2Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. 3Comparative study on skin temperature response to menstruation at acupuncture points in healthy volunteers and primary dysmenorrhea patients显示文摘OBJECTIVE:To assess skin temperature response to menstruation at acupuncture points in primary dysmenorrhea(PD) patients and healthy volunteers so as to explore acupuncture point specificity in reflecting diseases in the light of skin temperature.METHODS:Fifty-two PD patients and 49 healthy volunteers were recruited.Skin temperature measurements were performed with a skin temperature assessment device at 10 points.Absolute difference between skin temperature of the same point on the left and right side is used as main outcome measure.RESULTS:On the first day of menstruation, when menstrual pain attacking in PD patients, a significant increase in skin temperature difference was detected at Taixi(KI 3) compared with the healthy group(P < 0.01).A significant reduction in skin temperature difference was detected at Taixi(KI 3) in the first day of menstruation compared with those values in the third day after menstruation(P < 0.01)in the healthy group.On the third day after menstruation, a significant reduction in skin temperature difference was found at Zhongdu(LR 6) in PD group compared with the healthy group(P < 0.05).No significant differences of skin temperature were detected at other points(P > 0.05).CONCLUSION:The skin temperature difference at menstruation-relevant points in PD patients did not all change significantly more than those in women without PD.Significant difference was only found in Taixi(KI 3), the Yuan-source point of Kidney meridian.She Yanfen Ma Liangxiao Zhu Jiang Qi Conghui Wang Yanxia Tang Ling Li Chunhua Yuan Hongwen Liu Yuqi Song Jiashan 2017Journal of Traditional Chinese Medicine2017,37,2:8
4Grain refinement and orientation of AZ31B magnesium alloy in hot flow forming under different thickness reductions显示文摘An analysis of the hot flow forming of Mg-3.0 Al-1.0 Zn-0.3 Mn(AZ31 B) alloy was conducted by experiments and numerical simulations. The effects of different thickness reductions on the microstructure and mechanical properties were investigated at a temperature of 693 K, a spindle speed of 800 rev/min and a feed ratio of 0.1 mm/rev. Thickness reductions have great influence on the uniformity of microstructure along the radial direction(RD) and the grain sizes become refined and uniform when the thickness reduction reaches 45%. The c-axes of most grains are approximately parallel to the RD, with a slight inclination towards the axial direction(AD). The best mechanical properties with UTS of 280 MPa and YS of 175 MPa near the outer surface while 266 MPa and 153 MPa near the inner surface have been achieved due to grain refinement and texture. Moreover, the material flow behavior and stress/strain distributions for singlepass reductions were studied using the ABAQUS/Explicit software. The calculated results indicate that the materials mainly suffer from triaxial compressive stresses and undergo compressive plastic strain in RD and tensile strains in other directions. 6SARS-CoV-2 nsp12 attenuates type Ⅰ interferon production by inhibiting IRF3 nuclear translocation显示文摘SARS-CoV-2 is the pathogenic agent of COVID-19,which has evolved into a global pandemic.Compared with some other respiratory RNA viruses,SARS-CoV-2 is a poor inducer of type Ⅰ interferon(IFN).Here,we report that SARS-CoV-2 nsp12,the viral RNA-dependent RNA polymerase(RdRp),suppresses host antiviral responses.SARS-CoV-2 nsp12 attenuated Sendai virus(SeV)-or poly(I:C)-induced IFN-β promoter activation in a dose-dependent manner.It also inhibited IFN promoter activation triggered by RIG-I,MDA5,MAVS,and IRF3 overexpression.Nsp12 did not impair IRF3 phosphorylation but suppressed the nuclear translocation of IRF3.Mutational analyses suggested that this suppression was not dependent on the polymerase activity of nsp12.Given these findings,our study reveals that SARS-CoV-2 RdRp can antagonize host antiviral innate immunity and thus provides insights into viral pathogenesis.Wenjing Wang Zhuo Zhou Xia Xiao Zhongqin Tian Xiaojing Dong Conghui Wang Li Li Lili Ren Xiaobo Lei Zichun Xiang Jianwei Wang 2021Cellular & Molecular Immunology2021,18,4:4

