2Rapid determination of pesticide residues in Chinese materia medica using QuEChERS sample preparation followed by gas chromatography-mass spectrometry显示文摘Pesticide residue analysis plays an important role in the quality control of Chinese materia medica.This paper reports the development and validation of an analytical method for the quantitative determination of the residues of 39 pesticides in 12 different matrices of Chinese materia medica.Sample preparation utilized the QuEChERS method with acetonitrile:1% aqueous acetic acid(9:1,v/v)as extraction solvent followed by sample clean-up by dispersive solid phase extraction using primary secondary amine sorbent and graphitized carbon black.Extracts were then analysed by gas chromatography coupled with electron impact mass spectrometry in the selected ion monitoring mode.Limit of detection(LOD)and limit of quantitation(LOQ)values were in the ranges 0.5-50 ng/g and 1-100 ng/g,respectively.The recoveries of the 39 pesticides were in the range 75-112% with precision(as relative standard deviation,RSD)<15%.The results show that the modified QuEChERS method allows rapid and sensitive analysis of multiple pesticide residues in Chinese materia medica.Yichen Hu Li Wan Jinming Zhang Fang Yang Jiliang Cao 2012Acta Pharmaceutica Sinica B2012,2,3:11

4Predictive factors associated with gefitinib response in patients with advanced non-small-cell lung cancer(NSCLC)显示文摘Purpose: A number of different clinical characteristics have been reported to singly correlate with therapeutic activity of epidermal growth factor receptor(EGFR) tyrosine kinase inhibitors(TKIs) in advanced non-small-cell lung cancer(NSCLC). This study aimed to identify predictive factors associated with prognostic benefits of gefitinib.Patients and methods: EGFR gene typing in 33 advanced NSCLC patients received gefitinib(250 mg/day) were analyzed with mutant-enriched PCR assay. Gefitinib response was evaluated with potential predictive factors retrospectively.Results: The overall objective response rate(ORR) and median progression-free survival(PFS) in the 33 patients treated by gefitinib were 45.5% and 3.0(2.0-4.0) months. The ORR and median PFS in EGFR gene mutation patients were significantly higher/longer than those in EGFR gene wild-type patients(P<0.01). Similarly, the ORR and median PFS in non-smoker patients were significantly higher/longer than those in smoker patients(P<0.05, P<0.01, respectively). However, no difference for ORR and median PFS occurred between male and female patients. Logistic multivariate analysis showed that only EGFR mutated gene was significantly associated with the ORR(P<0.01). Both EGFR mutated gene and non-smoker were the major factors that contributed to PFS(P<0.05). 5Au nanorods decorated TiO_(2)nanobelts with enhanced full solar spectrum photocatalytic antibacterial activity and the sterilization file cabinet application显示文摘TiO_(2)photocatalysts have been widely studied and applied for removing bacteria,but its antibacterial efficiency is limited to the ultraviolet(UV)range of the solar spectrum.In this work,we use the gold(Au)nanorods to enhance the visible and near-infrared(NIR)light absorption of TiO_(2)NBs,a typical UV light photocatalyst,thus the enhancement of its full solar spectrum(UV,visible and NIR)photocatalytic antibacterial properties is achieved.Preliminary surface plasmon resonance(SPR)enhancement photocatalytic antibacterial mechanism is suggested.On one hand,transverse and longitudinal SPR of Au NRs is beneficial for visible and NIR light utilization.On the other hand,Au NRs combined with TiO_(2)NBs to form the heterostructure,which can improve the photogenerated carrier separation and direct electron transfer increases the hot electron concentration while Au NRs as the electron channel can well restrain charge recombination.finally produces the high yield of radical oxygen species and exhibits a superior antibacterial efficiency.Furthermore,we design a sterilization file cabinet with Au NR/TiO_(2)NB heterostructures as the photocatalytic coating plates.Our study reveals that Au NR/TiO_(2)NB heterostructure is a potential candidate for sterilization of bacteria and archives protection.Yingying Qin Yichen Guo Zhangqian Liang Yanjun Xue Xiaoli Zhang Lei Yang Jian Tian 2021Chinese Chemical Letters2021,32,4:6
