1Elimination of senescent cells byβ-galactosidase-targeted prodrug attenuates inflammation and restores physical function in aged mice显示文摘Cellular senescence,a persistent state of cell cycle arrest,accumulates in aged organisms,contributes to tissue dysfunction,and drives age-related phenotypes.The clearance of senescent cells is expected to decrease chronic,low-grade inflammation and improve tissue repair capacity,thus attenuating the decline of physical function in aged organisms.However,selective and effective clearance of senescent cells of different cell types has proven challenging.Herein,we developed a prodrug strategy to design a new compound based on the increased activity of lysosomalβ-galactosidase(β-gal),a primary characteristic of senescent cells.Our prodrug SSK1 is specifically activated byβ-gal and eliminates mouse and human senescent cells independently of senescence inducers and cell types.In aged mice,our compound effectively cleared senescent cells in different tissues,decreased the senescence-and age-associated gene signatures,attenuated low-grade local and systemic inflammation,and restored physical function.Our results demonstrate that lysosomalβ-gal can be effectively leveraged to selectively eliminate senescent cells,providing a novel strategy to develop anti-aging interventions.Yusheng Cai Huanhuan Zhou Yinhua Zhu Qi Sun Yin Ji Anqi Xue Yuting Wang Wenhan Chen Xiaojie Yu Longteng Wang Han Chen Cheng Li Tuoping Luo Hongkui Deng 2020Cell Research2020,30,7:12

6DNA N^6-methyladenine demethylase ALKBH1 enhances osteogenic differentiation of human MSCs显示文摘ALKBH1 was recently discovered as a demethylase for DNA N^6-methyladenine(N6-m A), a new epigenetic modification, and interacts with the core transcriptional pluripotency network of embryonic stem cells.However, the role of ALKBH1 and DNA N6-m A in regulating osteogenic differentiation is largely unknown.In this study, we demonstrated that the expression of ALKBH1 in human mesenchymal stem cells(MSCs)was upregulated during osteogenic induction. Knockdown of ALKBH1 increased the genomic DNA N6-m A levels and significantly reduced the expression of osteogenic-related genes, alkaline phosphatase activity, and mineralization. ALKBH1-depleted MSCs also exhibited a restricted capacity for bone formation in vivo.By contrast, the ectopic overexpression of ALKBH1 enhanced osteoblastic differentiation. Mechanically,we found that the depletion of ALKBH1 resulted in the accumulation of N6-m A on the promoter region of ATF4, which subsequently silenced ATF4 transcription. In addition, restoring the expression of ATP by adenovirus-mediated transduction successfully rescued osteogenic differentiation. 7Genome-and transcriptome-wide association studies provide insights into the genetic basis of natural variation of seed oil content in Brassica napus显示文摘Seed oil content(SOC)is a highly important and complex trait in oil crops.Here,we decipher the genetic basis of natural variation in SOC of Brassica napus by genome-and transcriptome-wide association studies using 505 inbred lines.We mapped reliable quantitative trait loci(QTLs)that control SOC in eight environments,evaluated the effect of each QTL on SOC,and analyzed selection in QTL regions during breeding.Six-hundred and ninety-two genes and four gene modules significantly associated with SOC were identified by analyzing population transcriptomes from seeds.A gene prioritization framework,POCKET(prioritizing the candidate genes by incorporating information on knowledge-based gene sets,effects of variants,genome-wide association studies,and transcriptome-wide association studies),was implemented to determine the causal genes in the QTL regions based on multi-omic datasets.A pair of homologous genes,BnPMT6s,in two QTLs were identified and experimentally demonstrated to negatively regulate SOC.This study provides rich genetic resources for improving SOC and valuable insights toward understanding the complex machinery that directs oil accumulation in the seeds of B.napus and other oil crops.Shan Tang Hu Zhao Shaoping Lu Liangqian Yu Guofang Zhang Yuting Zhang Qing-Yong Yang Yongming Zhou Xuemin Wang Wei Ma Weibo Xie Liang Guo 2021Molecular Plant2021,14,3:6