8Comparison of diploid and triploid Carassius auratus provides insights into adaptation to environmental change显示文摘Focusing on adaptation of aquatic organisms, especially fish, can help elucidate complex dynamics in freshwater ecology. The differences in genetic and epigenetic regulation between diploid and triploid Carassius auratus affect survival under eutrophication. To identify the underlying mechanisms that lead to better adaption of triploids than diploids, we compared mRNA and microRNA(miRNA) expressions in liver tissue of diploid and triploid individuals obtained from the Dongting lake water system in central China. Differential expression analysis revealed that 566 transcripts were significantly up-regulated, whereas758 were down-regulated in triploids; of these differentially expressed transcripts, 33 transcripts including cacna1 d, nfkb2, hspa1 and fgfr4 were involved in the MAPK signaling pathway, and eight transcripts were determined to be regulated by seven miRNAs. Additionally, four of 25 differential expressed(DE) transcripts(mhc1, irf7, nfkb2 and pik3 c) involving the viral carcinogenesis pathway were regulated by four miRNAs. Furthermore, genetic polymorphisms analysis showed that more heterozygous mutations were detected in triploids than diploids. The d N/d S results revealed that 21 genes were under positive selection(d N/d S>1) in C. auratus complex. 10Evidence for the paternal mitochondrial DNA in the crucian carp-like fish lineage with hybrid origin显示文摘In terms of taxonomic status,common carp(Cyprinus carpio,Cyprininae)and crucian carp(Carassius auratus,Cyprininae)are different species;however,in this study,a newborn homodiploid crucian carp-like fish(2n=100)(2nNCRC)lineage(F1–F3)was established from the interspecific hybridization of female common carp(2n=100)×male blunt snout bream(Megalobrama amblycephala,Cultrinae,2n=48).The phenotypes and genotypes of 2 n NCRC differed from those of its parents but were closely related to those of the existing diploid crucian carp.We further sequenced the whole mitochondrial(mt)genomes of the 2n NCRC lineage from F1 to F3.The paternal mt DNA fragments were stably embedded in the mt-genomes of F1–F3 generations of 2n NCRC to form chimeric DNA fragments.Along with this chimeric process,numerous base sites of F1–F3 generations of 2 n NCRC underwent mutations.Most of these mutation sites were consistent with the existing diploid crucian carp.Moreover,the mt DNA organization and nucleotide composition of 2n NCRC were more similar to those of the existing diploid crucian carp than those of the parents.The inheritable chimeric DNA fragments and mutant loci in the mt-genomes of different generations of 2nNCRC provided important evidence of the mt DNA change process in the newborn lineage derived from hybridization of different species.Our findings demonstrated for the first time that the paternal mt DNA were transmitted into the mt-genomes of homodiploid lineage,which provided new insights into the existence of paternal mt DNA in the mt DNA inheritance.Shi Wang Ni Jiao Lu Zhao Meiwen Zhang Pei Zhou Xuexue Huang Fangzhou Hu Conghui Yang Yuqin Shu Wuhui Li Chun Zhang Min Tao Bo Chen Ming Ma Shaojun Liu 2020Science China(Life Sciences)2020,63,1:2
11An integrated strategy towards the facile synthesis of core-shell SiC-derived carbon@N-doped carbon for high-performance supercapacitors显示文摘Porous active core-shell carbon material with excellent synergistic effect has been regarded as a prospective material for supercapacitors.Herein,we report an integrated method for the facile synthesis of carbide-derived carbon(CDC)encapsulated with porous N-doped carbon(CDC@NC)towards highperformance supercapacitors.Polydopamine(PDA)as nitrogen and carbon sources was simply coated on SiC nanospheres to form SiC@PDA,which was then directly transformed into CDC@NC via a onestep molten salt electro-etching/in-situ doping process.The synthesized CDC@NC with hierarchically porous structure has a high specific surface area of 1191 m^(2) g^(-1).The CDC core and NC shell are typical amorphous carbon and more ordered N-doped carbon,respectively.Benefitting from its unique dual porous structures,the CDC@NC demonstrates high specific capacitances of 255 and 193 F g^(-1) at 0.5 and20 A g^(-1),respectively.The reaction mechanism of the electro-etching/in-situ doping process has also been investigated through experimental characterizations and theoretical density functional theory calculations.It is suggested that the molten salt electro-etching/in-situ doping strategy is promising for the synthesis of active core-shell porous carbon materials with synergistic properties for supercapacitors without the need for additional doping/activation processes.Zhongya Pang Guangshi Li Xingli Zou Chenteng Sun Conghui Hu Wei Tang Li Ji Hsien-Yi Hsu Qian Xu Xionggang Lu 2021Journal of Energy Chemistry2021,30,5:2