63D printed silk-gelatin hydrogel scaffold with different porous structure and cell seeding strategy for cartilage regeneration显示文摘Hydrogel scaffolds are attractive for tissue defect repair and reorganization because of their human tissue-like characteristics.However,most hydrogels offer limited cell growth and tissue formation ability due to their submicron-or nano-sized gel networks,which restrict the supply of oxygen,nutrients and inhibit the proliferation and differentiation of encapsulated cells.In recent years,3D printed hydrogels have shown great potential to overcome this problem by introducing macro-pores within scaffolds.In this study,we fabricated a macroporous hydrogel scaffold through horseradish peroxidase(HRP)-mediated crosslinking of silk fibroin(SF)and tyramine-substituted gelatin(GT)by extrusion-based low-temperature 3D printing.Through physicochemical characterization,we found that this hydrogel has excellent structural stability,suitable mechanical properties,and an adjustable degradation rate,thus satisfying the requirements for cartilage reconstruction.Cell suspension and aggregate seeding methods were developed to assess the inoculation efficiency of the hydrogel.Moreover,the chondrogenic differentiation of stem cells was explored.Stem cells in the hydrogel differentiated into hyaline cartilage when the cell aggregate seeding method was used and into fibrocartilage when the cell suspension was used.Finally,the effect of the hydrogel and stem cells were investigated in a rabbit cartilage defect model.After implantation for 12 and 16 weeks,histological evaluation of the sections was performed.We found that the enzymatic cross-linked and methanol treatment SF5GT15 hydrogel combined with cell aggregates promoted articular cartilage regeneration.In summary,this 3D printed macroporous SF-GT hydrogel combined with stem cell aggregates possesses excellent potential for application in cartilage tissue repair and regeneration.Qingtao Li Sheng Xu Qi Feng Qiyuan Dai Longtao Yao Yichen Zhang Huichang Gao Hua Dong Dafu Chen Xiaodong Cao 2021Bioactive Materials2021,6,10:6
7Cell sheet formation enhances the therapeutic effects of human umbilical cord mesenchymal stem cells on myocardial infarction as a bioactive material显示文摘Stem cell-based therapy has been used to treat ischaemic heart diseases for two decades.However,optimal cell types and transplantation methods remain unclear.This study evaluated the therapeutic effects of human umbilical cord mesenchymal stem cell(hUCMSC)sheet on myocardial infarction(MI).Methods:hUCMSCs expressing luciferase were generated by lentiviral transduction for in vivo bio-luminescent imaging tracking of cells.We applied a temperature-responsive cell culture surface-based method to form the hUCMSC sheet.Cell retention was evaluated using an in vivo bio-luminescent imaging tracking system.Unbiased transcriptional profiling of infarcted hearts and further immunohistochemical assessment of monocyte and macrophage subtypes were used to determine the mechanisms underlying the therapeutic effects of the hUCMSC sheet.Echocardiography and pathological analyses of heart sections were performed to evaluate cardiac function,angiogenesis and left ventricular remodelling.Results:When transplanted to the infarcted mouse hearts,hUCMSC sheet significantly improved the retention and survival compared with cell suspension.At the early stage of MI,hUCMSC sheet modulated inflammation by decreasing Mcp1-positive monocytes and CD68-positive macrophages and increasing Cx3cr1-positive non-classical macrophages,preserving the cardiomyocytes from acute injury.Moreover,the extracellular matrix produced by hUCMSC sheet then served as bioactive scaffold for the host cells to graft and generate new epicardial tissue,providing mechanical support and routes for revascularsation.These effects of hUCMSC sheet treatment significantly improved the cardiac function at days 7 and 28 post-MI.Conclusions:hUCMSC sheet formation dramatically improved the biological functions of hUCMSCs,mitigating adverse post-MI remodelling by modulating the inflammatory response and providing bioactive scaffold upon transplantation into the heart.Translational perspective:Due to its excellent availability as well as superior local cellular retention and survival,allogenic transplantation of hUCMSC sheets can more effectively acquire the biological functions of hUCMSCs,such as modulating inflammation and enhancing angiogenesis.Moreover,the hUCMSC sheet method allows the transfer of an intact extracellular matrix without introducing exogenous or synthetic biomaterial,further improving its clinical applicability.Rui Guo Feng Wan Masatoshi Morimatsu Qing Xu Tian Feng Hang Yang Yichen Gong Shuhong Ma Yun Chang Siyao Zhang Youxu Jiang Heqing Wang Dehua Chang Hongjia Zhang Yunpeng Ling Feng Lan 2021Bioactive Materials2021,6,9:6
