| 2 | Green fluorescent protein (GFP) as a vital marker for studying the interaction of Phytophthora sojae and soybean显示文摘Transgenic Phytophthora sojae strains that produce green fluorescent protein (GFP) were obtained after stable DNA integration using the Hsp70 promoter and the Ham34 terminator of Bremia lactucae. The expression of GFP during different developmental stages of P. sojae was observed using fluorescent microscopy. Based on this reporter system, the histopathologic events caused by the pathogen in soybean leaves, hypocotyls and roots were monitored. Meanwhile, the difference in resistance between different soybean cultivars against P. sojae was analyzed microscopically in roots. The results indicate that GFP can be stably expressed in zoosporangia, zoospores, cysts, hyphae and oospores of P. sojae. Using the GFP marker, the infecting pathogens in leaves, hypocotyls and roots of host could be distinctly visualized. The germ tube length of cysts germinating on the roots of resistant cultivar Nannong 8848 was longer than that on the roots of susceptible cultivar Hefeng 35. | 3 | Derivation and characterization of human embryonic stem cell lines from poor quality embryos显示文摘Poor quality embryos discarded from in vitro fertilization (IVF) laboratories are good sources for deriving human embryonic stem cell (hESC) lines. In this study, 166 poor quality embryos donated from IVF centers on day 3 were cultured in a blastocyst medium for 2 days, and 32 early blastocysts were further cultured in a blastocyst optimum culture medium for additional 2 days so that the inner cell masses (ICMs) could be identified and isolated easily. The ICMs of 17 blastocysts were isolated by a mechanical method, while those of the other 15 blastocysts were isolated by immunosurgery. All isolated ICMs were inoculated onto a feeder layer for subcultivation. The rates of ICM at- tachment, primary ICM colony formation and the efficiency of hESC derivation were similar between the ICMs isolated by the two methods (P>0.05). As a result, four new hESC lines were established. Three cell lines had normal karyotypes and one had an unbalanced Robertsonian translocation. All cell lines showed normal hESC characteristics and had the differentiation ability. In conclusion, we established a stable and effective method for hESC isolation and culture, and it was confirmed that the mechanical isolation was an effective method to isolate ICMs from poor embryos. | 4 | Heavy ion and X-ray irradiation alter the cytoskeleton and cytomechanics of cortical neurons显示文摘Heavy ion beams with high linear energy transfer exhibit more beneficial physical and biological performance than conventional X-rays, thus improving the potential of this type of radiotherapy in the treatment of cancer. However, these two radiotherapy modalities both cause inevitable brain injury. The objective of this study was to evaluate the effects of heavy ion and X-ray irradiation on the cytoskeleton and cytomechanical properties of rat cortical neurons, as well as to determine the potential mechanism of neuronal injury after irradiation. Cortical neurons from 30 new-born mice were irradiated with heavy ion beams at a single dose of 2 Gy and X-rays at a single dose of 4 Gy; subsequent evaluation of their effects were carried out at 24 hours after irradiation. An immunofluorescence assay showed that after irradiation with both the heavy ion beam and X-rays, the number of primary neurons was significantly decreased, and there was evidence of apoptosis. Radiation-induced neuronal injury was more apparent after X-irradiation. Under atomic force microscopy, the neuronal membrane appeared rough and neuronal rigidity had increased. These cell changes were more apparent following exposure to X-rays. Our findings indicated that damage caused by heavy ion and X-ray irradiation resulted in the structural distortion and rearrangement of the cytoskeleton, and affected the cytomechanical properties of the cortical neurons. | 5 | Multi-fluid Eulerian simulation of binary particles mixing and gas–solids contacting in high solids-flux downer reactor equipped with a lateral particle feeding nozzle显示文摘The performance of binary particles mixing and gas-solids contacting,which is considered qualitatively to have a significant influence on the heat