2Selection and characterization of the novel anti-human PD-1 FV78 antibody from a targeted epitope mammalian cell-displayed antibody library显示文摘Currently,display-based methods are well established and widely used in antibody engineering for affinity maturation and structural stability improvement.We obtained a novel anti-human programmed death 1(PD-1)antibody using computer-aided design and a mammalian cell display technology platform.We used computer-aided modeling and distance geometry methods to predict and assign the key residues that contributed to the binding of human PD-L1 to PD-1.Then,we analyzed the sequence of nivolumab(an anti-human PD-1 antibody,referred to as MIL75 in the article)to determine the template for antibody design and library construction.We identified a series of potential substitutions on the obtained template and constructed a virtual epitope-targeted antibody library based on the physicochemical properties and each possible location of the assigned key residues.The virtual antibody libraries were displayed on the surface of mammalian cells as the antigen-binding fragments of full-length immunoglobulin G.Then,we used flow cytometry and sequencing approaches to sort and screen the candidates.Finally,we obtained a novel anti-human PD-1 antibody named FV78.FV78 competitively recognized the PD-1 epitopes that interacted with MIL75 and possessed an affinity comparable to MIL75.Our results implied that FV78 possessed equivalent bioactivity in vitro and in vivo compared with MIL75,which highlighted the probability and prospect of FV78 becoming a new potential antibody therapy.Longlong Luo Shi Wang Xiaoling Lang Tingting Zhou Jing Geng Xinying Li Chunxia Qiao Jiannan Feng Beifen Shen Ming Lv Yan Li 2018Cellular & Molecular Immunology2018,15,2:4

6Remodeling tumor immunosuppressive microenvironment via a novel bioactive nanovaccines potentiates the efficacy of cancer immunotherapy显示文摘The clinical outcomes of cancer nanovaccine have been largely impeded owing to the low antigen-specific T cell response rate and acquired resistance caused by the immunosuppressive tumor microenvironment(TME).Here,we reported a tumor acidity-responsive nanovaccine to remodel the immunosuppressive TME and expand the recruitment of tumor infiltrating lymphocytes(TILs)using hybrid micelles(HM),which encapsulated colony stimulating factor 1 receptor(CSF1-R)inhibitor BLZ-945 and indoleamine 2,3-dioxygenase(IDO)inhibitor NLG-919 in its core and displayed a model antigen ovalbumin(OVA)on its surface(denoted as BN@HM-OVA).The bioactive nanovaccine is coated with a polyethylene glycol(PEG)shell for extending nanoparticle circulation.The shell can be shed in response to the weakly acidic tumor microenvironment.The decrease in size and the increase in positive charge may cause the deep tumor penetration of drugs.We demonstrated that the bioactive nanovaccine dramatically enhance antigen presentation by dendritic cells(DCs)and drugs transportation into M1-like tumor-associated macrophages(TAMs)and tumor cells via size reduction and increasing positive charge caused by the weakly acidic TME.Such bioactive nanovaccine could remodel the immunosuppressive TME into an effector T cells favorable environment,leading to tumor growth inhibition in prophylactic and therapeutic E.G7-OVA tumor models.Furthermore,combining the bioactive nanovaccine with simultaneous anti-PD-1 antibody treatment leads to a long-term tumor inhibition,based on the optimal timing and sequence of PD-1 blockade against T cell receptor.This research provides a new strategy for the development of efficient cancer immunotherapy.Xiaoxue Xie Yi Feng Hanxi Zhang Qingqing Su Ting Song Geng Yang Ningxi Li Xiaodan Wei Tingting Li Xiang Qin Shun Li Chunhui Wu Xiaojuan Zhang Guixue Wang Yiyao Liu Hong Yang 2022Bioactive Materials2022,7,10:1