13Abnormal meiosis in fertile and sterile triploid cyprinid fish显示文摘Meiosis is the key process for producing mature gametes. A natural fertile triploid Carassius auratus population(3nDTCC) and an artificially derived sterile triploid crucian carp(3nCC) have been previously observed, providing suitable model organisms for investigating meiosis characteristics in triploid fish. In the present study, the microstructures and ultrastructures of spermatogenesis were studied in these fishes. Td T-mediated d UTP nick end labeling detection was performed to investigate the apoptosis of spermatocytes. Fluorescence in situ hybridization was employed to trace chromatin pairing. In addition, the m RNA expressions of cell cycle-related genes(i.e., cell division control 2 and cell cycle protein B) were determined by quantitative realtime polymerase chain reaction to illustrate the molecular mechanism of abnormal meiosis in the 3nCC. The results showed that the 3nCC undergoes an irregular prophase I, with the chromosomes distributed in a unipolar radial manner and exhibiting partial pairing, hindered metaphase I, and degenerated cells in the subsequent stages. Meanwhile, the 3nDTCC presented a relatively regular meiotic prophase I with complete conjugate chromosome pairs and chromosomes distributed along the karyotheca,which were presented as a ring structure by slicing. Only the spreads with 130–150 irregular chromosomes can be easily detected in the 3nDTCC, suggesting that it may undergo an abnormal metaphase I. 14Analysis of the formation process and performance of magnetic Fe_3O_4@Poly(4-vinylpyridine) absorbent prepared by in-situ synthesis显示文摘The morphology evolution of the mesoporous magnetic composite nanospheres Fe_3O_4@Poly(4-vinylpyridine) during the formation process and its absorption property of Congo red were studied in this study. A simple solvothermal method was applied for the fabrication of Fe_3O_4@Poly(4-vinylpyridine)composites with regular structure and uniform size distribution in the presence of 4-vinylpyridine as the structure inducer. The morphology, structure and magnetism performance were characterized and the adsorption model and mechanism were discussed. The results showed that the Fe_3O_4@Poly(4-vinylpyridine) composites were efficient adsorbent for the removal of Congo red from water and it could be reused by a magnetic separation. 16Functional analysis of flavanone 3-hydroxylase(F3H)from Dendrobium officinale,which confers abiotic stress tolerance显示文摘Flavonoids are important bioactive components in Dendrobium officinale,a medicinal orchid.They are involved in many biological activities,including protecting plants against biotic and abiotic stresses.Research on the key genes related to flavonoid biosynthesis in D.officinale is limited.In this study,one of the key flavonoid biosynthesis genes,flavanone 3-hydroxylase(F3H),was characterized from D.officinale.The open reading frame of DoF3H was 1134 bp long and it encoded a 377-amino acid protein.The DoF3H protein showed considerably high homology with F3H proteins from other plant species and shared a common evolutionary ancestor with other F3Hs.DoF3H transcripts were detected in different organs of adult plants and mainly accumulated in flowers,followed by roots,stems and leaves,a pattern that was similar to the content of flavonoids.Recombinant DoF3H protein,which was localized in the cytosol,could convert naringenin to dihydrokaempferol.The mRNA levels of DoF3H were significantly induced by salt and cold stresses.Furthermore,the heterologous expression of DoF3H in Escherichia coli conferred it higher tolerance to salt and cold stresses.These results provide insight into the molecular function of DoF3H in the biosynthesis of flavonoids,and provide a new application for improvement of abiotic tolerance in D.officinale.Can Si Wei Dong Jaime A.Teixeira da Silva Chunmei He Zhenming Yu Mingze Zhang Lei Huang Conghui Zhao Danqi Zeng Chuanmao Li Shengxiang Qiu Jun Duan 2023Horticultural Plant Journal2023,9,2:1
17Aerosol characterization in a city in central China plain and implications for emission control显示文摘Extensive studies on aerosol chemistry have been carried out in megacities in China,however,aerosol characterization in Central China Plain(CCP)is limited.Here we conducted real-time measurements of fine particle composition with a time-of-flight aerosol chemical speciation monitor in Kaifeng,Henan province in October 2019.Our results showed that nitrate and organics constituted the major fraction of non-refractory PM_(2.5)for the entire study,on average accounting for 34%and 33%,respectively.However,aerosol composition was substantially different among four periods due to different meteorological conditions and chemical processing.For instance,nitrate presented the lowest contribution during the first period