| 1 | Comparative Transcriptome Analysis Reveals Different Host Cell Responses to Acute and Persistent Foot-and-Mouth Disease Virus Infection显示文摘Foot-and-mouth disease virus(FMDV) rapidly causes cytopathic effects in susceptible cells. Incomplete viral clearance during the acute infection leads to persistent infection. The relationship between host gene expression and the persistent infection remains unclear. In this study, we analyzed the transcriptome profiles of BHK-21 cells acutely and persistently infected with FMDV to identify differences in gene expression. GO and KEGG enrichment analyses showed that the 8,378 differentially expressed genes were significantly enriched in categories including metabolism, biosynthesis, ribosome function, and endocytosis. In persistently infected BHK-21 cells, ribosome-and translation-related genes were significantly down-regulated. There were more differentially expressed immune-related genes during persistent infection than during acute infection. Two hundred and seventy-four genes were differentially expressed in both acutely and persistently infected BHK-21 cells. Among these genes, heat shock protein family B member 1(Hspb1) knockdown significantly inhibited FMDV replication. | 2 | Engineering Cu^(+)/CeZrO_(x) interfaces to promote CO_(2) hydrogenation to methanol显示文摘Cu-based catalysts are widely employed for CO_(2) hydrogenation to methanol,which is expected as a promising process to achieving carbon neutrality.However,most Cu-based catalysts still suffer from low methanol yield with a passable CO_(2) conversion and lack insight into its reaction mechanism for guiding the design of catalysts.In this work,Cu^(+)/CeZrO_(x) interfaces are engineered by employing a series of ceria-zirconia solid solution catalysts with various Ce/Zr ratios,forming a Cu^(+)-O_(v)-Ce^(3+)structure where Cu^(+)atoms are bonded to the oxygen vacancies(O_(v))of ceria.Compared to Cu/CeO_(2) and Cu/ZrO_(2),the optimized catalyst(i.e.,Cu_(0.3)Ce_(0.3)Zr_(0.7))exhibits a much higher mass-specific methanol formation rate(192g_(MeOH)/kg_(cat)/h)at 240℃and 3 MPa.Through a series of in-situ and ex-situ characterization,it is revealed that oxygen vacancies in solid solutions can effectively assist the activation of CO_(2) and tune the electronic state of copper to promote the formation of Cu^(+)/CeZrO_(x) interfaces,which stabilizes the key*CO intermediate,inhibits its desorption and facilitates its further hydrogenation to methanol via the reverse watergas-shift(RWGS)+CO-Hydro pathway.Therefore,the concentration of*CO or the apparent Cu^(+)/(Cu^(+)+Cu^(0))ratio could be employed as a quantitative descriptor of the methanol formation rate.This work is expected to give a deep insight into the mechanism of metal/support interfaces in CO_(2) hydrogenation to methanol,offering an effective strategy to develop new catalysts with high performance. | Jingpeng Zhang Xiaohang Sun Congyi Wu Wenquan Hang Xu Hu Dawei Qiao Binhang Yan | 2023 | Journal of Energy Chemistry2023,,2: | 3 |
| 3 | Indoleamine 2,3-dioxygenase and regulatory dendritic cells contribute to the allograft protection induced by infusion of donor-specific splenic stromal cells显示文摘It has been reported that splenic stromal cells(SSCs)are capable of directly supporting the development of CD11c ^(lo)CD45RB^(+) IL-10-producing dendritic cells(DCs)from lineage-negative c-kit^(+) progenitor cells in the absence of exogenous cytokines.In vitro,DCs that differentiate on stromal cells suppress mixed leukocyte reaction responses and induce primary alloreactive CD4^(+) T cells to differentiate into IL-10-producing Tr1 cells.However,the precise mechanisms by which these SSCs exert their regulatory functions in vivo remain undefined.Furthermore,their possible contribution to the development of allograft transplantation tolerance has yet to be examined.Here,we have used both murine skin and cardiac allograft transplantation models to explore whether in vivo alloresponses can be regulated by infusion with donor-derived SSCs and to investigate the possible mechanisms by which SSCs exert regulatory effects to prevent allograft rejection.We show that intravenous SSC infusion prolonged murine skin allograft survival.The prolonged graft survival is associated with augmentation of the generation of regulatory DC subsets and CD4^(+) CD25^(+) Foxp3^(+) regulatory T cells(Tregs),as well as upregulation of the production of suppressive cytokines IL-10 and transforming growth factor(TGF)-b.Moreover,we found that indoleamine 2,3-dioxygenase and SSC-derived regulatory DCs contribute to allograft protection by infusion of donor-specific SSCs.Our data suggest that donor-derived SSCs could be used as a therapeutic target to promote transplantation tolerance. | Li Liu Lihua Duan Min Gong Hong Dai Quan Gong Fang Zheng Zheng Tan Congyi Wang Feili Gong Min Fang | 2011 | Cellular & Molecular Immunology2011,8,1: | 2 |
| 4 | Selective electroreduction of carbon dioxide to formic acid on electrodeposited SnO_2@N-doped porous carbon catalysts显示文摘Electrocatalytic reduction of CO_2 is a promising route for energy storage and utilization. Herein we synthesized SnO_2 nanosheets and supported them on N-doped porous carbon (N-PC) by electrodeposition for the first time. The SnO_2 and N-PC in the SnO_2@N-PC composites had exellent synergistic effect for electrocatalytic reduction of CO_2 to HCOOH. The Faradaic efficiency of HCOOH could be as high as 94.1% with a current density of 28.4 mA cm-2 in ionic liquid-MeCN system. The reaction mechanism was proposed on the basis of some control experiments. | 10 | Construction of cytopathic PK-15 cell model of classical swine fever virus显示文摘No cytopathic effect (CPE) can be observed on classical swine fever virus (CSFV) infected cell culture in vitro. This brings an obstacle to the researches on reciprocity between CSFV and host cells. Based on the construction of full-length genomic infectious Cdna clone of Chinese CSFV standard virulent Shimen strain, partial deletion is intro- duced into genomic Cdna to obtain a 7.5 kb subgenomic Cdna. A new subgenomic CSFV is derived from transfection with the subgenomic Cdna on PK-15 cells pre-infected by CSFV Shimen virus. Typical CPE induced by this subgenomic virus is observed on PK-15 cells. Coexistence of wild- type and subgenomic virus in cytopathic cell culture is dem- onstrated by RT-PCR detection in cytopathic cells. | 19 | Ultrafast laser-chemical modification hybrid fabrication of hydrostatic bearings with a superhydrophobicity solid-liquid interface显示文摘Oil film vortex severely reduces the stability of hydrostatic bearings. A solid-liquid interface with drag and slip properties can weaken the oil film vortex of the bearing. Here, a combined picosecond laser ablation and chemical modification method is proposed to prepare surfaces with microbulge array structure on 6061 aluminum alloy substrates. Because of the low surface energy of the perfluorododecyltriethoxysilane modification and the bulge geometry of the microbulge array structure, the surface shows excellent superhydrophobicity. The optimum contact angle in air for water is 164°, and that for oil is 139°. Two surfaces with “lotus-leaf effect” and “rose-petal effect” were obtained by controlling the processing parameters. The drag reduction properties of superhydrophobic surfaces were systematically investigated with slip lengths of 22.26 and 36.25 μm for deionized water and VG5 lubricant, respectively. In addition, the superhydrophobic surface exhibits excellent mechanical durability and thermal stability.