8Preparation and application of streptavidin magnetic particles显示文摘Two kinds of streptavidin magnetic particles,namely streptavidin GoldMag particles and streptavidin amino terminal particles were prepared by the methods of physical adsorption and covalent interaction respectively.The streptavidin coated on magnetic particle surface,crucial to many applications,was greatly influenced by the choice of the different buffer.Compared with DynalbeadsM-270 streptavidin, the binding capacity for biotin of different streptavidin magnetic particles was determined by enzyme inhibition method,and the coupling capacity and activity of biotinylated oligonucleotide on their sur- face were also analyzed.The results indicated that the streptavidin GoldMag particle prepared by physical adsorption was stable in STE(NaCl-Tris-EDTA)buffer that was frequently used in nucleic acid hybridization and detection.The streptavidin amino terminal particles prepared by covalent interaction could be used both in STE buffer and PBS(phosphate buffered saline)buffer.The biotin binding ca- pacity for 1 mg of streptavidin GoldMag particles and streptavidin amino terminal particles was 4950 and 5115 pmol respectively.The capacity of biotinylated oligonucleotide(24 bp)coupled on 1 mg of GoldMag and amino terminal magnetic particles was 2839 and 2978 pmol separately.These data were about 6-7 times higher than those of DynabeadsM-270 streptavidin.The hybridization results with FITC-labeled complementary probe on magnetic particle surface demonstrated that the oligonucleotide coupled on streptavidin magnetic particles had high biological activity.ZHANG ZhiFeng ZHU HongLi TANG YiTong CUI Ting GENG TingTing CHEN Chao CUI YaLi 2007Science China Chemistry2007,50,1:1
9Genome-wide high-resolution mapping of mitotic DNA synthesis sites and common fragile sites by direct sequencing显示文摘Common fragile sites(CFSs)are genomic loci prone to the formation of breaks or gaps on metaphase chromosomes.They are hotspots for chromosome rearrangements and structural variations,which have been extensively implicated in carcinogenesis,aging,and other pathological processes.Although many CFSs were identified decades ago,a consensus is still lacking for why they are particularly unstable and sensitive to replication perturbations.This is in part due to the lack of high-resolution mapping data for the vast majority of the CFSs,which has hindered mechanistic interrogations.Here,we seek to map human CFSs with high resolution on a genome-wide scale by sequencing the sites of mitotic DNA synthesis(MiDASeq)that are specific for CFSs.We generated a nucleotide-resolution atlas of MiDAS sites(MDSs)that covered most of the known CFSs,and comprehensively analyzed their sequence characteristics and genomic features.Our data on MDSs tallied well with long-standing hypotheses to explain CFS fragility while highlighting the contributions of late replication timing and large transcription units.Notably,the MDSs also encompassed most of the recurrent double-strand break clusters previously identified in mouse neural stem/progenitor cells,thus bridging evolutionarily conserved break points across species.Moreover,MiDAseq provides an important resource that can stimulate future research on CFSs to further unravel the mechanisms and biological relevance underlying these labile genomic regions.Fang Ji Hongwei Liao Sheng Pan Liujian Ouyang Fang Jia Zaiyang Fu Fengjiao Zhang Xinwei Geng Xinming Wang Tingting Li Shuangying Liu Madiha Zahra Syeda Haixia Chen Wen Li Zhihua Chen Huahao Shen Songmin Ying 2020Cell Research2020,30,11:1

11Single-molecule investigation of human telomeric G-quadruplex interactions with Thioflavin T显示文摘G-quadruplex ligands have been accepted as potential therapeutic agents for anticancer treatment.Thioflavin T(ThT), a highly selective G-quadruplex ligand, can bind G-quadruplex with a fluorescent light-up signal change and high specificity against DNA duplex. However, there are still different opinions that ThT induces/stabilizes G-quadruplex foldings/topologies in human telomere sequence. Here, a sensitive single-molecule nanopore technology was utilized to analyze the interactions between human telomeric DNA(Tel DNA) and ThT. Both translocation time and current blockade were measured to investigate the translocation behaviors. Furthermore, the effects of metal ion(K^+ and Na^+) and pH on the translocation behaviors