1Sodium oligomannate therapeutically remodels gut microbiota and suppresses gut bacterial amino acids-shaped neuroinflammation to inhibit Alzheimer's disease progression显示文摘Recently,increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease(AD)progression,yet the role of gut microbiota in AD pathogenesis remains obscure.Herein,we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression.Using AD mouse models,we discovered that,during AD progression,the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine,which stimulates the differentiation and proliferation of pro-inflammatory T helper 1(Thl)cells.The brain-infiltrated peripheral Th1 immune cells are associated with the Ml microglia aaivation,contributing to AD-associated neuroinflammation.Importantly,the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment(MCI)due to AD.Furthermore,GV-971,a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China,suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation,harnesses neuroinflammation and reverses the cognition impairment.Together,our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota.Xinyi Wang Guangqiang Sun Teng Feng Jing Zhang Xun Huang Tao Wang Zuoquan Xie Xingkun Chu Jun Yang Huan Wang Shuaishuai Chang Yanxue Gong Lingfei Ruan Guanqun Zhang Siyuan Yan Wen Lian Chen Du Dabing Yang Qingli Zhang Feifei Lin Jia Liu Haiyan Zhang Changrong Ge Shifu Xiao Jian Ding Meiyu Geng 2019Cell Research2019,29,10:192
2Electro-acupuncture regulates the cholinergic anti-inflammatory pathway in a rat model of chronic obstructive pulmonary disease显示文摘Objective: Acupuncture has a definite therapeutic effect on chronic obstructive pulmonary disease(COPD), and the cholinergic anti-inflammatory pathway(CAP) has been shown to be involved in regulation of inflammation. In this study, we investigated whether electro-acupuncture(EA) affects the CAP in COPD.Methods: Sprague-Dawley rats were induced into COPD through exposure to cigarette smoke combined with lipopolysaccharide. EA treatment was applied at Zusanli(ST36) and Feishu(BL13) points for 30 min/d for 7 d. Seventy-two rats were randomly divided into six study groups, including normal, normal + EA,normal + α-bungarotoxin(a-BGT)(the antagonist of the nicotinic acetylcholine receptor a7 subunit(a7 nAChR)) + EA, COPD, COPD + EA, and COPD + α-BGT + EA. Lung function, pathology and vagus nerve discharge were tested. The levels of acetylcholine(ACh), acetylcholinesterase(AChE), interleukin-6(IL-6) and tumor necrosis factor-a(TNF-a) in bronchoalveolar lavage fluid(BALF) and lung tissue were measured by enzyme-linked immunosorbent assay. The mRNA and protein expression and immunoreactivity of a7 nAChR and its postreceptor inflammation signal pathway, including janus kinase 2(JAK2), signal transducers and activators of transcription 3(STAT3), nuclear factor-KB(NF-κB), were observed by quantitative reverse transcription-polymerase chain reaction, Western blot and immunohistochemistry.Results: Compared with normal rats, there were a significant decline in lung function and discharge of the vagus nerve(P < 0.01), a marked sign of lung inflammation and an increase of ACh, AChE, IL-6 and TNF-a level in BALF or lung tissue(P < 0.05, P < 0.01) and higher expression of a7 nAChR, JAK2, STAT3 and NF-κB(P<0.05, P<0.01) in the COPD rats. In rats receiving EA, the lung function and vagal discharge were enhanced(P < 0.01), lung inflammation was improved and the levels of ACh, AChE, IL-6 and TNF-a were decreased(P<0.01). Further, the expression of a7 nAChR, JAK2, STAT3 and NF-κB was downregulated(P < 0.05, P < 0.01). 4Atomically dispersed Au_1 catalyst towards efficient electrochemical synthesis of ammonia显示文摘Tremendous efforts have been devoted to explore energy-efficient strategies of ammonia synthesis to replace Haber–Bosch process which accounts for 1.4% of the annual energy consumption. In this study,atomically dispersed Au_1 catalyst is synthesized and applied in electrochemical synthesis of ammonia under ambient conditions. A high NH_4^+ Faradaic efficiency of 11.1% achieved by our Au_1 catalyst surpasses most of reported catalysts under comparable conditions. Benefiting from efficient atom utilization, an NH_4^+ yield rate of 1,305 lg h^(-1) mg_(Au)^(-1) has been reached, which is roughly 22.5 times as high as that by supported Au nanoparticles. We also demonstrate that by employing our Au_1 catalyst, NH4+can be electrochemically produced directly from N_2 and H_2 with an energy utilization rate of 4.02 mmol kJ^(-1). 