1Self-Protection against Triptolide-Induced Toxicity in Human Hepatic Cells via Nrf2-ARE-NQO1 Pathway显示文摘Objective: To find the signaling pathway of triptolide(TP)-induced liver injury and to reveal whether NF-E2-related factor 2(Nrf2) plays an important role in cellular self-protection. Methods: The L-02 and HepG2 cells were cultured and treated with various concentrations of TP. The cell viability was observed, and the cell medium was collected for detecting the aspartate aminotransferase(ALT), alanine aminotransferase(AST), lactate dehydrogenase(LDH), superoxide dismutase(SOD) and L-glutathione production(GSH) levels. Nrf2 and its downstream target NAD(P)H: quinine oxidoreductase 1(NQO1) and heme oxygenase-1(HO-1) expression, the nuclear translocation of Nrf2, and the binding ability of Nrf2 and antioxidant response element(ARE) were also identified. Meanwhile,shRNA was used to silence Nrf2 in L-02 cells to find out whether Nrf2 plays a protective role. Results: The viability of the L-02 and HepG2 cells treated with TP decreased in a doseand time-dependent manner, and TP(20–80 μg/mL) markedly induced the release of ALT, AST and LDH(P<0.05 or P<0.01), reduced the levels of SOD and GSH(P<0.01), and increased the intracellular reactive oxygen species. Meanwhile, TP augmented the Nrf2 expression in L-02 and HepG2 cells(P<0.05 or P<0.01), induced Nrf2 nuclear translocation, increased the Nrf2 ARE binding activity, and increased HO-1 and NQO1 expressions. Nrf2 knockdown revealed a more severe toxic effect of TP(P<0.05 or P<0.01). Conclusions: Human hepatic cells treated with TP induced oxidative stress, and led to cytotoxicity. 2Is the Chinese Medicinal Formula Guipi Decoction(归脾汤) Effectiveas AnAdjunctive Treatment for Depression?AMeta-Analysis of Randomized Controlled Trials显示文摘Objectives: To evaluate the effectiveness and safety of Guipi Decoction(归脾汤, GPD) as an adjunctive in the treatment of depression. Methods: A review of all relevant studies retrieved from a search of the following databases were conducted without any language restriction: Excerpt Medica Database(EMBASE), Pub Med, Cochrane Central Register of Controlled Trials, China National Knowledge Infrastructure(CNKI), VIP Information, Wanfang Data, and the Chinese Biomedical Literature Database. Papers published until February 2013 were taken into consideration. The analysis was performed using the Cochrane software Revman 5.1. Results: Nine randomized controlled trials involving 620 patients with depression were included in this review. The meta-analysis revealed that compared with antidepressant therapy alone, treatment with a combination of GPD and an antidepressant drug significantly improved the symptoms of depression [weighted mean difference(WMD): –3.09; 95% confidence interval(CI): –4.11 to –2.07] and increased the rates of effectiveness(OR: 4.75; 95% CI: 2.66–8.51) as well as recovery(OR: 1.73; 95% CI: 1.17–2.56). The adverse effects of GPD were not found to be significant in these studies. Conclusions: The findings of this meta-analysis were in keeping with the notion that GPD formulations were effective in the treatment of depression without causing any severe adverse effects. 3Xiaoyaosan improves depressive-like behaviors by regulating the NLRP3 signaling pathway in the rat cerebral cortex显示文摘Objective:To observe changes in the molecular expression of the NLR Family Pyrin Domain Containing Protein 3(NLRP3)pathway in depressed rats after treatment with Xiaoyaosan,and identify the regulatory mechanism of this compound.Methods:Male SpragueeD awley rats were randomly divided into five groups with 12 rats in each group,including the control group,model group,Fluoxetine group,Xiaoyaosan group,and MCC950 group.A depression model was generated by chronic immobilization stress(induced by 3 h of restraint immobilization every day),and the drugs were administered at the same time in each group for 21 days.The effects of Xiaoyaosan on behavioral changes of depressed rats were observed through macroscopic characterization,body