| 1 | Endostatin inhibits hypertrophic scarring in a rabbit ear model显示文摘Objective:The present study was designed to use an in vivo rabbit ear scar model to investigate the efficacy of systemic administration of endostatin in inhibiting scar formation.Methods:Eight male New Zealand white rabbits were randomly assigned to two groups.Scar model was established by making six full skin defect wounds in each ear.For the intervention group,intraperitoneal injection of endostatin was performed each day after the wound healed(about 15 d post wounding).For the control group,equal volume of saline was injected.Thickness of scars in each group was measured by sliding caliper and the scar microcirculatory perfusion was assessed by laser Doppler flowmetry on Days 15,21,28,and 35 post wounding.Rabbits were euthanatized and their scars were harvested for histological and proteomic analyses on Day 35 post wounding.Results:Macroscopically,scars of the control group were thicker than those of the intervention group.Significant differences between the two groups were observed on Days 21 and 35(p<0.05).Scar thickness,measured by scar elevation index(SEI) at Day 35 post wounding,was significantly reduced in the intervention group(1.09±0.19) compared with the controls(1.36±0.28).Microvessel density(MVD) observed in the intervention group(1.73±0.94) was significantly lower than that of the control group(5.63±1.78) on Day 35.The distribution of collagen fibers in scars treated with endostatin was relatively regular,while collagen fibers in untreated controls were thicker and showed disordered alignment.Western blot analysis showed that the expressions of type I collagen and Bcl-2 were depressed by injection of endostatin.Conclusions:Our results from the rabbit ear hypertrophic scar model indicate that systemic application of endostatin could inhibit local hypertrophic scar formation,possibly through reducing scar vascularization and angiogenesis.Our results indicated that endostatin may promote the apoptosis of endothelial cells and block their release of platelet-derived growth factor(PDGF) and fibroblast growth factor(FGF),thereby controlling collagen production by fibroblasts.Blood vessel-targeted treatment may be a promising strategy for scar therapy. | Hai-tao REN Hang HU Yuan LI Hong-fei JIANG Xin-lei HU Chun-mao HAN | 2013 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2013,14,3: | 17 |
| 2 | GPER agonist G1 suppresses neuronal apoptosis mediated by endoplasmic reticulum stress after cerebral ischemia/reperfusion injury显示文摘Studies have confirmed a strong association between activation of the endoplasmic reticulum stress pathway and cerebral ischemia/reperfusion(I/R) injury.In this study,three key proteins in the endoplasmic reticulum stress pathway(glucose-regulated protein 78,caspase-12,and C/EBP homologous protein) were selected to examine the potential mechanism of endoplasmic reticulum stress in the neuroprotective effect of G protein-coupled estrogen receptor.Female Sprague-Dawley rats received ovariectomy(OVX),and then cerebral I/R rat models(OVX+ I/R) were established by middle cerebral artery occlusion.Immediately after I/R,rat models were injected with 100 μg/kg E2(OVX + I/R +E2),or 100 μg/kg G protein-coupled estrogen receptor agonist G1(OVX + I/R + G1) in the lateral ventricle.Longa scoring was used to detect neurobehavioral changes in each group.Infarct volumes were measured by 2,3,5-triphenyltetrazolium chloride staining.Morphological changes in neurons were observed by Nissl staining.Terminal dexynucleotidyl transferase-mediated nick end-labeling staining revealed that compared with the OVX + I/R group,neurological function was remarkably improved,infarct volume was reduced,number of normal Nissl bodies was dramatically increased,and number of apoptotic neurons in the hippocampus was decreased after E2 and G1 intervention.To detect the expression and distribution of endoplasmic reticulum stress-related proteins in the endoplasmic reticulum,caspase-12 distribution and expression were detected by immunofluorescence,and mRNA and protein levels of glucose-regulated protein 78,caspase-12,and