Chinese deserts and sand fields in Last Glacial Maximum and Holocene Optimum显示文摘The Last Glacial Maximum (LGM, c. 26-16 ka) and the Holocene Optimum (HO, c. 9-5 ka) were characterized by cold-dry and warm-wet climates respectively in the recently geological Earth. How Chinese deserts and sand fields responded to these distinctive climatic changes is still not clear, however. To reconstruct environments of the deserts and sand fields during the LGM and HO is helpful to understand the forcing mechanisms of environment change in this arid region, and to test paleoclimatic modeling results. Through our long-term field and laboratory investigations, 400 optically stimulated luminescence (OSL) ages and more than 100 depositional records in the Chinese deserts and sand fields were obtained; on the basis of these data, we reconstruct spatial distributions of the deserts and sand fields during the LGM and HO. Our results show that the sand fields of Mu Us, Hunshandake, Horqin and Hulun Buir in northern and northeastern China had expanded 25%, 37%, 38% and 270%, respectively, during the LGM; the sand fields of Gonghe in the northeastern Qinghai-Tibetan Plateau had expanded 20%, and the deserts of Badain Jaran, Tengger in central northern China had expanded 39% and 29% separately during the LGM; the deserts of Taklimakan, Gurbantünggüt and Kumtag in northwestern China had expanded 10%-20% respectively, compared to their modern areas. On the other hand, all of the sand fields were nearly completely covered by vegetation during the HO; the deserts in northwestern and central northern China were reduced by around 5%-20% in area during this time. Lakes in this arid region were probably expanded during the HO but this conclusion needs more investigation. Compared with the geological distributions of deserts and sand fields, human activity has clearly changed (expanded) the area of active sand dunes at the present time. Our observations show that environmental conditions of Chinese deserts and sand fields are controlled by regional climate together with human activity. | LU HuaYu YI ShuangWen XU ZhiWei ZHOU YaLi ZENG Lin ZHU FangYing FENG Han DONG LiNa ZHUO HaiXin YU KaiFeng MASON Joseph WANG XiaoYong CHEN YingYong LU Qi WU Bo DONG ZhiBao QU JianJun WANG XunMing GUO ZhengTang | 2013 | Chinese Science Bulletin2013,58,23: | 40

Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously. Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians, pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. Moreover, it has prepared NGS criteria, the sequencing standard operation procedure(SOP), data analysis, report, and NGS platform certification and validation. | Xuchao Zhang Zhiyong Liang Shengyue Wang Shun Lu Yong Song Ying Cheng Jianming Ying Weiping Liu Yingyong Hou Yangqiu Li Yi Liu Jun Hou Xiufeng Liu Jianyong Shao Yanhong Tai Zheng Wang Li Fu Hui Li Xiaojun Zhou Hua Bai Mengzhao Wang You Lu Jinji Yang Wenzhao Zhong Qing Zhou Xuening Yang Jie Wang Cheng Huang Xiaoqing Liu Xiaoyan Zhou Shirong Zhang Hongxia Tian Yu Chen Ruibao Ren Ning Liao Chunyan Wu Zhongzheng Zhu Hongming Pan Yanhong Gu Liwei Wang Yunpeng Liu Suzhan Zhang Tianshu Liu Gong Chen Zhimin Shao Binghe Xu Qingyuan Zhang Ruihua Xu Lin Shen Yilong Wu | 2019 | Cancer Biology & Medicine2019,16,1: | 14