mass,open field experiments,and a sucrose preference test.The m RNA and protein expression of the NLRP3 signaling pathway was examined by fluorescence real-time quantitative PCR and Western blot assays.Results:The Xiaoyaosan group,Fluoxetine group,and MCC950 group rats showed improved depressive behavior and an increased weight of sucrose water consumption.The protein and mRNA expression levels of NLRP3,Caspase-1,and IL-1 b were also decreased in the Fluoxetine,Xiaoyaosan,and MCC950 groups.Conclusion:NLRP3,Caspase-1,and IL-1 b protein and mRNA expression levels were increased in the cortex of depressed rats,while Xiaoyaosan protected cortical tissue in these rats by decreasing NLRP3,Caspase-1,and IL-1 b protein and mRNA expression.Cong Chen Rong Yu Zhe Xue Zhiyi Yan Qinlai Bian Yajing Hou Yunzhi Chen Yueyun Liu Jiaxu Chen 2020Journal of Traditional Chinese Medical Sciences2020,7,3:5
4Metformin activates chaperone-mediated autophagy and improves disease pathologies in an Alzheimer disease mouse model显示文摘Chaperone-mediated autophagy(CMA)is a lysosome-dependent selective degradation pathway implicated in the pathogenesis of cancer and neurodegenerative diseases.However,the mechanisms that regulate CMA are not fully understood.Here,using unbiased drug screening approaches,we discover Metformin,a drug that is commonly the first medication prescribed for type 2 diabetes,can induce CMA.We delineate the mechanism of CMA induction by Metformin to be via activation of TAK1-IKKα/β signaling that leads to phosphorylation of Ser85 of the key mediator of CMA,Hsc70,and its activation.Notably,we find that amyloid-beta precursor protein(APP)is a CMA substrate and that it binds to Hsc70 in an IKKα/β-dependent manner.The inhibition of CMA-mediated degradation of APP enhances its cytotoxicity.Importantly,we find that in the APP/PS1 mouse model of Alzheimer's disease(AD),activation of CMA by Hsc70 overexpression or Metformin potently reduces the accumulated brain Aβplaque levels and reverses the molecular and behavioral AD phenotypes.Our study elucidates a novel mechanism of CMA regulation via Metformin-TAK1-IKKα/β-Hsc70 signaling and suggests Metformin as a new activator of CMA for diseases,such as AD,where such therapeutic intervention could be beneficial.Xiaoyan Xu Yaqin Sun Xufeng Cen Bing Shan Qingwei Zhao Tingxue Xie Zhe Wang Tingjun Hou Yu Xue Mengmeng Zhang Di Peng Qiming Sun Cong Yi Ayaz Najafov Hongguang Xia 2021Protein & Cell2021,12,10:5
5In vitro and in silico analysis of dual-function peptides derived from casein hydrolysate显示文摘There is no study on food-derived peptide with both anticoagulant and angiotensin I-converting enzyme inhibitory (ACEI) activities yet. In this work, the anticoagulant and ACEI activities of the casein hydrolysates released by pepsin digestion were evaluated for the first time to the best of our knowledge. Results indicated that the casein hydrolysate exhibited potent anticoagulant activity by prolonging the thrombin time (TT) and the activated partial thromboplastin time (APTT). Compared with control samples, at 10 mg/mL, the TT and APTT of casein hydrolysate were 186.0 % ± 6.6 % and 163.5 % ± 7.4 %, respectively. The casein hydrolysate also showed a strong ACEI activity with an IC50 value of 1.775 mg/mL. The components of the bioactive casein hydrolysate were analyzed by nanoscale liquid chromatography quadrupole time-of-flight tandem mass spectrometry (NanoLC-Q-TOF-MS/MS). Total of 115 peptides were identified, among which 34, 9, 55 and 17 peptides were derived from α_(s1-), α_(s2-), β-, and κ-casein, respectively. The results of PeptideRanker and PepSite 2 analysis showed that 6 peptides (FRQFYQL, NENLLRF, NPWDQVKR, PVVVPPFLQ, PVRGPFPIIV, and ARHPHPHLSF) have both ACEI and anticoagulant activities by binding to the active sites of ACE and thrombin. 