C/EBP homologous protein were determined by polymerase chain reaction and western blot assay.The results showed that compared with the OVX+ I/R group,E2 and G1 treatment obviously decreased mRNA and protein expression levels of glucose-regulated protein 78,C/EBP homologous protein,and caspase-12.However,the G protein-coupled estrogen receptor antagonist G15(OVX + I/R + E2 + G15) could eliminate the effect of E2 on cerebral I/R injury.These results confirm that E2 and G protein-coupled estrogen receptor can inhibit the expression of endoplasmic reticulum stress-related proteins and neuronal apoptosis in the hippocampus,thereby improving dysfunction caused by cerebral I/R injury.Every experimental protocol was approved by the Institutional Ethics Review Board at the First Affiliated Hospital of Shihezi University School of Medicine,China(approval No.SHZ A2017-171) on February 27,2017. | Zi-Wei Han Yue-Chen Chang Ying Zhou Hang Zhang Long Chen Yang Zhang Jun-Qiang Si Li Li | 2019 | Neural Regeneration Research2019,14,7: | 16 |

| 4 | Allyl isothiocyanate ameliorates lipid accumulation and inflammation in nonalcoholic fatty liver disease via the Sirt1/AMPK and NF-κB signaling pathways显示文摘BACKGROUND Allyl isothiocyanate(AITC),a classic anti-inflammatory and antitumorigenic agent,was recently identified as a potential treatment for obesity and insulin resistance.However,little is known about its direct impact on the liver.AIM To investigate the effect and underlying mechanism of AITC in nonalcoholic fatty liver disease(commonly referred to as NAFLD).METHODS To establish a mouse and cellular model of NAFLD,C57BL/6 mice were fed a high fat diet(HFD)for 8 wk,and AML-12 cells were treated with 200μM palmitate acid for 24 h.For AITC treatment,mice were administered AITC(100 mg/kg/d)orally and AML-12 cells were treated with AITC(20μmol/L).RESULTS AITC significantly ameliorated HFD-induced weight gain,hepatic lipid accumulation and inflammation in vivo.Furthermore,serum alanine aminotransferase and aspartate aminotransferase levels were markedly reduced in AITC-treated mice.Mechanistically,AITC significantly downregulated the protein levels of sterol regulatory elementbinding protein 1(SREBP1)and its lipogenesis target genes and upregulated the levels of proteins involved in fatty acidβ-oxidation,as well as the upstream mediators Sirtuin 1(Sirt1)and AMPactivated protein kinaseα(AMPKα),in the livers of HFD-fed mice.AITC also attenuated the nuclear factor kappa B(NF-κB)signaling pathway.Consistently,AITC relieved palmitate acid-induced lipid accumulation and inflammation in AML-12 cells in vitro through the Sirt1/AMPK and NF-κB signaling pathways.Importantly,further studies showed that the curative effect of AITC on lipid accumulation was abolished by siRNA-mediated knockdown of either Sirt1 or AMPKαin AML-12 cells.CONCLUSION AITC significantly ameliorates hepatic steatosis and inflammation by activating the Sirt1/AMPK pathway and inhibiting the NF-κB pathway.Therefore,AITC is a potential therapeutic agent for NAFLD. | Chun-Xiao Li Jian-Guo Gao Xing-Yong Wan Yi Chen Cheng-Fu Xu Ze-Min Feng Hang Zeng Yi-Ming Lin Han Ma Ping Xu Chao-Hui Yu You-Ming Li | 2019 | World Journal of Gastroenterology2019,25,34: | 14 |

| 6 | Morinda officinalis oligosaccharides increase serotonin in the brain and ameliorate depression via promoting 5-hydroxytryptophan production in the gut microbiota显示文摘Morinda officinalis oligosaccharides(MOO) are an oral drug approved in China for the treatment of depression in China. However, MOO is hardly absorbed so that their anti-depressant mechanism has not been elucidated. Here, we show that oral MOO acted on tryptophan → 5-hydroxytryptophan(5-HTP) → serotonin(5-HT) metabolic pathway in the gut microbiota. MOO could increase tryptophan hydroxylase levels in the gut microbiota which accelerated 5-HTP production from tryptophan;meanwhile, MOO inhibited 5-hydroxytryptophan decarboxylase activity, thus reduced 5-HT generation,and accumulated 5-HTP. The raised 5-HTP from the gut microbiota was absorbed to the blood, and then passed across the blood-brain barrier to improve 5-HT levels in