4Diabetes fuels periodontal lesions via GLUT1-driven macrophage inflammaging显示文摘Hyperglycemia induces chronic low-grade inflammation(inflammaging),which is a newly identified contributor to diabetes-related tissue lesions,including the inflammatory bone loss in periodontitis.It is also a secondary senescent pattern mediated by an increased burden of senescent cells and senescence-associated secretory phenotype(SASP).Macrophage is a key SASPspreading cell and may contribute to the maintenance of SASP response in the periodontal microenvironment.Using a transgenic diabetic model(BLKS/J-Lepr^(db)/lepr^(db)mice)we identified striking senescence of the periodontium in young(18-wk)-diabetic mice accompanied by amassed p16^(+)-macrophages and enhanced early SASP response.Exposed to high glucose in vitro,bone marrow-derived macrophage(BMDM)revealed a strong GLUT1 m RNA response driving the elevated-glucose uptake.GLUT1 is a representative and facilitative glucose transporter in macrophages with potential roles in hyperglycemia-induced inflammation.In this study,both GLUT1 and the downstream GTPase Rheb expression upregulated in the gingiva of diabetic mice with impaired condition.Furthermore,SASP release and p16/p21 signaling were proven to be triggered by m TOR phosphorylation in BMDM and antagonized by restricting glucose uptake in GLUT1^(-/-)BMDM.Taken together,our findings suggest that elevated-GLUT1 sensor responded to high glucose is important for macrophage senescence and SASP response,generated as a result of hyperglycemia,and it is a potential molecular mechanism for the exacerbation of periodontitis in diabetes.Qian Wang Lulingxiao Nie Pengfei Zhao Xinyi Zhou Yi Ding Qianming Chen Qi Wang 2021International Journal of Oral Science2021,13,3:5

7具有TSC2基因突变的儿童嗜酸性实性和囊性肾细胞癌临床病理学观察显示文摘目的:探讨具有TSC2基因突变的儿童嗜酸性实性和囊性肾细胞癌(ESC RCC)临床病理特征、免疫组织化学及分子遗传学特征和预后。方法:收集2017年至2018年2例ESC RCC临床资料,HE和免疫组织化学染色(EnVision)法进行形态学观察;荧光原位杂交检测TFE3、TFEB断裂重排基因;分子基因检测,PCR扩增,二代测序。结果:2例ESC RCC为男性,年龄分别9岁8个月和13岁,均发生在右肾。肿瘤大小5~7 cm,切面囊实性,灰红、灰白鱼肉样。显微镜下肿瘤具有纤维性包膜,肿瘤细胞突破胞膜。肿瘤细胞弥漫分布,圆形、多边性,胞质丰富,嗜伊红,可见大小不一的空泡,呈透明细胞样;部分区域类似乳头样结构,可见纤维轴心。细胞核圆形、泡状核,易见多核及巨核细胞,核分裂象未见;少量大小不一的囊样结构,囊壁可见单层瘤细胞被覆呈钉突样。间质见厚壁血管,灶状淋巴细胞浸润;较多嗜伊红坏死,可见钙化和胆固醇结晶。免疫组织化学阳性表达:PAX8(弥漫)、细胞角蛋白(CK)20、广谱细胞角蛋白(CKpan,局灶)、CD10(1例局灶/1例弥漫)、INI1、波形蛋白、CD68阳性,Ki-67阳性指数5%~10%。HMB45、S-100蛋白、Melan A、p53、结蛋白、TFE3、CK7、CK19、上皮细胞膜抗原(EMA)、CD56、嗜铬粒素A(CgA)、突触素、CD30、CD117、WT1、平滑肌肌动蛋白(SMA)阴性。分子遗传学检测:TFE3、TFEB断裂重排阴性。二代测序:例1,TSC2 p.Lys574Ter(0.198);例2,TSC2 p.Arg406Ter(0.355)。结论:ESC RCC儿童发病比较罕见,容易误诊,其具有独特的病理形态学特征、免疫表型和分子遗传学改变,预后较好。杨文萍 Kenneth Tou En Chang 徐红艳 Chik Hong Kuick Eileen Hui Qi Ng 黄慧 熊枫 吴艳 曾松涛 樊金星 Xinyi Loh 2020中华病理学杂志2020,49,7:3
8Hydrodynamic studies on two wiggling hydrofoils in an oblique arrangement显示文摘The propulsive performance of an oblique school of fish is numerically studied using an immersed boundary technique. The effect of the spacing and wiggling phase on the hydrodynamics of the system is investigated. The hydrodynamics of the system is deeply affected by the spacing between each fish in the school. When the horizontal separation is smaller than the length of the fish body,the downstream fish exhibits a larger thrust coefficient and greater propulsive efficiency than the isolated fish. However,the corresponding values for the upstream fish are smaller.The opposite behavior occurs when the horizontal separation increases beyond the length of fish body. 9Support Vector Regression for Bus Travel Time Prediction Using Wavelet Transform显示文摘In order to accurately predict bus travel time, a hybrid model based on combining wavelet transform technique with support vector regression(WT-SVR) model is employed. In this model, wavelet decomposition is used to extract important information of data at different levels and enhances the forecasting ability of the model. After wavelet transform different components are forecasted by their corresponding SVR predictors. The final prediction result is obtained by the summation of the predicted results for each component. The proposed hybrid model is examined by the data of bus route No.550 in Nanjing, China. The performance of WT-SVR model is evaluated by mean absolute error(MAE), mean absolute percent error(MAPE) and relative mean square error(RMSE), and also compared to regular SVR and ANN models. The results show that the prediction method based on wavelet transform and SVR has better tracking ability and dynamic behavior than regular SVR and ANN models. 