1Alteration of the microRNA expression profile and identification of miRNA/mRNA negative regulation pairs in neural tube defects显示文摘MicroRNAs (miRNAs) are small, noncoding transcripts, and ~22 nucleotides long, which belong to a class of endogenous small regulatory RNA molecules that target mRNAs and trigger either translational suppression or mRNA degradation [1–3]. Several studies have shown that miRNAs play crucial roles in neurogenesis and development of nervous system [4,5] and have distinct expression patterns within the developing brain and central nervous system (CNS)[6]. Abnormal expression of miRNAs has also been reported in dysregulated neural tube (NT) development including NT defects (NTDs)[7,8]. To date, however, all NTD-related miRNA profiles were obtained from different microarray platforms, which always contain limited probes, and the data obtained are less precise in quantitative analysis. No quantitatively profiled global miRNA expression in mouse NTD fetuses has been characterized so far. To gain further insight of the possible roles of miRNAs in NTDs, we used deep sequencing to carry out a comprehensive survey of miRNA expression in all-trans retinoic acid (RA)-induced NTD mouse fetuses collected from three different embryonic time points (E8.5, E9.5, and E10.5). 2Immune signature profiling identified prognostic factors for gastric cancer显示文摘Objective: Tumor microenvironment, especially the host immune system, plays a pivotal role in tumor initiation and progression. Profiling of immune signature within tumor might uncover biomarkers for targeted therapies and clinical outcomes. However, systematic analysis of immune-related genes in gastric cancer(GC) has not been reported.Methods: Expressions of a total of 718 immune-related genes were generated in 372 stomach adenocarcinoma(STAD) patients from The Cancer Genome Atlas(TCGA) database using RNA-sequencing data. Integrated bioinformatics analyses were performed to identify prognostic factors as well.Results: Survival analyses revealed 73 genes, which were significantly associated with patient’s overall survival(OS). Taken together with clinicopathological parameters, we established a predictive model, containing 10 immune-related genes, which were NRP1, C6, CXCR4, LBP, PNMA1, TLR5, ITGA6, MICB, PBK and TNFRSF18,with powerful efficiency in distinguishing satisfactory or poor survival of STAD patients. Moreover, the top 3 ranked prognostic genes, NRP1, TGFβ2 and MFGE8, were also significantly associated with patient’s OS by an independent validation achieved from Kaplan-Meier plotter database.Conclusions: We profiled prognostic immune signature and established prognostic predictive model for GC,which could reflect immune disorders within tumor microenvironment, and also may provide novel predictive and therapeutic targets for GC patients in the near future.Wenhui Yang Zhiyong Lai Yuan Li Jianbing Mu Mudan Yang Jun Xie Jun Xu 2019Chinese Journal of Cancer Research2019,31,3:5

4Excessive apoptosis and ROS induced by ethionine affect neural cell viability and differentiation显示文摘The central nervous system(CNS)diseases are still a major cause of morbidity and mortality throughout the world,which imposes heavy burden on the development of society.Ethionine is a non-proteinogenic amino acid having similar chemical structure and activity to that of methionine,with which it competes.Previous studies have confirmed that ethionine affects various cellular functions by inhibiting the biosynthesis of proteins,RNA,DNA,and phospholipids,or all of them.The relationship of ethionine with some CNS diseases,including neural tube defects,has been investigated recently.However,the detailed effects of ethionine on the nerve cell bioactivities and the underlying mechanisms have not been fully explored.Herein,we systematically investigated the influences of ethionine on the proliferation,differentiation,and apoptosis of neural stem cells(NSCs)and post-mitotic nerve cells.We demonstrated that ethionine inhibited cell viability by disrupting the balance between proliferation and apoptosis,prevented NSCs from differentiating into neurons and astrocytes,and blocked cell progression from G1 to S phase via reducing cyclin D1 function in nerve cells including NSCs,a mouse hippocampal neuron cell line(HT-22),and a mouse brain neuroma cell line(Neuro-2a).We speculated that the inhibitory effect of ethionine on cell viability and differentiation are associated with increased reactive oxygen species production.Our results also supported the concept that ethionine may be an underlying cause of abnormal folate metabolism-induced CNS diseases.Our findings may provide important direction for the application of abnormal folate metabolism-induced CNS diseases in future NSC-based therapies.Li Zhang Dandan Li Juan Zhang Ping Yan Xueqin Liu Lei Wang Ajab Khan Zhizhen Liu Jianbing Mu Jun Xu Bo Niu Jun Xie 2020Acta Biochimica et Biophysica Sinica2020,52,10:1

7Gully erosion and expansion mechanisms in loess tablelands and the scientific basis of gully consolidation and tableland protection显示文摘Gully erosion is serious in the tableland area of the Loess Plateau due to high-intensity human activities and extreme rainfall, which cause serious soil loss and an increasing tableland shrinkage rate. Severe gully erosion has exerted a notable negative impact on local agriculture, human life and socioeconomic development. In recent decades, progress has been made in soil and water conservation with the goal of reducing soil erosion and protecting loess tableland, but basic research on gully consolidation and tableland protection(GCTP) is lacking, especially regarding the mechanisms of gully erosion and expansion in loess tableland under the interactive impacts of hydrodynamics and human activities. In addition, there is a lack of a deep understanding of the underlying mechanisms of soil-water disasters and controlling factors of unreasonable GCTP projects.Currently, the problems of headcut erosion and tableland fragmentation remain serious. Based on this situation, the Dongzhi tableland, the largest tableland on the Loess Plateau, was adopted as an example, and we studied gully erosion and expansion mechanisms in the loess tableland and the scientific basis of GCTP projects. We obtained a series of novel findings, including the following:(1) vertical joints are widely developed in loess and impose a controlling effect on tableland edge erosion;(2) rapid urbanization and road network expansion intensify headcut erosion and are the main reasons for severe erosion and tableland shrinkage in the Dongzhi tableland;and(3) unreasonable drainage of surface runoff and a rise in the groundwater level are the key factors affecting GCTP project stability. Moreover, the mechanisms and modes of erosion disasters in the project driven by these two factors were explained. The systematic remediation idea of retention, storage, drainage and consolidation for the GCTP project was introduced, and the core is water control, which emphasizes the combination of soil and water conservation and geohazard prevention measures.