the brain. Additionally, pentasaccharide,as one of the main components in MOO, exerted the significant anti-depressant effect through a mechanism identical to that of MOO. | 7 | Assessing the Ecological Quality of Nanjing during Its Urbanization Process by Using Satellite,Meteorological,and Socioeconomic Data显示文摘Assessing the ecological status of different districts within a city undergoing urbanization is challenging given their complex surface types and fast pace of development.In this study,we utilized satellite data obtained from Landsat5/TM(Thematic Mapper)and Landsat 8/OLI(Operational Land Imager)images in conjunction with meteorological and socioeconomic data to construct a remote sensing ecological index(RSEI)for monitoring the ecological quality of Nanjing,Jiangsu Province.A higher RSEI value corresponded to better ecological quality.Five ratings were associated with RSEI values of city districts:very poor,poor,average,good,and excellent.In Nanjing,the percentage of areas evidencing good RSEI ratings decreased from 55.9%in 2000 to 48.0%in 2018,whereas there was a slight increase in areas with very poor RSEI ratings during this period.Of the 11 city districts,16.8%,21.8%,and 61.4%respectively evidenced the increasing,decreasing,and stable ecological quality relative to their quality in 2000.Of the11 administrative districts in Nanjing,the main urban districts evidenced increased RSEI values in 2018 compared with those in 2000,with the improved areas exceeding the ones that had deteriorated in these districts.However,the ecological quality of new urban and ed because of the urban expansion,with areas that had deteriorated exceeding the improved ones.Of the three protected ecological zones,the quality of Zijin Mountain National Forest Park was considerably better than that of Laoshan and Jiangxinzhou.Overall,the urbanization rate and RSEI evidenced a high negative correlation coefficient value(-0.76).The urbanization process of Nanjing induced a declining trend for the ecological quality,indicating the need of strong protection measures for the maintenance or improvement of its ecological environment. | Xin HANG Yachun LI Xiaochun LUO Min XU Xiuzhen HAN | 2020 | Journal of Meteorological Research2020,34,2: | 10 |
| 8 | Comorbidity Burden of Dementia:A Hospital-Based Retrospective Study from 2003 to 2012 in Seven Cities in China显示文摘Dementia is increasing dramatically and imposes a huge burden on society. To date, there is a lack of data on the health status of patients with dementia in China. In an attempt to investigate the comorbidity burden of dementia patients in China at the national level, we enrolled 2,938 patients with Alzheimer's disease(AD), vascular dementia(Va D), or other types of dementia, who were admitted to tertiary hospitals in seven regions of China from January2003 to December 2012. The Charlson Comorbidity Index(CCI) was used to evaluate the comorbidity burden of the patients with dementia. Among these patients, 53.4% had AD, 26.3% had Va D, and 20.3% had other types of dementia. The CCI was 3.0 ± 1.9 for all patients,3.4 ± 1.8 for those with Va D, and 3.0 ± 2.1 for those with AD. The CCI increased with age in all patients, andthe length of hospital stay and daily expenses rose with age and CCI. Males had a higher CCI and a longer stay than females. Moreover, patients admitted in the last 5 years of the study had a higher CCI than those admitted in the first 5 years. We found that the comorbidity burden of patients with dementia is heavy. | 9 | A novel anticancer property of Lycium barbarum polysaccharide in triggering ferroptosis of breast cancer cells显示文摘Breast cancer is one of the most malignant tumors and is associated with high mortality rates among women.Lycium barbarum polysaccharide(LBP)is an extract from the fruits of the traditional Chinese herb,L.barbarum.LBP is a promising anticancer drug,due to its high activity and low toxicity.Although it has anticancer properties,its mechanisms of action have not been fully established.Ferroptosis,which is a novel anticancer strategy,is a cell death mechanism that relies on iron-dependent lipid reactive oxygen