| 1 | Short-chain fatty acids act as antiinflammatory mediators by regulating prostaglandin E_2 and cytokines显示文摘AIM:To investigate the effect of short-chain fatty ac-ids (SCFAs) on production of prostaglandin E2 (PGE2),cytokines and chemokines in human monocytes.METHODS:Human neutrophils and monocytes were isolated from human whole blood by using 1-Step Polymorph and RosetteSep Human Monocyte Enrich-ment Cocktail,respectively. Human GPR41 and GPR43 mRNA expression was examined by quantitative real-time polymerase chain reaction. The calcium flux assay was used to examine the biological activities of SCFAs in human neutrophils and monocytes. The effect ofSCFAs on human monocytes and peripheral blood mononuclear cells (PBMC) was studied by measuring PGE2,cytokines and chemokines in the supernatant. The effect of SCFAs in vivo was examined by intraplantar injection into rat paws.RESULTS:Human GPR43 is highly expressed in human neutrophils and monocytes. SCFAs induce robust calcium flux in human neutrophils,but not in human monocytes. In this study,we show that SCFAs can in-duce human monocyte release of PGE2 and that this effect can be enhanced in the presence of lipopolysac-charide (LPS). In addition,we demonstrate that PGE2 production induced by SCFA was inhibited by pertussis toxin,suggesting the involvement of a receptor-mediated mechanism. Furthermore,SCFAs can specifi cally inhibit constitutive monocyte chemotactic protein-1 (MCP-1) production and LPS-induced interleukin-10 (IL-10) production in human monocytes without affecting the secretion of other cytokines and chemokines examined. Similar activities were observed in human PBMC for the release of PGE2,MCP-1 and IL-10 after SCFA treatment. In addition,SCFAs inhibit LPS-induced production of tumor necrosis factor-α and interferon-γ in human PBMC. Finally,we show that SCFAs and LPS can induce PGE2 production in vivo by intraplantar injection into rat paws (P < 0.01). | 2 | Phylogenetic reconstruction of Chirita and allies (Gesneriaceae) with taxonomic treatments显示文摘Chirita D. Don, a large genus in the subfamily Cyrtandroideae of Gesneriaceae, has been the subject of much debate whether it is a natural group or not. In addition, the highly heterogeneous Chirita has also been very problematic with regard to delimitation and subdivision. Here we used the nrDNA internal transcribed spacer and cpDNA trnL-F for molecular phylogenetic analaysis, combined with morphological data. Our results suggest that Chirita is an artificial, polyphyletic genus. The most important character that defines Chirita, the dorso-ventrally oblique and bilamellar stigma, has evolved convergently in different clades of diandrous Cyrtandroideae. Chirita sensu stricto only includes the species of Chirita sect. Chirita, whereas Chirita sect. Microchirita is an independent clade located at the basal node of the phylogenetic tree. Chirita sect. Liebigia is closely related to Didymocarpus with an entire stigma unlike other species of Chirita. The species of Chirita sect. Gibbosaccus, Chiritopsis, Primulina, and Wentsaiboea form a monophyletic group that is sister to a strongly supported clade comprising four monotypic genera Paralagarosolen, Calcareoboea, Petrocodon, and Tengia. | 3 | Intensity-modulated radiation therapy with concurrent chemotherapy for locally advanced cervical and upper thoracic esophageal cancer显示文摘AIM: To evaluate the dosimetry, efficacy and toxicity of intensity-modulated radiation therapy (IMRT) and concurrent chemotherapy for patients with locally advanced cervical and upper thoracic esophageal cancer. METHODS: A retrospective study was performed on 7 patients who were definitively treated with IMRT and concurrent chemotherapy. Patients who did not receive IMRT radiation and concurrent chemotherapy