| 2 | Molecular mechanism about lymphogenous metastasis of hepatocarcinoma cells in mice显示文摘AIM To investigate the correlation between lymphogenous metastasis and matrix metalloproteinases (MMPs) activity and the expression of Fas Iigand of tumor cells in lymph nodes.METHODES Fifty-six inbred 615-mice were equally divided into 2 groups and inoculated with Hca-F and Hca-P cells. Their lymph node metastatic rates were examined.Growth fraction of lymphocytes in host lymph nodes was detected by flow cytometry. The Hca-F and Hca-P cells were cultured with extract of lymph node, liver or spleen.The quantity of MMPs in these supernatants was examined by zymographic analysis. The expression of Fas ligand,PCNA, Bcl-2 protein of Hca-F and Hca-P cells in the mice were examined by immunohistochemistry. The apoptosis signals of macrophages in lymph nodes were observed with in situ DNA fragmentation.RESULTS On the 28th day post-inoculation, the lymph node metastatic rate of Hca-F was 80% (16/20), whereas that of Hca-P was 25% (5/20). The growth fraction of lymphocytes was as follows: in the Hca-F cells, the proliferating peak of lymphocytes appeared on the 14th day post-inoculation and then decreased rapidly, while in HcaP cells, the peak appeared on the 7th day post-inoculation and then kept at a high level. With the extract of lymph node, the quantity of the MMP-9 activity increased (P<0.01) and active MMP-9 and MMP-2 were produced by both Hca-F and Hca-P tumor cells, which did not produce MMPs without the extract of lymph node or with the extracts of the liver and spleen. The expression of Fas Ligand of Hca-F cells was stronger than that of Hca-P cells (P<0.01). The expressions of PCNA and Bcl-2 protein of Hca-F cells in the tumors of inoculated area were the same as that of Hca-P cells. In situ DNA fragmentation showed that the positive signals of macrophages were around HcaF cells.CONCLUSION Secretion of MMPs which was associated with metastatic ability of Hca-F and Hca-P tumor cells depends on the environment of lymph nodes. | 3 | 42,573 cases of hepatectomy in China: a multicenter retrospective investigation显示文摘Hepatectomy is currently routinely performed in most hospitals in China. China owns the largest population of liver diseases and the biggest number of liver resection cases. A nationwide multicenter retrospective investigation involving 112 hospitals was performed, and focused on liver resection for patients with hepatocellular carcinoma(HCC). 42,573 cases of hepatectomy were enrolled, and 18,275 valid cases of liver resection for HCC patients were selected for statistical analysis. The epidemiology of HCC, distribution of hepatectomy, postoperative complications and prognosis were finally analyzed. In the 18,275 HCC patients,81% had hepatitis B virus infection and 10% had hepatitis C virus infection. 38% of the HCC patients had normal Alphafetoprotein(AFP) level, and other 35% had an AFP level lower than 400 ng mL^(-1). In the study period, 97% of the hepatectomy for HCC were treated with open surgery, and 23.81% had vascular exclusion techniques. The operation time was(191.7±105.6) min,the blood loss was(546.0±562.8) m L, and blood transfusion was(543.0±1,035.2) m L. The median survival for HCC patients was 631 days, with 1-, 3-, and 5-year overall survival of 73.2%, 28.8% and 19.6%, respectively. | 4 | Comparison of quartz sand, anthracite, shale and biological ceramsite for adsorptive removal of phosphorus from aqueous solution显示文摘The choice of substrates with high phosphorus adsorption capacity is vital for sustainable phosphorus removal from waste water in constructed wetlands. In this study, four substrates were used: quartz sand, anthracite, shale and biological ceramsite. These substrate samples were characterized by Xray diffractometry and scanning electron microscopy studies for their mineral components(chemical components) and surface characteristics. The dynamic experimental results revealed the following ranking order for total phosphorus(TP) removal efficiency: anthracite > biological ceramsite > shale > quartz sand. The adsorptive removal capacities for TP using anthracite, biological ceramsite, shale and quartz sand were 85.87, 81.44, 59.65, and 55.98 mg/kg, respectively. Phosphorus desorption was also studied to analyze the substrates' adsorption efficiency in wastewater