| 4 | Single cell analysis reveals inhibition of angiogenesis attenuates the progression of heterotopic ossification in Mkx^(−/−)mice显示文摘Tendon heterotopic ossification(HO)is characterized by bone formation inside tendon tissue,which severely debilitates people in their daily life.Current therapies fail to promote functional tissue repair largely due to our limited understanding of HO pathogenesis.Here,we investigate the pathological mechanism and propose a potential treatment method for HO.Immunofluorescence assays showed that the Mohawk(MKX)expression level was decreased in human tendon HO tissue,coinciding with spontaneous HO and the upregulated expression of osteochondrogenic and angiogenic genes in the tendons of Mkx^(−/−)mice.Single-cell RNA sequencing analyses of wild-type and Mkx^(−/−)tendons identified three cell types and revealed the excessive activation of osteochondrogenic genes during the tenogenesis of Mkx^(−/−)tendon cells.Single-cell analysis revealed that the gene expression program of angiogenesis,which is strongly associated with bone formation,was activated in all cell types during HO.Moreover,inhibition of angiogenesis by the small-molecule inhibitor BIBF1120 attenuated bone formation and angiogenesis in the Achilles tendons of both Mkx mutant mice and a rat traumatic model of HO.These findings provide new insights into the cellular mechanisms of tendon HO and highlight the inhibition of angiogenesis with BIBF1120 as a potential treatment strategy for HO. | Junxin Lin Yuwei Yang Wenyan Zhou Chao Dai Xiao Chen Yuanhao Xie Shan Han Huanhuan Liu Yejun Hu Chenqi Tang Varitsara Bunpetch Dandan Zhang Yishan Chen Xiaohui Zou Di Chen Wanlu Liu Hongwei Ouyang | 2022 | Bone Research2022,10,4: | 1 |
| 5 | Polydopamine directed MnO@C microstructures as electrode for lithium ion battery显示文摘In this work,a facile process was reported to fabricate amorphous carbon-coated MnO micropeanuts(MPs)with 1.8μm in length and 1.0μm in width using hydrothermal reaction followed by heat treatment in the oxygen-free environment.With Mn Cl_2 and KMnO_4 dissolved in the mixture of ethylene glycol and water,MnCO_3 MP precursors were obtained via the hydrothermal reaction with dopamine as surfactant.Then MnCO_3 MP was annealed at 600°C in the N_2 atmosphere and was transformed into MnO MP,and simultaneously the formed polydopamine during the hydrothermal reaction was carbonized to produce amorphous carbon-coating on the MnO MP surface.In contrast,MnCO_3 nanoparticle(NP)precursor was formed without the addition of dopamine and MnO NP agglomerates were prepared after pyrolysis.The carbonization of polydopamine during thermolysis improves the electrical conductivity and thermal stability of the MnO MP and thus its electrochemical performance as electrode materials for lithium ion battery.Hopefully,this facile strategy for fabricating and designing carbon-coated materials would inspire more novel nanostructures and applications thereof. | Delu Li Yejun Zhang Lun Li Feng Hu Hongchao Yang Changhong Wang Qiangbin Wang | 2016 | Science China Chemistry2016,59,1: | 0 |
| 6 | Characterization of Evolutionarily Conserved MicroRNAs in Amphioxus显示文摘Amphioxus is an extant species closest to the ancestry of vertebrates.Observation of microRNA(miRNA) distribution of amphioxus would lend some hints for evolutionary research of vertebrates.In this study,using the publicly available scaffold data of the Florida amphioxus(Branchiostoma floridae) genome,we screened and characterized homologs of miRNAs that had been identified in other species.In total,68 pieces of such homologs were obtained and classified into 33 families.Most of these miRNAs were distributed as clusters in genome.Inter-species comparison showed that many miRNAs,which had been thought as vertebrate-or mammal-specific before,were also present in amphioxus,while some miRNAs that had been considered as protostome-specific before also existed in amphioxus.Compared with ciona,amphioxus had an apparent miRNA gene expansion,but phylogenetic analysis showed that the duplicated miRNAs or clusters of amphioxus had a higher homology level than those duplicated ones in vertebrates. | Lei Wang Lan Jiang Songnian Hu Yejun Wang | 2010 | Genomics, Proteomics & Bioinformatics2010,8,1: | 0 |