5Pathways of China's PM;air quality 2015–2060 in the context of carbon neutrality显示文摘Clean air policies in China have substantially reduced particulate matter(PM;) air pollution in recent years,primarily by curbing end-of-pipe emissions.However,reaching the level of the World Health Organization(WHO) guidelines may instead depend upon the air quality co-benefits of ambitious climate action.Here,we assess pathways of Chinese PM;air quality from 2015 to 2060 under a combination of scenarios that link global and Chinese climate mitigation pathways(i.e.global 2℃-and 1.5℃-pathways,National Determined Contributions(NDC) pledges and carbon neutrality goals) to local clean air policies.We find that China can achieve both its near-term climate goals(peak emissions) and PM;air quality annual standard(35 μg/m;) by 2030 by fulfilling its NDC pledges and continuing air pollution control policies.However,the benefits of end-of-pipe control reductions are mostly exhausted by 2030,and reducing PM;exposure of the majority of the Chinese population to below 10 μg/m;by 2060 will likely require more ambitious climate mitigation efforts such as China’ s carbon neutrality goals and global1.5℃-pathways.Our results thus highlight that China’s carbon neutrality goals will play a critical role in reducing air pollution exposure to the level of the WHO guidelines and protecting public health.Jing Cheng Dan Tong Qiang Zhang Yang Liu Yu Lei Gang Yan Liu Yan Sha Yu Ryna Yiyun Cui Leon Clarke Guannan Geng Bo Zheng Xiaoye Zhang Steven JDavis Kebin He 2021National Science Review2021,8,12:23
6Asiatic acid inhibits lung cancer cell growth in vitro and in vivo by destroying mitochondria显示文摘Asiatic acid(AA), a pentacyclic triterpene found in Centella asiatica, displays significant antiproliferative effects on cancer cells in vitro although the underlying mechanism of this effect remains unknown. This study investigated the efficacy and mechanism of action of AA against lung cancer both in vivo and in vitro. Using the MTT assay, AA was found to induce apoptosis in a dose- and timedependent manner, an effect enhanced by pretreatment with an autophagy inhibitor. It also elevated expression of microtubule-associated protein 1 light chain 3(LC3) and decreased the expression of p62.Furthermore, exposure to AA resulted in collapse of the mitochondrial membrane potential and generation of reactive oxygen species(ROS), suggesting mitochondria are the target of AA. In the mouse lung cancer xenograft model, oral administration of AA significantly inhibited tumor volume and weight accompanied by significant apoptosis of lung cancer cells. In addition, it led to a significant decrease in the expression of proliferating cell nuclear antigen(PCNA). 8A drug-eluting Balloon for the trEatment of coronarY bifurcatiON lesions in the side branch:a prospective multicenter ranDomized (BEYOND)clinical trial in China显示文摘Background:Treatment of coronary bifurcation lesions remains challenging;a simple strategy has been preferred as of late,but the disadvantage is ostium stenosis or even occlusion of the side branch(SB).Only a few single-center studies investigating the combination of a drug-eluting stent in the main branch followed by a drug-eluting balloon in the SB have been reported.This prospective,multicenter,randomized study aimed to investigate the safety and efficacy of a paclitaxel-eluting balloon(PEB)compared with regular balloon angioplasty(BA)in the treatment of non-left main coronary artery bifurcation lesions.Methods:Between December 2014 and November 2015,a total of 222 consecutive patients with bifurcation lesions were enrolled in this study at ten Chinese centers.Patients were randomly allocated at a 1:1 ratio to a PEB group(n=113)and a BA group(n=109).The primary efficacy endpoint was angiographic target lesion stenosis at 9 months.Secondary efficacy and safety endpoints included target lesion revascularization,target vessel revascularization,target lesion failure,major adverse cardiac and cerebral events(MACCEs),all-cause death,cardiac death,non-fatal myocardial infarction,and thrombosis in target lesions.The main analyses performed in this clinical trial included case shedding analysis,base-value equilibrium analysis,effectiveness analysis,and safety analysis.SAS version 9.4 was used for the statistical analyses.Results:At the 9-month angiographic follow-up,the difference in the primary efficacy endpoint of target lesion stenosis between the PEB(28.7%±18.7%)and BA groups(40.0%±19.0%)was-11.3%(95%confidence interval:-16.3%to-6.3%,Psuperiority<0.0001)in the intention-to-treat analysis,and similar results were recorded in the per-protocol analysis,demonstrating the superiority of PEB to BA.Late lumen loss was significantly lower in the PEB group than in the BA group(-0.06±0.32 vs.0.18±0.34 mm,P<0.0001).For intention-to-treat,there were no significant differences between PEB and BA in the 9-month percentages of MACCEs(0.9%vs.3.7%,P=0.16)or non-fatal myocardial infarctions(0 vs.0.9%,P=0.49).There were no clinical events of target lesion revascularization,target vessel revascularization,target lesion failure,all-cause death,cardiac death or target lesion thrombosis in either group.Conclusions:In de novo non-left main coronary artery bifurcations treated with provisional T stenting,SB dilation with the PEB group demonstrated better angiographic results than treatment with regular BA at the 9-month follow-up in terms of reduced target lesion stenosis.Trial registration:ClinicalTrials.gov,NCT02325817;http://gffzzd3f38a9638994581s559v00fb96bp6wqp.ffgz.tsg.suse.edu.cn.Quan-Min Jing Xin Zhao Ya-Ling Han Ling-Ling Gao Yang Zheng Zhan-Quan Li Ping Yang Hong-Liang Cong Chuan-Yu Gao Tie-Min Jiang Hui Li Jun-Xia Li Dong-Mei Wang Geng Wang Zhan-Chun Cong Zhong Zhang 2020Chinese Medical Journal2020,,8:17