species(ROS)accumulation.In this study,human breast cancer cells(Michigan Cancer Foundation-7(MCF-7)and MD Anderson-Metastatic Breast-231(MDA-MB-231))were treated with LBP.LBP inhibited their viability and proliferation in association with high levels of ferroptosis.Therefore,we aimed to ascertain whether LBP reduced cell viability through ferroptosis.We found that the structure and function of mitochondria,lipid peroxidation,and expression of solute carrier family 7 member 11(SLC7 A11,also known as x CT,the light-chain subunit of cystine/glutamate antiporter system X_(c)^(-))and glutathione peroxidase 4(GPX4)were altered by LBP.Moreover,the ferroptosis inhibitor,Ferrostatin-1(Fer-1),rescued LBP-induced ferroptosis-associated events including reduced cell viability and glutathione(GSH)production,accumulation of intracellular free divalent iron ions and malondialdehyde(MDA),and down-regulation of the expression of x CT and GPX4.Erastin(x CT inhibitor)and RSL3(GPX4 inhibitor)inhibited the expression of x CT and GPX4,respectively,which was lower after the co-treatment of LBP with Erastin and RSL3.These results suggest that LBP effectively prevents breast cancer cell proliferation and promotes ferroptosis via the x CT/GPX4 pathway.Therefore,LBP exhibits novel anticancer properties by triggering ferroptosis,and may be a potential therapeutic option for breast cancer. | Xing DU Jingjing ZHANG Ling LIU Bo XU Hang HAN Wenjie DAI Xiuying PEI Xufeng FU Shaozhang HOU | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,4: | 10 |
| 10 | Origin and Evolution of Core Components Responsible for Monitoring Light Environment Changes during Plant Terrestrialization显示文摘Light serves as the source of energy as well as an information signal for photosynthetic plants. During evolution, plants have acquired the ability to monitor environmental light radiation and adjust their developmental patterns to optimally utilize light energy for photosynthesis. The mechanisms of light perception and signal transduction have been comprehensively studied in past decades, mostly in a few model plants, including Arabidopsis thaliana. However, systematic analyses of the origin and evolution of core components involved in light perception and signaling are still lacking. In this study, we took advantage of the recently sequenced genomes and transcriptomes covering all the main Archaeplastida clades in the public domain to identify orthologous genes of core components involved in light perception and signaling and to reconstruct their evolutionary history. Our analyses suggested that acclimation to different distribution of light quality in new environments led to the origination of specific light signaling pathways in plants. The UVR8 (UV Resistance Locus 8) signaling pathway originated during the movement of plants from the deeper sea to shallow water and enabled plants to deal with ultraviolet B light (UV-B). After acquisition of UV-B adaptation, origination of the phytochrome signaling pathway helped plants to colonize water surface where red light became the prominent light energy source. The seedling emergence pathway, which is mediated by a combination of light and phytohormone signals that orchestrate plant growth pattern transitions, originated before the emergence of seed plants. | 11 | A comprehensive analysis of the response of the fungal community structure to long-term continuous cropping in three typical upland crops显示文摘Certain agricultural management practices are known to affect the soil microbial community structure;however,knowledge of the response of the fungal community structure to the long-term continuous cropping and rotation of soybean,maize and wheat in the same agroecosystem is limited.We assessed the fungal abundance,composition and diversity among soybean rotation,maize rotation and wheat rotation systems and among long-term continuous cropping systems of soybean,maize and wheat as the effect of crop types on fungal community structure.We compared these fungal parameters of same crop between long-term crop rotation and continuous cropping systems as the effect of cropping systems on fungal community