that osthole is effective in inhibiting the proliferation of MDA-MB 435 cells,The mitochondrion-mediated apoptotic pathway was involved in apoptosis induced by osthole,as indicated by activation of caspase-9 and caspase-3 followed by PARP degradation.The mechanism underlying its effect on the induction of G1 phase arrest was due to the up-regulation of p53 and p21 and down-regulation of Cdk2 and cyclin D1 expression.Were observed taken together,these findings suggest that the anticancer efficacy of osthole is mediated via induction of cell cycle arrest and apoptosis in human breast cancer cells and osthole may be a potential chemotherapeutic agent against human breast cancer.Lintao Wang Yanyan Peng Kaikai Shi Haixiao Wang Jianlei Lu Yanli Li Changyan Ma 2015The Journal of Biomedical Research2015,29,2:12
2Microstructure and mechanical properties of ultra-fine grained MoNbTaTiV refractory high-entropy alloy fabricated by spark plasma sintering显示文摘The MoNbTaTiV refractory high-entropy alloy(RHEA)with ultra-fine grains and homogeneous microstructure was successfully fabricated by mechanical alloying(MA)and spark plasma sintering(SPS).The microstructural evolutions,mechanical properties and strengthening mechanisms of the alloys were systematically investigated.The nanocrystalline mechanically alloyed powders with simple bodycentered cubic(BCC)phase were obtained after 40 h MA process.Afterward,the powders were sintered using SPS in the temperature range from 1500℃to 1700℃.The bulk alloys were consisted of submicron scale BCC matrix and face-centered cubic(FCC)precipitation phases.The bulk alloy sintered at 1600℃had an average grain size of 0.58μm and an FCC precipitation phase of 0.18μm,exhibiting outstanding micro-hardness of 542 HV,compressive yield strength of 2208 MPa,fracture strength of 3238 MPa and acceptable plastic strain of 24.9%at room temperature.The enhanced mechanical properties of the MoNbTaTiV RHEA fabricated by MA and SPS were mainly attributed to the grain boundary strengthening and the interstitial solid solution strengthening.It is expectable that the MA and SPS processes are the promising methods to synthesize ultra-fine grains and homogenous microstructural RHEA with excellent mechanical properties.Qing Liu Guofeng Wang Xiaochong Sui Yongkang Liu Xiao Li Jianlei Yang 2019Journal of Materials Science & Technology2019,35,11:11
3Allogeneic Vγ9Vδ2 T-cell immunotherapy exhibits promising clinical safety and prolongs the survival of patients with late-stage lung or liver cancer显示文摘Vγ9Vδ2 T cells are promising candidates for cellular tumor immunotherapy.Due to their HLA-independent mode of action,allogeneic Vγ9Vδ2 T cells can be considered for clinical application.To apply allogeneic Vγ9Vδ2 T cells in adoptive immunotherapy,the methodology used to obtain adequate cell numbers with optimal effector function in vitro needs to be optimized,and clinical safety and efficacy also need to be proven.Therefore,we developed a novel formula to improve the expansion of peripheralγδT cells from healthy donors.Then,we used a humanized mouse model to validate the therapeutic efficacy of expandedγδT cells in vivo;furthermore,the expandedγδT cells were adoptively transferred into late-stage liver and lung cancer patients.We found that the expanded cells possessed significantly improved immune effector functions,including proliferation,differentiation,and cancer cell killing,both in vitro and in the humanized mouse model.Furthermore,a phase I clinical trial in 132 late-stage cancer patients with a total of 414 cell infusions unequivocally validated the clinical safety of allogeneic Vγ9Vδ2 T cells.Among these 132 patients,8 liver cancer patients and 10 lung cancer patients who received≥5 cell infusions showed greatly prolonged survival,which preliminarily verified the efficacy of allogeneic Vγ9Vδ2 T-cell therapy.Our clinical studies underscore the safety and efficacy of allogeneic Vγ9Vδ2 T-cell immunotherapy,which will inspire further clinical investigations and eventually benefit cancer patients.Yan Xu Zheng Xiang Mohammed Alnaggar Léonce Kouakanou Jiawei Li Junyi He Jiashuang Yang Yi Hu Yan Chen Li Lin Jianlei Hao Jingxia Li Jibing Chen Man Li Qingling Wu Christian Peters Qinghua Zhou Jianshuang Li Yingqing Liang Xiaohua Wang Baohui Han Meili Ma Dieter Kabelitz Kecheng Xu Wenwei Tu Yangzhe Wu Zhinan Yin 2021Cellular & Molecular Immunology2021,18,2:8

