1Activation of mTORC1 in subchondral bone preosteoblasts promotes osteoarthritis by stimulating bone sclerosis and secretion of CXCL12显示文摘Increasing evidences show that aberrant subchondral bone remodeling plays an important role in the development of osteoarthritis(OA).However,how subchondral bone formation is activated and the mechanism by which increased subchondral bone turnover promotes cartilage degeneration during OA remains unclear.Here,we show that the mechanistic target of rapamycin complex 1(mTORC1)pathway is activated in subchondral bone preosteoblasts(Osterix+)from OA patients and mice.Constitutive activation of mTORC1 in preosteoblasts by deletion of the mTORC1 upstream inhibitor,tuberous sclerosis 1,induced aberrant subchondral bone formation,and sclerosis with little-to-no effects on articular cartilage integrity,but accelerated posttraumatic OA development in mice.In contrast,inhibition of mTORC1 in preosteoblasts by disruption of Raptor(mTORC1-specific component)reduced subchondral bone formation and cartilage degeneration,and attenuated post-traumatic OA in mice.Mechanistically,mTORC1 activation promoted preosteoblast expansion and Cxcl12 secretion,which induced subchondral bone remodeling and cartilage degeneration during OA.A Cxcl12-neutralizing antibody reduced cartilage degeneration and alleviated OA in mice.Altogether,these findings demonstrate that mTORC1 activation in subchondral preosteoblasts is not sufficient to induce OA,but can induce aberrant subchondral bone formation and secrete of Cxcl12 to accelerate disease progression following surgical destabilization of the joint.Pharmaceutical inhibition of the pathway presents a promising therapeutic approach for OA treatment.Chuangxin Lin Liangliang Liu Chun Zeng Zhong-Kai Cui Yuhui Chen Pinling Lai Hong Wang Yan Shao Haiyan Zhang Rongkai Zhang Chang Zhao Hang Fang Daozhang Cai Xiaochun Bai 2019Bone Research2019,7,1:12
2Perovskite light-emitting/detecting bifunctional fibres for wearable LiFi communication显示文摘Light fidelity(LiFi),which is emerging as a compelling technology paradigm shifting the common means of highcapacity wireless communication technologies,requires wearable and full-duplex compact design because of its great significance in smart wearables as well as the‘Internet of Things’.However,the construction of the key component of wearable full-duplex LiFi,light-emitting/detecting bifunctional fibres,is still challenging because of the conflicting process between carrier separation and recombination,as well as the highly dynamic film-forming process.Here,we demonstrate light-emitting/detecting bifunctional fibres enabled by perovskite QDs with hybrid components.The hybrid perovskite inks endow fibres with super-smooth QD films.This,combined with the small exciton binding energy and high carrier mobility of perovskite QDs,enables successful integration of electroluminescence and photodetection into monofilaments.The bifunctional fibres possess the narrowest electroluminescence full width at half maximum of ~19 nm and,more importantly,the capability for simultaneously transmitting and receiving information.The successful fabrication of narrow emission full-duplex LiFi fibres paves the way for the fabrication and integration of low crosstalk interoperable smart wearables.Qingsong Shan Changting Wei Yan Jiang Jizhong Song Yousheng Zou Leimeng Xu Tao Fang Tiantian Wang Yuhui Dong Jiaxin Liu Boning Han Fengjuan Zhang Jiawei Chen Yongjin Wang Haibo Zeng 2020Light(Science & Applications)2020,9,1:7

4Measuring loblolly pine crowns with drone imagery through deep learning显示文摘In modeling forest stand growth and yield,crown width,a measure for stand density,is among the parameters that allows for estimating stand timber volumes.However,accurately measuring tree crown size in the field,in particular for mature trees,is challenging.This study demonstrated a novel method of applying machine learning algorithms to aerial imagery acquired by an unmanned aerial vehicle(UAV)to identify tree crowns and their widths in two loblolly pine plantations in eastern Texas,USA.An ortho mosaic image derived from UAV-captured aerial photos was acquired for each plantation(a young stand before canopy closure,a mature stand with a closed canopy).For each site,the images were split into two subsets:one for training and one for validation purposes.Three widely used object detection methods in deep learning,the Faster region-based convolutional neural network(Faster R-CNN),You Only Look Once version 3(YOLOv3),and single shot detection(SSD),were applied to the training data,respectively.Each was used to train the model for performing crown recognition and crown extraction.Each model output was evaluated using an independent test data set.All three models were successful in detecting tree