1Microfluidic Electrospray Niacin Metal-Organic Frameworks Encapsulated Microcapsules for Wound Healing显示文摘Niacin metal-organic frameworks(MOFs)encapsulated microcapsules with alginate shells and copper-/zinc-niacin framework cores were in situ synthesized by using a microfuidic electrospray approach for wound healing.As the alginate shells were bacteriaresponsively degradable,the niacin MOFs encapsulated microcapsules could intelligently,controllably,and programmably release calcium,copper,and zinc ions,depending on the degree of infections.Te released ions could not only kill microbes by destroying their membrane and inducing the outfow of nutrient substance,but also activate copper/zinc superoxide dismutase(Cu/ZnSOD)to eliminate oxygen free radicals and rescue the cells from oxidative stress injury.Furthermore,the simultaneously released niacin could promote hemangiectasis and absorption of functional metal ions.Tus,the niacin MOFs encapsulated microcapsules were imparted with outstanding antibacterial,antioxidant,and angiogenesis properties.Based on an in vivo study,we have also demonstrated that the chronic wound healing process of an infected full-thickness skin defect model could be signifcantly enhanced by using the niacin MOFs encapsulated microcapsules as therapeutic agent.Terefore,the microfuidic electrospray niacin MOFs encapsulated microcapsules are potential for clinical applications.Guopu Chen Yunru Yu Xiuwen Wu Gefei Wang Guosheng Gu Feng Wang Jianan Ren Huidan Zhang Yuanjin Zhao 2019Research2019,,1:21

3Cultivar-dependent rhizobacteria community and cadmium accumulation in rice: Effects on cadmium availability in soils and iron-plaque formation显示文摘The association between the rhizospheric microbial community and Cd accumulation in rice is poorly understood.A field trial was conducted to investigate the different rhizobacterial communities of two rice cultivars with high Cd accumulation(HA)and low Cd accumulation(LA)at four growth stages.Results showed that the Cd content in the roots of the HA cultivar was 1.23-27.53 higher than that of the LA cultivar(0.08-10.5μg/plant)at four stages.The LA cultivar had a significantly lower Cd availability in rhizosphere and a higher quantity of iron plaque(IP)on the root surface than the HA cultivar at four stages.This resulted in the reduction of Cd concentration in IPs and Cd translocation from IP-to-root.Microbial analysis indicated that the LA cultivar formed a distinct rhizobacterial community from the HA cultivar and had lessα-diversity.The rhizosphere of the LA cultivar was enriched in specific bacterial taxa(e.g.,Massilia and Bacillus)involved in Cd immobilization by phosphate precipitation and IP formation by iron oxidization.However,the rhizosphere in the HA cultivar assembled abundant sulfur-oxidizing bacteria(e.g.,Sulfuricurvum)and iron reduction bacteria(Geobacter).They promoted Cd mobilization and reduced IP formation via the metal redox process.This study reveals a potential approach in which specific rhizobacteria decrease or increase Cd accumulation in rice on contaminated soil and provides a new perspective for secure rice production.Feng Zhang Di Peng Lu Liu Huidan Jiang Lianyang Bai 2022Journal of Environmental Sciences2022,34,6:2
4Loss-of-function of KMT5B leads to neurodevelopmental disorder and impairs neuronal development and neurogenesis显示文摘Autism spectrum disorder(ASD)is a group of neurodevelopmental disorders that cause severe social,communication,and behavioral problems.Recent studies show that the variants of a histone methyltransferase gene KMT5B cause neurodevelopmental disorders(NDDs),including ASD,and the knockout of Kmt5b in mice is embryonic lethal.However,the detailed genotype-phenotype correlations and functional effects of KMT5B in neurodevelopment are unclear.By targeted sequencing of a large Chinese ASD cohort,analyzing published genome-wide sequencing data,and mining literature,we curated 39 KMT5B variants identified from NDD individuals.A genotype-phenotype correlation analysis for 10 individuals with KMT5B pathogenic variants reveals common symptoms,including ASD,intellectual disability,languages problem,and macrocephaly.In vitro knockdown of the expression of Kmt5b in cultured mouse primary cortical neurons leads to a decrease in neuronal dendritic complexity and an increase in dendritic spine density,which can be rescued by expression of human KMT5B but not that of pathogenic de novo missense mutants.In vivo knockdown of the Kmt5b expression in the mouse embryonic cerebral cortex by in utero electroporation results in decreased proliferation and accelerated migration of neural progenitor cells.Our findings reveal essential roles of histone methyltransferase KMT5B in neuronal development,prenatal neurogenesis,and neuronal migration.Guodong Chen Lin Han Senwei Tan Xiangbin Jia Huidan Wu Yingting Quan Qiumeng Zhang Bin Yu Zhengmao Hu Kun Xia Hui Guo 2022Journal of Genetics and Genomics2022,49,9:1

7A cell marker-based clustering strategy(cmCluster)for precise cell type identification of scRNA-seq data显示文摘Background:The precise and efficient analysis of single-cell transcriptome data provides powerful support for studying the diversity of cell functions at the single-cell level.The most important and challenging steps are cell clustering and recognition of cell populations.While the precision of clustering and annotation are considered separately in most current studies,it is worth attempting to develop an extensive and flexible strategy to balance clustering accuracy and biological explanation comprehensively.Methods:The cell marker-based clustering strategy(cmCluster),which is a modified Louvain clustering method,aims to search the optimal clusters through genetic algorithm(GA)and grid search based on the cell type annotation results.Results:By applying cmCluster on a set of single-cell transcriptome data,the results showed that it was beneficial for the recognition of cell populations and explanation of biological function even on the occasion of incomplete cell type information or multiple data resources.In addition,cmCluster also produced clear boundaries and appropriate subtypes with potential marker genes.The relevant code is available in GitHub website(huangyuwei301/cmCluster).Conclusions:We speculate that cmCluster provides researchers effective screening strategies to improve the accuracy of subsequent biological analysis,reduce artificial bias,and facilitate the comparison and analysis of multiple studies.Yuwei Huang Huidan Chang Xiaoyi Chen Jiayue Meng Mengyao Han Tao Huang Liyun Yuan Guoqing Zhang 2023Quantitative Biology2023,11,2:0
