1Prevalence and evolution of drug resistance HIV-1 variants in Henan, China显示文摘To understand the prevalence and evolution of drug resistant HIV strains in Henan China after the implementation of free antiretroviral therapy for AIDS patients. 45 drug nave AIDS patients, 118 AIDS patients who received three months antiretroviral therapy and 124 AIDS patients who received six months antiretroviral treatment were recruited in the southern part of Henan province. Information on general condition, antiretroviral medicines, adherence and clinical syndromes were collected by face to face interview. Meanwhile, 14ml EDTA anticoagulant blood was drawn. CD4/CD8 T cell count, viral load and genotypic drug resistance were tested. The rates of clinical improvement were 55.1% and 50.8% respectively three months and six months after antiretroviral therapy. The mean CD4 cell count after antiretroviral therapy was significantly higher than in drug nave patients. The prevalence rate of drug resistant HIV strains were 13. 9%, 45.4% and 62.7% in drug nave patients, three month treatment patients and six month treatment patients, respectively. The number of resistance mutation codons and the frequency of mutations increased significantly with continued antiretroviral therapy. The mutation sites were primarily at the 103, 106 and 215 codons in the three-month treatment group and they increased to 15 codon mutations in the six-month treatment group. From this result, the evolution of drug resistant strains was inferred to begin with the high level NNRTI resistant strain, and then develop low level resistant strains to NRTIs. The HIV strains with high level resistance to NVP and low level resistance to AZT and DDI were highly prevalent because of the AZT+DDI+NVP combination therapy. These HIV strains were also cross resistant to DLV, EFV, DDC and D4T. Poor adherence to therapy was believed to be the main reason for the emergence and prevalence of drug resistant HIV strains. The prevalence of drug resistant HIV strains was increased with the continu- ation of antiretroviral therapy in the southern part of Henan province. 2The Epitope Study on the SARS-CoV Nucleocapsid Protein显示文摘The nucleocapsid protein (N protein) has been found to be an antigenic protein in a number of coronaviruses. Whether the N protein in severe acute respiratory syndrome-associated coronavirus (SARS-CoV) is antigenic remains to be elucidated. Using Western blot and Enzyme-linked Immunosorbent Assay (ELISA),the recombinant N proteins and the synthesized peptides derived from the N protein were screened in sera from SARS patients. All patient sera in this study displayed strong positive immunoreactivities against the recombinant N proteins,whereas normal sera gave negative immunoresponses to these proteins, indicating that the N protein of SARS-CoV is an antigenic protein. Furthermore, the epitope sites in the N protein were determined by competition experiments, in which the recombinant proteins or the synthesized peptides competed against the SARS-CoV proteins to bind to the antibodies raised in SARS sera. One epitope site located at the C-terminus was confirmed as the most antigenic region in this protein. A detailed screening of peptide with ELISA demonstrated that the amino sequence from Codons 371 to 407 was the epitope site at the C-terminus of the N protein. 4Whole genome sequencing of 10K patients with acute ischaemic stroke or transient ischaemic attack: design, methods and baseline patient characteristics显示文摘Background and purpose Stroke is the second leading cause of death worldwide and the leading cause of mortality and long-term disability in China,but its underlying risk genes and pathways are far from being comprehensively understood.We here describe the design and methods of whole genome sequencing(WGS)for 10914 patients with acute ischaemic stroke or transient ischaemic attack from the Third China National Stroke Registry(CNSR-III).Methods Baseline clinical characteristics of the included patients in this study were reported.DNA was extracted from white blood cells of participants.Libraries are constructed using qualified DNA,and WGS is conducted on BGISEQ-500 platform.The average depth is intended to be greater than 30×for each subject.Afterwards,Sentieon software is applied to process the sequencing data under the Genome Analysis Toolkit best practice guidance to call genotypes of single nucleotide variants(SNVs)and insertion-deletions.For each included subject,21 fingerprint SNVs are genotyped by MassARRAY assays to verify that DNA sample and sequencing data originate from the same individual.The copy number variations and structural variations are also called for each patient.All of the genetic variants are annotated and predicted by bioinformatics software or by reviewing public databases.Results The average age of the included 10914 patients was 62.2±11.3 years,and 31.4%patients were women.Most of the baseline clinical characteristics of the 10914 and the excluded patients were balanced.Conclusions The WGS data together with abundant clinical and imaging data of CNSR-III could provide opportunity to elucidate the molecular mechanisms and discover novel therapeutic targets for stroke.Si Cheng Zhe Xu Yang Liu Jinxi Lin Yong Jiang Yilong Wang Xia Meng Anxin Wang Xinying Huang Zhimin Wang Guohua Chen Songdi Wu Zhengchang Jia Yongming Chen Xuerong Qiu Jun Wu Binbin Song Weizhong Ji Zhongping An Wenjun Xue Lili Zhao Yu Geng Hongyan Li Hao Li Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:8
