1Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
2Design Property Network-Based Change Propagation Prediction Approach for Mechanical Product Development显示文摘Design changes are unavoidable during mechanical product development; whereas the avalanche propagation of design change imposes severely negative impacts on the design cycle. To improve the validity of the change propagation prediction, a mathematical programming model is presented to predict the change propagation impact quantitatively. As the foundation of change propagation prediction, a design change analysis model(DCAM)is built in the form of design property network. In DCAM,the connections of the design properties are identified as the design specification, which conform to the small-world network theory. To quantify the change propagation impact, change propagation intensity(CPI) is defined as a quantitative and much more objective assessment metric.According to the characteristics of DCAM, CPI is defined and indicated by four assessment factors: propagation likelihood, node degree, long-chain linkage, and design margin. Furthermore, the optimal change propagation path is searched with the evolutionary ant colony optimization(ACO) algorithm, which corresponds to the minimized maximum of accumulated CPI. In practice, the change impact of a gear box is successfully analyzed. 3Characterization of flavor fingerprinting of red sufu during fermentation and the comparison of volatiles of typical products显示文摘Traditional Chinese red sufu is a popular condiment,and typical flavor is an important indicator of sensory qualities in commercial products.In this study,the volatile aroma analysis of Wangzhihe red sufu(WRS)from different fermentation stages and four typical red sufu products from different locations was carried out by gas chromatography-mass spectrometry(GC–MS),gas chromatography-mass spectrometry/olfactory(GC–MS/O),electronic nose(E-nose)and sensory evaluation.Results showed that 106 volatile compounds were identified in Wangzhihe red sufu during fermentation process,in which phenolics and alcohols were dominant at molded and salted phetze stages,while esters and alcohols became predominate at post-fermentation stage.The volatile aroma substances varied at each fermentation stage.Furthermore,86 volatiles,including 16 aroma-active compounds,were detected in four typical red sufu products.The multivariate analysis results showed the difference in samples from different fermentation stages and typical red sufu products according to GC–MS and E-nose analysis.Combined with flavor omics,discriminant model was established for effective discrimination of samples from different fermentation stages and locations,as well as references to sufu maturity extent.The study presented a new strategy for quality evaluation of red sufu,which could be the supplement to quality evaluation standards.Peng Wang Xiaoran Ma Wenping Wang Dandan Xu Xin Zhang Jian Zhang Yong Sun 2019Food Science and Human Wellness2019,8,4:7
4Multiobjective optimization using an immunodominance and clonal selection inspired algorithm显示文摘Based on the mechanisms of immunodominance and clonal selection theory,we propose a new multiobjective optimization algorithm,immune dominance clonal multiobjective algorithm(IDCMA).IDCMA is unique in that its fitness values of current dominated individuals are assigned as the values of a custom distance measure,termed as Ab-Ab affinity,between the dominated individuals and one of the nondominated individuals found so far.According to the values of Ab-Ab affin-ity,all dominated individuals(antibodies) are divided into two kinds,subdominant antibodies and cryptic antibodies.Moreover,local search only applies to the sub-dominant antibodies,while the cryptic antibodies are redundant and have no func-tion during local search,but they can become subdominant(active) antibodies during the subsequent evolution.Furthermore,a new immune operation,clonal proliferation is provided to enhance local search.Using the clonal proliferation operation,IDCMA reproduces individuals and selects their improved maturated progenies after local search,so single individuals can exploit their surrounding space effectively and the newcomers yield a broader exploration of the search space.The performance comparison of IDCMA with MISA,NSGA-Ⅱ,SPEA,PAES,NSGA,VEGA,NPGA,and HLGA in solving six well-known multiobjective function optimization problems and nine multiobjective 0/1 knapsack problems shows that IDCMA has a good performance in converging to approximate Pareto-optimal fronts with a good distribution.GONG MaoGuo JIAO LiCheng MA WenPing DU HaiFeng 2008Science in China(Series F)2008,51,8:6