crowns with an accuracy greater than 93%,except the Faster R-CNN model that failed on the mature site.On the young site,the SSD model performed the best for crown extraction with a coefficient of determination(R^(2))of 0.92,followed by Faster R-CNN(0.88)and YOLOv3(0.62).As to the mature site,the SSD model achieved a R^(2)as high as 0.94,follow by YOLOv3(0.69).These deep leaning algorithms,in particular the SSD model,proved to be successfully in identifying tree crowns and estimating crown widths with satisfactory accuracy.For the purpose of forest inventory on loblolly pine plantations,using UAV-captured imagery paired with the SSD object detention application is a cost-effective alternative to traditional ground measurement.Xiongwei Lou Yanxiao Huang Luming Fang Siqi Huang Haili Gao Laibang Yang Yuhui Weng I.-K.uai Hung 2022Journal of Forestry Research2022,33,1:3
5Development of T. aestivum L.eH. californicum Alien Chromosome Lines and Assignment of Homoeologous Groups of Hordeum californicum Chromosomes显示文摘Hordeum californicum(2n=2x=14, HH) is resistant to several wheat diseases and tolerant to lower nitrogen. In this study, a molecular karyotype of H. californicum chromosomes in the Triticum aestivum L. cv. Chinese Spring(CS)eH. californicum amphidiploid(2n=6x=56, AABBDDHH) was established. By genomic in situ hybridization(GISH) and multicolor fluorescent in situ hybridization(FISH) using repetitive DNA clones(pTa71, pTa794 and pSc119.2) as probes, the H. californicum chromosomes could be differentiated from each other and from the wheat chromosomes unequivocally. Based on molecular karyotype and marker analyses, 12 wheatealien chromosome lines, including four disomic addition lines(DAH1, DAH3, DAH5 and DAH6), five telosomic addition lines(MtH7L,MtH1 S, MtH1 L, DtH6 S and DtH6L), one multiple addition line involving H. californicum chromosome H2, one disomic substitution line(DSH4) and one translocation line(TH7S/1BL), were identified from the progenies derived from the crosses of CSeH. californicum amphidiploid with common wheat varieties. A total of 482 EST(expressed sequence tag) or SSR(simple sequence repeat) markers specific for individual H. californicum chromosomes were identified, and 47, 50, 45, 49, 21, 51 and 40 markers were assigned to chromosomes H1, H2, H3, H4, H5, H6 and H7, respectively. According to the chromosome allocation of these markers, chromosomes H2,H3, H4, H5, and H7 of H. californicum have relationship with wheat homoeologous groups 5, 2, 6, 3, and 1, and hence could be designated as 5Hc, 2Hc, 6Hc, 3Hcand 1Hc, respectively. 7PD0325901, an ERK inhibitor, enhances the efficacy of PD-1 inhibitor in non-small cell lung carcinoma显示文摘ERK pathway regulated the programmed death ligand-1(PD-L1)expression which was linked to the response of programmed death-1(PD-1)/PD-L1 blockade therapy.So it is deducible that ERK inhibitor could enhance the efficacy of PD-1 inhibitor in cancer immunotherapy.In this study,PD0325901,an oral potent ERK inhibitor,strongly enhanced the efficacy of PD-1 antibody in vitro and in vivo models in non-small cell lung carcinoma(NSCLC)cells.Mechanistically,PD0325901 or shRNA-ERK1/2 significantly downregulated the PD-L1 expression in NSCLC cells and increased the CD3+T cells infiltration and functions in tumor tissue.There was a positive correlation between the p-ERK1/2 expression and PD-L1 expression in patients with NSCLC.And the patients with low p-ERK1/2 expression were observed a high response rate of PD-1/PD-L1 blockage therapy.Our results demonstrate that PD0325901,an ERK inhibitor,can enhance the efficacy of PD-1 blockage against NSCLC in vitro and in vivo models.And the combination of ERK inhibitor such as PD0325901 and PD-1/PD-L1 blockage is a promising regimen and encouraged to be further confirmed in the treatment of patients with NSCLC.Min Luo Yuhui Xia Fang Wang Hong Zhang Danting Su Chaoyue Su Chuan Yang Shaocong Wu Sainan An Suxia Lin Liwu Fu 2021Acta Pharmaceutica Sinica B2021,11,10:1

13Chinese herbal medicine combined with cognitive–behavioural therapy for avoidant paruresis:a controlled trial显示文摘Background Avoidant paruresis is a common clinical condition in urology and psychosomatic medicine.However,it has limited treatment options that are safe and effective with few side effects.Aims Our study aimed to investigate the effectiveness and safety of the Chinese herbal Yangxin Tongquan decoction combined with cognitive-behavioural therapy(CBT)for avoidant paruresis.Methods Sixty-eight patients with avoidant paruresis were divided into a treatment group(33 patients)and a control group(35 patients).The control group was assigned 10 weeks of CBT and systematic desensitisation.In addition to CBT and systematic desensitisation,the treatment group was given the Chinese herbal Yangxin Tongquan