5On-board measurements of gaseous pollutant emission characteristics under real driving conditions from light-duty diesel vehicles in Chinese cities显示文摘A total of 15 light-duty diesel vehicles(LDDVs) were tested with the goal of understanding the emission factors of real-world vehicles by conducting on-board emission measurements. The emission characteristics of hydrocarbons(HC) and nitrogen oxides(NOx) at different speeds, chemical species profiles and ozone formation potential(OFP) of volatile organic compounds(VOCs) emitted from diesel vehicles with different emission standards were analyzed. The results demonstrated that emission reductions of HC and NOxhad been achieved as the control technology became more rigorous from Stage I to Stage IV. It was also found that the HC and NOxemissions and percentage of O2 dropped with the increase of speed, while the percentage of CO2 increased. The abundance of alkanes was significantly higher in diesel vehicle emissions, approximately accounting for 41.1%–45.2%, followed by aromatics and alkenes. The most abundant species were propene,ethane, n-decane, n-undecane, and n-dodecane. The maximum incremental reactivity(MIR)method was adopted to evaluate the contributions of individual VOCs to OFP. The results indicated that the largest contributors to O3 production were alkenes and aromatics, which accounted for 87.7%–91.5%. Propene, ethene, 1,2,4-trimethylbenzene, 1-butene, and1,2,3-trimethylbenzene were the top five VOC species based on their OFP, and accounted for 54.0%-64.8% of the total OFP. The threshold dilution factor was applied to analyze the possibility of VOC stench pollution. 6TAZ inhibits osteoclastogenesis by attenuating TAK1/NF-κB signaling显示文摘Osteoporosis is an osteolytic disorder commonly associated with excessive osteoclast formation.Transcriptional coactivator with PDZ-binding motif(TAZ)is a key downstream effector of the Hippo signaling pathway;it was suggested to be involved in the regulation of bone homeostasis.However,the exact role of TAZ in osteoclasts has not yet been established.In this study,we demonstrated that global knockout and osteoclast-specific knockout of TAZ led to a low-bone mass phenotype due to elevated osteoclast formation,which was further evidenced by in vitro osteoclast formation assays.Moreover,the overexpression of TAZ inhibited RANKL-induced osteoclast formation,whereas silencing of TAZ reduced it.Mechanistically,TAZ bound to TGF-activated kinase 1(TAK1)and reciprocally inhibited NF-κB signaling,suppressing osteoclast differentiation.Collectively,our findings highlight an essential role of TAZ in the regulation of osteoclastogenesis in osteoporosis and its underlying mechanism.Wanlei Yang Xuanyuan Lu Tan Zhang Weiqi Han Jianlei Li Wei He Yewei Jia Kangxian Zhao An Qin Yu Qian 2021Bone Research2021,9,3:6

8Toll-like receptor 7 deficiency suppresses type 1 diabetes development by modulating B-cell differentiation and function显示文摘Innate immunity mediated by Toll-like receptors(TLRs),which can recognize pathogen molecular patterns,plays a critical role in type 1 diabetes development.TLR7 is a pattern recognition receptor that senses single-stranded RNAs from viruses and host tissue cells;however,its role in type 1 diabetes development remains unclear.In our study,we discovered that Tlr7-deficient(Tlr7^(−/−))nonobese diabetic(NOD)mice,a model of human type 1 diabetes,exhibited a significantly delayed onset and reduced incidence of type 1 diabetes compared with Tlr7-sufficient(Tlr7^(+/+))NOD mice.Mechanistic investigations showed that Tlr7 deficiency significantly altered B-cell differentiation and immunoglobulin production.Moreover,Tlr7^(−/−)NOD B cells were found to suppress diabetogenic CD4^(+)T-cell responses and protect immunodeficient NOD mice from developing diabetes induced by diabetogenic T cells.In addition,we found that Tlr7 deficiency suppressed the antigen-presenting functions of B cells and inhibited cytotoxic CD8^(+)T-cell activation by downregulating the expression of both nonclassical and classical MHC class I(MHC-I)molecules on B cells.Our data suggest that TLR7 contributes to type 1 diabetes development by regulating B-cell functions and subsequent interactions with T cells.Therefore,therapeutically targeting TLR7 may prove beneficial for disease protection.Juan Huang Jian Peng James Alexander Pearson Georgios Efthimiou Youjia Hu Ningwen Tai Yanpeng Xing Luyao Zhang Jianlei Gu Jianping Jiang Hongyu Zhao Zhiguang Zhou F.Susan Wong Li Wen 2021Cellular & Molecular Immunology2021,18,2:3