11High neutrophil-lymphocyte ratio indicates poor prognosis for acute-on-chronic liver failure after liver transplantation显示文摘AIM:To investigate the significance of pre-transplant neutrophil-lymphocyte ratio(NLR) in determining the prognosis of liver transplant(LT) recipients with acuteon-chronic liver failure(ACLF).METHODS:Data were collected from the liver transplantation data bank.The NLR values and other conventional inflammatory markers were evaluated for their ability to predict the prognosis of 153 patients with ACLF after LT.The NLR cut-off value was based on a receiver operating characteristic curve analysis.A Kaplan-Meier curve analysis and univariate and multivariate Cox regression models were used to define the independent risk factors for poor outcomes.RESULTS:The optimal NLR cut-off value was 4.6.Out of 153 patients,83(54.2%) had an NLR ≥ 4.6.The 1-,3-,and 5-year overall survival rates were 94.3%,92.5% and 92.5%,respectively,in the normal NLR group and 74.7%,71.8% and 69.8%,respectively,in patients with high NLRs(P < 0.001).Furthermore,there was a significant difference in infectious complications after LT between the high and normal NLR groups.There were no significant differences for other complications.In the multivariate Cox regression model,a high NLR was defined as a significant predictor of poor outcomes for LT.CONCLUSION:A high NLR is a convenient and available predictor for prognosis of LT patients and can potentially optimize the current criteria for LT in ACLF.Bing-Yi Lin Lin Zhou Lei Geng Zhi-Yun Zheng Jun-Jun Jia Jing Zhang Jia Yao Shu-Sen Zheng 2015World Journal of Gastroenterology2015,21,11:14
12Chemical screen identifies a geroprotective role of quercetin in premature aging显示文摘Aging increases the risk of various diseases. The main goal of aging research is to find therapies that attenuate aging and alleviate aging-related diseases. In this study, we screened a natural product library for geroprotective compounds using Werner syndrome (WS) human mesenchymal stem cells (hMSCs), a premature aging model that we recently established. Ten candidate compounds were identified and quercetin was investigated in detail due to its leading effects. Mechanistic studies revealed that quercetin alleviated senescence via the enhancement of cell proliferation and restoration of heterochromatin architecture in WS hMSCs. RNA-sequencing analysis revealed the transcriptional commonalities and differences in the geroprotective effects by quercetin and Vitamin C. Besides WS hMSCs, quercetin also attenuated cellular senescence in Hutchinson-Gilford progeria syndrome (HGPS) and physiological-aging hMSCs. 13Impact of Long-Term Atrazine Use on Groundwater Safety in Jilin Province, China显示文摘The long-lasting application of representative herbicide atrazine (ATR) has given rise to the accumulation of its residues in the groundwater. To investigate the impact of long-term ATR use on groundwater safety, the residues of ATR and its metabolites, desethylatrazine (DEA), deisopropylatrazine (DIA) and hydroxyatrazine (HA) were monitored in groundwater and top soil at the major corn growing region of Qian'an and Gongzhuling in Jilin Province, China. The residues of the target compounds were analyzed by UPLC-MS/MS. The limits of detection (LODs) of ATR, DEA, DIA, and HA were 0.5, 0.5, 5, and 0.5 ng L-1 in groundwater and 0.33, 0.33, 3.3, and 0.33 μg kg-1 in soil. The target compounds were found in 94% of groundwater samples and 100% of soil samples. The compounds detected most frequently in groundwater were ATR (89%), DEA (64%) and HA (17%), whereas in soil were ATR (97%), DEA (36%) and HA (97%). DIA was not detected in any determined groundwater and soil sample. Average residues were 106.8 ng L-1 for ATR, 0.9 ng L-1 for DEA and 0.3 ng L-1 for HA in groundwater, whereas 11.1 μg kg-1 for ATR, 0.4 μg kg-1 for DEA and 7.8 μg kg-1 for HA in soil. ATR residues detected in groundwater samples were below standards for drinking water quality (GB5749-2006, 2 μg L-1), while the total residues of ATR and its chloro-s-triazine metabolites (DEA and DIA) were below current WHO (World Health Organization) guideline value (GV, 0.1 mg L-1). In addition, concentrations of HA in groundwater were determined below current WHO GV (0.2 mg L-1). The results indicated that ATR is safe to be used in Jilin Province under the current application scheme. 