decoction during the 10-week study.The Shy Bladder Syndrome Scale(SBS)and the Self-rating Anxiety Scale(SAS)were administered before and after treatment to measure any change.Results The overall efficacy in the treatment group(n=30)was 80.0%vs 62.5%in the control group(n=33).Comparing pretreatment and post-treatment measures,both groups showed improvement in SBS scores and SAS scores(treatment group:t_((SBS))=8.397,p_((SBS))<0.001,t_((SAS))=8.216,p_((SAS))<0.001;control group:t_((SBS))=6.802,p_((SBS))<0.001,t_((SAS))=5.171,p_((SAS))<0.001).Moreover,both groups’SBS and SAS scores changed significantly over time(SBS scores:F_(time)=118.299,p<0.001;SAS scores:F_(time)=92.114,p<0.001).However,the treatment group performed better than the control group(SBS scores:F_(time*group)=5.709,p=0.020;SAS scores:F_(time*group)=7.235,p=0.009).Conclusions The Chinese herbal Yangxin Tongquan decoction combined with cognitive-behavioural psychotherapy positively affects the treatment of avoidant paruresis without significant adverse effects.Shijie Liang Ying Zhou Chao Yu Xiang Gao Fangbin Ji Qianyuan Fang Zhihang Zhang Libin Yang Youkang Zhao Yuhui Zhao Renwu Yin Kaimin Chen Zheng Lu 2023General Psychiatry2023,36,5:0

15Pyrocincholic acid 3β-O-β-D-quinovopyranosyl-28-O-β-Dglucopyranoside suppresses adipogenesis and regulates lipid metabolism in 3T3-L1 adipocytes显示文摘Obesity is crucially involved in many metabolic diseases,such as type 2 diabetes,cardiovascular disease and cancer.Regulating the number or size of adipocytes has been suggested to be a potential treatment for obesity.In this study,we investigated the effect of pyrocincholic acid 3β-O-β-D-quinovopyranosyl-28-O-β-D-glucopyranoside(PAQG),a 27-nor-oleanolic acid saponin extracted from Metadina trichotoma,on adipogenesis and lipid metabolism in 3T3-L1 adipocytes.The 3T3-L1 pre-adipocytes were incubated with vehicle or PAQG for 6 days in differentiation process.PAQG significantly reduced the adipogenesis,adiponectin secretion and the expression level of key transcription factors related to adipogenesis,such as PPARc,C/EBPb,C/EBPa,and FABP4.Moreover,PAQG increased the levels of FFA and glycerol in medium and reduced TG level in mature adipocytes.Interestingly,PAQG not only promoted the activation of AMPK and genes involved in fatty oxidation including PDK4 and CPT1a,but also inhibited those genes involved in fatty acid biosynthesis,such as SREBP1c,FAS,ACCa and SCD1.In conclusion,PAQG inhibits the differentiation and regulates lipid metabolism of 3T3-L1 cells via AMPK pathway,suggesting that PAQG may be a novel and promising natural product for the treatment of obesity and hyperlipidemia.Li Peng Yanting Lu Yuhui Xu Jing Hu Fang Wang Yumei Zhang Wenyong Xiong 2017Natural Products and Bioprospecting2017,7,3:0
16Robust Benchmark Structural Variant Calls of An Asian Using State-of-the-art Long-read Sequencing Technologies显示文摘The importance of structural variants(SVs)for human phenotypes and diseases is now recognized.Although a variety of SV detection platforms and strategies that vary in sensitivity and specificity have been developed,few benchmarking procedures are available to confidently assess their performances in biological and clinical research.To facilitate the validation and application of these SV detection approaches,we established an Asian reference material by characterizing the genome of an Epstein-Barr virus(EBV)-immortalized B lymphocyte line along with identified benchmark regions and high-confidence SV calls.We established a high-confidence SV callset with 8938 SVs by integrating four alignment-based SV callers,including 109×Pacific Bio sciences(PacBio)continuous long reads(CLRs),22×PacBio circular consensus sequencing(CCS)reads,104×Oxford Nanopore Technologies(ONT)long reads,and 114×Bionano optical mapping platform,and one de novo assembly-based SV caller using CCS reads.A total of 544 randomly selected SVs were validated by PCR amplification and Sanger sequencing,demonstrating the robustness of our SV calls.Combining trio-binning-based haplotype assemblies,we established an SV benchmark for identifying false negatives and false positives by constructing the continuous high-confidence regions(CHCRs),which covered 1.46 gigabase pairs(Gb)and 6882 SVs supported by at least one diploid haplotype assembly.Establishing high-confidence SV calls for a benchmark sample that has been characterized by multiple technologies provides a valuable resource for investigating SVs in human biology,disease,and clinical research.Xiao Du Lili Li Fan Liang Sanyang Liu Wenxin Zhang Shuai Sun Yuhui Sun Fei Fan Linying Wang Xinming Liang Weijin Qiu Guangyi Fan Ou Wang Weifei Yang Jiezhong Zhang Yuhui Xiao Yang Wang Depeng Wang Shoufang Qu Fang Chen Jie Huang 2022Genomics, Proteomics & Bioinformatics2022,20,1:0
