1A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 2A cyclodextrin-based nanoformulation achieves co-delivery of ginsenoside Rg3 and quercetin for chemo-immunotherapy in colorectal cancer显示文摘The immune checkpoint blockade therapy has profoundly revolutionized the field of cancer immunotherapy. However, despite great promise for a variety of cancers, the efficacy of immune checkpoint inhibitors is still low in colorectal cancer(CRC). This is mainly due to the immunosuppressive feature of the tumor microenvironment(TME). Emerging evidence reveals that certain chemotherapeutic drugs induce immunogenic cell death(ICD), demonstrating great potential for remodeling the immunosuppressive TME. In this study, the potential of ginsenoside Rg3(Rg3) as an ICD inducer against CRC cells was confirmed using in vitro and in vivo experimental approaches. The ICD efficacy of Rg3 could be significantly enhanced by quercetin(QTN) that elicited reactive oxygen species(ROS). To amelioratein vivo delivery barriers associated with chemotherapeutic drugs, a folate(FA)-targeted polyethylene glycol(PEG)-modified amphiphilic cyclodextrin nanoparticle(NP) was developed for co-encapsulation of Rg3 and QTN. The resultant nanoformulation(CD-PEG-FA.Rg3.QTN) significantly prolonged blood circulation and enhanced tumor targeting in an orthotopic CRC mouse model, resulting in the conversion of immunosuppressive TME. Furthermore, the CD-PEG-FA.Rg3.QTN achieved significantly longer survival of animals in combination with Anti-PD-L1. 3Precision integration of grating-based polarizers onto focal plane arrays of near-infrared photovoltaic detectors for enhanced contrast polarimetric imaging显示文摘Polarimetric imaging enhances the ability to distinguish objects from a bright background by detecting their particular polarization status,which offers another degree of freedom in infrared remote sensing.However,to scale up by monolithically integrating grating-based polarizers onto a focal plane array(FPA)of infrared detectors,fundamental technical obstacles must be overcome,including reductions of the extinction ratio by the misalignment between the polarizer and the detector,grating line width fluctuations,the line edge roughness,etc.This paper reports the authors’latest achievements in overcoming those problems by solving key technical issues regarding the integration of large-scale polarizers onto the chips of FPAs with individual indium gallium arsenide/indium phosphide(In Ga As/In P)sensors as the basic building blocks.Polarimetric and photovoltaic chips with divisions of the focal plane of 540×4 pixels and 320×256 superpixels have been successfully manufactured.Polarimetric imaging with enhanced contrast has been demonstrated.The progress made in this work has opened up a broad avenue toward industrialization of high quality polarimetric imaging in infrared wavelengths.Bo Feng Yifang Chen Duo Sun Zongyao Yang Bo Yang Xue Li Tao Li 2021International Journal of Extreme Manufacturing2021,3,3:4
4High-throughput screening identifies established drugs as SARS-CoV-2 PLpro inhibitors显示文摘A new coronavirus(SARS-CoV-2)has been identified as the etiologic agent for the COVID-19 outbreak.Currently,effective treatment options remain very limited for this disease;therefore,there is an urgent need to identify new anti-COVID-19 agents.In this study,we screened over 6,000 compounds that included approved drugs,drug candidates in clinical trials,and pharmacologically active compounds to identify leads that target the SARS-CoV-2 papain-like protease(PLpro).Together with main protease(Mpro),PLpro is responsible for processing the viral replicase polyprotein into functional units.There-fore,it is an attractive target for antiviral drug develop-ment.Here we discovered four compounds,YM155,cryptotanshinone,tanshinone I and GRL0617 that inhibit SARS-CoV-2 PLpro with IC50 values ranging from 1.39 to 5.63 pmol/L.These compounds also exhibit strong antiviral activities in cell-based assays.YM155,an anti-cancer drug candidate in clinical trials,has the most potent antiviral activity with an EC50 value of 170 nmol/L.In addition,we have determined the crystal structures of this enzyme and its complex with YM155,revealing a unique binding mode.YM155 simultaneously targets three'hot'spots on PLpro,including the substrate-binding pocket,the interferon stimulating gene product 15(ISG15)binding site and zinc finger motif.Our results demonstrate the efficacy of this screening and repur-posing strategy,which has led to the discovery of new drug leads with clinical potential for COVID-19 treatments.Yao Zhao Xiaoyu Du Yinkai Duan Xiaoyan Pan Yifang Sun Tian You Lin Han Zhenming Jin Weijuan Shang Jing Yu Hangtian Guo Qianying Liu Yan Wu Chao Peng Jun Wang Chenghao Zhu Xiuna Yang Kailin Yang Ying Lei Luke W.Guddat Wenqing Xu Gengfu Xiao Lei Sun Leike Zhang Zihe Rao Haitao Yang 2021Protein & Cell2021,12,11:4
