1Applications of magnetic nanoparticles in surface-enhanced Raman scattering(SERS)detection of environmental pollutants显示文摘Environmental pollution, a major problem worldwide, poses considerable threat to human health and ecological environment. Efficient and reliable detection technologies, which focus on the appearance of emerging environmental and trace pollutants, are urgently needed. Surface-enhanced Raman scattering(SERS) has become an attractive analytical tool for sensing trace targets in environmental field because of its inherent molecular fingerprint specificity and high sensitivity. In this review, we focused on the recent developments in the integration of magnetic nanoparticles(MNPs) with SERS for facilitating sensitive detection of environmental pollutants. An overview and classification of different types of MNPs for SERS detection were initially provided, enabling us to categorize the huge amount of literature that was available in the interdisciplinary research field of MNPs based SERS technology. Then, the basic working principles and applications of MNPs in SERS detection were presented. Subsequently, the detection technologies integrating MNPs with SERS that eventually were used for the detection of various environmental pollutions were reviewed. Finally, the advantages of MNP-basedSERS detection technology for environmental pollutants were concluded, and the current challenges and future outlook of this technology in practical applications were highlighted. 2The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 3Comparison of selected inflammation-based prognostic markers in relapsed or refractory metastatic colorectal cancer patients显示文摘AIM: To investigate the impact of systemic inflammationbased prognostic markers on overall survival in relapsed/refractory metastatic colorectal cancer(m CRC) patients. METHODS: To investigate prognostic markers in m CRC patients,this study was performed with patients who have experienced relapsed/refractory m CRC with standard chemotherapy or were inapplicable to conventional treatment modality because of poor performance status,age,or comorbidity. We reviewed the medical records of 177 m CRC patients managed with Korean Medicine(KM) treatment modality using an anticancer agent of Rhus verniciflua Stokes extract from June 2006 to April 2013. The clinicopathologic characteristics,laboratory test,the systemic inflammation markers including the modified Glasgow prognostic score(m GPS),neutrophil lymphocyte ratio(NLR),platelet lymphocyte ratio(PLR),lymphocyte monocyte ratio(LMR),and prognostic nutritional index(PNI) were analyzed. The overall survival of patients was calculated with the Kaplan-Meier method and the statistical significance was compared using with the log-rank test. To compare the impact of systemic inflammation based markers,the hazard ratio(HR) of m GPS,NLR,PLR,LMR,and PNI for overall survival were evaluated with the Cox proportional hazards regression.RESULTS: The majority of m CRC patients had relapsed/refractory to standard chemotherapy; 128 patients(72.3%) had undergone more than second line chemotherapy,and the median time from diagnosis of m CRC to initiation of KM was 9.4 mo. The median overall survival of enrolled patients was 8.3 mo. On univariate analyses,the inflammation markers of higher m GPS(P < 0.001),NLR ≥ 5(P < 0.001),PLR > 300(P = 0.004),LMR ≤ 3.4(P < 0.001),and PNI ≤ 45.3(P = 0.001) were significantly associated with decreased survival time. On stepwise multivariate proportional hazards model,m GPS at 2 vs 0(HR = 3.212,95%CI: 1.437-7.716,P = 0.004),and LMR ≤ 3.4(HR = 1.658,95%CI: 1.092-2.518,P = 0.018) as independent predictors associated with poor overall survival along with carbohydrate antigen 19-9(HR = 1.482,95%CI: 1.007-2.182,P = 0.046),AST ≥ 40(HR = 2.377,95%CI: 1.359-4.155,P = 0.002),and the treatment duration for KM less than 2.9 mo(HR = 1.718,95%CI: 1.160-2.543,P = 0.007).CONCLUSION: These results indicate that the inflammatory markers,m GPS and LMR are independent prognostic factors for predicting overall survival in relapsed/refractory m CRC patients.Anna Song Wankyu Eo Sookyung Lee 2015World Journal of Gastroenterology2015,21,43:8
4The deubiquitinase OTUB1 augments NF-κB-dependent immune responses in dendritic cells in infection and inflammation by stabilizing UBC13显示文摘Dendritic cells(DCs)are indispensable for defense against pathogens but may also contribute to immunopathology.Activation of DCs upon the sensing of pathogens by Toll-like receptors(TLRs)is largely mediated by pattern recognition receptor/nuclear factor-κB(NF-κB)signaling and depends on the appropriate ubiquitination of the respective signaling molecules.However,the ubiquitinating and deubiquitinating enzymes involved and their interactions are only incompletely understood.Here,we reveal that the deubiquitinase OTU domain,ubiquitin aldehyde binding 1(OTUB1)is upregulated in DCs upon murine Toxoplasma gondii infection and lipopolysaccharide challenge.Stimulation of DCs with the TLR11/12 ligand T.gondii profilin and the TLR4 ligand lipopolysaccharide induced an increase in NF-κB activation in OTUB1-competent cells,resulting in elevated interleukin-6(IL-6),IL-12,and tumor necrosis factor(TNF)production,which was also observed upon the specific stimulation of TLR2,TLR3,TLR7,and TLR9.Mechanistically,OTUB1 promoted NF-κB activity in DCs by K48-linked deubiquitination and stabilization of the E2-conjugating enzyme UBC13,resulting in increased K63-linked ubiquitination of IRAK1(IL-1 receptor-associated kinase 1)and TRAF6(TNF receptor-associated factor 6).Consequently,DC-specific deletion of OTUB1 impaired the production of cytokines,in particular IL-12,by DCs over the first 2 days of T.gondii infection,resulting in the diminished production of protective interferon-γ(IFN-γ)by natural killer cells,impaired control of parasite replication,and,finally,death from chronic T.encephalitis,all of which could be prevented by low-dose IL-12 treatment in the first 3 days of infection.In contrast,impaired OTUB1-deficient DC activation and cytokine production by OTUB1-deficient DCs protected mice from lipopolysaccharide-induced immunopathology.Collectively,these findings identify OTUB1 as a potent novel regulator of DCs during infectious and inflammatory diseases.Floriana Mulas Xu Wang Shanshan Song Gopala Nishanth Wenjing Yi Anna Brunn Pia-Katharina Larsen Berend Isermann Ulrich Kalinke Antonio Barragan Michael Naumann Martina Deckert Dirk Schlüter 2021Cellular & Molecular Immunology2021,18,6:4
