| 1 | Chromosome-Level Reference Genome and Population Genomic Analysis Provide Insights into the Evolution and Improvement of Domesticated Mulberry (Morns alba)显示文摘Mulberry(Morns spp.)is the sole plant consumed by the domesticated silkworm.However,the genome of domesticated mulberry has not yet been sequenced,and the ploidy level of this species remains unclear.Here,we report a high-quality,chromosome-level domesticated mulberry(Morus alba)genome.Analysis of genomic data and karyotype analyses confirmed that M.alba is a diploid with 28 chromosomes(2/7=2x=28).Population genomic analysis based on resequencing of 134 mulberry accessions classified domesticated mulberry into three geographical groups,namely,Taihu Basin of southeastern China(Hu mulberry),northern and southwestern China,and Japan.Hu mulberry had the lowest nucleotide diversity among these accessions and demonstrated obvious signatures of selection associated with environmental adaptation.Further phylogenetic analysis supports a previous proposal that multiple domesticated mulberry accessions previously classified as different species actually belong to one species.This study expands our understanding of genome evolution of the genus Morus and population structure of domesticated mulberry,which would facilitate mulberry breeding and improvement. | Feng Jiao Rongsong Luo Xuelei Dai Hui Liu Gang Yu Shuhua Han Xin Lu Chao Su Qi Chen Qinxia Song Caiting Meng Fanghong Li Hongmei Sun Rui Zhang Tian Hui Yonghua Qian Aichun Zhao Yu Jiang | 2020 | Molecular Plant2020,13,7: | 10 |
| 2 | Different contribution of SBP and DBP variability to vascular events in patients with stroke显示文摘Background High blood pressure variability(BPV)is a novel risk factor for cardiovascular disease.However,the heterogeneity of systolic blood pressure variability(SBPV)and diastolic blood pressure variability(DBPV)for different vascular events remains unclear.This study aims to investigate whether SBPV or DBPV has different contribution to vascular events in patients with acute ischaemic stroke(IS)or transient ischaemic attack(TIA).Methods Data from the BOSS(blood pressure and clinical outcome in TIA or IS)study were examined for vascular events at 3-month and 1-year follow-up.BPV was defined as the SD and coefficient of variation(CV)of day-to day measurements within 3 months after IS/TIA.Vascular events include cardiovascular events(myocardial infarction,unstable angina,cardiac death and congestive heart failure)and cerebrovascular events(ischaemic or haemorrhagic stroke).Logistic regression model was used to test the associations between BPV and vascular events.results Of 2325 patients with IS or TIA,103(4.43%)experienced a recurrent stroke and 64(2.75%)had cardiovascular events within 3 months.Day-to day SBPV was only associated with stroke recurrence(BPVSD:OR,1.72,95%CI 1.09 to 2.71;BPVCV:1.86,95%CI 1.19 to 2.92),but not cardiovascular events(BPVSD:1.67,95%CI 0.94 to 2.94;BPVCV:1.51,95%CI 0.86 to 2.64).However,DBPV seems to be related to both stroke(BPVSD:1.60,95%CI 1.02 to 2.49;BPVCV:1.53,95%CI 0.99 to 2.37)and cardiovascular events(BPVSD:2.48,95%CI 1.37 to 4.48;BPVCV:1.92,95%CI 1.09 to 3.36).Similar results were found at 1 year.Conclusions For patients with IS/TIA,stroke recurrence was associated with both SBPV and DBPV;however,cardiovascular events seem to be only related to DBPV. | Liye Dai Aichun Cheng Xiwa Hao Jie Xu Yingting Zuo Anxin Wang Xia Meng Hao Li Yilong Wang Xingquan Zhao Yongjun Wang | 2020 | Stroke & Vascular Neurology2020,5,2: | 6 |