8Subvoxel light-sheet microscopy for high-resolution high-throughput volumetric imaging of large biomedical specimens显示文摘A key challenge when imaging whole biomedical specimens is how to quickly obtain massive cellular information over a large field of view(FOV).We report a subvoxel light-sheet microscopy(SLSM)method enabling high-throughput volumetric imaging of mesoscale specimens at cellular resolution.A nonaxial,continuous scanning strategy is developed to rapidly acquire a stack of large-FOV images with three-dimensional(3-D)nanoscale shifts encoded.Then,by adopting a subvoxel-resolving procedure,the SLSM method models these low-resolution,cross-correlated images in the spatial domain and can iteratively recover a 3-D image with improved resolution throughout the sample.This technique can surpass the optical limit of a conventional light-sheet microscope by more than three times,with high acquisition speeds of gigavoxels per minute.By fast reconstruction of 3-D cultured cells,intact organs,and live embryos,SLSM method presents a convenient way to circumvent the trade-off between mapping large-scale tissue(>100 mm3)and observing single cell(∼1-μm resolution).It also eliminates the need of complicated mechanical stitching or modulated illumination,using a simple light-sheet setup and fast graphics processing unit-based computation to achieve high-throughput,high-resolution 3-D microscopy,which could be tailored for a wide range of biomedical applications in pathology,histology,neuroscience,etc.Peng Fei Jun Nie Juhyun Lee Yichen Ding Shuoran Li Hao Zhang Masaya Hagiwara Tingting Yu Tatiana Segura Chih-Ming Ho Dan Zhu Tzung KHsiaic 2019Advanced Photonics2019,1,1:6
9Hedgehog signaling in gastrointestinal carcinogenesis and the gastrointestinal tumor microenvironment显示文摘The Hedgehog(HH) signaling pathway plays important roles in gastrointestinal carcinogenesis and the gastrointestinal tumor microenvironment(TME). Aberrant HH signaling activation may accelerate the growth of gastrointestinal tumors and lead to tumor immune tolerance and drug resistance.The interaction between HH signaling and the TME is intimately involved in these processes, for example, tumor growth, tumor immune tolerance, inflammation, and drug resistance. Evidence indicates that inflammatory factors in the TME, such as interleukin 6(IL-6) and interferon-g(IFN-g), macrophages, and T cell-dependent immune responses, play a vital role in tumor growth by affecting the HH signaling pathway. Moreover, inhibition of proliferating cancer-associated fibroblasts(CAFs) and inflammatory factors can normalize the TME by suppressing HH signaling. Furthermore, aberrant HH signaling activation is favorable to both the proliferation of cancer stem cells(CSCs) and the drug resistance of gastrointestinal tumors. 103D Bioprinting:A Novel Avenue for Manufacturing Tissues and Organs显示文摘Three-dimensional(3D)bioprinting is a rapidly growing technology that has been widely used in tissue engineering,disease studies,and drug screening.It provides the unprecedented capacity of depositing various types of biomaterials,cells,and biomolecules in a layer-by-layer fashion,with precisely controlled spatial distribution.This technology is expected to address the organ-shortage issue in the future.In this review,we first introduce three categories of 3D bioprinting strategies:inkjet-based printing(IBP),extrusion-based printing(EBP),and light-based printing(LBP).Biomaterials and cells,which are normally referred to as“bioinks,”are then discussed.We also systematically describe the recent advancements of 3D bioprinting in fabricating cell-laden artificial tissues and organs with solid or hollow structures,including cartilage,bone,skin,muscle,vascular network,and so on.The development of organs-onchips utilizing 3D bioprinting technology for drug discovery and toxicity testing is reviewed as well.Finally,the main challenges in current studies and an outlook of the future research of 3D bioprinting are discussed.Bin Zhang Lei Gao Liang Ma Yichen Luo Huayong Yang Zhanfeng Cui 2019Engineering2019,5,4:5
11Facile one-step synthesis of graphitic carbon nitride-modified biochar for the removal of reactive red 120 through adsorption and photocatalytic degradation显示文摘Converting waste biomass into value-added biochar has been considered as a green and sustainable strategy for resource management and pollution control.In this study,graphitic carbon nitride(g-C_(3)N_(4))modified biochars(BCs)were produced through one-pot pyrolysis of urea and hickory chips in differential ratios at 520℃.The resulting BC/g-C_(3)N_(4)composites were evaluated in laboratory for their physicochemical,adsorptive,and photocatalytic properties.The characterization tests showed the successful synthesis of the BC/g-C_(3)N_(4)composites that introduced g-C_(3)N_(4)structure,N-containing surface func-tional groups,reduced surface area,and better thermal stability to the biochar.After modification,the BC/g-C_(3)N_(4)composites showed better adsorption ability to reactive red 120(RR120)than the pristine BC,due to the strong electrostatic attrition between N-containing functional groups of g-C_(3)N_(4)on biochar surface and anionic RR120.The BC/g-C_(3)N_(4)composites also inherited g-C_(3)N_(4)’s photocatalytic activity,which is visible light responsive to generate free radicals for RR120 degradation.In addition,the composites with higher urea modification ratios were more effective in the degradation of RR120.Overall,this study demonstrates the feasibility and promising potential of combining biochar and photocatalyst for the removal of aqueous dye.Because of the synergistic adsorption and photodegradation ability,BC/g-C_(3)N_(4)composites present a novel and cost-effective solution for the removal of aqueous dye and other photodegradable contaminants under natural conditions.Yulin Zheng Yichen Yang Yue Zhang Weixin Zou Yidan Luo Lin Dong Bin Gao 2019Biochar2019,1,1:5