12Biological Characteristics of Aeromonas Strains in Nanchang Area显示文摘[Objectives]This study was conducted to find effective measures to prevent and control Pelteobagrus fulvidraco hemorrhage.[Methods]Typical disease materials were collected for bacterial isolation and culture and biological characteristics detection,and morphological characteristics were observed by pure culture method.Biochemical characteristics were identified by micro biochemical method.Molecular identification was performed by PCR amplification method.Drug resistance was detected by disk diffusion method.The agar diffusion method was used for the detection of in-vitro inhibitory effect.The immunogenicity of the isolates was detected by subcutaneous immunoassay.The virulence factor was detected by spot inoculation.The intraperitoneal injection was used for the detection of pathogenicity.[Results]The isolates were round,grayish white,and identified as Aeromonas.They fermented maltose,glucose,etc.,did not ferment lactose,and were negative for urea and H2S and positive for lysine and arginine.The target fragment appeared at 1 500,and the sequences shared the highest homology with Aeromonas.The isolates were highly susceptible to amikacin and gentamicin,and resistant to penicillin G and tetracycline.Eight lactic acid bacteria strains produced 1.1-1.9 cm of inhibition zone against the isolates.After the second vaccination,the titer reached 2^13;both hemolysin and protease were produced;and the test mice died within 24 h.[Conclusions]The isolates are Aeromonas,and have strong drug resistance,immunogenicity and pathogenicity.Inactivated emusion-oil vaccines of Aeromonas combined with the co-use of lactic acid bacteria and susceptible drugs can prevent the outbreak of diseases caused by Aeromonas.Xinyi SHAN Dongxu WEN Xiaolan LIU Wangming TAO Mengjuan ZHANG Wei QI Fengwei ZHANG Zhixiu ZHU 2019Agricultural Biotechnology2019,8,3:1

14Distribution of aromatic amines,phenols,chlorobenzenes,and naphthalenes in the surface sediment of the Dianchi Lake,China显示文摘Organic pollutants are widespread environmental pollutants with high toxicity,persistence,and bioaccumulation.Our aim was to investigate the distribution of aromatic amines,phenols,chlorobenzenes,and naphthalenes in the surface sediment of the Dianchi Lake,China.Nineteen surface sediment samples were collected from the Dianchi Lake,and 40 types of organic pollutants were analyzed via gas chromatography-mass spectrometry.The total organic pollutant concentrations in the surface sediment of the Dianchi Lake varied from 27.4 to 1.62×10^3 ng/g.The concentrations of phenols were much higher than those in other water bodies but still within a controllable range,whereas the concentrations of the other organic pollutant classes were similar or even lower.The detection ratio of 3-or 4-methylphenol was the highest(100.00%)among the pollutants.The average total organic pollutant concentrations decreased in the following order:Caohai(540 ng/g)>the middle of Waihai(488 ng/g)>the edge of Waihai(351 ng/g)>Haigeng Dam(90.4 ng/g).Pearson analysis showed a strong correlation among 1-methylnaphthalene,2-methylnaphthalene,1,3-dinitronaphthalene,and 1,4-dinitronaphthalene(P<0.01).Caohai,the north lakeshore of Waihai and the south of Waihai showed higher risk because of high concentration;meanwhile,1,4-dichlorobenzene,3-or 4-methylphenol and 1,2,4-trichlorobenzene were mor e likely to cause risks.Xinyi Hu Ting Yang Chen Liu Jun Jin Bingli Gao Xuejun Wang Min Qi Baokai Wei Yuyu Zhan Tan Chen Hongtao Wang Yanting Liu Dongrui Bai Zhu Rao Nan Zhan 2020Frontiers of Environmental Science & Engineering2020,14,4:1