| 3 | Environmental archaeology on Longshan Culture (4500-4000 aBP) at Yuhuicun Site in Bengbu, Anhui Province显示文摘Based on the samples collected from the west wall of the ABYT2004 at Yuhuicun Site in Bengbu,Anhui Province,the multi-environmental substitute indexes such as the grain-size,Rb/Sr,zircon crystal morphology and magnetic susceptibility were analyzed.The age of archaeological strata was determined by AMS14C dating.According to the results,combined with the information of the ancient human activities and the cultural heritage data obtained from the Longshan cultural archaeological strata,it can be concluded that(1) Before 4500 aBP,the climate was humid and water level was high in the Huaihe River and its tributaries,the drainage basin and its surrounding areas were even flooded,so the site area was submerged and there were no human activities around the site.(2) About 4500 aBP,the climate began to become drier and water level of the Huaihe River and its tributaries began to decline,thus floodplain gradually appeared,Yuhuicun Site and Longshan Culture began to appear.(3) Around 4100 aBP,the climate was humid and rainfall was abundant,which resulted in frequent flood disasters in the whole Huaihe River Basin.(4) After 4100 aBP,the climate gradually transited to be cold and dry,agricultural production was constrained to reduce the food source,Yuhuicun Site was almost abandoned.(5) The environmental information in sedimentary strata and historical records based on ancient books agree with each other.In addition,Yuhuicun Site was related to Dayu Zhishui(Yu the Great in taming the floods) in ancient books. | ZHANG Guangsheng ZHU Cheng WANG Jihuai ZHU Guangyao MA Chunmei ZHENG Chaogui ZHAO Lanhui LI Zhongxuan LI Lan JIN Aichun | 2010 | Journal of Geographical Sciences2010,20,3: | 3 |
| 4 | Chromosome-level Genomes Reveal the Genetic Basis of Descending Dysploidy and Sex Determination in Morus Plants显示文摘Multiple plant lineages have independently evolved sex chromosomes and variable karyotypes to maintain their sessile lifestyles through constant biological innovation.Morus notabilis,a dioecious mulberry species,has the fewest chromosomes among Morus spp.,but the genetic basis of sex determination and karyotype evolution in this species has not been identified.In this study,three high-quality genome assemblies were generated for Morus spp.[including dioecious M.notabilis(male and female)and Morus yunnanensis(female)]with genome sizes of 301–329 Mb and were grouped into six pseudochromosomes.Using a combination of genomic approaches,we found that the putative ancestral karyotype of Morus species was close to 14 protochromosomes,and that several chromosome fusion events resulted in descending dysploidy(2n=2x=12).We also characterized a~6.2-Mb sex-determining region on chromosome 3.Four potential male-specific genes,a partially duplicated DNA helicase gene(named MSDH)and three Ty3_Gypsy long terminal repeat retrotransposons(named MSTG1/2/3),were identified in the Y-linked area and considered to be strong candidate genes for sex determination or differentiation.Population genomic analysis showed that Guangdong accessions in China were genetically similar to Japanese accessions of mulberry.In addition,genomic areas containing selective sweeps that distinguish domesticated mulberry from wild populations in terms of flowering and disease resistance were identified.Our study provides an important genetic resource for sex identification research and molecular breeding in mulberry. | Zhongqiang Xia Xuelei Dai Wei Fan Changying Liu Meirong Zhang Peipei Bian Yuping Zhou Liang Li Baozhong Zhu Shuman Liu Zhengang Li Xiling Wang Maode Yu Zhonghuai Xiang Yu Jiang Aichun Zhao | 2022 | Genomics, Proteomics & Bioinformatics2022,20,6: | 1 |