9A new dynamic method for measuring hydrogen partial pressure in molten aluminum alloy显示文摘Hydrogen partial pressure is an important parameter to calculate hydrogen concentration levels in molten aluminum alloy. A new dynamic method for measuring hydrogen partial pressure in molten aluminum alloy is studied. Dynamic and rapid measurement is realized through changing the volume of the vacuum chamber and calculating the pressure difference ΔP between the theoretical and measured pressures in the vacuum chamber. Positive ΔP indicates hydrogen transmits from melt to vacuum chamber and negative ΔP means the reverse. When ΔP is equal to zero, hydrogen transmitted from both sides reached a state of dynamical equilibrium and the pressure in the vacuum chamber is equal to the hydrogen partial pressure in the molten aluminum alloy. 10Simulation of the Equatorially Asymmetric Mode of the Hadley Circulation in CMIP5 Models显示文摘The tropical Hadley circulation(HC) plays an important role in influencing the climate in the tropics and extra-tropics.The realism of the climatological characteristics, spatial structure, and temporal evolution of the long-term variation of the principal mode of the annual mean HC(i.e., the equatorially asymmetric mode, EAM) was examined in model simulations from the Coupled Model Intercomparison Project Phase 5(CMIP5). The results showed that all the models are moderately successful in capturing the HC's climatological features, including the spatial pattern, meridional extent, and intensity, but not the spatial or temporal variation of the EAM. The possible reasons for the poor simulation of the long-term variability of the EAM were explored. None of the models can successfully capture the differences in the warming rate between the tropical Southern Hemisphere(SH) and Northern Hemisphere(NH), which is considered to be an important driver for the variation of the AM. Most of the models produce a faster warming in the NH than in the SH, which is the reverse of the observed trend.This leads to a reversed trend in the meridional gradient between the SH and NH, and contributes to the poor simulation of EAM variability. Thus, this aspect of the models should be improved to provide better simulations of the variability of the HC. 13Performance of rectangular closed diaphragm walls in gently sloping liquefiable deposits subjected to different earthquake ground motions显示文摘The performance of rectangular closed diaphragm walls(RCDW)subjected to earthquake ground motions is extremely complicated in gently sloping liquefiable deposits and requires further investigation.A nonlinear finite element(FE)model was developed to investigate the seismic performance of an RCDW in the OpenSees platform.Initially,the feasibility of the FE model to simulate the seismic behavior of the RCDW was validated by comparing the numerical results with the experimental data.The numerical results agree well with the centrifuge test data.Then,the calibrated model was used to study the seismic performance of the RCDW subjected to different ground motions in gently sloping liquefiable deposits.The numerical results indicate that the soil-RCDW system under near-fault ground motions is more likely to be damaged than that under far-fault ground motions.The difference between the maximum and minimum(Dmax-min)the RCDW displacement and the maximum excess pore pressure in the soil core increased as the Arias intensity increased.The seismic response of the soil-RCDW system was strongly affected by the frequency content,durations,energy distribution and initial directions of the ground motions.Moreover,the modified specific energy density(MSED)has a good linear relationship with the Dmax-min of the RCDW displacement.Zhang Jianlei Cheng Qiangong Li Yan Qiu Yuheng Wang Yufeng Wu Jiujiang 2021Earthquake Engineering and Engineering Vibration2021,20,4:2