transfer in internal heated circulating fluidized beds,is carefully investigated by means of a numerical approach in the newly developed high solids-flux downer lignite pyrolyzer(φ0.1 m×6.5 m).Since binary particles are used in this system,a reasonably validated 3 D,transient,multi-fluid model,in which three heat transfer modes relating to the convection,conduction and radiation are considered,is adopted to simulate the flow behavior,temperature profiles as well as volatile contents.The simulation results showed that the solids stream impinges the left wall surface initially and turns towards the right wall in the further downward direction and then shrinks during this process resulting in that the solids concentrate a little more at the central region.In the further downward section of the downer,the particle flow disperses near the right wall and develops uniformly.Meanwhile,the coal phase is slowly heated in the downer and it is found that most of the heat absorbed by the coal is from the convection heat transfer mode.To explore the heat transfer mechanism more quantitatively,two indexes(mixing index and contacting index)are proposed,and it is found that the mixing index initially increased fast and later remained at a relatively flat state.For the contact index,it shows a trend with a first rising and then falling,finally rising continuously.Also,it is found that the convection heat transfer is closely correlated to the contacting status of gas-coal which indicates that the improving of the gas-coal contacting efficiency should be an effective way to strengthen the coal particle heating process. | Qiang Zheng Jingxuan Yang Wenhao Lian Baoping Zhang Xueer Pan Zhonglin Zhang Xiaogang Hao Guoqing Guan | 2021 | Chinese Journal of Chemical Engineering2021,34,7: | 2 |

| 9 | Epidemiological characteristics and clinical manifestations of pediatric patients with COVID-19 in China: A multicenter retrospective study显示文摘Importance:The Coronavirus disease 2019(COVID-19)global pandemic poses a considerable challenge for pediatricians.Objective:This study aimed to identify the epidemiological characteristics and clinical features of pediatric patients with COVID-19 in China.Methods:This multicenter retrospective study included pediatric patients from 46 hospitals in China,covering 12 provinces and two municipalities.Epidemiological,demographic,clinical,laboratory,treatment,and outcome data were analyzed.Results:In total,211 pediatric patients with COVID-19 were included in this study.The median age was 7.0 years(range:22 days to 18 years).Approximately 16.3%of the patients exhibited asymptomatic infections,23.0%had upper respiratory tract infections,and 60.7%had pneumonia,including two with severe pneumonia and one with critical illness.Approximately 78.7%of the pediatric patients occurred in familial clusters.The most three common symptoms or signs at onset in children with COVID-19 were fever(54.5%),cough(49.3%),and pharyngeal congestion(20.8%).Only 17.6%of the patients presented with decreased lymphocyte count,whereas 13.6%had increased lymphocyte count.Among the patients with pneumonia who exhibited abnormal chest computed tomography findings,18.2%(23/127)of the patients had no other symptoms.Generally,the chest radiographs showed abnormalities that affected both lungs(49.6%);ground-glass opacity(47.2%)was the most common manifestation.The cure and improvement rates were 86.7%(183/211)and 13.3%(28/211),respectively.Only one patient with an underlying condition received invasive mechanical ventilation;none of the patients died.Interpretation:Similar to adults,children of all age groups are susceptible to COVID-19.Fortunately,most pediatric patients have mild symptoms or remain asymptomatic,despite the high incidence of pneumonia.Decreased proportions of white blood cells and lymphocytes are less frequent in children than in adults. | Yan Bai Liwei Gao Xianfeng Wang Lili Zhong Jingfeng Li Shenggang Ding Yuejie Zheng Jun Liu Yuxia Cui Lina Wang Mingfeng Han Jun Liu Xinping Jiang Min Jiang Junhua Li Ning Chen Yunxiao Shang Hourong Zhou Yi Xu Gen Lu Xing Chen Jizhi Xu Qihong Fan Yu Tang Jiang Wu Cen Li Xiaoxiang Yang Chunxi Chen Yonghong Yang Gary Wing-Kin Wong Adong Shen Tianyou Wang Runming Jin Baoping Xu Kunling Shen | 2021 | Pediatric Investigation2021,5,3: | 0 |
