2An in vivo study with an MRI tracer method reveals the biophysical properties of interstitial fluid in the rat brain显示文摘The nature of brain interstitial fluid (ISF) has long been a subject of controversy. Most of the previous studies on brain ISF were carried out in vitro. In the present study, a novel method was developed to characterize ISF in the living rat brain by magnetic resonance (MR) imaging using gadolinium-diethylenetriaminepentaacetic acid (Gd-DTPA) as a tracer. Sprague Dawley rats (n=8) were subjected to MR scanning before and after the introduction of Gd-DTPA into the caudate nucleus. A one-way drainage of brain ISF was demonstrated on the dynamic MR images. According to the traditional diffusion model, the diffusion and clearance rate constants of the tracer within brain extracellular space (ECS) were derived as (3.38±1.07)×10-4 mm 2 s-1 and (7.60±4.18)×10-5 s-1 . Both diffusion and bulk flow contributed to the drainage of ISF from the caudate nucleus, which demonstrated an ISF-cerebrospinal fluid confluence in the subarachnoid space at the lateral and ventral surface of the brain cortex at 3 h after the injection. 3Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. 5A novel nonviral nanoparticle gene vector: Poly-L-lysine- silica nanoparticles显示文摘DNA delivery is a core technology for gene structure and function research as well as clinical settings. The ability to safely and efficiently targeted transfer foreign DNA into cells is a fundamental goal in biotechnology. With the development of nanobiotechnology, nanoparticle gene vectors brought about new hope to reach the goal. In our research, silica nanoparticles (SiNP) were synthesized first in a microemulsion system polyoxyethylene nonylphenyl ether (OP-10)/cyclohexane/ammonium hydroxide, at the same time the effects of SiNP size and its distribution were elucidated by orthogonal analysis; then poly-L-lysine (PLL) was linked on the surface of SiNP by nanoparticle surface energy and electrostatically binding; lastly a novel complex nanomate-rial-poly-L-lysine-silica nanoparticles (PLL-SiNP) wasprepared. The analysis of plasmid DNA binding and DNase I enzymatic degradation discovered that PLL-SiNP could bind DNA, and protect it against enzymatic degradation. 10Microglia modulation with 1070-nm light attenuates Aβ burden and cognitive impairment in Alzheimer’s disease mouse model显示文摘Photobiomodulation,by utilizing low-power light in the visible and near-infrared spectra to trigger biological responses in cells and tissues,has been considered as a possible therapeutic strategy for Alzheimer’s disease(AD),while its specific mechanisms have remained elusive.Here,we demonstrate that cognitive and memory impairment in an AD mouse model can be ameliorated by 1070-nm light via reducing cerebralβ-amyloid(Aβ)burden,the hallmark of AD.The glial cells,including microglia and astrocytes,play important roles in Aβclearance.Our results show that 1070-nm light pulsed at 10 Hz triggers microglia rather than astrocyte responses in AD mice.The 1070-nm lightinduced microglia responses with alteration in morphology and increased colocalization with Aβare sufficient to reduce Aβload in AD mice.Moreover,1070-nm light pulsed at 10 Hz can reduce perivascular microglia and promote angiogenesis to further enhance Aβclearance.Our study confirms the important roles of microglia and cerebral vessels in the use of 1070-nm light for the treatment of AD mice and provides a framework for developing a novel therapeutic approach for AD.Lechan Tao Qi Liu Fuli Zhang Yuting Fu Xi Zhu Xiaofu Weng Hongbin Han Yong Huang Yuanzhen Suo Liang Chen Xiaoling Gao Xunbin Wei 2021Light(Science & Applications)2021,10,10:4

