| 1 | Monoamine alterations and rotational asymmetry in a rat model of Parkinson's disease following lateral ventricle transplantation of human amniotic epithelial cells显示文摘BACKGROUND: Human amniotic epithelial cells (HAECs) can differentiate into neurons, astrocytes,and oligodendrocytes. They biologically secrete many active neurotrophins and have the capacity tometabolize dopamine enzymes. These features underlie a theoretical basis for the treatment ofParkinson's disease (PD).OBJECTIVE: To investigate the survival and differentiation of transplanted HAECs in the lateralventricle of PD model rats, and to explore its effect on circling behavior, as well as levels ofdopamine (DA), the metabolite homovanillic acid, dihydroxyphenyl acetic acid,5-hydroxyindoleacetic acid, and 5-hydroxytryptamine in the striatum.DESIGN, TIME AND SETTING: A randomized, controlled, animal study was performed at theInstitute of Biochemistry and Cell Biology, Shanghai Institute for Biological Sciences, ChineseAcademy of Sciences, and Shanghai Celstar Institute of Biotechnology from May 2007 to December2008.MATERIALS: HAECs were derived from the placental chorion following caesarean delivery at theShanghai International Maternal and Child Health Hospital. 6-hydroxydopamine (6-OHDA), andmouse anti-human Vimentin monoclonal antibody were purchased from Sigma, USA; mouseanti-human nestin and tyrosine hydroxylase (TH) monoclonal antibodies were purchased fromChemicon, USA.METHODS: A total of 114 healthy, adult, Sprague Dawley rats were randomly assigned to twogroups: PD model [n = 90, stereotactic microinjection of 2 μL 6-OHDA (3.5 μg/μL) into the striatum]and control (n = 24, no treatment). The 51 successful PD model rats were randomly divided into 3subgroups (n = 17): HAEC, PBS, and model. The HAEC and PBS groups were respectively injectedwith 10 μL PBS solution containing 1 x 105/mL HAECs or 10 μL PBS into the lateral ventricle. Themodel group was not treated.MAIN OUTCOME MEASURES: TH protein expression in the striatum was evaluated byimmunohistochemistry 5 weeks after HAEC transplantation. At 10 weeks, HAEC survival in thelateral ventricle was investigated by immunofluorescent staining; differentiation of HAECs in thelateral and third ventricles was examined by TH immunohistochemistry; concentrations of DA,homovanillic acid, dihydroxyphenyl acetic acid, 5-hydroxyindoleacetic acid, and 5-hydroxytryptaminein the striatum, as well as DA concentration in the cerebrospinal fluid, were measured withhigh-performance liquid chromatography-electrochemical detection. Circling behavior of PD modelrats was consecutively observed for 10 weeks following intraperitoneal injection of amphetamine 1week after successful model establishment.RESULTS: In the HAEC group, the number of TH-positive cells significantly increased in thestriatum, and circling behavior significantly decreased, compared with the PBS and model groups(P < 0.01). In addition, monoamine concentrations in the striatum, as well as DA concentrations inthe cerebrospinal fluid, significantly increased, compared with the PBS group (P < 0.05-0.01 ).Moreover, a large number of nestin-, vimentin-, and TH-positive cells were observed in the lateraland third ventricles following HAEC injection. CONCLUSION: HAECs survived for 10 weeks with no overgrowth following transplantation into thelateral ventricle of PD model rats. Moreover, the cells differentiated into dopaminergic neurons,which increased DA secretion. | 5 | The novel amyloid-beta peptide aptamer inhibits intracellular amyloid-beta peptide toxicity显示文摘Amyloid β peptide binding alcohol dehydrogenase (ABAD) decoy peptide (DP) can competitively antagonize binding of amyloid β peptide to ABAD and inhibit the cytotoxic effects of amyloid β peptide. Based on peptide aptamers, the present study inserted ABAD-DP into the disulfide bond of human thioredoxin (TRX) using molecular cloning technique to construct a fusion gene that can express the TRX1-ABAD-DP-TRX2 aptamer. Moreover, adeno-associated virus was used to allow its stable expression. Immunofluorescent staining revealed the co-expression of the transduced fusion gene TRX1-ABAD-DP-TRX2 and amyloid β peptide in NIH-3T3 cells, indicating that the TRX1-ABAD-DP-TRX2 aptamer can bind amyloid β peptide within cells. In addition, cell morphology and MTT results suggested that TRX1-ABAD-DP-TRX2 attenuated amyloid β peptide-induced SH-SY5Y cell injury and improved cell viability. These findings confirmed the possibility of constructing TRX-based peptide aptamer using ABAD-DP. | 16 | In vitro selection of G-rich RNA aptamers that target HIV-1 integrase显示文摘Aptamers that interact with various HIV-1 proteins,such as reverse transcriptase,Rev,Tat protein,and nuclear capsule protein,have been prepared through SELEX (systematic evolution of ligands by ex-ponential enrichment) technique. However,there are few reports about the DNA or RNA aptamers that target HIV-1 integrase. In this investigation,we selected alternative RNA aptamers specific for the HIV-1 integrase by using a different binding buffer containing 10 mmol·L-1 MgCl2 and 100 mmol·L-1 KCl. Aptamer IN1,IN2,IN3 had similar and the highest Kd values from 145 to 239 nmol·L-1. Structural studies showed that they formed similar stem-loop structure. Deletion of any stem structure resulted in diminished affinity. In addition,structure probing study with antisense DNA indicated that the stem-loop structure in the random region was critical for integrase binding. Although aptamer IN1 failed to form G-quartet structure,it might directly interact with the DDE motif of integrase,which is the virus DNA-binding site,because G-quadruplex T40214 competitively inhibited the interaction between IN1 and integrase. | 18 | Visual-feature-assisted mobile robot localization in a long corridor environment显示文摘Localization plays a vital role in the mobile robot navigation system and is a fundamental capability for autonomous movement.In an indoor environment,the current mainstream localization scheme uses two-dimensional(2D)laser light detection and ranging(LiDAR)to build an occupancy grid map with simultaneous localization and mapping(SLAM)technology;it then locates the robot based on the known grid map.However,such solutions work effectively only in those areas with salient geometrical features.For areas with repeated,symmetrical,or similar structures,such as a long corridor,the conventional particle filtering method will fail.To solve this crucial problem,this paper presents a novel coarse-to-fine paradigm that uses visual features to assist mobile robot localization in a long corridor.First,the mobile robot is remote-controlled to move from the starting position to the end along a middle line.In the moving process,a grid map is built using the laser-based SLAM method.At the same time,a visual map consisting of special images which are keyframes is created according to a keyframe selection strategy.The keyframes are associated with the robot’s poses through timestamps.Second,a moving strategy is proposed,based on the extracted range features of the laser scans,to decide on an initial rough position.This is vital for the mobile robot because it gives instructions on where the robot needs to move to adjust its pose.Third,the mobile robot captures images in a proper perspective according to the moving strategy and matches them with the image map to achieve a coarse localization.Finally,an improved particle filtering method is presented to achieve fine localization.Experimental results show that our method is effective and robust for global localization.The localization success rate reaches 98.8%while the average moving distance is only 0.31 m.In addition,the method works well when the mobile robot is kidnapped to another position in the corridor. | Gengyu GE Yi ZHANG Wei WANG Lihe HU Yang WANG Qin JIANG | 2023 | Frontiers of Information Technology & Electronic Engineering2023,24,6: | 0 |
