1New eolian red clay sequence on the western Chinese Loess Plateau linked to onset of Asian desertification about 25 Ma ago显示文摘The expansion of inland Asia deserts has considerably influenced the environmental, social and economic activities in Asia. Aridification of inland Asia, especially timing of the initiation of Asian desertification, is a contentious topic in paleoclimatology. Late Cenozoic eolian loess-red clay sequences on the Chinese Loess Plateau, which possess abundant paleoclimatic and paleo-environmental information, can be regarded as an indicator of inland Asia desertification. Here we present a detailed magnetostratigraphic investigation of a new red clay sequence about 654 m in Zhuanglang located at the western Chinese Loess Plateau. Sedimentological, geochemical, mineralogical, and quartz morphological lines of evidence show that the red clay is of eolian origin. Magnetostratigraphic correlations indicate that this core sequence spans from 25.6 to 4.8 Ma, and typical eolian red clay appears as early as 25 Ma. This extends the lower limit of the red clay on the Chinese Loess Plateau from the previously thought early Miocene back into the late Oligocene. This new red clay record further implies that the inland Asia desertification was initiated at least by the late Oligocene. 2QTL editing confers opposing yield performance in different rice varieties显示文摘Grain yield is one of the most important and complex trait for genetic improvement in crops; it is known to be controlled by a number of genes known as quantitative trait loci(QTLs). In the past decade, many yield-contributing QTLs have been identified in crops.However, it remains unclear whether those QTLs confer the same yield performance in different genetic backgrounds. 3Rapid generation of genetic diversity by multiplex CRISPR/Cas9 genome editing in rice显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated endonuclease 9(CRISPR/Cas9) system has emerged as a promising technology for specific genome editing in many species. Here we constructed one vector targeting eight agronomic genes in rice using the CRISPR/Cas9 multiplex genome editing system. By subsequent genetic transformation and DNA sequencing, we found that the eight target genes have high mutation efficiencies in the T_0 generation. Both heterozygous and homozygous mutations of all editing genes were obtained in T_0 plants. In addition, homozygous sextuple, septuple, and octuple mutants were identified. As the abundant genotypes in T_0 transgenic plants, various phenotypes related to the editing genes were observed. 7Heteroepitaxial Growth and Characterization of 3C-SiC Films on Si Substrates Using LPVCVD显示文摘3C-SiC films have been deposited on Si (111) substrates by the Iow-pressure vertical chemical vapor deposition (LPVCVD) with gas mixtures of SiH4, C3H8 and Ha. The growth mechanism of SiC films can be obtained through the observations using field emission scanning electron microscope (FESEM). It is found that the growth process varies from surface control to diffusion control when the deposition temperature increases from 1270 to 1350℃. The X-ray diffraction (XRD) patterns show that the SiC films have good crystallinity and strong preferred orientation.The results of the high resolution transmission electron microscopy (HRTEM) image and the transmission electron diffraction (TED) pattern indicate a peculiar superlattice structure of the film. 8Neuroprotective effect of Longshengzhi capsule following permanent middle cerebral artery occlusion in rats显示文摘Objective:Longshengzhi capsule(LSZC)is an optimized preparation based on the traditional Chinese Medicine formula Buyanghuanwu Decoction(BYHWD),and is approved by the China Food and Drug Administration for treating stroke-induced disability and vascular diseases.Herein,we examined the pharmacodynamics,anti-apoptotic and anti-oxidant actions,and potential mechanisms of action of LSZC following stroke in rats.Methods:Permanent middle cerebral artery occlusion(MCAO)was used as an ischemic stroke model.LSZC was administered intragastrically.We examined the survival rate,bodyweight,and neurological deficits in stroke rats.Brain infarct size was determined by 2,3,5-triphenyltetrazolium chloride staining.Brain pathology was examined using hematoxylin and eosin staining,Nissl staining,and TdT-mediated dUTP Nick-End Labeling staining.Malondialdehyde,catalase,superoxide dismutase,and glutathione levels