Onset of Xiashu loess deposition in southern China by 0.9 Ma and its implications for regional aridification显示文摘The Xiashu loess is a typical Quaternary eolian deposit in southern China and represents an important terrestrial paleoclimate archive in this low-latitude monsoon region. However, the chronological framework of Xiashu loess deposition has yet to be established. Determining the timing of the onset of Xiashu loess deposition will allow researchers to better understand late Quaternary aridification across the Asian continent, the evolution of the East Asian monsoon and regional environmental changes in subtropical regions. Therefore, in this study, a systematic chronological study of the Xiashu loess is conducted to answer this question. For the first time, magnetostratigraphic classification reveals that the Matuyama/Brunhes(M/B) reversal is present in the Xiashu loess at two sites in Jiangsu Province, the Qingshan profile at Yizheng and the Dagang core in Zhenjiang.Based on the results of magnetostratigraphy and optically stimulated luminescence(OSL) dating, the age of the lower boundary of the Xiashu loess is estimated to be approximately 0.9 Ma. Consequently, this Xiashu loess deposit is the oldest reported to date and is comparable in age to the red soil deposit in Xuancheng, Anhui Province. The onset of Xiashu loess deposition by 0.9 Ma represents the further expansion of arid range in Asia in the late Quaternary in response to significant aridification and winter monsoon strengthening in this subtropical region. We suggest that these climate changes were primarily driven by global cooling and an increase in high-latitude ice volume in the Northern Hemisphere and that the initiation of Xiashu loess accumulation was a regional response of southern China to the 0.9 Ma global cooling event. | Xusheng LI Zhiyong HAN Huayu LU Yingyong CHEN Yang LI Xiaokang YUAN Yuwen ZHOU Mengyao JIANG Cunjuan LV | 2018 | Science China Earth Sciences2018,61,3: | 11
Newly discovered Palaeolithic artefacts from loess deposits and their ages in Lantian, central China显示文摘Eight new Palaeolithic open-air sites were identified and 770 stone artefacts were collected from 2009 to 2011 in the Lantian area of the Bahe River valley, central China. Because the famous Homo erectus fossils were unearthed at the Gongwangling and Chenjiawo localities, and more than 30 Palaeolithic open-air sites were investigated in the 1960s in this region, the catchment of Bahe River is regarded as one of the most important hominin sites from the late early Pleistocene to the middle Pleistocene. These eight newly discovered open-air sites are located at the second(n = 6), third(n = 1) or higher terraces(n = 1) of the Bahe River. The Diaozhai section on the second terrace was sampled in detail. Two samples were collected for optically stimulated luminescence dating(OSL). The OSL results suggest that a buried lithic artefact layer at the Diaozhai site spans approximately 70–30 ka. The lithic assemblage analysis suggests that the stone artefacts were made of local pebbles/cobbles such as greywacke, quartz, sandstone and igneous rocks. The main percussion techniques that were used were direct hard hammer percussion and bi-polar techniques. The lithic artefacts comprise hammer stones, cores, flakes, retouched tools and flaking debris. Acheulian-type large cutting tools(LCTs) such as hand-axes, picks and cleavers were indentified in the Lantian region as well. This is the first time Acheulian-type LCTs from the late Pleistocene have been identified in this region. This study distinguishes age gaps between Western world and East Asian Acheulian-type tools. | Shejiang Wang Huayu Lu Hongyan Zhang Xuefeng Sun Shuangwen Yi Yingyong Chen Gaike Zhang Luda Xing Weigang Sun | 2014 | Chinese Science Bulletin2014,59,7: | 7

An invasive zone in human liver cancer identified by Stereo-seq promotes hepatocyte–tumor cell crosstalk, local immunosuppression and tumor progression显示文摘Dissecting and understanding the cancer ecosystem, especially that around the tumor margins, which have strong implications for tumor cell infiltration and invasion, are essential for exploring the mechanisms of tumor metastasis and developing effective new treatments.Using a novel tumor border scanning and digitization model enabled by nanoscale resolution-SpaTial Enhanced REsolution Omics-sequencing(Stereo-seq),we identified a 500µm-wide zone