1MiR-30a Positively Regulates the Inflammatory Response of Microglia in Experimental Autoimmune Encephalomyelitis显示文摘Multiple sclerosis(MS) is a classical inflammatory demyelinating disease of the central nervous system(CNS). Microglia are the main resident immune cells in the CNS and are closely associated with the pathogenesis of MS.In the present study, we found that mi R-30 a was highly expressed in jellyfish-like microglia in chronic active lesions of MS patients, as well as in the microglia of mice with experimental autoimmune encephalomyelitis(EAE) at the chronic phase. In vitro, the conditioned supernatant of mouse microglia overexpressing miR-30 a promoted the apoptosis of oligodendrocyte precursor cells(OPCs), and inhibited OPC differentiation. In vivo, overexpressing miR-30 a in transplanted microglia exacerbated the progression of EAE.Overexpression and knock-down experiments in primary cultured mouse microglia showed that mi R-30 a increased the expression of IL-1 b and i NOS, which are pro-inflammatory, while inhibiting the expression of Ym-1 and CD206.Mechanistically, mi R-30 a inhibited the expression of Ppargc1 b, which is the co-activator of peroxisome proliferator-activated receptor gamma, resulting in pro-inflammatory effects. 3Identification of Differentially-expressed Genes in Intestinal Gastric Cancer by Microarray Analysis显示文摘Gastric cancer(GC) is one of the most frequent malignant tumors. In order to systematically characterize the cellular and molecular mechanisms of intestinal GC development,in this study, we used 22 K oligonucleotide microarrays and bioinformatics analysis to evaluate the gene expression profiles of GC in 45 tissue samples, including 20 intestinal GC tissue samples,20 normal appearing tissues(NATs) adjacent to tumors and 5 noncancerous gastric mucosa tissue samples. These profiles allowed us to explore the transcriptional characteristics of GC and determine the change patterns in gene expression that may be of clinical significance. 1519 and 1255 differentially-expressed genes(DEGs) were identified in intestinal GC tissues and NATs, respectively,as determined by Bayesian analysis(P < 0.001). 10Screening responsive or resistant biomarkers of immune checkpoint inhibitors based on online databases显示文摘Immune checkpoint inhibitors are a promising strategy in the treatment of cancer, especially advanced types. However, not all patients are responsive to immune checkpoint inhibitors. The response rate depends on the immune microenvironment, tumor mutational burden (TMB), expression level of immune checkpoint proteins, and molecular subtypes of cancers. Along with the Cancer Genome Project, various open access databases, including The Cancer Genome Atlas and Gene Expression Omnibus, provide large volumes of data, which allow researchers to explore responsive or resistant biomarkers of immune checkpoint inhibitors? 15A quick and reliable image-based AI algorithm for evaluating cellular senescence of gastric organoids显示文摘Objective:Organoids are a powerful tool with broad application prospects in biomedicine.Notably,they provide alternatives to animal models for testing potential drugs before clinical trials.However,the number of passages for which organoids maintain cellular vitality ex vivo remains unclear.Methods:Herein,we constructed 55 gastric organoids from 35 individuals,serially passaged the organoids,and captured microscopic images for phenotypic evaluation.Senescence-associatedβ-galactosidase(SA-β-Gal),cell diameter in suspension,and gene expression reflecting cell cycle regulation were examined.The YOLOv3 object detection algorithm integrated with a convolutional block attention module(CBAM)was used to evaluate organoid vitality.Results:SA-β-Gal staining intensity;single-cell diameter;and expression of p15,p16,p21,CCNA2,CCNE2,and LMNB1 reflected the progression of aging in organoids during passaging.The CBAM-YOLOv3 algorithm precisely evaluated aging organoids on the basis of organoid average diameter,organoid number,and number×diameter,and the findings positively correlated with SA-β-Gal staining and single-cell diameter.Organoids derived from normal gastric mucosa had limited passaging ability(passages 1–5),before aging,whereas tumor organoids showed unlimited passaging potential for more than 45 passages(511 days)without showing clear senescence.Conclusions:Given the lack of indicators for evaluating organoid growth status,we established a reliable approach for integrated analysis of phenotypic parameters that uses an artificial intelligence algorithm to indicate organoid vitality.This method enables precise evaluation of organoid status in biomedical studies and monitoring of living biobanks.Ruixin Yang Yutong Du Wingyan Kwan Ranlin Yan Qimeng Shi Lu Zang Zhenggang Zhu Jianming Zhang Chen Li Yingyan Yu 2023Cancer Biology & Medicine2023,20,7:1
16Multi-target combinatory strategy to overcome tumor immune escape显示文摘Immune therapy has become the fourth approach after surgery,chemotherapy,and radiotherapy in cancer treatment.Many immune checkpoints were identified in the last decade since ipilimumab,which is the first immune checkpoint inhibitor to cytotoxic T-lymphocyte associated protein 4,had been approved by the US Food and Drug Administration(FDA)for the treatment of unresectable or metastatic melanoma in 2011.The use of several antibody drugs that target PD1/PD-L1 for various cancer treatments has been approved by the FDA.However,fewer people are benefitting from immune checkpoint inhibitor treatment in solid cancers.Approximately 80%of patients do not respond appropriately because of primary or acquired therapeutic resistance.Along with the characterization of more immune checkpoints,the combinatory treatment of multi-immune checkpoint inhibitors becomes a new option when monotherapy could not receive a good response.In this work,the author focuses on the combination therapy of multiple immune checkpoints(does not include targeted therapy of oncogenes or chemotherapy),introduces the current progression of multiple immune checkpoints and their related inhibitors,and discusses the advantages of combination therapy,as well as the risk of immune-related adverse events.Yingyan Yu 2022Frontiers of Medicine2022,16,2:1