were studied. Based on the single-molecule level analysis, there are some conclusions:(1) In the electrolyte solution containing 50 mmol/L KCI and 450 mmol/L NaCl, ThT can bind strongly with Tel DNA but nearly does not change the G-quadruplex form;(2) In the presence of high concentration K^+,the ThT binding induces the structural change of hybrid G-quadruplex into antiparallel topology with an enhanced structural stability;(3) In either alkaline or acidic buffer, G-quadruplex form will change in certain degree. All above conclusions are helpful to deeply understand the interaction behaviors between Tel DNA and ThT. 12A study on historical location and evolution of Lop Nor in China with maps and DEM显示文摘Lop Sea,located at the east end of the Tarim Basin,Northwest China,dried up permanently,which is the terminal lake of the Tarim River.Lop Sea was considered as the lake basin of Lop Nor since Quaternary.However,the possibility that Lop Nor was away from the Lop Sea in historical time is crucial to be discussed to interpret the proxy records in sediment profiles.To obtain a general view of the evolution of Lop Nor and Lop Sea in a historical period,several approaches were adopted in this paper.First,the Qianlong Thirteen-Row Atlas,an ancient imperial atlas of the Qing Dynasty,which was completed around 1760,indicated that the Tarim River formed a relatively large lake at its modern upstream region.Second,a Digital Elevation Model(DEM)with a 10-m spatial resolution and a relative precision of 0.42 m was derived from TanDEM-X/Terra SAR-X satellite image pairs using the interferometry method,which was verified using ICESat-GLAS laser footprints and a local DEM acquired by a drone.Finally,based on the spatial analysis of historical documents,expedition reports,sediment profiles and archaeological evidence,it can be deduced that the lacustrine deposition was discontinued in the Lop Sea.Six episodes in the evolutionary history of the drainage system in eastern Tarim Basin were summarized.The proved depositional condition variations could be used for future interpretation of proxy records in sediment.The high-accurate DEM provided a reference for the location of further fieldwork in the Lop Sea.The method proposed in this paper may be efficient for the research of inland lakes or rivers in global arid regions.ZHANG Tingting SHAO Yun GENG Yuyang GONG Huaze YANG Lan 2021Journal of Arid Land2021,13,6:0
13The paleoclimatic environment reconstruction of Lop Nur in NW China in UAV spectroscopy显示文摘The change in the ecological environment in the arid core area is a critical issue in the context of global warming.To study the paleoclimate evolution,precise identification of minerals deposited in Asia’s arid hinterland,Lop Nur Salt Lake,NW China was conducted.The hyperspectral data of the salt crust was sampled to identify the species and content of sedimentary minerals,and the multispectral photos were used to reconstruct the salt crust morphology using the unmanned aerial vehicles platform.The SUnSAL(sparse unmixing by variable splitting and augmented Lagrangian)method was employed to inverse the sedimentary mineral components along the shoreline.The heterogeneity of salt and clay minerals in bright and dark ear-shaped strips was evaluated.The paleoclimatic environment associated with salt lake extinction was reconstructed by analyzing paleoclimate records of sediments,spectral reflectance and morphology of the salt crust.Results show that:(1)the variations in the micro-geomorphology of the salt crust are obviously the reason for the formation of bright and dark ear-shaped strips and the differences in the species and relative content of the sedimentary minerals are the microscopic reason.The high ratio of sedimentary salt minerals to clay minerals(RS/C)contributes to the high reflectivity,and the salt crust presents a bright texture.The low RS/C results in the low reflectivity,salt crust presents a dark texture;(2)the bright and dark ear-shaped strips represent warm-arid and cold-humid climates.The shape of the Lop Nur Lake shoreline evolved due to alternating warm-dry and cold-humid paleoclimate changes.Lan YANG Tingting ZHANG Huaze GONG Yuyang GENG Guangjin TIAN 2023Journal of Oceanology and Limnology2023,41,4:0