structure.The fungal abundance and composition were measured by quantitative real-time PCR and Illumina MiSeq sequencing.The results revealed that long-term continuous soybean cropping increased the soil fungal abundance compared with soybean rotation,and the fungal abundance was decreased in long-term continuous maize cropping compared with maize rotation.The long-term continuous soybean cropping also exhibited increased soil fungal diversity.The variation in the fungal community structure among the three crops was greater than that between long-term continuous cropping and rotation cropping.Mortierella,Guehomyces and Alternaria were the most important contributors to the dissimilarity of the fungal communities between the continuous cropping and rotation cropping of soybean,maize and wheat.There were 11 potential pathogen and 11 potential biocontrol fungi identified,and the relative abundance of most of the potential pathogenic fungi increased during the long-term continuous cropping of all three crops.The relative abundance of most biocontrol fungi increased in long-term continuous soybean cropping but decreased in long-term continuous maize and wheat cropping.Our results indicate that the response of the soil fungal community structure to long-term continuous cropping varies based upon crop types. | LIU Hang PAN Feng-juan HAN Xiao-zeng SONG Feng-bin ZHANG Zhi-ming YAN Jun XU Yan-li | 2020 | Journal of Integrative Agriculture2020,19,3: | 8 |
| 12 | Classification of the Gut Microbiota of Patients in Intensive Care Units During Development of Sepsis and Septic Shock显示文摘The gut microbiota of intensive care unit(ICU)patients displays extreme dysbiosis associated with increased susceptibility to organ failure,sepsis,and septic shock.However,such dysbiosis is difficult to characterize owing to the high dimensional complexity of the gut microbiota.We tested whether the concept of enterotype can be applied to the gut microbiota of ICU patients to describe the dysbiosis.We collected 131 fecal samples from 64 ICU patients diagnosed with sepsis or septic shock and performed 16S rRNA gene sequencing to dissect their gut microbiota compositions.During the development of sepsis or septic shock and during various medical treatments,the ICU patients always exhibited two dysbiotic microbiota patterns,or ICU-enterotypes,which could not be explained by host properties such as age,sex,and body mass index,or external stressors such as infection site and antibiotic use.ICU-enterotype I(ICU E1)comprised predominantly Bacteroides and an unclassified genus of Enterobacteriaceae,while ICU-enterotype II(ICU E2)comprised predominantly Enterococcus.Among more critically ill patients with Acute Physiology and Chronic Health Evaluation II(APACHE II)scores>18,septic shock was more likely to occur with ICU E1(P=0.041).Additionally,ICU E1 was correlated with high serum lactate levels(P=0.007).Therefore,different patterns of dysbiosis were correlated with different clinical outcomes,suggesting that ICU-enterotypes should be diagnosed as independent clinical indices.Thus,the microbial-based human index classifier we propose is precise and effective for timely monitoring of ICU-enterotypes of individual patients.This work is a first step toward precision medicine for septic patients based on their gut microbiota profiles. | Wanglin Liu Mingyue Cheng Jinman Li Peng Zhang Hang Fan Qinghe Hu Maozhen Han Longxiang Su Huaiwu He Yigang Tong Kang Ning Yun Long | 2020 | Genomics, Proteomics & Bioinformatics2020,18,6: | 7 |