9Engineering rock mechanics practices in the underground powerhouse at JinpingⅠhydropower station显示文摘Based on the analyses of data obtained from the underground powerhouse at Jinping Ⅰ hydropower station,a comprehensive review of engineering rock mechanics practice in the underground powerhouse is first conducted.The distribution of strata,lithology,and initial geo-stress,the excavation process and corresponding rock mass support measures,the deformation and failure characteristics of the surrounding rock mass,the stress characteristics of anchorage structures in the cavern complex,and numerical simulations of surrounding rock mass stability and anchor support performance are presented.The results indicate that the underground powerhouse of Jinping Ⅰ hydropower station is characterized by high to extremely high geo-stresses during rock excavation.Excessive surrounding rock mass deformation and high stress of anchorage structures,surrounding rock mass unloading damage,and local cracking failure of surrounding rock masses,etc.,are mainly caused by rock mass excavation.Deformations of surrounding rock masses and stresses in anchorage structures here are larger than those found elsewhere:20%of extensometers in the main powerhouse record more than 50 mm with the maximum at around 250 mm observed in the downstream sidewall of the transformer hall.There are about 25%of the anchor bolts having recorded stresses of more than 200 MPa.Jinping Ⅰ hydropower plant is the first to have an underground powerhouse construction conducted in host rocks under extremely high geo-stress conditions,with the ratio of rock mass strength to geo-stress of less than 2.0.The results can provide a reference to underground powerhouse construction in similar geological conditions.Aiqing Wu Jimin Wang Zhong Zhou Shuling Huang Xiuli Ding Zhihong Dong Yuting Zhang 2016Journal of Rock Mechanics and Geotechnical Engineering2016,8,5:4
10Cold Stress-induced Glucosyltransferase CsUGT78A15 is Involved in the Formation of Eugenol Glucoside in Camellia sinensis显示文摘Eugenol is a natural phenolic compound known for its health-promoting properties and its ability to add a floral scent to tea plants.Plant eugenol glycosides have been identified and shown to make important contributions to fruit floral quality.However,the details of their biosynthesis and metabolism in tea plants are still unknown.Here,eugenol glucoside was unambiguously identified as a native metabolite in the tea plant,and its biosynthesis was shown to be induced by low temperature treatment.Through the analysis of UGTs induced by low temperature,the glycosyltransferase CsUGT78A15 was identified in tea,and its encoded protein was shown to catalyze the glucosylation of eugenol.Vmax/Km ratios showed that eugenol was the most suitable substrate for CsUGT78A15.Sugar donor preference analysis showed that CsUGT78A15 had a higher selectivity for glucose,followed by galactose and glucuronic acid.The expression of CsUGT78A15was correlatedwith the accumulation of eugenol glucoside in different tissues and genotypes of tea.Down-regulation of CsUGT78A15 led to a decreased eugenol glucoside content under cold stress,indicating that CsUGT78A15 plays an important role in the biosynthesis of eugenol glucoside under cold stress.The identification of eugenol glucoside in the tea plant and the discovery of a cold stress-induced eugenol glucosyltransferase in tea provide the foundation for the improvement of tea flavor under cold stress and the biotechnological production of eugenol glucoside.Mingyue Zhao Binbin Cai Jieyang Jin Na Zhang Tingting Jing Jingming Wang Yuting Pan Zixiang Zhou Yifan Zhao Yingying Feng Feng Yu Mengting Zhang Yating Li Zhonghua Liu Chuankui Song 2020Horticultural Plant Journal2020,6,6:4