were not included in this analysis. IMRT plans were evaluated to assess the tumor coverage and normal tissue avoidance. Treatment response was evaluated and toxicities were assessed. RESULTS: Five- to nine-beam IMRT were used to deliver a total dose of 59.4-66 Gy (median: 64.8 Gy) to the primary tumor with 6-MV photons. The minimum dose received by the planning tumor volume (PTV) of the gross tumor volume boost was 91.2%-98.2% of the prescription dose (standard deviation [SD]: 3.7%-5.7%). The minimum dose received by the PTV of the clinical tumor volume was 93.8%-104.8% (SD: 4.3%-11.1%) of the prescribed dose. With a median follow-up of 15 mo (range: 3-21 mo), all 6 evaluable patients achieved complete response. Of them, 2 developed localrecurrences and 2 had distant metastases, 3 survived with no evidence of disease. After treatment, 2 patients developed esophageal stricture requiring frequent dilation and 1 patient developed tracheal-esophageal fi stula. CONCLUSION: Concurrent IMRT and chemotherapy resulted in an excellent early response in patients with locally advanced cervical and upper thoracic esophageal cancer. However, local and distant recurrence and toxicity remain to be a problem. | 5 | Efficient Targeted Genome Modification in Maize Using CRISPR/Cas9 System显示文摘CRISPR(clustered regularly interspaced short palindromic repeats)/Cas9 system, which is a newly developed technology for targeted genome modification, has been successfully used in a number of species. In this study, we applied this technology to carry out targeted genome modification in maize. A marker gene Zmzb7 was chosen for targeting. The sg RNA-Cas9 construct was transformed into maize protoplasts, and indel(insertion and deletion) mutations could be detected. A mutant seedling with an expected albino phenotype was obtained from screening 120 seedlings generated from 10 callus events. Mutation efficiency in maize heterochromatic regions was also investigated. Twelve sites with different expression levels in maize centromeres or pericentromere regions were selected. The sg RNACas9 constructs were transformed into protoplasts followed by sequencing the transformed protoplast genomic DNA. The results show that the genes in heterochromatic regions could be targeted by the CRISPR/Cas9 system efficiently, no matter whether they are expressed or not. Meanwhile, off-target mutations were not found in the similar sites having no PAM(protospacer adjacent motif) or having more than two mismatches. Plant adaptation to long-day conditions at higher latitudes requires early flowering and a reduction or loss of photoperiod sensitivity;adaptation to short-day conditions at lower latitudes involves delayed flowering, which prolongs vegetative growth for maximum yield potential. Due to the influence of numerous major loci and quantitative trait loci(QTLs), soybean has broad adaptability across latitudes. Forward genetic approaches have uncovered the molecular basis for several of these major maturity genes and QTLs. Moreover, the molecular characterization of orthologs of Arabidopsis thaliana flowering genes has enriched our understanding of the photoperiodic flowering pathway in soybean. Building on early insights into the importance of the photoreceptor phytochrome A, several circadian clock components have been integrated into the genetic network controlling flowering in soybean: E1, a repressor of FLOWERING LOCUS T orthologs, plays a central role in this network. | 16 | Hijacking antibody-induced CTLA-4 lysosomal degradation for safer and more effective cancer immunotherapy显示文摘It remains unclear why the clinically used anti-CTLA-4 antibodies, popularly called checkpoint inhibitors, have severe immunotherapy-related adverse effects (irAEs) and yet suboptimal cancer