5Advances in crop phenotyping and multi-environment trials显示文摘Efficient evaluation of crop phenotypes is a prerequisite for breeding, cultivar adoption, genomics and phenomics study. Plant genotyping is developing rapidly through the use of high-throughput sequencing techniques,while plant phenotyping has lagged far behind and it has become the rate-limiting factor in genetics, large-scale breeding and development of new cultivars. In this paper,we consider crop phenotyping technology under three categories. The first is high-throughput phenotyping techniques in controlled environments such as greenhouses or specifically designed platforms. The second is a phenotypic strengthening test in semi-controlled environments, especially for traits that are difficult to be tested in multi-environment trials(MET), such as lodging, drought and disease resistance. The third is MET in uncontrolled environments, in which crop plants are managed according to farmer's cultural practices. 6The Chinese Hα Solar Explorer(CHASE) mission: An overview显示文摘The Chinese Hα Solar Explorer(CHASE), dubbed “Xihe”—Goddess of the Sun, was launched on October 14, 2021 as the first solar space mission of China National Space Administration(CNSA). The CHASE mission is designed to test a newly developed satellite platform and to acquire the spectroscopic observations in the Hα waveband. The Hα Imaging Spectrograph(HIS)is the scientific payload of the CHASE satellite. It consists of two observational modes: raster scanning mode and continuum imaging mode. The raster scanning mode obtains full-Sun or region-of-interest spectral images from 6559.7 to 6565.9 ? and from 6567.8 to 6570.6 ? with 0.024 ? pixel spectral resolution and 1 min temporal resolution. The continuum imaging mode obtains photospheric images in continuum around 6689 ? with the full width at half maximum of 13.4 ?. The CHASE mission will advance our understanding of the dynamics of solar activity in the photosphere and chromosphere. 9Experimental and numerical investigations of cavitation evolution in a high-speed centrifugal pump with inducer显示文摘Along with the anti-cavitation performance,the high speed and the high power density,are the main trends in the development of centrifugal pumps.At present,the most effective method is to install an inducer in front of the impeller.However,the tip leakage of the inducer results in the vortex cavitation at the blade leading edge of the inducer,and the cavitating flow inside the inducer seriously interferes with the hydraulic behavior of the inducer as well as the impeller with the development of the cavitation,thus to badly affect the operational reliability of the high-speed centrifugal pump.In the present paper,the cavitating flow in a high-speed centrifugal pump with an inducer is investigated by numerical simulations and visual experiments for different cavitation numbers.A typical evolution process of the cavitation is shown,including the inception,the development and the deterioration.A general description of the pump head-drop phenomenon is made through the study of the local and global flow fields,and the relationship between the vapor distribution and the static pressure distribution along the inducer is determined to describe the evolution of the cavitation.This paper intends to provide the foundation for studying the overall cavitation state of a high-speed centrifugal pump,and designing the inducer with a better cavitation resistance.Yu-ying Huan Yao-yao Liu Xiao-jun Li Zu-chao Zhu Jing-tian Qu Lin Zhe An-da Han 2021Journal of Hydrodynamics2021,33,1:6
10Altered microRNA profiles of testicular biopsies from patients with nonobstructive azoospermia显示文摘Many studies have shown that microRNAs(miRNAs)play vital roles during the spermatogenesis.However,little is known about the altered miRNA profiles of testicular tissues in nonobstructive azoospermia(NOA).Using microarray technology,the miRNA expression profiles of testicular biopsies from patients with NOA and of normal testicular tissues were determined.Bioinformatics analyses were conducted to predict the enriched biological processes and functions of identified miRNAs.The microarray data were validated by quantitative reverse transcriptase polymerase chain reaction(qRT-PCR),the results of which were then validated with a larger sample size.Correlations between the miRNA expression levels and clinical characteristics were analyzed.Receiver operating characteristic(ROC)curve analysis was used to evaluate the diagnostic ability of miRNAs for azoospermia.Hierarchical clustering showed that 129 miRNAs were significantly differentially expressed between the NOA and control groups.Bioinformatics analysis indicated that the differentially expressed miRNAs were involved