were examined by commercial kits.Expression of Nrf2,heme oxygenase-1,Bax,Bcl-2,cleaved caspase-3,and caspase-3 proteins in brain tissue was measured by Western blot.Results:LSZC markedly improved the survival rate and bodyweight,and reduced infarct volume and neurological deficit scores,in MCAO stroke rats.LSZC also significantly attenuated oxidative stress,as indicated by decreased expression of malondialdehyde,and upregulation of Nrf2,heme oxygenase-1,catalase,superoxide dismutase,and glutathione.Moreover,LSZC significantly decreased apoptosis,including a decrease in Bax and cleaved caspase-3 expression,and an increase in Bcl-2,as well as a reduction in numbers of apoptotic neurons.Conclusion:LSZC treatment is neuroprotective against ischemic stroke,potentially via reducing oxidative stress and apoptosis.The Nrf2 and apoptotic signaling pathways may play important roles in the antioxidant and anti-apoptotic actions of LSZC.Xiaoli Sun Shuzhen Guo Yong Wang Bangze Fu Junjie Liu Yili Zhang Ruocong Yang Chun Li Jian Gao Li Gao Wei Wang 2020Journal of Traditional Chinese Medical Sciences2020,7,1:4

11Fast integration and accumulation of beneficial breeding alleles through an AB–NAMIC strategy in wheat显示文摘Wheat(Triticum aestivum)is among the most important staple crops for safeguarding the food security of the growing world population.To bridge the gap between genebank diversity and breeding programs,we developed an advanced backcross-nested association mapping plus inter-crossed population(AB-NAMIC)by crossing three popular wheat cultivars as recurrent founders to 20 germplasm lines from a mini core collection.Selective backcrossing combined with selection against undesirable traits and extensive crossing within and between sub-populations created new opportunities to detect unknown genes and increase the frequency of beneficial alleles in the AB-NAMIC population.We performed phenotyping of 590 AB-NAMIC lines and a natural panel of 476 cultivars for six consecutive growing seasons and genotyped these 1066 lines with a 660K SNP array.Genome-wide association studies of both panels for plant development and yield traits demonstrated improved power to detect rare alleles and loci with medium genetic effects in AB-NAMIC.Notably,genome-wide association studies in AB-NAMIC detected the candidate gene TaSWEET6-7B(TraesCS7B03G1216700),which has high homology to the rice SWEET6b gene and exerts strong effects on adaptation and yield traits.The commercial release of two derived AB-NAMIC lines attests to its direct applicability in wheat improvement.Valuable information on genome-wide association studymapping,candidate genes,and their haplotypes for breeding traits are available through WheatGAB.Our research provides an excellent framework for fast-tracking exploration and accumulation of beneficial alleles stored in genebanks.Chengzhi Jiao Chenyang Hao Tian Li Abhishek Bohra Lanfen Wang Jian Hou Hongxia Liu Hong Liu Jing Zhao Yamei Wang Yunchuan Liu Zhiwei Wang Xin Jing Xiue Wang Rajeev K.Varshney Junjie Fu Xueyong Zhang 2023Plant Communications2023,4,3:3

13QTL analysis across multiple environments reveals promising chromosome regions associated with yield-related traits in maize under drought conditions显示文摘Drought is one of the most critical abiotic stresses influencing maize yield. Improving maize cultivars with drought tolerance using marker-assisted selection requires a better understanding of its genetic basis. In this study, a doubled haploid(DH) population consisting of 217 lines was created by crossing the inbred lines Han 21(drought-tolerant) and Ye 478(drought-sensitive). The population was genotyped with a 6 K SNP assay and 756 SNP(single nucleotide polymorphism) markers were used to construct a linkage map with a length of 1344 c M. Grain yield(GY), ear setting percentage(ESP), and anthesis–silking interval(ASI) were recorded in seven environments under well-watered(WW) and water-stressed(WS) regimes. High phenotypic variation was observed for all traits under both water regimes. Using the LSMEAN(least-squares mean) values from all environments for each trait, 18 QTL were detected, with 9 associated with the WW and 9 with the WS regime. Four chromosome regions,Chr. 3: 219.8–223.7 Mb, Chr. 5: 191.5–194.7 Mb, Chr. 7: 132.2–135.6 Mb, and Chr. 10: 88.2–89.4 Mb, harbored at least 2 QTL in each region, and QTL co-located in a region inherited favorable alleles from the same parent. A set of 64 drought-tolerant BC_3F_6 lines showed preferential accumulation of the favorable alleles in these four regions, supporting an association between the four regions and maize drought tolerance. QTL-by-environment interaction analysis revealed 28 ed QTL(environment-dependent QTL) associated with the WS regime and 22 associated with the WW regime for GY, ESP, and ASI. All WS QTL and55.6% of WW QTL were located in the ed QTL regions. 