14Numerical study on evolution of subharmonic varicose low-speed streaks in turbulent channel flow显示文摘The evolution of two spanwise-aligned low-speed streaks in a wall turbulent flow, triggered by the instability of the subharmonic varicose(SV) mode, is studied by a direct numerical simulation(DNS) method in a small spatial-periodic channel. The results show that the SV low-speed streaks are self-sustained at the early stage, and then transform into subharmonic sinuous(SS) low-speed streaks. Initially, the streamwise vortex sheets are formed by shearing, and then evolve into zigzag vortex sheets due to the mutual induction. As the intensification of the SV low-speed streaks becomes prominent,the tilted streamwise vortex tubes and the V-like streamwise vortex tubes can be formed simultaneously by increasing +? u? x. When the SV low-speed streaks break down, new zigzag streamwise vortices will be generated, thus giving birth to the next sustaining cycle of the SV low-speed streaks. When the second breakdown happens, new secondary V-like streamwise vortices instead of zigzag streamwise vortices will be generated. 16A new strategy to achieve enhanced upconverted circularly polarized luminescence in chiral perovskite nanocrystals显示文摘Achieving large luminescence dissymmetry factors(pium)is challenging in the research field of circularly polarized luminescence(CPL).While various approaches have been developed to construct organic systems with CPL activity,there is still a lack of effective methods for fabricating CPL active inorganic materials.Herein,we propose an approach for endowing upconversion nanoparticles(UCNPs)and perovskite nanocrystal(PKNC)hybrid nanomaterials with upconverted circularly polarized luminescence(UC-CPL)activity.Chiral cesium lead bromides(CsPbBr_(3))PKNCs were synthesized by a chiral-ligand-assistant method.Meanwhile,UCNP could be embedded into the chiral PKNC,enabling a photon upconvesion feature to the PKNC.The embedded UCNPs in PKNCs were confirmed by electron tomography.Consequently,various CPL activities,including prompt CPL,UC-CPL,and energy transfer enhanced circularly polarized luminescence(ET-CPL),were realized.The chiral perovskite nanocrystals could reabsorb the chiral energy generated from UCNPs,showing energy transfer enhanced CPL activity with four times magnification of the circular polarization.These findings provide a meaningful strategy for designing chiral photon upconversion inorganic nanomaterials with highly efficient UC-CPL activity.Xue Jin Minghao Zhou Jianlei Han Bin Li Tianyong Zhang Shuang Jiang Pengfei Duan 2022Nano Research2022,15,2:1
17Relationships among the Monsoon-like Southwest Australian Circulation,the Southern Annular Mode, and Winter Rainfall over Southwest Western Australia显示文摘This study examines the relationships among the monsoon-like southwest Australian circulation(SWAC), the Southern Annular Mode(SAM), and southwest Western Australia winter rainfall(SWR), based on observed rainfall, reanalysis datasets, and the results of numerical modeling. By decomposing the SWAC into two components using a linear model, i.e.the component related to SAM(RSAM) and the component unrelated to SAM(SWACI*), we find it is the SWACI* that shows a significant influence on SWR. Similarly, it is the component of SAM associated with SWAC that exhibits an impact on SWR, whereas the component unrelated to SAM. A similar result is obtained in terms of the circulation associated with SWAC and the SAM. These facts suggest the SAM plays an indirect role in influencing SWR, and raise the possibility that SWAC acts as a bridge between the SAM and SWR, by which the SAM passes its influences onto SWR. This is due to the fact that the variations of SWAC are closely linked to the thermal contrast between land and sea across the southern Indian Ocean and southwest Australia. By contrast, the SAM does not significantly relate to this thermal structure, particularly for the component unrelated to SWAC. The variations of surface sea temperature over the southern Indian Ocean contribute to the favored rainfall circulation patterns. This finding is supported by the numerical modeling results.