in spermatogenesis,cell cycle,and mitotic prometaphase.In the subsequent qRT-PCR assays,the selected miRNA expression levels were consistent with the microarray results,and similar validated results were obtained with a larger sample size.Some clinical characteristics were significantly associated with the expression of certain miRNAs.In particular,we identified a combination of two miRNAs(miR-10b-3p and miR-34b-5p)that could serve as a predictive biomarker of azoospermia.This study provides altered miRNA profiles of testicular biopsies from NOA patients and examines the roles of miRNAs in spermatogenesis.These profiles may be useful for predicting and diagnosing the presence of testicular sperm in individuals with azoospermia.Hai-Tao Zhang Zhe Zhang Kai Hong Wen-Hao Tang De-Feng Liu Jia-Ming Mao Yu-Zhuo Yang Hao-Cheng Lin Hui Jiang 2020Asian Journal of Andrology2020,22,1:4
11Effect of Inlet Guide Vanes on the Performance of Small Axial Flow Fan显示文摘Effects of the inlet guide vanes on the static characteristics, aerodynamic noise and internal flow characteristics of a small axial flow fan are studied in this work. The inlet guide vanes with different outlet angle are designed,which are mounted on the casing and located at the upstream of the impeller of the prototype fan. Both steady and unsteady flow simulations are performed. The steady flow is simulated by the calculations of Navier-Stokes equations coupled with RNG k-epsilon turbulence model, while the unsteady flow is computed with large eddy simulation. According to the theoretical analysis, the inlet guide vanes with outlet angle of 60° are regarded as the optimal inlet guide vanes. The static characteristic experiment is carried out in a standard test rig and the aerodynamic noise is tested in a semi-anechoic room. Then, performances of the fan with optimal inlet guide vanes are compared with those of the prototype fan. The results show that there is reasonable agreement between the simulation results and the experimental data. It is found that the static characteristics of small axial flow fan is improved obviously after installing the optimal inlet guide vanes. 12An NIR photothermal-responsive hybrid hydrogel for enhanced wound healing显示文摘Moderately regulating vascularization and immune microenvironment of wound site is necessary to achieve scarless wound healing of the skin.Herein,we have prepared an angiogenesis-promoting and scar-preventing band-aid with a core-shell structure,that consists of MXene-loaded nanofibers(MNFs)as the core and dopamine-hyaluronic acid hydrogel(H)as the shell(MNFs@V-H@DA)to encapsulate a growth factor(vascular endothelial growth factor,VEGF,abbreviated as V)and H2S donor(diallyl trisulfide,DATS,abbreviated as DA).The continuous release of DA from this system produced H2S,which would successfully induce macrophages to polarize into M2-lile phenotype,regulating the immune microenvironment and inhibiting an excessive inflammatory response at the wound sites.It is conducive to the proliferation of skin cells,facilitating the wound healing.In addition,an appropriate amount of VEGF can be released from the MXene nanofibrous skeleton by adjusting the time of near-infrared(NIR)light exposure,preventing excessive neovascularization and extracellular matrix deposition at the wound sites.Collectively,this NIR photothermal-responsive band-aid achieved scarless wound healing through gradient-controlled vascularization and a related immune sequential reaction of damaged skin tissue.Lin Jin Xiaoqing Guo Di Gao Yan Liu Jiahua Ni Zhiming Zhang Yiqiao Huang Guibin Xu Zhe Yang Xingcai Zhang Xianhan Jiang 2022Bioactive Materials2022,7,10:4

19Effect of the Uneven Circumferential Blade Space on the Performance of Small Axial Flow Fan显示文摘Effects of the uneven circumferential blade space on static characteristics and aerodynamic noise of a small axial flow fan are studied in this work.The blade angle modulation is adopted to design a series of unequally spaced fans,which have different maximum of modulation angular displacement.The steady flow is simulated by the calculations of Navier-Stokes equations coupled with RNG k-epsilon turbulence model,while the unsteady flow is computed with large eddy simulation.According to theoretical analysis,a fan with a maximum of modulation angular displacement of 6° is regarded as the optimal unequally spaced fan.The experiment of static characteristic is carried out in a standard wind tunnel and the aerodynamic noise of both fans is tested in a semi-anechoic room.Then,performances of the optimal unequally spaced fan are compared with those of the prototype fan.The results show that there is reasonable agreement between the simulation results and the experimental data.It is found that the discrete noise of the optimal unequally spaced fan is lower than that of the prototype fan at the near field monitoring point.This can be explained that the total pressure fluctuation of the optimal unequally spaced fan is much more regular than that of the prototype fan.LIU Yang LIN Zhe LIN Peifeng JIN Yingzi Toshiaki Setoguchi Heuy Dong Kim 2016Journal of Thermal Science2016,25,6:2