| 13 | Berberine treats atherosclerosis via a vitamine-like effect downregulating Choline-TMA-TMAO production pathway in gut microbiota显示文摘Trimethylamine-N-oxide(TMAO)derived from the gut microbiota is an atherogenic metabolite.This study investigates whether or not berberine(BBR)could reduce TMAO production in the gut microbiota and treat atherosclerosis.Effects of BBR on TMAO production in the gut microbiota,as well as on plaque development in atherosclerosis were investigated in the culture of animal intestinal bacterial,HFD-fed animals and atherosclerotic patients,respectively.We found that oral BBR in animals lowers TMAO biosynthesis in intestine through interacting with the enzyme/co-enzyme of choline-trimethylamine lyase(CutC)and flavincontaining monooxygenase(FMO)in the gut microbiota.This action was performed by BBR’s metabolite dihydroberberine(a reductive BBR by nitroreductase in the gut microbiota),via a vitamine-like effect down-regulating Choline-TMA-TMAO production pathway.Oral BBR decreased TMAO production in animal intestine,lowered blood TMAO and interrupted plaque formation in blood vessels in the HFD-fed hamsters.Moreover,21 patients with atherosclerosis exhibited the average decrease of plaque score by 3.2%after oral BBR(0.5 g,bid)for 4 months(^(*)P<0.05,n=21);whereas the plaque score in patients treated with rosuvastatin plus aspirin,or clopidogrel sulfate or ticagrelor(4 months,n=12)increased by 1.9%.TMA and TMAO in patients decreased by 38 and 29%in faeces(^(*)P<0.05;^(*)P<0.05),and 37 and 35%in plasma(^(***)P<0.001;^(*)P<0.05),after 4 months on BBR.BBR might treat atherosclerotic plaque at least partially through decreasing TMAO in a mode of action similar to that of vitamins. | Shu-Rong Ma Qian Tong Yuan Lin Li-Bin Pan Jie Fu Ran Peng Xian-Feng Zhang Zhen-Xiong Zhao Yang Li Jin-Bo Yu Lin Cong Pei Han Zheng-Wei Zhang Hang Yu Yan Wang Jian-Dong Jiang | 2022 | Signal Transduction and Targeted Therapy2022,7,8: | 6 |
| 14 | Elevated skin autofluorescence is strongly associated with foot ulcers in patients with diabetes: a cross-sectional, observational study of Chinese subjects显示文摘Objective: This study was designed to evaluate the association between skin autofluorescence (AF), an indicator of advanced glycation end-products (AGEs), and foot ulcers in subjects with diabetes. Methods: In this study, 195 Chinese diabetic subjects were examined. Their feet were examined regardless of whether an ulcer was present or not. Skin AF was measured with an AGE reader. Demographic characteristics and blood data were recorded. Results: The mean values of skin AF were 2.29±0.47 for subjects without foot ulcers, and 2.80±0.69 for those with foot ulcers, a significant difference (P<0.05). Skin AF was strongly correlated with age and duration of diabetes. After adjusting for these factors, multivariate logistic regression showed that skin AF was independently associated with foot ulcerations. Conclusions: Skin AF is independently associated with diabetic foot ulcerations. | 16 | Phase 1 studies comparing safety, tolerability, pharmacokinetics and pharmacodynamics of HLX01(a rituximab biosimilar) to reference rituximab in Chinese patients with CD20-positive B-cell lymphoma显示文摘Objective: This study aimed to compare the pharmacokinetic, pharmacodynamic and safety profiles of HLX01(a rituximab biosimilar) and reference rituximab sourced from China(Mab Thera?;rituximab-CN).Methods: Here we report the results of two phase 1 studies. In the phase 1 a, open-label, dose-escalation study(NCT03218072, CTR20140400), eligible patients received 250, 375 and 500 mg/m^(2) HLX01 sequentially at 7-day intervals, after confirming no dose-limiting toxicity(DLT). In the phase 1 b, double-blind study(NCT02584920,CTR20140764), eligible patients were given a single dose of 375 mg/m^(2) HLX01 or rituximab-CN. The primary endpoints included safety and tolerability parameters for the phase 1 a and the area under the plasma concentrationtime curve from time zero to day 91(AUC0-91 d) for the phase 1 b study. Equivalence was concluded if 90%confidence interval(90% CI) for the geometric least squares mean ratio(GLSMR) fell in the pre-specified equivalence criteria(80%-125%).Results: Between June 20, 2014 and January 5, 2015, 12 patients were enrolled in the phase 1 a study. The pharmacokinetics of HLX01 showed dose proportionality and accumulation to steady state. HLX01 was well tolerated, with no serious adverse events(AEs), discontinuations or DLTs. Between November 8, 2014 and August13, 2015, 87 eligible patients were enrolled in the phase 1 b study, including 43 who received HLX01 and 44 who were treated with rituximab-CN. The equivalence endpoint was met with GLSMR for AUC0-91 d being 89.6%(90% CI: 80.4%-99.8%).