5Optimal stimulation parameters for Renzhong (DU 26) electro-acupuncture for improving motor function in a rat model of middle cerebral artery occlusion显示文摘The selection of electro-acupuncture parameters remains poorly unified between clinical studies.The present study observed the effects of electro-acupuncturing Renzhong (DU 26) with different stimulation parameters on motor function recovery following middle artery occlusion injury in rats.Results showed an optimal stimulation parameter for Renzhong electro-acupuncture that was low frequency and mild current (2 Hz,1 mA) significantly improved cortical excitability and conductive function,and promoted recovery in a rat model of motor function in middle artery occlusion.Frequency had a greater impact than current or interaction,and played a critical role in electro-acupuncture therapy.Lin Han Shu Wang Jinquan Ma Wenping Yao Yan Shen Xuemin Shi 2011Neural Regeneration Research2011,6,12:5
6Zinc Anode for Mild Aqueous Zinc-Ion Batteries: Challenges, Strategies, and Perspectives显示文摘The rapid advance of mild aqueous zinc-ion batteries(ZIBs)is driving the development of the energy storage system market.But the thorny issues of Zn anodes,mainly including dendrite growth,hydrogen evolution,and corrosion,severely reduce the performance of ZIBs.To commercialize ZIBs,researchers must overcome formidable challenges.Research about mild aqueous ZIBs is still developing.Various technical and scientific obstacles to designing Zn anodes with high stripping efficiency and long cycling life have not been resolved.Moreover,the performance of Zn anodes is a complex scientific issue determined by various parameters,most of which are often ignored,failing to achieve the maximum performance of the cell.This review proposes a comprehensive overview of existing Zn anode issues and the corresponding strategies,frontiers,and development trends to deeply comprehend the essence and inner connection of degradation mechanism and performance.First,the formation mechanism of dendrite growth,hydrogen evolution,corrosion,and their influence on the anode are analyzed.Furthermore,various strategies for constructing stable Zn anodes are summarized and discussed in detail from multiple perspectives.These strategies are mainly divided into interface modification,structural anode,alloying anode,intercalation anode,liquid electrolyte,non-liquid electrolyte,separator design,and other strategies.Finally,research directions and prospects are put forward for Zn anodes.This contribution highlights the latest developments and provides new insights into the advanced Zn anode for future research.Jinzhang Yang Bosi Yin Ying Sun Hongge Pan Wenping Sun Baohua Jia Siwen Zhang Tianyi Ma 2022Nano-Micro Letters2022,14,3:5
7CMTM4 inhibits cell proliferation and migration via AKT, ERK1/2,and STAT3 pathway in colorectal cancer显示文摘CMTM4(CKLF-like MARVEL transmembrane domain containing 4),a potential tumor suppressor gene,is involved in several types of malignancies.It has been reported to be downregulated and exhibit anti-tumorigenic activities by regulating cell growth and cell cycle in clear cell renal cell carcinoma.It has also been identified as a tumor suppressor in hepatocellular carcinoma(HCC),and its negative expression is a risk factor for poor prognosis of HCC patients.In the present study,an integrated bioinformatics analysis based on The Cancer Genome Atlas(TCGA)database showed that CMTM4 was frequently reduced in colorectal cancer(CRC)and high expression of CMTM4 was associated with increased overall survival rates.Based on these findings,we adopted gain-of-function and lost-of-function strategies using SW480 and HT29 CRC cell lines which have relatively low and high endogenous CMTM4 levels,respectively.We observed impeded cell proliferation and migration upon overexpression of CMTM4 in SW480 cells,and the opposite effects were observed upon knockdown of CMTM4 in HT-29 cells.Cell signaling pathways essential for CRC progression were then examined,and the phosphorylation levels of AKT,ERK1/2,and STAT3 were found to be decreased by CMTM4 overexpression in SW480 cells and elevated by CMTM4 silencing in HT29 cells.Their inhibitors were used to validate that the three signaling pathways contributed to the inhibitory effects of CMTM4 on CRC cells.Taken together,our results suggest that CMTM4 plays a tumor suppressive role in CRC.Hui Xue Ting Li Pingzhang Wang Xiaoning Mo Hejun Zhang Shigang Ding Dalong Ma Wenping Lv Jing Zhang Wenling Han 2019Acta Biochimica et Biophysica Sinica2019,51,9:5
