2Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
3Identification of novel cis-elements bound by BplMYB46 involved in abiotic stress responses and secondary wall deposition显示文摘Transcription factors(TFs)play vital roles in various biological processes by binding to cis-acting elements to control expressions of their target genes.The MYB TF BplMYB46,from Betula platyphylla,is involved in abiotic stress responses and secondary wall deposition.In the present study,we used a TF-centered yeast onehybrid technology(TF-centered Y1H)to identify the cisacting elements bound by BplMYB46.We screened a shortinsert random library and identified three cis-elements bound by BplMYB46:an E-box(CA(A/T/C)(A/G/C)TG)and two novel motifs,a TC-box(T(G/A)TCG(C/G))and a GT-box(A(G/T)T(A/C)GT(T/G)C).Chromatin immunoprecipitation(ChIP)and effector-reporter coexpression assays in Nicotiana tabacum confirmed binding of BplMYB46 to the TC-box,GT-box,and E-box motifs in the promoters of the phenylalanine ammonia lyase(PAL),peroxidase(POD),and superoxide dismutase(SOD)genes,which function in abiotic stress tolerance and secondary wall biosynthesis.This finding improves our understanding of potential regulatory mechanisms in the response to abiotic stress and secondary wall deposition of BplMYB46 in B.platyphylla.Huiyan Guo Liuqiang Wang Chuanping Yang Yiming Zhang Chunrui Zhang Chao Wang 2018Journal of Integrative Plant Biology2018,60,10:7
4Clonal variations in nutritional components of Pinus koreansis seeds collected from seed orchards in Northeastern China显示文摘From four Korean pine(Pinus koraiensis)orchards, 60 clones were selected and analyzed for the fatty acid and amino acid components of the seeds to reveal the variations and correlations of the seed characteristics among the clonal source orchards and clones. The nutritional components of the seeds of the P. koraiensis trees exhibited rich genetic variation; the variation coefficient of the fatty acids was 2.24–66.83 %, while the variation coefficient of the amino acids was 14.70–38.88 %. Relatively high genetic-improvement potential exists for the nutritional components of the seeds. The phenotypic differentiation of the fatty acid and amino acid components reveals that variation within the population(85.18 %) was the primary source for the variation of the fatty acid components; variation among the orchards(63.08 %) was the primary source of the variation of the amino acid components. Data drawn from various clonal source orchards all showed that the seed characteristics were highly controlled by heritability(h2[ 80 %), and the seed characteristics of the P. koraiensis trees exhibited a similar genetic gain trend. The principal components were analyzed to obtain the comprehensive principal component value for each clonal seed orchard. Twelve clones were selected based on a clonal selection rate of 20 %. Correlation and multiple stepwise-regression analyses were conducted, considering different location conditions, to reveal the stable correlations between the seed characteristics to facilitate improvements of the seed yield of P.koraiensis trees and the clonal selection. Species of real characteristics in P. koraiensis were controlled by higher heritability. 5Recent advances in fermentative biohydrogen production显示文摘Hydrogen energy, as a kind of clean energy with great potential, has been a hotspot for study worldwide. Based on the recent research on biohydrogen production, this paper gives a brief review on the following aspects: fermentative hydrogen production process and the engineering control statagy, key factors affecting the effciency of hydrogen production, such as substrates, cysteine, metal ions, anaerobic fermentation terminal products, and formic acid and ammonia. Moreover, anaerobic fermentative hydrogen-producing strain and reg- ulation and control of enzyme gene in fermentative hydrogen production are also discussed. 6Analysis of three types of triterpenoids in tetraploid white birches(Betula platyphylla Suk.) and selection of plus trees显示文摘Betulin, oleanolic acid, and betulinic acid are naturally occurring pentacyclic triterpenoids that have significant medicinal value. Considerable amounts of these triterpenoids are available in the outer bark of white birch.In this study, we used ultrasound-assisted extraction(UAE)to extract triterpenoids from birch bark rapidly and with high efficiency. Using high performance liquid chromatography(HPLC), three types of triterpenoids were separated and detected. We examined the differences among triterpenoids extracted from diploid versus tetraploid white birch. Then, we used factor analysis to screen out tetraploid white birches with comprehensively excellent performance. The results indicate that the optimum conditions for extraction include the use of ethanol as an extraction solvent, a solid-to-liquid ratio of 0.1 g/10 ml,ultrasonic power set at 100 W, a temperature of 60 °C and an extraction time of 15 min. A reversed-phase C18 column(4.6 mm × 250 mm × 5 lm) with a column temperature of 30 °C and the mobile phase composed of A(acetonitrile) and B(0.1 % aqueous phosphoric acid, v/v)at a flow rate of 0.5 ml/min were used, and the detection wavelength was 195 nm. No significant difference was observed between diploid and tetraploid white birch in terms of the content of three types of triterpenoids(at a confidence level of 0.05). 7Two novel eukaryotic translation initiation factor 5A genes from Populus simonii×P.nigra confer tolerance to abiotic stresses in Saccharomyces cerevisiae显示文摘The role of plant e IF5 A proteins in multiple biological processes, such as protein synthesis regulation,translation elongation, m RNA turnover, programmed cell death and stress tolerance is well known. Toward using these powerful proteins to increase stress tolerance in agricultural plants, in the present study, we cloned and characterized Psne IF5A2 and Psne IF5A4 from young poplar(P. simonii 9 P. nigra) leaves. The deduced amino acid sequences of Psne IF5A2 and Psne IF5A4 were 98 %similar to each other, and they are orthologs of e IF5A1 in Arabidopsis. In a subcellular localization analysis,Psne IF5A2 and Psne IF5A4 proteins were localized in the nucleus and cytoplasm. q RT-PCR analysis showed that Psne IF5A2 and Psne IF5A4 were transcribed in poplar flowers, stem, leaves, and roots. In addition, they were also induced by abiotic stresses. Transgenic yeast expressing Psne IF5A2 and Psne IF5A4 had increased salt, heavy metal, osmotic, oxidative tolerance. 9Screening of Transgenic Soybean Transformed by Means of Pollen-tube Using Kanamycin显示文摘Kanamycin was used to screen T0 seeds of the variety Dongnong 46 transformed by means of pollen-tube method. The results showed that 400 mg·L-1 kanamycin could inhibit growth of non-transgenic plants, and 2 positive plants were gotten combined with Gus dyeing and PCR detection. 14Use of Real- time RT-PCR Analysis for mRNA Expression of Tobacco Ferritin Gene (NtFer1)显示文摘To understand the use of real-time reverse transcription-polymerase chain reaction (real-time RT-PCR) for detecting the relative abundance of mRNA, the expression of a tobacco ferritin gene (NtFer1) was detected by Northern blot and real-time RT-PCR. The results indicated that both of the two methods were able to detect mRNA expression of NtFer1 clearly and similarly, namely NtFer1 expression was responsive to iron-overload, and the abundance of NtFer1 mRNA was greatly increased after iron loaded for 6 h. To compare the effect and sensitivity of two methods, results revealed that Northern blot need 30 ?滋g of total RNA and at least 3 days for the total protocol performance, whereas real-time RT-PCR only need 2 ?滋g of total RNA and 1.5 h. The real-time RT-PCR is rather sensitive and effective than Northern blot.
