| 1 | Immunity-and-matrix-regulatory cells derived from human embryonic stem cells safely and effectively treat mouse lung injury and fibrosis显示文摘Lung injury and fibrosis represent the most significant outcomes of severe and acute lung disorders,including COVID-19.However,there are still no effective drugs to treat lung injury and fibrosis.In this study,we report the generation of clinical-grade human embryonic stem cells(hESCs)-derived immunity-and matrix-regulatory cells(IMRCs)produced under good manufacturing practice requirements,that can treat lung injury and fibrosis in vivo.We generate IMRCs by sequentially differentiating hESCs with serum-free reagents.IMRCs possess a unique gene expression profile distinct from that of umbilical cord mesenchymal stem cells(UCMSCs),such as higher expression levels of proliferative,immunomodulatory and anti-fibrotic genes.Moreover,intravenous delivery of IMRCs inhibits both pulmonary inflammation and fibrosis in mouse models of lung injury,and significantly improves the survival rate of the recipient mice in a dose-dependent manner,likely through paracrine regulatory mechanisms.IMRCs are superior to both primary UCMSCs and the FDA-approved drug pirfenidone,with an excellent efficacy and safety profile in mice and monkeys.In light of public health crises involving pneumonia,acute lung injury and acute respiratory distress syndrome,our findings suggest that IMRCs are ready for clinical trials on lung disorders. | Jun Wu Dingyun Song Zhongwen Li Baojie Guo Yani Xiao Wenjing Liu Lingmin Liang Chunjing Feng Tingting Gao Yanxia Chen Ying Li Zai Wang Jianyan Wen Shengnan Yang Peipei Liu Lei Wang Yukai Wang Liang Peng Glyn Nigel Stacey Zheng Hu Guihai Feng Wei Li Yan Huo Ronghua Jin Ng Shyh-Chang Qi Zhou Liu Wang Baoyang Hu Huaping Dai Jie Hao | 2020 | Cell Research2020,30,9: | 7 |
| 2 | Study of the Retinal Imaging and Output in Premature Baby under a Computer-assisted Binocular Indirect Ophthalmoscope显示文摘Purpose:To explore a clear retinal imaging and output and enhance the development of retinopathy of prematurity (ROP) screening,which is safe and effective for ROP screening in premature infants. Methods:A computer-assisted binocular indirect ophthalmoscope imaging and output system was equipped with camera and image processing hardware and connected to computers. The process of fundus examination was videotaped (photograph) and output. Simulated eyes were utilized to debug video head and acquire stable and clear fundus images by binocular indirect ophthalmoscope for premature infants. Results:Fundus imaging output technique was sucessfully established. The common reasons of unclear imaging and corresponding solutions were summarized. This technique can capture and output stable and clear fundus images of premature infants. | 4 | Enhanced carbon acquisition and use efficiency alleviate microbial carbon relative to nitrogen limitation under soil acidification显示文摘Background:Soil microbial communities cope with an imbalanced supply of resources by adjusting their element acquisition and utilization strategies.Although soil pH has long been considered an essential driver of microbial growth and community composition,little is known about how soil acidification affects microbial acquisition and utilization of carbon(C)and nitrogen(N).To close the knowledge gap,we simulated soil acidification and created a pH gradient by adding eight levels of elemental sulfur(S)to the soil in a meadow steppe.Results:We found that S-induced soil acidification strongly enhanced the ratio of fungi to bacteria(F:B)and microbial biomass C to N(MBC:MBN)and subsequently decreased the C:N imbalance between microbial biomass and their resources.The linear decrease in the C:N imbalance with decreasing soil pH implied a conversion from N limitation to C limitation.To cope with enhanced C versus N limitation,soil microbial communities regulated the relative production of enzymes by increasing the ratio ofβ-glucosidase(BG,C-acquiring enzyme)to leucine aminopeptidase(LAP,N-acquiring enzyme),even though both enzymatic activities decreased with S addition.Structural equation modeling(SEM)suggested that higher C limitation and C:N-acquiring enzyme stimulated microbial carbon-use efficiency(CUE),which counteracted the negative effect of metal stress(i.e.,aluminum and manganese)under soil acidification.Conclusions:Overall,these results highlight the importance of stoichiometric controls in microbial adaption to soil acidification,which may help predict soil microbial responses to future acid deposition. | Tianpeng Li Ruzhen Wang Jiangping Cai Yani Meng Zhirui Wang Xue Feng Heyong Liu Ronald F.Turco Yong Jiang | 2021 | Ecological Processes2021,10,1: | 2 |