12Alterations in gut microbiota and metabolites associated with altitude-induced cardiac hypertrophy in rats during hypobaric hypoxia challenge显示文摘The gut microbiota is involved in host responses to high altitude.However,the dynamics of intestinal microecology and their association with altitude-related illness are poorly understood.Here,we used a rat model of hypobaric hypoxia challenge to mimic plateau exposure and monitored the gut microbiome,short-chain fatty acids(SCFAs),and bile acids(BAs)over 28 d.We identified weight loss,polycythemia,and pathological cardiac hypertrophy in hypoxic rats,accompanied by a large compositional shift in the gut microbiota,which is mainly driven by the bacterial families of Prevotellaceae,Porphyromonadaceae,and Streptococcaceae.The aberrant gut microbiota was characterized by increased abundance of the Parabacteroides,Alistipes,and Lactococcus genera and a larger Bacteroides to Prevotella ratio.Trans-omics analyses showed that the gut microbiome was significantly correlated with the metabolic abnormalities of SCFAs and BAs in feces,suggesting an interaction network remodeling of the microbiome-metabolome after the hypobaric hypoxia challenge.Interestingly,the transplantation of fecal microbiota significantly increased the diversity of the gut microbiota,partially inhibited the increased abundance of the Bacteroides and Alistipes genera,restored the decrease of plasma propionate,and moderately ameliorated cardiac hypertrophy in hypoxic rats.Our results provide an insight into the longitudinal changes in intestinal microecology during the hypobaric hypoxia challenge.Abnormalities in the gut microbiota and microbial metabolites contribute to the development of high-altitude heart disease in rats.Zhiyuan Pan Yichen Hu Zongyu Huang Ni Han Yan Li Xiaomei Zhuang Jiye Yin Hui Peng Quansheng Gao Wenpeng Zhang Yong Huang Yujun Cui Yujing Bi Zhenjiang Zech Xu Ruifu Yang 2022Science China(Life Sciences)2022,65,10:5

14Novel HDAC5-interacting motifs of Tbx3 are essential for the suppression of E-cadherin expression and for the promotion of metastasis in hepatocellular carcinoma显示文摘Tbx3,a transcriptional repressor,is essential in the organogenesis of vertebrates,stem cell self-renewal and differentiation,and the carcinogenesis of multiple tumor types.However,the mechanism by which Tbx3 participates in the metastasis of hepatocellular carcinoma(HCC)remains largely unknown.In this study,we show that Tbx3 was dramatically upregulated in clinical HCC samples and that elevated expression of Tbx3 promoted cancer progression.To determine the underlying mechanism,systematic glycine scan mutagenesis and deletion assays were performed.We identified two critical motifs,^(585)LFSYPYT^(591)and^(604)HRH^(606),that contribute to the repression of transcriptional activity.These motifs are also essential for Tbx3 to promote cell migration and metastasis both in vitro and in vivo via the suppression of E-cadherin expression.More importantly,Tbx3 directly interacts with HDAC5 via these motifs,and an HDAC inhibitor blocks Tbx3-mediated cell migration and the downregulation of E-cadherin in HCC.As Tbx3 is involved in the carcinogenesis of multiple types of human cancers,our findings suggest an important target for anticancer drug development.Liang Dong Qi Dong Ying Chen Yichen Li Bao Zhang Fanghang Zhou Xiaoming Lyu George GChen Paul Lai Hsiang-fu Kung Ming-Liang He 2018Signal Transduction and Targeted Therapy2018,3,1:3
15Nickel and indium core-shell co-catalysts loaded silicon nanowire arrays for efficient photoelectrocatalytic reduction of CO_(2) to formate显示文摘Developing an efficient artificial photosynthetic system for transforming carbon dioxide and storing solar energy in the form of chemical bonds is one of the greatest challenges in modern chemistry.However,the limited choice of catalysts with wide light absorption range,long-term stability and excellent selectivity for CO_(2) reduction makes the process sluggish.Here,a core-shell-structured nonnoble-metal Ni@In co-catalyst loaded p-type silicon nanowire arrays(SiNWs)for efficient CO_(2) reduction to formate is demonstrated.The formation rate and Faradaic efficiency of formate over the Ni@In/SiNWs catalyst reach 58μmol h^(-1) cm^(-2) and 87% under the irradiation of one simulated sunlight(AM 1.5 G,100 mW cm^(-2)),respectively,which are about 24 and 12 times those over the pristine SiNWs.The enhanced photoelectrocatalytic performance for CO_(2) reduction is attributed to the rational combination of Ni capable of effectively extracting the photogenerated electrons and In responsible for the selective activation of CO_(2).Wenchao Ma Mingcan Xie Shunji Xie Longfu Wei Yichen Cai Qinghong Zhang Ye Wang 2021Journal of Energy Chemistry2021,30,3:3