12Epithelial-mesenchymal transition status of circulating tumor cells in breast cancer and its clinical relevance显示文摘Objective:Circulating tumor cells(CTCs)play a critical role in cancer metastasis,but their prevalence and significance remain unclear.This study attempted to track the epithelial-mesenchymal transition(EMT)status of CTCs in breast cancer patients and investigate their clinical relevance.Methods:In this study,the established negFACS-IF:E/M platform was applied to isolate rare CTCs and characterize their EMT status in breast cancer.A total of 89 breast cancer patients were recruited,including stage 0–III(n=60)and late stage(n=29)cases.Results:Using the negFACS-IF:E/M platform,it was found that in human epidermal growth factor receptor 2(HER2)+patients,mesenchymal CTCs usually exhibited a high percentage of HER2+cells.Stage IV breast cancer patients had considerably more CTCs than stage 0–III patients.Among stage 0–III breast cancers,the HER2 subtype included a significantly higher percentage of mesenchymal and biphenotypic(epithelial and mesenchymal)CTCs than the luminal A or B subtypes.Among stage IV patients,CTCs were predominantly epithelial in cases with local recurrence and were more mesenchymal in cases with distant metastasis.By applying a support vector machine(SVM)algorithm,the EMT status of CTCs could distinguish between breast cancer cases with metastasis/local recurrence and those without recurrence.Conclusions:The negFACS-IF:E/M platform provides a flexible and generally acceptable method for the highly sensitive and specific detection of CTCs and their EMT traits in breast cancer.This study demonstrated that the EMT status of CTCs had high clinical relevance in breast cancer,especially in predicting the distant metastasis or local recurrence of breast cancer.Jiaojiao Zhou Xuan Zhu Shijie Wu Jingxin Guo Kun Zhang Chunjing Xu Huihui Chen Yuxi Jin Yuting Sun Shu Zheng Yiding Chen 2020Cancer Biology & Medicine2020,17,1:3
13A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults.Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors,built and examined the operating characteristics of a point-based algorithm for HCC risk prediction.Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells,concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively. 14Generation of purple-violet chrysanthemums via anthocyanin B-ring hydroxylation and glucosylation introduced from Osteospermum hybrid F3'5'H and Clitoria ternatea A3'5'GT显示文摘Chrysanthemums possess no metabolic pathway to synthesize delphinidin because of the lack of endogenous F3'5'H gene encoding the key enzyme in its biosynthetic pathway;therefore,there are no blue or blue-purple chrysanthemums occurring naturally.Currently,the introduction of exogenous F3'5'H into chrysanthemums is an efficient method for breeding bluish chrysanthemums.In this study,we explored the effects of the introduction of mutant CmF3'H(generated via site-directed mutagenesis,T485S,CmF3'Hm)and exogenous Osteospermum hybrid F3'5'H(OhF3'5'H)genes combined with Clitoria ternatea A3'5'GT(CtA3'5'GT)on delphinidin synthesis in chrysanthemum.Among the F3'5'H transgenic lines,those overexpressing endogenous CmF3'Hm could not generate blue flower color,although red color was changed to light pink due to CtA3'5'GT function.Meanwhile,OhF3'5'H introduction promoted the accumulation of delphinidin and its derivatives in chrysanthemum,changing the flower color from red-purple to purple-violet.These results indicate the applicability of exogenous OhF3'5'H and CtA3'5'GT transformation for promoting delphinidin synthesis during the molecular breeding of violet/blue chrysanthemums.Xiaoying Han Yuting Luo Jiaoyang Lin Huiying Wu Hao Sun Lijie Zhou Sumei Chen Zhiyong Guan Weimin Fang Fei Zhang Fadi Chen Jiafu Jiang 2021Ornamental Plant Research2021,1,1:3
15Modulating the optical and electrical properties of MAPbBr_(3) single crystals via voltage regulation engineering and application in memristors显示文摘Defect density is one of the most significant characteristics of perovskite single crystals(PSCs)that determines their optical and electrical properties,but few strategies are available to tune this property.Here,we demonstrate that voltage regulation is an efficient method to tune defect density,as well as the optical and electrical properties of PSCs.A three-step carrier transport model of MAPbBr_(3) PSCs is proposed to explore the defect regulation mechanism and carrier transport dynamics via an applied bias.Dynamic and steady-state photoluminescence measurements subsequently show that the surface defect density,average carrier lifetime,and photoluminescence intensity can be efficiently tuned by the applied bias.In particular,when the regulation voltage is 20 V(electrical poling intensity is 0.167 Vμm^(−1)),the surface defect density of MAPbBr_(3) PSCs is reduced by 24.27%,the carrier lifetime is prolonged by 32.04%,and the PL intensity is increased by 112.96%.Furthermore,a voltage-regulated MAPbBr_(3) PSC memristor device shows an adjustable multiresistance,weak ion migration effect and greatly enhanced device stability.Voltage regulation is a promising engineering technique for developing advanced perovskite optoelectronic devices.Jun Xing Chen Zhao Yuting Zou Wenchi Kong Zhi Yu Yuwei Shan Qingfeng Dong Ding Zhou Weili Yu Chunlei Guo 2020Light(Science & Applications)2020,9,1:3