15De novo combination therapy with lamivudine and adefovir dipivoxil in chronic hepatitis B patients显示文摘AIM:To investigate the appropriate time for combination therapy in HBeAg positive chronic hepatitis B(CHB) patients with decompensated cirrhosis.METHODS:Thirty HBeAg positive CHB patients with decompensated cirrhosis were enrolled in the study.All of the patients were given 48 wk combination therapy with lamivudine(LAM) and adefovir dipivoxil(ADV) .Briefly,10 patients were given the de novo combination therapy with LAM and ADV,whereas the other 20 patients received ADV in addition to LAM after hepatitis B virus(HBV) genetic mutation.RESULTS:Serum alanine aminotransferase and total bilirubin were both improved in the two groups at 4,12,24 and 48 wk after treatment.Serum albumin was also improved at 24 and 48 wk after combination therapy in both groups.The serum HBV DNA level wasstill detectable in every patient in the two groups at 4 and 12 wk after combination treatment.However,in the de novo combination group,serum HBV DNA levels in 4(40%) and 9(90%) patients was decreased to below 1×10 3 copies/mL at 24 and 48 wk after the combination treatment,respectively.In parallel,serum HBV DNA levels in 2(20%) and 8(40%) patients in the add-on combination group became undetectable at 24 and 48 wk after combination treatment,respectively.Furthermore,6(60%) patients in the de novo combination group achieved HBeAg seroconversion after 48 wk treatment,whereas only 4(20%) patients in the add-on combination group achieved seroconversion.Child-Pugh score of patients in the de novo combination group was better than that of patients in the add-on combination group after 48 wk treatment.Moreover,patients in the de novo combination group had a significantly decreased serum creatinine level and elevated red blood cell counts.CONCLUSION:De novo combination therapy with LAM and ADV was better than add-on combination therapy in terms of Child-Pugh score,virus inhibition and renal function.Xiao-Hong Fan Jian-Zhang Geng Li-Fen Wang Ying-Ying Zheng Hai-Ying Lu Jing Li Xiao-Yuan Xu 2011World Journal of Gastroenterology2011,17,43:12
16Ultrasensitive skin‐like wearable optical sensors based on glass micro/nanofibers显示文摘Electronic skin,a class of wearable electronic sensors that mimic the functionalities of human skin,has made remarkable success in applications including health monitoring,human-machine interaction and electronic-biological interfaces.While electronic skin continues to achieve higher sensitivity and faster response,its ultimate performance is fundamentally limited by the nature of low-frequency AC currents.Herein,highly sensitive skin-like wearable optical sensors are demonstrated by embedding glass micro/nanofibers(MNFs)in thin layers of polydimethylsiloxane(PDMS).Enabled by the transition from guided modes into radiation modes of the waveguiding MNFs upon external stimuli,the skin-like optical sensors show ultrahigh sensitivity(1870 k·Pa^-1),low detection limit(7 mPa)and fast response(10μs)for pressure sensing,significantly exceeding the performance metrics of state-of-the-art electronic skins.Electromagnetic interference(EMI)-free detection of high-frequency vibrations,wrist pulse and human voice are realized.Moreover,a five-sensor optical data glove and a 2×2-MNF tactile sensor are demonstrated.These initial results pave the way toward a new category of optical devices ranging from ultrasensitive wearable sensors to optical skins.Lei Zhang Jing Pan Zhang Zhang Hao Wu Ni Yao Dawei Cai Yingxin Xu Jin Zhang Guofei Sun Liqiang Wang Weidong Geng Wenguang Jin Wei Fang Dawei Di Limin Tong 2020Opto-Electronic Advances2020,3,3:12

19De novo assembly of a Chinese soybean genome显示文摘Soybean was domesticated in China and has become one of the most important oilseed crops. Due to bottlenecks in their introduction and dissemination, soybeans from different geographic areas exhibit extensive genetic diversity. Asia is the largest soybean market; therefore, a high-quality soybean reference genome from this area is critical for soybean research and breeding.Here, we report the de novo assembly and sequence analysis of a Chinese soybean genome for 'Zhonghuang 13' by a combination of SMRT, Hi-C and optical mapping data. The assembled genome size is 1.025 Gb with a contig N50 of 3.46 Mb and a scaffold N50 of 51.87 Mb. Comparisons between this genome and the previously reported reference genome(cv. Williams82) uncovered more than 250,000 structure variations. A total of 52,051 protein coding genes and 36,429 transposable elements were annotated for this genome, and a gene co-expression network including 39,967 genes was also established.