[Method]In the cyanophyte enrichment area of northern Dianchi Lake,phytoplankton community,chlorophyll a,total nitrogen (TN) and total phosphorus (TP) concentration had been monitored monthly from January 2007 to December 2011,and the temporal and spatial variation of phytoplankton community and main pollution indicators were analyzed. [Result]From January 2007 to December 2011,algal diversity in the area increased slightly,and algal diversity in the alongshore region was less than that in the offshore region; chlorophyll a,TN and TP concentration decreased in the alongshore region,but it was otherwise in the offshore region; chlorophyll a,TN and TP concentration in the alongshore region were higher than those in the offshore region. | 9 | Effect of Ti layer thickness on microstructure and magnetic properties of Ti/Co/Ti films显示文摘TiCoTi granular films were prepared by DC facing-target magnetron sputtering system onto glass substrates and subsequently in situ annealing in vacuum. Structural of Ti (t nm)/Co (40 nm)/Ti (t nm) films were investigated in detail, which shows that the majority Co nanograins are formed as the hexagonal-close-packed (HCP) structure. Vibrating sample magnetometer (VSM) and scanning probe microscope (SPM) were applied to study the magnetic properties, morphologies and domain structures of these samples. It has been found that the structure and magnetic properties of the Ti/Co/Ti films depend strongly on the Ti layer thickness. The out-of-plane coercivities (Hc) of the film is maximum about 78.8 kA·m-1 when t=5 nm with annealing at 300 ℃; the distributing of grains of the sample is uniformity; and the average size of particles is about 13 nm. | 17 | Boronic acid-containing diarylpyrimidine derivatives as novel HIV-1NNRTIs: Design, synthesis and biological evaluation显示文摘Drug resistance remains to be a serious problem with type Ⅰ human immunodeficiency virus(HIV-1) nonnucleoside reverse transcriptase inhibitors(NNRTIs). A series of novel boronic acid-containing diarylpyrimidine(DAPY) derivatives were designed via bioisosterism and scaffold-hopping strategies,taking advantage of the ability of a boronic acid group to form multiple hydrogen bonds. The target compounds were synthesized and evaluated for their anti-HIV activities and cytotoxicity in MT-4 cells.Compound 10 j yielded the most potent activity and turned out to be a single-digit nanomolar inhibitor towards the HIV-1 ⅢB [wild-type(WT) strain], L100 I and K103 N strains, with 50% effective concentration(EC_(50)) values of 7.19–9.85 nmol/L. Moreover, 10 j inhibited the double-mutant strain RES056 with an EC_(50) value of 77.9 nmol/L, which was 3.3-more potent than that of EFV(EC_(50)= 260 nmol/L) and comparable to that of ETR(EC_(50)= 32.2 nmol/L). 10j acted like classical NNRTIs with high affinity for WT HIV-1 reverse transcriptase(RT) with 50% inhibition concentration(IC_(50)) value of 0.1837 μmol/L. Furthermore,molecular dynamics simulation indicated that 10 j was proposed as a promising molecule for fighting against HIV-1 infection through inhibiting RT activity. | 19 | Semi-quantitative design of synergetic surficial/interfacial sites for the semi-continuous oxidation of glycerol显示文摘Qualitatively identifying the dominant catalytic site for each step of a semi-continuous reaction and semi-quantitatively correlating such different sites to the catalytic performance is of great significance toward the integration of multiple well-optimized sites on a heterogeneous catalyst.Herein,a series of structurally defined TiO_(x)-based catalysts were synthesized to provide a feasible approach to investigate the aforementioned issues using the semi-continuous oxidation of glycerol as a model reaction.Detailed investigations have verified the simultaneous presence of two kinds of Pt active sites:1)Negatively charged Pt bound to the oxygen vacancies of modified TiO_(x)in the form of Pt^(δ−)-O_(v)-Ti^(3+) sites and 2)metallic Pt(Pt_(0)site)located away from the inter-face.Meanwhile,the proportion of surficial and interfacial sites varies over this series of catalysts.Combined in situ FTIR experiments revealed that the reaction network was well-tuned via a site cooperation mechanism:The surficial Pt_(0)sites dissociatively adsorb the OH group of glycerol with a monodentate bonding geometry and the Pt^(δ−)-O_(v)-Ti^(3+) sites dissociate the C=O bond of the aldehyde group in a bidentate form.Furthermore,CO-FTIR spectroscopy confirmed a correlation between the reaction rate/product selectivity and the fraction of surficial/interfacial sites.A rational proportion of surficial and interfacial sites is key to enabling a high yield of glyceric acid.The most active catalyst with 32%surface sites and 68%interfacial sites exhibited 90.0%glycerol conversion and 68.5%GLYA selectivity.These findings provide a deeper understanding of the structure-activity relationships using qualitative identification and semi-quantitative analysis. | Mingyu Gao Pengfei Yang Xinyi Zhang Yani Zhang Dianqing Li Junting Feng | 2022 | Fundamental Research2022,2,3: | 0 |