12Tribological Performance of TiB_2-Ni Composite Coating Deposited by APS显示文摘TiB2-Ni composite powders were prepared by pressurized hydrogen reduction cladding with different proportion of TiB2.The coatings were then prepared by APS.The microstructures and the phase composition of the powders as well as the deposited coatings were analyzed by scanning electron microscopy(SEM)and X-ray diffraction(XRD).The coatings were tested using a ring-on-disc tribometer from ambient temperature to 300°C.A sprayed Cr3C2-NiCr coating was tested as a reference.The morphologies of the worn surface of the coatings were observed and analyzed.It is found that the TiB2-Ni composite coatings present higher friction coefficient than that of the Cr3C2-NiCr coating at room temperature due to the adhesive wear mechanism,whereas the main wear mechanism of Cr3C2-NiCr coating is rupture and exfoliation.However,at high temperature,the friction coefficient of the TiB2-Ni composite coating decreases as a result of B2O3 solid lubricant,which alleviates the adhesive wear on coatings.Furthermore,the TiB2-Ni composite coating greatly reduces the mass loss of the boron cast iron.Zhu Hongbin,Li Hui,Li Zhuoxin Beijing University of Technology,Beijing 100124,China 2012稀有金属材料与工程2012,41,S1:2
13Discrete twinning dynamics and size-dependent dislocation-to twin transition in body-centred cubic tungsten显示文摘Body-centred cubic(BCC) metals are known to have unstable intrinsic stacking faults and high resistance to deformation twinning, which can strongly influence their twinning behaviour. Though twinning mechanisms of BCC metals have been investigated for more than 60 years, the atomistic level dynamics of twinning remains under debate, especially regarding its impact on competition between twinning and slip. Here, we investigate the atomistic level dynamics of twinning in BCC tungsten(W) nanowires using in situ nanomechanical testing. Quantitative experimental studies directly visualize that deformation twins in W nanowires have a minimum size of six-layers and grow in increments of approximately three-layers at a time, in contrast to the layer-by-layer growth of deformation twins in face-centred cubic metals. These unique twinning dynamics induces a strong competition with ordinary dislocation slip,as exhibited by a size-dependent dislocation-to-twin transition in W nanowires, with a transition size of ~40 nm. 15Cyclical helping hands: seasonal tailwinds differentially affect migrating Oriental Storks (Ciconia boyciana) travel speed显示文摘Background:The Oriental Stork(Ciconia boyciana)breeds in southeastern Siberia and parts of northeast China,and winters mainly in southeast China.Although the autumn migration pattern of Oriental Storks has been previ-ously described,differences between spring and autumn migration travel speed in relation to wind assistance were unknown.Methods:Using GPS/GSM transmitters,we tracked the full migrations of 18 Oriental Storks during 2015-2018 to compare differences in autumn and spring migration patterns,and combined the satellite telemetry data with the National Center for Environmental Prediction Reanalysis data to explain the relationship between 850 mbar wind vec-tors and seasonal differences in travel speed.Results:Differences in tailwinds contributed to significant differences in daily average Oriental Storks travel speed in spring(258.11±64.8 km/day)compared to autumn(172.23±49.7 km/day,p<0.001).Storks stopped significantly more often in autumn than spring(1.78±1.1 versus 1.06±0.9,p<0.05),but stopover duration(15.52±12.4 versus 16.30±15.1 days,respectively,p=0.3)did not differ significantly.Tailwinds at 850 mbar pressure level(extracted from the National Center of Environmental Prediction Reanalysis data archive)significantly affected daily flying speed dur-ing spring and autumn migration.Tailwind conditions in spring(mean 4.40±5.6 m/s)were always more favourable than in autumn when they received no net benefit(0.48±5.6 m/s,p<0.001).Despite mean spring migration dura-tion being less than autumn(27.52±15.9 versus 32.77±13.4 days,p=0.17),large individual variation meant that this duration did not differ significantly from each other.Conclusions:For long distance migratory soaring birds(such as storks),relative duration of spring and autumn migration likely relates to the interaction between imperative for earliest arrival to breeding grounds and seasonal meteorological conditions experienced en route.Shujuan Fan Qingshan Zhao Hongbin Li Baoguang Zhu Shubin Dong Yanbo Xie Lei Cao Anthony David Fox 2020Avian Research2020,11,2:2