16Interactions between commensal bacteria and viral infection:insights for viral disease control in farmed animals显示文摘Viral diseases cause serious economic loss in farmed animals industry.However,the efficacy of remedies for viral infection in farmed animals is limited,and treatment strategies are generally lacking for aquatic animals.Interactions of commensal microbiota and viral infection have been studied in recent years,demonstrating a third player in the interaction between hosts and viruses.Here,we discuss recent developments in the research of interactions between commensal bacteria and viral infection,including both promotion and inhibition effect of commensal bacteria on viral pathogenesis,as well as the impact of viral infection on commensal microbiota.The antiviral effect of commensal bacteria is mostly achieved through priming or regulation of the host immune responses,involving differential microbial components and host signaling pathways,and gives rise to various antiviral probiotics.Moreover,we summarize studies related to the interaction between commensal bacteria and viral infection in farmed animals,including pigs,chickens,fish and invertebrate species.Further studies in this area will deepen our understanding of antiviral immunity of farmed animals in the context of commensal microbiota,and promote the development of novel strategies for treatment of viral diseases in farmed animals.Chao Ran Yu Li Xufa Ma Yadong Xie Mingxu Xie Yuting Zhang Wei Zhou Yalin Yang Zhen Zhang Li Zhou Kaijian Wei Zhigang Zhou 2021Science China(Life Sciences)2021,64,9:2

19Laparoscopic vs.open surgery for gastrointestinal stromal tumors of esophagogastric junction:A multicenter,retrospective cohort analysis with propensity score weighting显示文摘Objective:Laparoscopic resection is increasingly performed for gastrointestinal stromal tumors(GISTs).However,the laparoscopic approach for GISTs located in the esophagogastric junction(EGJ-GIST)is surgically challenging.This study compares the efficacy of laparoscopic surgery and the open procedure for EGJ-GIST through the propensity score weighting(PSW)method.Methods:Between April 2006 and April 2018,1,824 surgical patients were diagnosed with primary gastric GIST at four medical centers in South China.Of these patients,228 were identified as EGJ-GISTs and retrospectively reviewed clinicopathological characteristics,operative information,and long-term outcomes.PSW was used to create the balanced cohorts.Results:PSW was carried out in laparoscopic and open-surgery cohorts according to year of surgery,sex,age,body mass index(BMI),tumor size,mitotic rates and recurrence risk.After PSW,438 patients consisting of 213 laparoscopic(L group)and 225 open surgery(O group)patients were enrolled.After PSW,the following measures in the L group were superior to those in the O group:median operative time[interquartile range(IQR)]:100.0(64.5-141.5)vs.149.0(104.0-197.5)min,P<0.001;median blood loss(IQR):30.0(10.0-50.0)vs.50.0(20.0-100.0)mL,P=0.002;median time to liquid intake(IQR):3.0(2.0-4.0)vs.4.0(3.0-5.0)d,P<0.001;median hospital stay(IQR):6.0(4.0-8.0)vs.7.0(5.0-12.0)d,P<0.001;and postoperative complications(10.3%vs.22.7%,P=0.001).The median follow-up was 55(range,2-153)months in the entire cohort.No significant differences were detected in either relapse-free survival(RFS)[hazard ratio(HR):0.372,95%confidence interval(95%CI):0.072-1.910,P=0.236]or overall survival(OS)(HR:0.400,95%CI:0.119-1.343,P=0.138)between the two groups.Conclusions:Laparoscopic surgery for EGJ-GIST is associated with the advantages of shorter operative time,reduced blood loss,shorter time to liquid intake,and shorter length of stay,all without compromising postoperative outcomes and long-term survival.Wenjun Xiong Yuting Xu Tao Chen Xingyu Feng Rui Zhou Jin Wan Yong Li Guoxin Li Wei Wang 2021Chinese Journal of Cancer Research2021,33,1:2