16Immediate and long-term outcomes after treat-all among people living with HIV in China:an interrupted time series analysis显示文摘Background In 2003,China implemented free antiretroviral therapy(ART)for people living with HIV(PLHIV),establishing an eligibility threshold of CD4<200 cells/μl.Subsequently,the entry criteria were revised in 2012(eligibility threshold:CD4≤350 cells/μl),2014(CD4≤500 cells/μl),and 2016(treat-all).However,the impact of treat-all policy on HIV care and treatment indicators in China is unknown.We aimed to elucidate the immediate and long-term impact of the implementation of treat-all policy in China.Methods Anonymized programmatic data on ART initiation and collection in PLHIV who newly started ART were retrieved between 1 January 2015 and 31 December 2019,from two provincial and municipal Centers for Disease Control and Prevention and ten major infectious disease hospitals specialized in HIV care in China.We used Poisson and quasi-Poisson segmented regression models to estimate the immediate and long-term impact of treat-all on three key indicators:monthly proportion of 30-day ART initiation,mean CD4 counts(cells/μl)at ART initiation,and mean estimated time from infection to diagnosis(year).We built separate models according to gender,age,route of transmission and region.Results Monthly data on ART initiation and collection were available for 75,516 individuals[gender:83.8%males;age:median 39 years,interquartile range(IQR):28-53;region:18.5%Northern China,10.9%Northeastern China,17.5%Southern China,49.2%Southwestern China].In the first month of treat-all,compared with the contemporaneous counterfactual,there was a significant increase in proportion of 30-day ART initiation[+12.6%,incidence rate ratio(IRR)=1.126,95%CI:1.033-1.229;P=0.007]and mean estimated time from infection to diagnosis(+7.0%,IRR=1.070,95%CI:1.021-1.120;P=0.004),while there was no significant change in mean CD4 at ART initiation(IRR=0.990,95%CI:0.956-1.026;P=0.585).By December 2019,the three outcomes were not significantly different from expected levels.In the stratified analysis,compared with the contemporaneous counterfactual,mean CD4 at ART initiation showed significant increases in Northern China(+3.3%,IRR=1.033,95%CI:1.001-1.065;P=0.041)and Northeastern China(+8.0%,IRR=1.080,95%CI:1.003-1.164;P=0.042)in the first month of treat-all;mean estimated time from infection to diagnosis showed significant increases in male(+5.6%,IRR=1.056,95%CI:1.010-1.104;P=0.016),female(+14.8%,IRR=1.148,95%CI:1.062-1.240;P<0.001),aged 26-35(+5.3%,IRR=1.053,95%CI:1.001-1.109;P=0.048)and>50(+7.8%,IRR=1.078,95%CI:1.000-1.161;P=0.046),heterosexual transmission(+12.4%,IRR=1.124,95%CI:1.042-1.213;P=0.002)and Southwestern China(+12.9%,IRR=1.129,95%CI:1.055-1.208;P<0.001)in the first month of treat-all.Conclusions The implementation of treat-all policy in China was associated with a positive effect on HIV care and treatment outcomes.To advance the work of rapid ART,efforts should be made to streamline the testing and ART initiation process,provide comprehensive support services,and address the issue of uneven distribution of medical resources.Xinsheng Wu Guohui Wu Ping Ma Rugang Wang Linghua Li Yinghui Sun Junjie Xu Yuwei Li Tong Zhang Quanmin Li Yuecheng Yang Lijing Wang Xiaoli Xin Ying Qiao Bingxue Fang Zhen Lu Xinyi Zhou Yuanyi Chen Qi Liu Gengfeng Fu Hongxia Wei Xiaojie Huang Bin Su Hui Wang Huachun Zou 2023Infectious Diseases of Poverty2023,12,4:0