15Low Concordance Between Blood Pressures Measured in Periodic Health Examinations and in a Workplace-Based Hypertension Management Program显示文摘Poor adherence to standard protocols of blood pressure(BP)measurement in routine clinical practice leads to higher readings than“research-quality”measurements.Whether this phenomenon exists in periodic health examinations was unknown.We aimed to explore the concordance between BP measurements in periodic health examinations and those measured following a standard measurement protocol.We used data from the Kailuan Study,an ongoing longitudinal cohort study in China,of which participants received biennial health examinations in health management centers.In addition,BPs were measured following standard protocols in a workplace-based hypertension management program nested in the Kailuan Study.We compared BP readings of the same person between the two settings using generalized linear mixed-effects models.A total of 3988 men(the mean age was 44.9 years)had at least two BP measurements both in health examinations and management program with a time interval between the two settings that less than 90 days.The mean systolic blood pressures(SBP)and diastolic blood pressures(DBP)in health examinations were 4.2(95%CI 3.9–4.5)mm Hg and 3.3(95%CI 3.1–3.5)mm Hg higher than those in the management program,respectively.Bland–Altman analyses showed the wide agreement inter-vals ranging from-27.7-to 36.5-mm Hg for SBP and-18.3-to 24.7-mm Hg for DBP.In conclusion,BP measurements in periodic health examinations were generally higher than BPs measured following a standard protocol.Our findings highlight the importance of standard BP measurement to avoid overestimation of hypertension prevalence and treatment initiation.Jun-Xiang Chen Yan-Feng Zhou Tingting Geng Simiao Chen Shuohua Chen Guodong Wang Yan-Bo Zhang Yi Wang Zhou-Zheng Tu Gang Liu Shouling Wu An Pan 2022Phenomics2022,2,6:0
16Enhanced genome editing to ameliorate a genetic metabolic liver disease through co-delivery of adeno-associated virus receptor显示文摘Genome editing through adeno-associated viral(AAV) vectors is a promising gene therapy strategy for various diseases,especially genetic disorders. However, homologous recombination(HR) efficiency is extremely low in adult animal models. We assumed that increasing AAV transduction efficiency could increase genome editing activity, especially HR efficiency, for in vivo gene therapy. Firstly, a mouse phenylketonuria(PKU) model carrying a pathogenic R408W mutation in phenylalanine hydroxylase(Pah) was generated. Through co-delivery of the general AAV receptor(AAVR), we found that AAVR could dramatically increase AAV transduction efficiency in vitro and in vivo. Furthermore, co-delivery of SaCas9/sgRNA/donor templates with AAVR via AAV8 vectors increased indel rate over 2-fold and HR rate over 15-fold for the correction of the single mutation in Pah;mice. Moreover, AAVR co-injection successfully increased the site-specific insertion rate of a 1.4 kb Pah cDNA by 11-fold, bringing the HR rate up to 7.3% without detectable global off-target effects. Insertion of Pah cDNA significantly decreased the Phe level and ameliorated PKU symptoms. 17Fasting Blood Glucose but not TMAO is Associated with in-Stent Restenosis in Patients with Acute Coronary Syndrome显示文摘Background:The purpose of our study was to assess whether the occurrence of ISR might be associated with plasma TMAO levels in patients with ACS after DES implantation.Methods:This was a single center retrospective case-control study,in which 64 symptomatic patients with repeated coronary angiography after PCI and 15 patients with ISR were included in the ISR group,and 49 patients without ISR were included in the non-ISR group.High-performance liquid chromatography with tandem mass spectrometry was used to measure plasma TMAO levels.Results:No significant differences were observed in plasma TMAO between the ISR and non-ISR groups.Plasma TMAO levels showed no significant