treatment as well as the substrates' ability to be reused for treatment. It was noted that the removal performance for the different forms of phosphorus was dependent on the nature of the substrate and the adsorption mechanism. A comparative analysis showed that the removal of particulate phosphorus was much easier using shale. Whereas anthracite had the highest soluble reactive phosphorus(SRP) adsorptive capacity, biological ceramsite had the highest dissolved organic phosphorus(DOP) removal capacity. Phosphorus removal by shale and biological ceramsite was mainly through chemical adsorption, precipitation or biological adsorption. | 6 | Sevoflurane postconditioning protects against myocardial ischemia/reperfusion injury by restoring autophagic flux via an NO-dependent mechanism显示文摘Volatile anesthetics improve postischemic cardiac function and reduce infarction even when administered for only a brief-time at the onset of reperfusion.A recent study showed that sevoflurane postconditioning (SPC)attenuated myocardial reperfusion injury, but the underlying mechanisms remain unclear,in this study,we examined the effects of sevoflurane on nitric oxide (NO)release and autophagic flux during the myocardial ischemia/reperfusion (I/R)injury in rats in vivo and ex vivo.Male rats were subjected to 30 min ischemia and 2 h reperfusion in the presence or absence of sevoflurane (1.0 minimum alveolar concentration)during the first 15 min of reperfusion.We found that SPC significantly improved hemodynamic performance after reperfusion,alleviated postischemic myocardial infarction,reduced nicotinamide adenine dinucleotide content loss,and cytochrome c.release in heart tissues.Furthermore,SPC significantly increased the phosphorylation of endothelial nitric oxide synthase (NOS)and neuronal nitric oxide synthase,and elevated myocardial NOS activity and NO production.All these effects were abolished by treatment with an NOS inhibitor NG-nitro-L-arginine methyl ester (L-NAME,10 mg/kg,i.v.).We also observed myocardial I/R-induced accumulation of autophagosomes in heart tissues,as evidenced by increased ratios of microtubule:associated protein 1 light chain 3 Ⅱ/Ⅰ,upregulation of Beclin 1 and P62,and reduced lysosome-assciated membrane protein-2 expression.SPC significantly attenuated I/R- impaired autophagic flux,which were blocked by L-NAME.Moreover,pretreatment with the autophagic flUX blocker chloroquine (10 mg/kg,i.p.)increased autophagosome accumulation in SPC-treated heart following I/R and blocked SPC-induced cardioprotection.The same results were also observed in a rat model of myocardial I/R injury ex vivo,suggesting that SPC protects rat hearts against myocardial reperfusion injury by restoring I/R-impaired autophagic flux yia an NO-dependent mechanism. | Shi-gang Qiao Ying Sun Bo Sun An Wang Jia Qiu Lei Hong Jian-zhong An Chen Wang Hui-ling Zhang | 2019 | Acta Pharmacologica Sinica2019,40,1: | 21 |
| 7 | Outcomes and possible risk factors associated with axis alignment and rotational stability after implantation of the Toric implantable collamer lens for high myopic astigmatism显示文摘·AIM:To assess the visual outcomes and possible risk factors associated with axis alignment and rotational stability after implantation of Toric implantable collamer lens (TICL) for the correction of high myopic astigmatism. ·METHODS:In this prospective, nonrandomized clinical study, 54 consecutive eyes of 29 patients with high myopic astigmatism received TICL implantation. To evaluate postoperative axis deviation from the intended axis, a digital anterior segment photograph was taken. The ultrasound biomicroscopy(UBM) was used to observe footplate-position. ·RESULTS:After mean follow-up of 8.6 months, mean manifest refractive cylinder (MRC) decreased 79.3% from (-1.88±1.49)D preoperatively to (0.39±0.61)D postoperatively. MRC within 1.00D occurred in 68.5% (37/54) of eyes, whereas 48.1% (26/54) had MRC within 0.50D. Mean manifest refraction spherical equivalent (MRSE) changed from (-12.08±4.22)D preoperatively to (-0.41±0.61)D postoperatively. Uncorrected binocular vision of 20/20 or better occurred in 72.2%(39/54) of patients compared with binocular best-corrected visual acuity (BCVA) of 20/20 or better in 44.4% (24/54) preoperatively. The mean difference between intended and achieved TICL axes was (6.96±8.37)°. Footplates