5All-trans retinoic acid(ATRA)inhibits insufficient radiofrequency ablation(IRFA)-induced enrichment of tumor-initiating cells in hepatocellular carcinoma显示文摘Objective:Local recurrence of hepatocellular carcinoma(HCC)after radiofrequency ablation(RFA)treatment remains a serious problem.Tumor-initiating cells(TICs)are thought to be responsible for tumor relapse.Here,we investigated the effect of the TIC differentiation inducer,all-trans retinoic acid(ATRA),on RFA and explored the potential molecular mechanisms.Methods:The proportions of CD133+and epithelial cell adhesion molecule(Ep CAM);TICs in recurrent HCC after RFA and primary HCC were first determined in clinic.Then,the effect of heat intervention or insufficient RFA(IRFA)on the malignant potential of HCC cells,including cell migration,sphere formation ability,tumor growth,the proportion of CD133+and Ep CAM+TICs and expression of stem cell-related genes,was evaluated in vitro and in vivo.Finally,the effect of ATRA on the tumor growth and the proportion of TICs was evaluated.Results:In clinical data,a higher proportion of CD133+and Ep CAM+TICs was found in recurrent tumors than in primary tumors.In vitro heat intervention promoted the cell migration and sphere formation ability.Additionally,it increased the proportion of CD133+and Ep CAM+TICs and the expression of stem cell-related genes.In addition,after IRFA the residual tumors in xenografts grew faster and had more TICs than untreated tumors.ATRA remarkably inhibited residual tumor growth after IRFA by elimination of TICs though the PI3 K/AKT pathway.Combination treatment with ATRA resulted in longer survival outcomes in mouse xenografts than RFA alone.Conclusions:ATRA,as a TIC differentiation inducer,could help to improve the effect of RFA treatment,which was partially attributed to its effect against TICs.The data indicated its potential as an alternative drug in the development of better therapeutic strategies for use in combination with RFA.Song Wang Jingtao Liu Hao Wu Anna Jiang Kun Zhao Kun Yan Wei Wu Haibo Han Yanhua Zhang Wei Yang 2021Chinese Journal of Cancer Research2021,33,6:2
6Structural variation and parallel evolution of apomixis in citrus during domestication and diversification显示文摘Apomixis,or asexual seed formation,is prevalent in Citrinae via a mechanism termed nucellar or adventitious embryony.Here,multiple embryos of a maternal genotype form directly from nucellar cells in the ovule and can outcompete the developing zygotic embryo as they utilize the sexually derived endosperm for growth.Whilst nucellar embryony enables the propagation of clonal plants of maternal genetic constitution,it is also a barrier to effective breeding through hybridization.To address the genetics and evolution of apomixis in Citrinae,a chromosome-level genome of the Hongkong kumquat(Fortunella hindsii)was assembled following a genome-wide variation map including structural variants(SVs)based on 234 Citrinae accessions.This map revealed that hybrid citrus cultivars shelter genome-wide deleterious mutations and SVs into heterozygous states free from recessive selection,which may explain the capability of nucellar embryony in most cultivars during Citrinae diversification.Analyses revealed that parallel evolution may explain the repeated origin of apomixis in different genera of Citrinae.Within Fortunella,we found that apomixis of some varieties originated via introgression.In apomictic Fortunella,the locus associated with apomixis contains the FhRWP gene,encoding an RWP-RK domain-containing protein previously shown to be required for nucellar embryogenesis in Citrus.We found the heterozygous SV in the FhRWP and CitRWP promoters from apomictic Citrus and Fortunella,due to either two or three miniature inverted transposon element(MITE)insertions.A transcription factor,FhARID,encoding an AT-rich interaction domain-containing protein binds to the MITEs in the promoter of apomictic varieties,which facilitates induction of nucellar embryogenesis.This study provides evolutionary genomic and molecular insights into apomixis in Citrinae and has potential ramifications for citrus breeding.Nan Wang Xietian Song Junli Ye Siqi Zhang Zhen Cao Chenqiao Zhu Jianbing Hu Yin Zhou Yue Huang Shuo Cao Zhongjie Liu Xiaomeng Wu Lijun Chai Wenwu Guo Qiang Xu Brandon S.Gaut Anna M.G.Koltunow Yongfeng Zhou Xiuxin Deng 2022National Science Review2022,9,10:1

20Dynamic carbon surface chemistry: Revealing the role of carbon in electrolytic water oxidation显示文摘Carbon materials have been widely used as electrodes, but the mechanistic roles are still not clear due to the complexity of the carbon surface chemistry. Herein we clarify that intrinsic material properties of carbon have to be activated by extrinsic factors. Pure carbon has no catalytic activity when used as electrode for electrocatalytic water oxidation. The evolution of oxygen functional groups on the carbon surface with increasing potential and the subsequent formation of real active sites with iron impurities from the electrolyte have been confirmed. These in-situ formed active sites protect the carbon from deep oxidation.
