| 1 | Whole-genome sequencing provides insights into the genetic diversity and domestication of bitter gourd(Momordica spp.)显示文摘Bitter gourd(Momordica charantia)is a popular cultivated vegetable in Asian and African countries.To reveal the characteristics of the genomic structure,evolutionary trajectory,and genetic basis underlying the domestication of bitter gourd,we performed whole-genome sequencing of the cultivar Dali-11 and the wild small-fruited line TR and resequencing of 187 bitter gourd germplasms from 16 countries.The major gene clusters(Bi clusters)for the biosynthesis of cucurbitane triterpenoids,which confer a bitter taste,are highly conserved in cucumber,melon,and watermelon.Comparative analysis among cucurbit genomes revealed that the Bi cluster involved in cucurbitane triterpenoid biosynthesis is absent in bitter gourd.Phylogenetic analysis revealed that the TR group,including 21 bitter gourd germplasms,may belong to a new species or subspecies independent from M.charantia.Furthermore,we found that the remaining 166 M.charantia germplasms are geographically differentiated,and we identified 710,412,and 290 candidate domestication genes in the South Asia,Southeast Asia,and China populations,respectively.This study provides new insights into bitter gourd genetic diversity and domestication and will facilitate the future genomics-enabled improvement of bitter gourd. | Junjie Cui Yan Yang Shaobo Luo Le Wang Rukui Huang Qingfang Wen Xiaoxia Han Nansheng Miao Jiaowen Cheng Ziji Liu Changyuan Zhang Chengcheng Feng Haisheng Zhu Jianwen Su Xinjian Wan Fang Hu Yu Niu Xiaoming Zheng Yulan Yang Dai Shan Zhensheng Dong Weiming He Narinder P.S.Dhillon Kailin Hu | 2020 | Horticulture Research2020,7,1: | 7 |
| 2 | First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. | 3 | First results on ^(76)Ge neutrinoless double beta decay from CDEX-1 experiment显示文摘We report the first results on ^(76)Ge neutrinoless double beta decay from stage one of the China dark-matter experiment(CDEX).A p-type point-contact high-purity germanium detector with a mass of 994 g has been installed to detect neutrinoless double beta decay events, as well as to directly detect dark matter particles. An exposure of 304 kg d has been analyzed over a wide spectral band from 500 keV to 3 MeV. The average event rate obtained was about 0.012 counts per keV per kg per day over the 2.039 MeV energy range. The half-life of ^(76)Ge neutrinoless double beta decay derived based on this result is T^(0ν)1/2>6.4×10^(22) yr(90% C.L.). | 4 | High-risk phenotypes of genetic disease in a Neonatal Intensive Care Unit population显示文摘To the Editor: Genetic diseases contribute to 35% of deaths during the first year of life and are a significant cause of intensive care.[1] A previous study based on the China Neonatal Genomes Project investigated the genetic causes of early infant deaths and found that >25% of deceased neonates with genetic diagnoses can be cured if diagnosed in time.[2] Therefore, it is crucial to target and diagnose neonates with genetic diseases as early as possible. According to our experience, the typical phenotypes, such as special facial features or multiple congenital anomalies (MCAs), indicate a high risk of genetic disease and lead physicians to perform genetic testing in neonates as early as possible. However, in practice, infants without typical phenotypes typically undergo a long and costly diagnostic process before genetic diagnoses are confirmed. Moreover, a recent survey by the American College of Medical Genetics and Genomics (ACMG) and other national professional organizations indicated that there are insufficient numbers of qualified geneticists to fulfil genetic service needs.[3] The ACMG published the general clinical features for genetic testing indications. For example, patients with phenotypes or family history data that strongly implicate a genetic cause may undergo genetic testing.[1] However, the study indicated that many genetic conditions arise de novo or are inherited with no family history.[1] A previous study attempted to apply the non-phenotype-driven panel approach in neonates admitted to the neonate intensive care unit (NICU).[4] However, at present, the diagnostic yield is only 3.45% (1/29).[4] In addition, the economic and ethical issues associated with genomic screening remain challenging. | 9 | Selenium-enriched Cardamine violifolia protects against sepsis-induced intestinal injury by regulating mitochondrial fusion in weaned pigs显示文摘Sepsis is a life-threatening organ dysfunction caused by the dysregulated response of the host to an infection, and treatments are limited. Recently, a novel selenium source, selenium-enriched Cardamine violifolia(SEC) has attracted much attention due to its anti-inflammatory and antioxidant properties, but little is known about its role in the treatment of sepsis. Here, we found that SEC alleviated LPS-induced intestinal damage, as indicated by improved intestinal morphology, and increased disaccharidase activity and tight junction protein expression. Moreover, SEC ameliorated the LPS-induced release of pro-inflammatory cytokines, as indicated by decreased IL-6 level in the plasma and jejunum. Moreover, SEC improved intestinal antioxidant functions by regulating oxidative stress indicators and selenoproteins. In vitro, TNF-α-challenged IPEC-1 cells were examined and showed that selenium-enriched peptides, which are the main functional components extracted from Cardamine violifolia(CSP), increased cell viability, decreased lactate dehydrogenase activity and improved cell barrier function. Mechanistically, SEC ameliorated LPS/TNF-α-induced perturbations in mitochondrial dynamics in the jejunum and IPEC-1 cells. Moreover, CSPmediated cell barrier function is primarily dependent on the mitochondrial fusion protein MFN2 but not MFN1. | 10 | Single molecular insight into steric effect on C-terminal amino acids with various hydrogen bonding sites显示文摘Amino acids are basic units to construct a protein with the assistance of various interactions.During this building process,steric hindrance derived from amino acid side groups or side chains is a factor that could not be ignored.In this contribution,adsorption behaviors of C-terminal amino acid derivatives with amino acid residues fused in 3,4,9,10-perylenetetracarboxylic dianhydride were investigated by scanning tunneling microscopy(STM)and density functional theory(DFT)calculations at various liquid/solid interfaces.STM results at 1-phenyloctane/HOPG interface show that N,N'-3,4,9,10-perylenedicarboximide(GP)and N,N'-methyl-3,4,9,10-perylenedicarboximide(AP)formed linear and herringbone structures,respectively.The driving force could be attributed to different H-bonding sites induced by steric hindrance at side groups.N,N'-Benzyl-3,4,9,10-perylenedicarboximide(PP)generates both linear and herringbone structures because steric hindrance changes the H-bonding sites between PP molecules,whereas N,N'-isopropyl-3,4,9,10-perylenedicarboximide(LP)failed to be imaged because of strong steric hindrance coming from larger side group.To further investigate the impact of steric hindrance,we utilized octanoic acid(OA)as solvent to capture the adsorption details of LP and PP.We found that OA molecules drag PP and LP molecules in a different direction to generate linear structure,impeding the molecular rotation.The structure–solvent relationship shows that the steric hindrance is brought by the large side group,which makes it easier to recognize OA molecules at the interface.These results demonstrate that steric effect plays a significant role in altering interaction sites of the compounds during the adsorption process at the liquid/solid interface. | Yunzhi Xie Chunhua Liu Linxiu Cheng Yulan Fan Huifang Li Wei Liu Lei Zhu Xun Li Ke Deng Qingdao Zeng Shoufa Han | 2022 | Chinese Chemical Letters2022,33,10: | 1 |