8Intelligent multi-user detection using an artificial immune system显示文摘Artificial immune systems (AIS) are a kind of new computational intelligence methods which draw inspiration from the human immune system. In this study, we introduce an AIS-based optimization algorithm, called clonal selection algorithm, to solve the multi-user detection problem in code-division multipleaccess communications system based on the maximum-likelihood decision rule. Through proportional cloning, hypermutation, clonal selection and clonal death, the new method performs a greedy search which reproduces individuals and selects their improved maturated progenies after the affinity maturation process. Theoretical analysis indicates that the clonal selection algorithm is suitable for solving the multi-user detection problem. 9Characterization of NPR1 Genes from Norton and Cabernet Sauvignon Grapevine显示文摘Non-expressor of pathogenesis-related genes 1 (NPR1) plays a significant role in the defense responses of plants to pathogens by regulating the expression of defense-related genes. In the present study, we isolated two NPR1 genes from Vitis aestivalis cv. Norton and Vitis vinifera cv. Cabernet Sauvignon, which were referred to as VaNPR1.1 and VvNPR1. 1-CS, respectively. They encode a protein of 584 amino acids with a predicted molecular weight of 64.8 kDa and a theoretical isoelectric point (pI) of 5.74. The predicted amino acid sequences of VaNPR1.1 and VvNPR1.1-CS differ by only one amino acid. Over-expression of VaNPR1.1 gene in Arabidopsis npr1-1 mutant plants restores the transcriptional expression of AtPR-1 gene, though not to the full scale. This result demonstrated that a grapevine VaNPR1.1 possesses a similar function to the Arabidopsis NPR1 in the regulation of defense-related genes. Over-expression of VaNPR1.1 in transgenic Arabidopsis plant increased tolerance to salinity, but had no effect on the drought tolerance. 10Immunodominance and clonal selection inspired multiobjective clustering显示文摘The biological immune system is a highly parallel and distributed adaptive system. The information processing abilities of the immune system provide important insights into the field of computation. Based on immunodominance in the biological immune system and the clonal selection mechanism, a novel data mining method, Immune Dominance Clonal Multiobjective Clustering algorithm (IDCMC), is presented. The algorithm divides an individual population into three sub-populations according to three different measurements, and adopts different evolution and selection strategies for each sub-population. The update of each sub-population, however, is not carried out in isolation. The periodic combination operation of the analysis of the three sub-populations represents considerable advantages in its global search ability. The clustering task is a multiobjective optimization problem, which is more robust with respect to the variety of cluster structures of different datasets than a single-objective clustering algorithm. In addition, the new algorithm can determine the number of clusters automatically, which should identify the most promising clustering solutions in the candidate set. 12Homeobox B8 Targets Sterile Alpha Motif Domain-Containing Protein 9 and Drives Glioma Progression显示文摘Gliomas are the most commonly occurring tumors of the central nervous system. Glioblastoma multiforme (GBM) is the most malignant and aggressive brain cancer in adults. Further understanding of the mechanisms underlying the aggressive nature of GBM is urgently needed. Here we identified homeobox B8(HOXB8), a member of the homeobox family, as a crucial contributor to the aggressiveness of GBM. Data mining of publicly accessible RNA sequence datasets and our patient cohorts confirmed a higher expression of HOXB8 in the tumor tissue of GBM patients, and a strong positive correlation between the expression level and pathological grading of tumors and a negative correlation between the expression level and the overall survival rate. We next showed that HOXB8 promotes the proliferation and migration of glioblastoma cells and is crucial for the activation of the PI3K/AKT pathway and expression of epithelial–mesenchymal transition-related genes, possibly through direct binding to the promoter of SAMD9 (Sterile Alpha Motif Domain-Containing Protein 9) and activating its transcription. 16A Survey on Quantum Cryptography显示文摘Quantum cryptography(QC) as an important technology to protect the security of the future network communication, has gained extensive attention from both academia and industry in recent years. This paper provides a concise up-to-date review of QC, including Quantum key distribution(QKD), Quantum authentication, Quantum public-key cryptography(QPKC), and so on. Our aim is to pave a comprehensive and solid starting ground for interested readers. We firstly clarify the definition of QC.