centered around the tumor border in patients with liver cancer, referred to as“the invasive zone”.We detected strong immunosuppression, metabolic reprogramming, and severely damaged hepatocytes in this zone.We also identified a subpopulation of damaged hepatocytes with increased expression of serum amyloid A1 and A2(referred to collectively as SAAs)located close to the border on the paratumor side.Overexpression of CXCL6 in adjacent malignant cells could induce activation of the JAK-STAT3 pathway in nearby hepatocytes, which subsequently caused SAAs’overexpression in these hepatocytes.Furthermore, overexpression and secretion of SAAs by hepatocytes in the invasive zone could lead to the recruitment of macrophages and M2 polarization, further promoting local immunosuppression, potentially resulting in tumor progression.Clinical association analysis in additional five independent cohorts of patients with primary and secondary liver cancer(n=423)showed that patients with overexpression of SAAs in the invasive zone had a worse prognosis.Further in vivo experiments using mouse liver tumor models in situ confirmed that the knockdown of genes encoding SAAs in hepatocytes decreased macrophage accumulation around the tumor border and delayed tumor growth. The identification and characterization of a novel invasive zone in human cancer patients not only add an important layer of understanding regarding the mechanisms of tumor invasion and metastasis, but may also pave the way for developing novel therapeutic strategies for advanced liver cancer and other solid tumors. | Liang Wu Jiayan Yan Yinqi Bai Feiyu Chen Xuanxuan Zou Jiangshan Xu Ao Huang Liangzhen Hou Yu Zhong Zehua Jing Qichao Yu Xiaorui Zhou Zhifeng Jiang Chunqing Wang Mengnan Cheng Yuan Ji Yingyong Hou Rongkui Luo Qinqin Li Liang Wu Jianwen Cheng Pengxiang Wang Dezhen Guo Waidong Huang Junjie Lei Shang Liu Yizhen Yan Yiling Chen Sha Liao Yuxiang Li Haixiang Sun Na Yao Xiangyu Zhang Shiyu Zhang Xi Chen Yang Yu Yao Li Fengming Liu Zheng Wang Shaolai Zhou Huanming Yang Shuang Yang Xun Xu Longqi Liu Qiang Gao Zhaoyou Tang Xiangdong Wang Jian Wang Jia Fan Shiping Liu Xinrong Yang Ao Chen Jian Zhou | 2023 | Cell Research2023,33,8: | 1

Identification of TAZ as the essential molecular switch in orchestrating SCLC phenotypic transition and metastasis显示文摘Small-cell lung cancer(SCLC)is a recalcitrant cancer characterized by high metastasis. However, the exact cell type contributing to metastasis remains elusive.Using a Rb1^(L/L)/Trp53^(L/L) mouse model,we identify the NCAM^(hi)CD44^(lo/–) subpopulation as the SCLC metastasizing cell(SMC),which is progressively transitioned from the non-metastasizing NCAM^(lo)CD44^(hi) cell(non-SMC).Integrative chromatin accessibility and gene expression profiling studies reveal the important role of the SWI/SNF complex, and knockout of its central component,Brg1,significantly inhibits such phenotypic transition and metastasis.Mechanistically,TAZ is silenced by the SWI/SNF complex during SCLC malignant progression, and its knockdown promotes SMC transition and metastasis.Importantly, ectopic TAZ expression reversely drives SMC-to-non-SMC transition and alleviates metastasis.Single-cell RNA-sequencing analyses identify SMC as the dominant subpopulation in human SCLC metastasis, and immunostaining data show a positive correlation between TAZ and patient prognosis. These data uncover high SCLC plasticity and identify TAZ as the key molecular switch in orchestrating SCLC phenotypic transition and metastasis. | Yujuan Jin Qiqi Zhao Weikang Zhu Yan Feng Tian Xiao Peng Zhang Liyan Jiang Yingyong Hou Chenchen Guo Hsinyi Huang Yabin Chen Xinyuan Tong Jiayu Cao Fei Li Xueliang Zhu Jun Qin Dong Gao Xin-Yuan Liu Hua Zhang Luonan Chen Roman KThomas Kwok-Kin Wong Lei Zhang Yong Wang Liang Hu Hongbin Ji | 2022 | National Science Review2022,9,7: | 0