16A New Efficient Certificate-Based Signature Scheme显示文摘Certificate-based cryptography is a new kind of public key algorithm, which combines the merits of traditional Public key infrastructure(PKI) and identitybased cryptography. It removes the inherent key escrow problem in the identity-based cryptography and eliminates the certificate revocation problem and third-party queries in the traditional PKI. In this paper, we propose an efficient certificate-based signature scheme based on bilinear pairings. Under the strong security model of certificatebased signature scheme, we prove that our scheme is existentially unforgeable against adaptive chosen message and identity attacks in the random oracle. In our scheme, only two pairing operations are needed in the signing and verification processes. 18Toward the Next Generation of Retinal Neuroprosthesis: Visual Computation with Spikes显示文摘A neuroprosthesis is a type of precision medical device that is intended to manipulate the neuronal signals of the brain in a closed-loop fashion,while simultaneously receiving stimuli from the environment and controlling some part of a human brain or body.Incoming visual information can be processed by the brain in millisecond intervals.The retina computes visual scenes and sends its output to the cortex in the form of neuronal spikes for further computation.Thus,the neuronal signal of interest for a retinal neuroprosthesis is the neuronal spike.Closed-loop computation in a neuroprosthesis includes two stages:encoding a stimulus as a neuronal signal,and decoding it back into a stimulus.In this paper,we review some of the recent progress that has been achieved in visual computation models that use spikes to analyze natural scenes that include static images and dynamic videos.We hypothesize that in order to obtain a better understanding of the computational principles in the retina,a hypercircuit view of the retina is necessary,in which the different functional network motifs that have been revealed in the cortex neuronal network are taken into consideration when interacting with the retina.The different building blocks of the retina,which include a diversity of cell types and synaptic connections-both chemical synapses and electrical synapses(gap junctions)-make the retina an ideal neuronal network for adapting the computational techniques that have been developed in artificial intelligence to model the encoding and decoding of visual scenes.An overall systems approach to visual computation with neuronal spikes is necessary in order to advance the next generation of retinal neuroprosthesis as an artificial visual system.Zhaofei Yu Jian K.Liu Shanshan Jia Yichen Zhang Yajing Zheng Yonghong Tian Tiejun Huang 2020Engineering2020,6,4:3

20Diurnal and Seasonal Variations of Thermal Stratification and Vertical Mixing in a Shallow Fresh Water Lake显示文摘Among several influential factors, the geographical position and depth of a lake determine its thermal structure. In temperate zones, shallow lakes show significant differences in thermal stratification compared to deep lakes. Here,the variation in thermal stratification in Lake Taihu, a shallow fresh water lake, is studied systematically. Lake Taihu is a warm polymictic lake whose thermal stratification varies in short cycles of one day to a few days. The thermal stratification in Lake Taihu has shallow depths in the upper region and a large amplitude in the temperature gradient,the maximum of which exceeds 5°C m–1. The water temperature in the entire layer changes in a relatively consistent manner. Therefore, compared to a deep lake at similar latitude, the thermal stratification in Lake Taihu exhibits small seasonal differences, but the wide variation in the short term becomes important. Shallow polymictic lakes share the characteristic of diurnal mixing. Prominent differences on the duration and frequency of long-lasting thermal stratification are found in these lakes, which may result from the differences of local climate, lake depth, and fetch. A prominent response of thermal stratification to weather conditions is found, being controlled by the stratifying effect of solar radiation and the mixing effect of wind disturbance. Other than the diurnal stratification and convection, the representative responses of thermal stratification to these two factors with contrary effects are also discussed. When solar radiation increases, stronger wind is required to prevent the lake from becoming stratified. A daily average wind speed greater than 6 m s–1 can maintain the mixed state in Lake Taihu. Moreover, wind-induced convection is detected during thermal stratification. Due to lack of solar radiation, convection occurs more easily in nighttime than in daytime.