5Exploring the design space of immersive urban analytics显示文摘Recent years have witnessed the rapid development and wide adoption of immersive head-mounted devices,such as HTC VIVE,Oculus Rift,and Microsoft HoloLens.These immersive devices have the potential to significantly extend the methodology of urban visual analytics by providing critical 3D context information and creating a sense of presence.In this paper,we propose a theoretical model to characterize the visualizations in immersive urban analytics.Furthermore,based on our comprehensive and concise model,we contribute a typology of combination methods of 2D and 3D visualizations that distinguishes between linked views,embedded views,and mixed views.We also propose a supporting guideline to assist users in selecting a proper view under certain circumstances by considering visual geometry and spatial distribution of the 2D and 3D visualizations.Finally,based on existing work,possible future research opportunities are explored and discussed.Zhutian Chen Yifang Wang Tianchen Sun Xiang Gao Wei Chen Zhigeng Pan Huamin Qu Yingcai Wu 2017Visual Informatics2017,1,2:3

17Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 18Sustained release of vascular endothelial growth factor A and basic fibroblast growth factor from nanofiber membranes reduces oxygen/glucose deprivation-induced injury to neurovascular units显示文摘Upregulation of vascular endothelial growth factor A/basic fibroblast growth factor(VEGFA/b FGF)expression in the penumbra of cerebral ischemia can increase vascular volume,reduce lesion volume,and enhance neural cell proliferation and differentiation,thereby exerting neuroprotective effects.However,the beneficial effects of endogenous VEGFA/b FGF are limited as their expression is only transiently increased.In this study,we generated multilayered nanofiber membranes loaded with VEGFA/b FGF using layer-by-layer self-assembly and electrospinning techniques.We found that a membrane containing 10 layers had an ideal ultrastructure and could efficiently and stably release growth factors for more than 1 month.This 10-layered nanofiber membrane promoted brain microvascular endothelial cell tube formation and proliferation,inhibited neuronal apoptosis,upregulated the expression of tight junction proteins,and improved the viability of various cellular components of neurovascular units under conditions of oxygen/glucose deprivation.Furthermore,this nanofiber membrane decreased the expression of Janus kinase-2/signal transducer and activator of transcription-3(JAK2/STAT3),Bax/Bcl-2,and cleaved caspase-3.Therefore,this nanofiber membrane exhibits a neuroprotective effect on oxygen/glucose-deprived neurovascular units by inhibiting the JAK2/STAT3 pathway.Yifang Wu Jun Sun Qi Lin Dapeng Wang Jian Hai 2024Neural Regeneration Research2024,19,4:0
19Orogenic Movements during the Paleozoic Period: Development of the Granitoid Formations in the Northwestern Region of Spain’s Iberian Peninsula显示文摘In the present study,Paleozoic Variscan orogenesis was a model of the oroclinal flexion accompanied by extensive magmatism,which could be divided into the following two types:post-tectonic and syn-tectonic tonalite granite,and leuco-granite which were controlled by the tectonic characteristics of the intrusions.It was observed that a very high majority of the samples had displayed discontinuities in their structures,that were later utilized to define the granitoid morphology and development characteristics of the rock during the intrusion phases.Furthermore,it was determined that the tectonics associated with the Alpine orogeny results in the new generation of faults and fractures during the Paleogene Period had produced the development of the Sierras.Due to different weathering processes,the depressions which had resulted in the present granitoid reliefs were found to be exclusively related to the structural development processes during the geological history(either tectonic or magmatic)of the granite,and not as normally interpreted.Juan Ramon Vidal Romani Zhaojun Song Huimin Liu Yifang Sun Haonan Li 2020Journal of Earth Science2020,31,3:0
20Synthesis and optical properties of novel apatite-type NaCa_(3)Bi(PO_(4))_(3)F:Dy^(3+)yellow-emitting fluorophosphate phosphors for white LEDs显示文摘In this study,novel yellow-emitting fluorophosphate NaCa_(3)Bi(PO_(4))_(3)F phosphors doped with different concentrations of Dy^(3+)ions were first obtained via high-temperature solid-state reaction.The crystal structure,phase purity,particle morphology,photoluminescence(PL)properties,thermal stability,and luminescence decay curves of the resulting phosphors were then characterized in detail.Under the excitation of 349 nm,the three dominant peaks of the NaCa_(3)Bi(PO_(4))_(3)F:Dy^(3+)are centered at 480 nm(^(4)F_(9/2)-^(6)H_(15/2)),577 nm(^(4)F_(9/2)-^(6)H_(13/2)),and 662 nm(^(4)F_(9/2)-^(6)H_(11/2)).The optimal doping concentration of Dy^(3+)ions in the NaCa_(3)Bi(PO_(4))_(3)F:xDy^(3+)phosphors is x=5 mol%.The phosphors show excellent thermal stability with high activation energy(Ea=0.32 eV).Eventually,the synthesized white lightemitting diode(w-LED)demonstrates the Commission International de L’Eclairage(CIE)chromaticity coordinates of(0.341,0.334),a good correlated color temperature(CCT)of 5083 K,and a high color rendering index(Ra)of 92.Revealing its potential as yellow-emitting phosphors,the feasibility of the fabricated apatite-type NaCa_(3)Bi(PO_(4))_(3)F:Dy^(3+)fluorophosphate phosphors was confirmed for wLEDs.Yuan Wang Guangliang Lu Yang Qiu Wen Sun Siqi Qin Yifang Lin Bin Deng Dan zhang Ruijin Yu 2022Journal of Rare Earths2022,40,12:0