6Single-End Adaptive Optics Compensation for Emulated Turbulence in a Bi-Directional 10-Mbit/s per Channel Free-Space Quantum Communication Link Using Orbital-Angular-Momentum Encoding显示文摘A single-end adaptive-optics(AO)module is experimentally demonstrated to mitigate the emulated atmospheric turbulence efects in a bi-directional quantum communication link,which employs orbital angular momentum(OAM)for data encoding.A classical Gaussian beam is used as a probe to detect the turbulence-induced wavefront distortion in the forward direction of the link.Based on the detected wavefront distortion,an AO system located on one end of the link is used to simultaneously compensate for the forward and backward channels.Specifcally,with emulated turbulence and when the probe is turned on,the mode purity of photons carrying OAMℓ=1 is improved by∼21%with AO mitigation.We also measured the performance when encoding data using OAM{ℓ=−1,+2}and{ℓ=−2,+1}in the forward and backward channels,respectively,at 10 Mbit/s per channel with one photon per pulse on average.For this case,we found that the AO system could reduce the turbulence efects increased quantum-symbolerror-rate(QSER)by∼76%and∼74%,for both channels in the uni-directional and bi-directional cases,respectively.Similar QSER improvement is observed for the opposite direction channels in the bi-directional case.Cong Liu Kai Pang Zhe Zhao Peicheng Liao Runzhou Zhang Haoqian Song Yinwen Cao Jing Du Long Li Hao Song Yongxiong Ren Guodong Xie Yifan Zhao Jiapeng Zhao Seyed M.H.Rafsanjani Ari N.Willner Jeffrey H.Shapiro Robert W.Boyd Moshe Tur Alan E.Willner 2019Research2019,,1:3
7Orbital-angular-momentum-based reconfigurable optical switching and routing显示文摘Orbital angular momentum(OAM) has gained interest due to its potential to increase capacity in optical communication systems as well as an additional domain for reconfigurable networks. This is due to the following:(i) coaxially propagated OAM beams with different charges are mutually orthogonal,(ii) OAM beams can be efficiently multiplexed and demultiplexed, and(iii) OAM charges can be efficiently manipulated. Therefore, multiple data-carrying OAM beams could have the potential capability for reconfigurable optical switching and routing. 11Design of an RFQ for direct plasma injection scheme显示文摘A high current radio frequency quadrupole (RFQ) is being studied at the Institute of Modern Physics, Chinese Academy of Sciences (IMP, CAS) for the direct plasma injection scheme (DPIS). Because of the strong space charge of beams from laser ion source, the beam dynamics design of the RFQ has been carried out with a new code, which can deal with space charge effectively. The design of the RFQ structure is performed with an electromagnetic simulation code and the determination of parameters of the structure has been done to maximize the shunt impedance when the frequency is kept fixed. 12Twist2 is a valuable prognostic biomarker for colorectal cancer显示文摘AIM: To investigate the significance of Twist2 for colorectal cancer (CRC). METHODS: In this study, 93 CRC patients were included who received curative surgery in Eastern Hepatobiliary Surgery Hospital from January 1999 to December 2010. Records of patients' clinicopathological characteristics and follow up data were reviewed. Formalin-fixed, paraffin-embedded tissue blocks were used to observe the protein expression of Twist2 and E-cadherin by immunohistochemistry. Two independent pathologists who were blinded to the clinical information performed semiquantitative scoring of immunostaining. A total score of 3-6 (sum of extent + intensity) was considered as Twist2-positive expression. The expression of E-cadherin was divided into two levels (preserved and reduced). An exploratory statistical analysis was conducted to determine the association between Twist2 expression and clinicopathological parameters, as well as E-cadherin expression. Furthermore, the variables associated with prognosis were analyzed by Cox's proportional hazards model. Kaplan-Meier analysis was used to plot survival curves according to different expression levels of Twist2. RESULTS: Twist2-positive expression was observed in 66 (71.0%) samples and mainly located in the cytoplasm. Forty-three (46.2%) samples showed reduced expression of E-cadherin. There were no significant correlations between Twist2 expression and any of the clinicopathological parameters. However, Twist2-positive expression was significantly associated with reduced expression of E-cadherin (P=0.040). Multivariate analysis revealed that bad M-stage [hazard ratio (HR)=7.694, 95%CI: 2.927-20.224,P < 0.001] and Twist2-positive (HR=5.744, 95%CI: 1.347-24.298,P=0.018) were the independent risk factors for poor overall survival (OS), while Twist2-positive (HR=3.264, 95%CI: 1.455-7.375, P=0.004), bad N-stage (HR=2.149, 95%CI: 1.226-3.767, P=0.008) and bad M-stage (HR=10.907, 95%CI: 4.937-24.096, P < 0.001) were independently associated with poor disease-free survival (DFS). Survival curves showed a definite trend for Twist2-negative patients to have longer OS and DFS than Twist2-negative patients, not only overall, but also for patients in different stages, especially for DFS of patients in stage Ⅲ (P=0.033) and Ⅳ (P=0.026). 17Accurate and explainable machine learning for the power factors of diamond-like thermoelectric materials显示文摘The application of machine learning(ML)-based methods to the study of thermoelectric(TE)materials is promising.Although conventional ML algorithms can achieve high prediction performance,their lack of interpretability severely obstructs researchers from extracting material-oriented insights from ML models.In this work,high ML-based prediction performance was achieved with respect to TE power factors(PFs),and the results were well understood by the SHapley Additive exPlanations(SHAP),a method to identify the correlations between targets and descriptors.We designed a robust PF prediction model for diamond-like compounds via a stacking technique,and the model achieved a coefficient of determination value above 0.95 on the test set.From the SHAP analysis,the PFs were negatively correlated with electronegativity and positively correlated with the descriptor“volume per atom”based on the previously reported dataset.TE domain knowledge was adopted to understand these correlations.This work shows that ML models can achieve high accuracy while exhibiting good interpretability,making them useful for materials scientists.Zhe Yang Ye Sheng Cong Zhu Jianyue Ni Zhenyu Zhu Jinyang Xi Wu Zhang Jiong Yang 2022Journal of Materiomics2022,8,3:1

19PSPC1 regulates CHK1 phosphorylation through phase separation and participates in mouse oocyte maturation显示文摘Liquid–liquid phase separation (LLPS) underlies the formation of membraneless compartments in mammal cells. However, there are few reports that focus on the correlation of mouse oocyte maturation with LLPS. Previous studies have reported that paraspeckle component 1 (PSPC1) is related to the occurrence and development of tumors, but whether PSPC1 functions in mouse oocyte maturation is still unclear. Sequence analysis of PSPC1 protein showed that it contains a prion-like domain (PrLD) that is required for phase separation of proteins. In this study, we found that PSPC1 could undergo phase separation. Moreover, the loss of PrLD domain of PSPC1 could greatly weaken its phase separation ability. The immunofluorescence assays showed that PSPC1 is present in mouse oocytes in the germinal vesicle (GV) stage. Knockdown of PSPC1 significantly impeded the maturation of mouse oocytes in vitro. CHK1 has been reported to play important roles in the GV stage of mouse oocytes. Co-IP experiment revealed that PSPC1 could interact with phosphatase serine/threonine-protein phosphatase 5 (PPP5C), which regulates CHK1 phosphorylation. Western blot analysis revealed that PSPC1 could regulate the phosphorylation of CHK1 through PPP5C;however, PSPC1 without PrLD domain was inactive, suggesting that the lack of phase separation ability led to the abnormal function of PSPC1 in regulating CHK1 phosphorylation. Thus, we conclude that PSPC1 may undergo phase separation to regulate the phosphorylation level of CHK1 via PPP5C and participate in mouse oocyte maturation.