15Dynamical economic dispatch using distributed barrier function-based optimization algorithm显示文摘In this paper, we study the distributed economic dispatch problem where the supply demand balance, capacity constraints and ramp-rate constraints are considered. In order to accommodate varying power load and power storage in reality, we introduce two variables where each variable is time-varing and has its own dynamics. The renewable power is also considered. Barrier functions are introduced to deal with the local constraints by means of imposing penalty terms into the objective function to ensure that the optimal solution satisfies the corresponding constraints. Based on the Langrange dual theory, the primal optimization problem is transformed into the dual problem, which is solved by the primary-dual algorithm proposed in this paper. Under the assumption that the communication graph is an undirected and connected graph, we analyze the convergence of the proposed algorithm. 19Precipitate-stabilized surface enabling high-performance Na_(0.67)Ni_(0.33-x)Mn_(0.67)Zn_(x)O_(2) for sodium-ion battery显示文摘Electrode interfacial degradations are the key challenges for high-performance rechargeable batteries,usually mitigated through surface modification/coating strategies.Herein,we report a novel mechanism to enhance the surface stability of P2 layered cathodes by introducing a high density of dopant-enriched precipitates.Based on microscopic analysis,we show that forming a high density of precipitates at the grain surface can effectively suppress surface cracking and corrosion,which not only improves the surface/interface stability but also effectively suppresses the intergranular cracking issue.Increasing the doping level can lead to a greater density of precipitates at the surface region,which results in higher surface stability and increased cycling stability of the P2 layered cathode for a sodium-ion battery.We further reveal that prolonged cycling can induce the formation of a precipitate-free surface region due to the loss of Zn dopant and Na.Our in-depth microanalysis reveals cycling-induced dynamic structural evolution of the P2 layered cathodes,highlighting that dopant segregation-induced precipitation is a new approach to achieving high interfacial stability.Kuan Wang Zhengfeng Zhang Sulan Cheng Xiao Han Junjie Fu Manling Sui Pengfei Yan 2022eScience2022,2,5:2
20MKP-1 reduces Aβ generation and alleviates cognitive impairments in Alzheimer’s disease models显示文摘Mitogen-activated protein kinase(MAPK)phosphatase 1(MKP-1)is an essential negative regulator of MAPKs by dephosphorylating MAPKs at both tyrosine and threonine residues.Dysregulation of the MAPK signaling pathway has been associated with Alzheimer’s disease(AD).However,the role of MKP-1 in AD pathogenesis remains elusive.Here,we report that MKP-1 levels were decreased in the brain tissues of patients with AD and an AD mouse model.The reduction in MKP-1 gene expression appeared to be a result of transcriptional inhibition via transcription factor specificity protein 1(Sp1)cis-acting binding elements in the MKP-1 gene promoter.Amyloid-β(Aβ)-induced Sp1 activation decreased MKP-1 expression.However,upregulation of MKP-1 inhibited the expression of both Aβprecursor protein(APP)andβ-site APP-cleaving enzyme 1 by inactivating the extracellular signal-regulated kinase 1/2(ERK)/MAPK signaling pathway.Furthermore,upregulation of MKP-1 reduced Aβproduction and plaque formation and improved hippocampal long-term potentiation(LTP)and cognitive deficits in APP/PS1 transgenic mice.Our results demonstrate that MKP-1 impairment facilitates the pathogenesis of AD,whereas upregulation of MKP-1 plays a neuroprotective role to reduce Alzheimer-related phenotypes.Thus,this study suggests that MKP-1 is a novel molecule for AD treatment.Yehong Du Yexiang Du Yun Zhang Zhilin Huang Min Fu Junjie Li Yayan Pang Peng Lei Yu Tian Wang Weihong Song Guiqiong He Zhifang Dong 2019Signal Transduction and Targeted Therapy2019,4,1:2