due to evaporative loss associated with high temperature(T),and then rapidly increased during polluted periods as a function of relative humidity(RH).Positive matrix factorization analysis showed the dominance of secondary organic aerosol(SOA)in OA,and also the changes in OA composition under different T and RH levels.In addition,this study is unique with two periods of local emission controls.Back trajectory and coefficient of divergence analysis showed that air pollution in CCP was overall homogeneously distributed.As a result,the effectiveness of local emission controls in this region was strongly affected by meteorological conditions and regional transport.We found that one of the periods with emission control even showed the highest concentrations for the entire study.Our results point towards the limited effect of local emission controls in mitigating air pollution in CCP,and highlight the importance of joint emission controls under unfavorable meteorological conditions.Zhijie Li Lu Lei Yanpei Li Chun Chen Qingqing Wang Wei Zhou Jiaxing Sun Conghui Xie Yele Sun 2021Journal of Environmental Sciences2021,33,6:1
18Ccdc57 is required for straightening the body axis by regulating ciliary motility in the brain ventricle of zebrafish显示文摘Recently,cilia defects have been proposed to contribute to scoliosis.Here,we demonstrate that coiled-coil domain-containing 57(Ccdc57)plays an essential role in straightening the body axis of zebrafish by regulating ciliary beating in the brain ventricle(BV).Zygotic ccdc57(Zccdc57)mutant zebrafish developes scoliosis without significant changes in their bone density and calcification,and the maternal-zygotic ccdc57(MZccdc57)mutant embryos display curved bodies since the long-pec stage.The expression of ccdc57 is enriched in ciliated tissues and immunofluorescence analysis reveals colocalization of Ccdc57-HA with acetylated a-tubulin,implicating it in having a role in ciliary function.Further examination reveals that it is the coordinated cilia beating of multiple cilia bundles(MCB)in the MZccdc57 mutant embryos that is affected at 48 hours post fertilization,when the compromised cerebrospinal fluid flow and curved body axis have already occurred.Either ccdc57 m RNA injection or epinephrine treatment reverses the spinal curvature in MZccdc57 mutant larvae from ventrally curly to straight or even dorsally curly and significantly upregulates urotensin signaling.This study reveals the role of ccdc57 in maintaining coordinated cilia beating of MCB in the BV.Lu Li Juan Li Yuan Ou Jiaxin Wu Huilin Li Xin Wang Liying Tang Xiangyan Dai Conghui Yang Zehong Wei Zhan Yin Yuqin Shu 2023Journal of Genetics and Genomics2023,50,4:0

To explore the genetic diversity and identify the source region of introductions of Pelophylax nigromaculatus to the Xinjiang Uyghur Autonomous Region, we sequenced 695 bp of the mitochondrial Cyt b gene in 140 individuals of P.nigromaculatus and identified 42 haplotypes in Heilongjiang, Beijing, Jiangsu, Shaanxi, Zhejiang provinces, Chongqing and the Xinjiang Uyghur Autonomous Region. We detected only four mitochondrial haplotypes in 20 specimens from Yining city in Ili Kazak Autonomous Prefecture. We traced the origin of Yining P. nigromaculatus to the Beijing and Chongqing area. 20Defect-modulated synthesis and optoelectronic properties in chemical vapor deposited CsPbBr_(3) microplates显示文摘The remarkable optoelectronic features of halide perovskite promote their potential applications in semiconductor devices beyond solar cells,which require high-quality single crystals with controlled defect levels.Herein,we investigated the synthesis mechanism of chemical vapor deposited single-crystalline all-inorganic perovskite microplates(MPs),and reported a defect-modulated photocurrent which is closely related to the growth sequence of the MPs.The MP synthesis initiates from island-like nano-disks,and subsequently transits to a layer-by-layer fashion,resulting in a defect-rich area at the center of the MPs.At elevated temperatures,these central defects may be thermally activated and become highly mobile,leading to photoluminescence quenching and degression of local and overall optoelectronic attributes,as evidenced by the spatial resolved optical and electrical scanning probe microscopy.Overall,this work shines light on the formation,proliferation and dynamics of defects in perovskites,and offers guidance for preparation of high-quality perovskites micro-crystals for functional semiconductor devices with high temperature stability.Yanan Bao Hengshan Wang Meiqi An Huayi Tang Jing Li Jianliang Li Conghui Tan Yingmin Luo Jiao Xu Yiming Yang 2024Nano Research2024,17,5:0