| 5 | QTL mapping of economically important traits in Silkworm (Bombyx mori)显示文摘A backcrossed population (BC1) was derived from a cross between C100 and Dazao. AFLP technique was employed for mapping the QTLs. The QTLs for the whole cocoon weight, cocoon shell weight, ratio of cocoon shell, weight of pupae etc. were analyzed and 11 QTLs were detected based on the constructed linkage map. Two QTLs for whole cocoon weight were local-ized on linkage group 6 and 19; three QTLs for cocoon shell weight were localized on linkage group 3, 14 and 19; three QTLs for ratio of cocoon shell were localized on the linkage group 2, 11 and 15, and three QTLs for the weight of pupae were localized on linkage 2, 14 and 19. | 7 | Quantitative detection of Cymbidium mosaic virus by real time PCR显示文摘The technique of SYBR Green-based quantitative real-time reverse transcription polymerase chain reaction(real-time RT-PCR)was applied to quantitative detect a 764 bp nucleotide sequence containing total coat protein(cp)gene of Cymbidium mosaic virus(CyMV).The plasmid containing the target sequence was constructed to prepare the standard curve and detect the sensitivity.The standard curve was drawn based on the linear relationship between the logarithm(base 10)of the quantity of target sequence and cycle threshold[C(T)].While the concentration of plasmid DNA falling within the range of 2.6×10^(7)to 2.6×10^(2)copies per tube established a regression equation,y=-0.3583x+10.32,and related coefficient:r^(2)=0.995.The real-time RT-PCR assay for CyMV had a minimum detectable quantity of two copies per tube.The naturally infected samples of Phalaenopsis sp.and the artificially inoculated samples of Arachnis sp.with trace CyMV were quantitatively detected using this method.CyMVin the positive samples of Phalaenopsis sp.and Arachnis sp.was confirmed by DNA sequencing and cp gene homeology blast.The results showed that CyMV extracted from the leaves of orchid in Hangzhou,Zhejiang Province,China,could be derived from Kunming city(KM),Yunnan Province,China.This method characterized by high sensitivity,specificity,and precision is suitable for early diagnosis and quantitative detection of CyMV. | Aichun LIU Yun ZHAO Songlin RUAN Guozheng SHEN | 2009 | Frontiers in Biology2009,4,3: | 1 |
| 8 | An efficient and safe strategy for germ cell-specific automatic excision of foreign DNA in F1 hybrid transgenic silkworms显示文摘The safety of transgenic technology is a major obstacle in the popularization and use of transgenic silkworms and their products.In sericulture,only the first filial generation(F1)hybrid eggs produced by cross-breeding Japanese and Chinese original strains are usually used for the large-scale breeding of silkworms,but this may result in uncontrolled transgene dispersal during the popularization and application of the F1 hybrid transgenic eggs.To address this issue,we developed a safe and efficient strategy using the GAL4/Upstream activating sequence(UAS)system,the FLP/flippase recognition target(FRT)system,and the gonad-specific expression gene promoters(RSHP1p and Nanosp)for the germ cell-specific automatic excision of foreign DNA in the F1 hybrid transgenic silkworms.We established 2 types of activator strains,R1p::GAL4-Gr and Nsp::GAL4-Gr,containing the testis-specific GAL4 gene expression cassettes driven by RSHP1p or Nanosp,respectively,and 1 type of effector strain,UAS::FLP-Rg,containing the UAS-linked FLP gene expression cassette.The FLP recombinase-mediated sperm-specific complete excision of FRT-flanked target DNA in the F1 double-transgenic silkworms resulting from the hybridization of R1p::GAL4-Gr and UAS::FLP-Rg was 100%,whereas the complete excision efficiency resulting from the hybridization of Nsp::GAL4-Gr and UAS::FLP-Rg ranged from 13.73%to 80.3%.Additionally,we identified a gene,sw11114,that is expressed in both testis and ovary of Bombyx mori,and can be used to establish novel gonad-specific expression systems in transgenic silkworms.This strategy has the potential to fundamentally solve the safety issue in the production of F1 transgenic silkworm eggs and provides an important reference for the safety of transgenic technology in other insect species. | Dingpei Long Rongpeng Liu Yang Huang Anyao Fu Yuli Zhang Zhanzhang Hao Qiang Li Hanfu Xu Zhonghuai Xiang Aichun Zhao | 2024 | Insect Science2024,31,1: | 0 |