17Surface charge property governing co-transport of illite colloids and Eu(Ⅲ) in saturated porous media显示文摘The transport of colloids and radionuclides is sophisticated because of the variety of charge properties between colloidal particles and host subsurface media, which causes great difficulty in establishing a reliable model of radionuclides migration by taking the colloid phase into consideration. In this work,the co-transport of illite colloids(IC) and Eu(Ⅲ) in the quartz sand and iron-coated sand porous media was investigated by column experiments to address the predominant mechanism of charge properties on co-transport. Results showed that Eu(Ⅲ) transport was driven by the illite colloids and electrostatic interaction was critical in governing the co-transport patterns. The promotion of Eu(Ⅲ) transport by IC was attenuated in the iron-coated sand systems;more IC-Eu(Ⅲ) complexes were retained uniformly in the column. The pore throat shrinkage caused by electrostatic attachment between aggregated IC and iron oxides exacerbated the physical straining and size exclusion effect of IC-Eu(Ⅲ) complexes. An aggravated irreversible retention of IC-Eu(Ⅲ) was detected in iron-coated sand column due to the electrostatic attraction of IC-Eu(Ⅲ) to host media. 18Supercontinuum fiber laser-based coherent anti-Stokes Raman scattering microscopy for label-free chemical imaging显示文摘Coherent anti-Stokes Raman scattering(CA RS)microscopy can resolve the chemical compo-nents and distribution of living biological systems in a label-firee manner and is favored in several disciplines.Current CA RS microscopes typically use bulky,high-performance solid-state lasers,which are expensive and sensitive to environmental changes.With their relatively low cost and environmental sensitivity,supercontinum fiber(SF)lasers with a small footprint have found increasing use in biomedical applications.Upon these features,in this paper,we homebuilt a low-cost CARS microscope based on a SF laser module(scCA RS microscope).This SF laser module is specially customized by adding a time synchronized seed source channel to the SF laser to form a dual-channel output laser.The performance of the scCARS microscope is evaluated with dimethyl sulfoxide,whose results confirm a spatial resolution of better than 500nm and a detection sensitivity of millimolar concentrations.The dual-color imaging capability is further demonstrated by imaging different species of mixed microspheres.We finally explore the potential of our scCARS microscope by mapping lipid droplets in different cancer cells and corneal stromal lenses.Nan Wang Jiaojiao Zhang Wangting Zhou Siting Liu Jing Li Xinyi Xu Qi Zeng Yong Li Shouping Zhu Xueli Chen 2022Journal of Innovative Optical Health Sciences2022,15,4:0
19Signatures of functional constraint at Fgfr1a Genes in schizothoracine fishes (Pisces: Cypriniformes): The dermal skeleton variation adapted to high-altitude environments显示文摘Fishes harbor a huge resource of phenotypic diversity and are useful for understanding the genetic basis of mor­phological variation.However,it is difficult to transfer classical genetic mapping to most non-model species.In this study,we performed a comparative sequence analysis of Fgfr1a to first interpret the evolution of this candi­date scale-loss gene in 15 schizothoracine fishes with various scale phenotypes.While considerable amino acid(AA)substitutions were observed,molecular evolution analysis indicates that the overall coding regions were subject to functional constraint.We also identified extra copies of Fgfr1a in 4 scale-loss fishes and detected ac­celerated evolution in one AA substitution specific to these duplicates.We speculate that Fgfr1a had accumulat­ed mutations in the ancestral lineage of scale-loss schizothoracine fishes before experiencing duplication events,which was further followed by the diversification of species.In silico mutation analysis predicted deleterious ef­fects of the mutations while no disruptive molecular mechanism was detected.Collectively,our results highlight the important role of Fgfr1a gene in the adaptive evolution of schizothoracine fishes during their radiation in the Qinghai-Tibetan Plateau.Xinyi GUO Ling XIE Xuze ZHANG Yinfa JI Juan CHEN Bo PANG Yongtao XU Delin QI Songchang GUO 2016Integrative Zoology2016,11,2:0