16Isolation and Identification of Photobacterium damselae subsp.damselae(PDD) from Tongue Sole显示文摘In May 2016,an epizootic occured among cultured tongue soles caused mass deaths in a fish farm in Qinhuangdao,China.In order to find out the etiological agent,a bacterial strain was isolated from ascites and other tissues of sick tongue sole aseptically collected.The isolate was identified as Photobacterium damselae subsp.damsela(PDD) by isolation culture,Gram staining,physiological identification,morohological observation,biochemical identification and 16S rDNA sequence analysis.The results showed that the isolate shared 99.6% homology with the reference strain in GenBank.The animal regression test displayed that the isolate had very strong pathogenicity to tongue sole.The LD_(50) was 3.1 × 10~4 CFU/mL,and it showed pathogenicity to mammals.The antimicrobial susceptibility test showed the isolate was highly sensitive to nrofloxacin,Norfloxacin,Ciprofloxacin,Mequindox;moderately sensitive to Cefradine,Doxycycline;and insensitive to Gentamicin,Ceftriaxone,Tilmicosin,etc..Yang Nan Zhang Zhiqiang Wu Tonglei Zhu Jianxin Fu Yanfang Han Hongsheng Wang Hongbin Shi Qiumei Gao Guisheng 2018Animal Husbandry and Feed Science2018,10,2:2
17Evaluation of SARS-CoV-2-Neutralizing Nanobody Using Virus Receptor Binding Domain-Administered Model Mice显示文摘Due to the rapid spread of coronavirus disease 2019(COVID-19),there is an urgent requirement for the development of additional diagnostic tools for further analysis of the disease.The isolated nanobody Nb11-59 binds to the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)reeptor-binding domain[RBD]with high afinity to neutralize the virus and block the angiotensin-converting enzyme 2-(ACE2-)RBD interaction.Here,we introduce a novel nanobody-based radiotracer named^(68)Ga-Nb1159.The radiotracer retained high affinity for the RBD and showed reliable radiochemical characteristics both in vitro and in vivo.Predinical positron emission tomography(PET)studies of Ga-Nb1159 in mice revealed is rapid dearance from circulation and robust uptake into the renal and urinary systems.Fortunately,^(68)Ga-Nb1159 could speifcally reveal the distribution of the RBD in mice.This sudy also helped to evaluate the pharmacodynamic efects of the neutralizing nanobody.Moreover,^(68)Ga-Nb1159 may be a promising l0ol lo explore the distribution of the RBD and improve the understanding of the virus.In particular,this study identified a novel molecular radioagent and established a reliable evaluation method for speifially investigating the RBD through noninvasive and visual PET technology.Song Liu Guanghui Li Lei Ding Jin Ding Qian Zhang Dan Li Xingguo Hou Xiangxing Kong Jing Zou Shiming Zhang Hongbin Han Yakun Wan Zhi Yang Hua Zhu 2022Research2022,,2:2
18In Situ Analysis for Herbal Pieces of Aconitum Plants by Using Direct Analysis in Real Time Mass Spectrometry显示文摘In this study,an extend application was developed to in situ analyze the herbal pieces of Aconitum plants by Direct Analysis in Real Time Mass Spectrometry(DART-MS).Nearly all aconitine-type alkaloids can be desorbed and ionized in this method,including diester diterpenoid aconitines(DDAs),monoester diterpenoid aconitines(MDAs)and some other diterpenoid aconitines.The spectra of in situ analysis for the herbal pieces of aconitum plants are similar with that of their extracts.Radix Aconiti and Radix Aconiti Kusnezoffii can be distinguished from each other by the intensity differences of character fragment ions from MDAs,such as m/z 586,572,and 556.The qualified and unqualified herbal pieces can be also identified by the relative abundances of DDAs.The RSD of the relative abundances of some character ions at m/z 556,586,and 590 were 13.53%,4.03%,and 12.03%,respectively.So this in situ analytical method can identify both the types of Aconitum preparata and their quality.It provides the following advantages in the analysis of Chinese herbs:fast detection without much pretreatment,high-throughput analysis,and reduction of pollution without any organic solvent.Feng Zhou Hongbin Zhu Shu Liu Kang Ma Fengrui Song Zhiqiang Liu 2015Chinese Journal of Chemistry2015,33,2:1