correlation with ISR,but were significantly positively correlated with diabetes mellitus,serum HbA1c levels and serum creatinine levels;moreover,they were significantly negatively correlated with female sex.ISR was significantly positively correlated with diabetes mellitus,fasting blood glucose levels,the neutro-phil to lymphocyte ratio and syntax score;in addition,it was significantly negatively correlated with platelets.Logistic regression analysis indicated that fasting blood glucose was the only independent predictor of ISR.Conclusion:Plasma TMAO may not be associated with ISR and plaque burden in patients with ACS after DES im-plantation,whereas FBG may predict the development of ISR in these patients.Boda Zhou Yajun Xue Jie Zhou Shenjie Sun Tingting Lv Ou Zhang Yu Geng Guobin Miao Ping Zhang 2022Cardiovascular Innovations and Applications2022,,1:0

19The S100 calcium binding protein A11 promotes liver fibrogenesis by targeting TGF-βsignaling显示文摘Liver fibrosis is a key transformation stage and also a reversible pathological process in various types of chronic liver diseases.However,the pathogenesis of liver fibrosis still remains elusive.Here,we report that the calcium binding protein A11(S100A11)is consistently upregulated in the integrated data from GSE liver fibrosis and tree shrew liver proteomics.S100A11 is also experimentally activated in liver fibrosis in mouse,rat,tree shrew,and human with liver fibrosis.While overexpression of S100A11 in vivo and in vitro exacerbates liver fibrosis,the inhibition of S100A11 improves liver fibrosis.Mechanistically,S100A11 activates hepatic stellate cells(HSCs)and the fibrogenesis process via the regulation of the deacetylation of Smad3 in the TGF-βsignaling pathway.S100A11 physically interacts with SIRT6,a deacetylase of Smad2/3,which may competitively inhibit the interaction between SIRT6 and Smad2/3.The subsequent release and activation of Smad2/3 promote the activation of HSCs and fibrogenesis.Additionally,a significant elevation of S100A11 in serum is observed in clinical patients.Our study uncovers S100A11 as a novel profibrogenic factor in liver fibrosis,which may represent both a potential biomarker and a promising therapy target for treating liver fibrosis and fibrosis-related liver diseases.Tingting Zhu Linqiang Zhang Chengbin Li Xiaoqiong Tan Jing Liu Huiqin Li Qijing Fan Zhiguo Zhang Mingfeng Zhan Lin Fu Jinbo Luo Jiawei Geng Yingjie Wu Xiaoju Zou Bin Liang 2022Journal of Genetics and Genomics2022,49,4:0
20Tirapazamine encapsulated hyaluronic acid nanomicelles realized targeted and efficient photo-bioreductive cascading cancer therapy显示文摘Currently,architecting a ratio nal and efficient nanoplatform combing with multi-therapeutic modalities is highly obligatory for advanced cancer treatment.In order to remedy the self-limiting hypoxic dilemma of photodynamic therapy(PDT),herein,a facile photosensitizer(i.e.,chlorin e6,Ce6) and bioreductive prodrug(i.e.,tirapazamine,TPZ)-coloaded hyaluronic acid(HA) nanomicelles(denoted as TPZ@HA-Ce6)was developed for the cascading mode of photo-bioreductive cancer therapy.Taking the typical advantage of Ce6 coupled HA conjugate,TPZ was easily and successfully accommodated into the hydrophobic core of HA-Ce6 nanomicelles,yielding TPZ@HA-Ce6.It showed good dispersibility and stability with the hydrodynamic size of ca.170 nm.It targeted the CD44 overexpressed cancer cells by receptor-mediated endocytosis way and killed them effectively with singlet oxygen and the subsequent TPZ radicals resulting from the oxygen depletion of PDT.The later was further verified by the hypoxia probe in vivo.Using murine mammary carcinoma 4 T1 model,TPZ@HA-Ce6 nanomicelles exhibited cascading and synergistic anticancer effect of PDT and TPZ bioreductive therapy compared with each monotherapy.This work suggests the promising prospect of the hybrid hyaluronic nanomicelles for highly efficient cancer combination treatment.Chunhui Wu Qiuyue Liu Yikun Wang Zhengxin Xie Honglin Huang Ningxi Li Xiaodan Wei Geng Yang Tingting Li Hong Yang Shun Li Xiang Qin Yiyao Liu 2021Chinese Chemical Letters2021,32,8:0