immunotherapeutic effects (CITE). Here we report that while irAE-prone Ipilimumab and TremeIgG1 rapidly direct cell surface CTLA-4 for lysosomal degradation, the non-irAE-prone antibodies we generated, HL12 or HL32, dissociate from CTLA-4 after endocytosis and allow CTLA-4 recycling to cell surface by the LRBA-dependent mechanism. Disrupting CTLA-4 recycling results in robust CTLA-4 downregulation by all anti-CTLA-4 antibodies and confers toxicity to a non-irAE-prone anti-CTLA-4 mAb. Conversely, increasing the pH sensitivity of TremeIgG1 by introducing designed tyrosine-to-histidine mutations prevents antibody-triggered lysosomal CTLA-4 downregulation and dramatically attenuates irAE. Surprisingly, by avoiding CTLA-4 downregulation and due to their increased bioavailability, pH-sensitive anti-CTLA-4 antibodies are more effective in intratumor regulatory T-cell depletion and rejection of large established tumors. | 19 | 应用最大似然率及随机效应模型探讨非小细胞肺癌放化疗中急性放射性食管炎与照射剂量和疗程的关系显示文摘背景与目的:局部晚期非小细胞肺癌的治疗疗效差,增加治疗强度如同期放化疗是提高疗效的方法,但毒性反应较大。本研究探讨非小细胞肺癌同期放化疗的治疗方式中,应用最大似然率及随机效应模型探讨非小细胞肺癌放化疗中急性放射性食管炎的发生率与照射剂量和疗程时间因素的关系。方法:本研究中的39例病例资料来自于1998年3月-2000年11月间在M.D.Anderson癌症治疗中心进入随机临床Ⅲ期的同期放化疗治疗研究不能手术的Ⅱ期和Ⅲ期非小细胞肺癌患者。所有的患者在放疗前均接受顺铂和口服依托泊苷(VP-16)的化疗。放疗方案为每次1.2Gy,bid,前后野对照治疗至肿瘤剂量达40~50Gy后,采用避开脊髓的斜野放疗至总剂量69.6Gy。病例分布为amifostine治疗组,19例;非amifostine治疗组,20例。还原三维治疗计划设计并计算了剂量-体积直方图(DVHs)。治疗开始后每周对急性放射性食管炎程度评估1次,在放疗结束后1个月再随访评估1次。评分的标准是根据美国放射治疗研究组(RTOG)的急性反应的标准。采用了多因素的随机效应模型及最大似然率的分析以研究在这39例病例中急性放射性食管炎的严重程度与照射剂量和疗程时间因素的关系,并考虑了不同病例个体间的敏感性差异。结果:在amifostine治疗组有11%(2/19),非amifostine治疗组有30%(6/20)的患者发生Ⅲ度的急性食管炎。在这两组中,急性食管炎的发生率在开始治疗后第4周(累计剂量为48Gy)达高峰并呈一稳定状态。随治疗后期剂量的进一步增加,急性食管炎发生的危险性并不明显增加。原因可能是食管粘膜上皮组织的修复和在治疗后的上皮细胞加速再增殖。疗程中的周剂量或累计周剂量要比整个疗程的总剂量更好地预测急性食管炎的发生。结论:肿瘤累积剂量和患者的内在敏感性与Ⅲ度急性放射性食管炎的发生率有关。欲将发生急性食管炎的危险性降低至10%以下,需限制第4周的累计剂量在36Gy以下,或9Gy/周。这可能为开展IMRT等新技术的治疗提供有用的参考信息。 | 章真 Helen Liu Zhongxing Liao Ritsuko Komaki James D Cox | 2006 | 中国癌症杂志2006,16,12: | 5 |
| 20 | Supplementation of different fat sources affects growth performance and carcass composition of finishing pigs显示文摘Background: There are various fat sources with different energy values and fatty acid compositions that may affect growth performance and carcass composition of grow-finishing pigs. A higher net energy was recently reported in choice white grease compared with soybean oil. Therefore, two experiments were conducted to determine whether practical responses confirm that difference between choice white grease and soybean oil, and to extend the observations to other fat sources.Results: In Exp. 1, pigs fed fats had lower(P < 0.05) average daily feed intake in phase II and overall period,greater(P < 0.05) gain:feed in phase I, phase II, and overall period than pigs fed the control diet. Pigs fed fats tended(P = 0.057) to have thicker backfat depth at the last rib than those fed control. Pigs fed 6% fats had greater(P < 0.01) gain:feed in phase II and overall period than pigs fed 3% fats. During phase I, pigs fed choice white grease grew faster(P < 0.05) than pigs fed soybean oil. In Exp. 2, pigs fed dietary fats(soybean oil, choice white grease, animal-vegetable blend, palm oil, or tallow) had greater(P < 0.01) gain:feed in each phase and overall period, greater(P < 0.01) average daily gain in phase I, but lower(P < 0.01) average daily feed intake in phase II an overall than pigs fed the control diets. The choice white grease also increased(P < 0.05) average daily gain during phase I compared with soybean oil. Pigs fed palm oil had thicker(P < 0.05) backfat depth at the 10 thrib than those fed soybean oil, animal-vegetable blend, or tallow.Conclusions: Inclusion of 6% dietary fat improved feed efficiency of finishing pigs, while different fats produced different practical results that may be consistent with their different energy values.