of TICLs were in the ciliary sulcus in 22 eyes (46.3%), below the ciliary sulcus in 32 eyes (53.7%). The angle of TICL rotation had significant correlation with the footplates-position (t =2.127; P =0.045) and the postoperative TICL vaulting (r =-0.516; P =0.000). ·CONCLUSION:The results of our study further support the safety, efficacy and predictability of TICL for the correct high myopic astigmatism. | 13 | The geothermal formation mechanism in the Gonghe Basin: Discussion and analysis from the geological background显示文摘The Gonghe Basin,a Cenozoic down-warped basin,is located in the northeastern part of the Qinghai-Xizang (Tibetan) Plateau,and spread over important nodes of the transfer of multiple blocks in the central orogenic belt in the NWW direction.It is also called “Qin Kun Fork” and “Gonghe Gap”.The basin has a high heat flow value and obvious thermal anomaly.The geothermal resources are mainly hot dry rock and underground hot water.In recent years,the mechanism of geothermal formation within the basin has been controversial.On the basis of understanding the knowledge of predecessors,this paper proposes the geothermal formation mechanism of the “heat source–heat transfer–heat reservoir and caprock–thermal system”of the Gonghe Basin from the perspective of a geological background through data integrationintegrated research-expert,discussion-graph,compilation-field verification and other processes: (1) Heat source: geophysical exploration and radioisotope calculations show that the heat source of heat in the basin has both the contribution of mantle and the participation of the earth's crust,but mainly the contribution of the deep mantle.(2) Heat transfer: The petrological properties of the basin and the exposed structure position of the surface hot springs show that one transfer mode is the material of the mantle source upwells and invades from the bottom,directly injecting heat;the other is that the deep fault conducts the deep heat of the basin to the middle and lower parts of the earth's crust,then the secondary fracture transfers the heat to the shallow part.(3) Heat reservoir and caprock: First,the convective strip-shaped heat reservoir exposed by the hot springs on the peripheral fault zone of the basin;second,the underlying hot dry rock layered heat reservoir and the upper new generation heat reservoir and caprock in the basin revealed by drilling data.(4) Thermal system: Based on the characteristics of the “heat source-heat transfer-heat reservoir and caprock”,it is preliminarily believed that the Gonghe Basin belongs to the non-magmatic heat source hydrothermal geothermal system (type II21) and the dry heat geothermal system (type II22).Its favorable structural position and special geological evolutionary history have given birth to a unique environment for the formation of the geothermal system.There may be a cumulative effect of heat accumulation in the eastern part of the basin,which is expected to become a favorable exploration area for hot dry rocks. | Yan-fang Feng Xiao-xu Zhang Bo Zhang Jiang-tao Liu Yang-gang Wang De-long Jia Li-rong Hao Zhao-yu Kong | 2018 | China Geology2018,1,3: | 15 |

This strain was identified by gene sequence identification. The antioxidant and anti-inflammatory capacity of ferulic acid released from wheat bran by this A. niger strain(FA-WB) were evaluated. Methods Molecular identification techniques based on PCR analysis of specific genomic sequences were conducted; antioxidant ability was examined using oxygen radical absorbance capacity(ORAC), cellular antioxidant activity(CAA) assays, and erythrocyte hemolysis assays. RAW264.7 cells were used as a model to detect anti-inflammatory activity. Results The filamentous fungal isolate was identified to be A. niger. ORAC and CAA assay showed that FA-WB had better antioxidant activity than that of the ferulic acid standard. The erythrocyte hemolysis assay results suggested that FA-WB could attenuate AAPH-induced oxidative stress through inhibition of reactive oxy gen species(ROS) generation. FA-WB could significantly restore the AAPH-induced increase in intracellular antioxidant enzyme activities to normal levels as well as inhibit the intracellular malondialdehyde formation. TNF-?, IL-6, and NO levels indicated that FA-WB can inhibit the inflammation induced by lipopolysaccharide(LPS).
