1Plankton community composition in the Three Gorges Reservoir Region revealed by PCR-DGGE and its relationships with environmental factors显示文摘To explore the relationships between community composition and the environment in a reservoir ecosystem, plankton communities from the Three Gorges Reservoir Region were studied by PCR-denaturing gradient gel electrophoresis fingerprinting. Bacterial and eukaryotic operational taxonomic units (OTUs), generated by DGGE analysis of the PCR-amplified 16S and 18S rRNA genes, were used as surrogates for the dominant 'biodiversity units'. OTU composition among the sites was heterogeneous; 46.7% of the total bacterial OTUs (45) and 64.1% of the eukaryotic OTUs (39) were identified in less than half of the sampling sites. Unweighted pair group method with arithmetic averages (UPGMA) clustering of the OTUs suggested that the plankton communities in the Xiangxi Rive sites were not always significantly different from those from the Yangtze River sites, despite clear differences in their environmental characterizations. Canonical correspondence analysis (CCA) was applied to further investigate the relationships between OTU composition and the environmental factors. 2Impacts of algal blooms removal by chitosan-modified soils on zooplankton community in Taihu Lake,China显示文摘It is important to assess the eflect on zooplankton when perform the environmental protection or restoration technology,especially removing algal blooms,because algae were the major primary producer in algal lakes.The influence on zooplankton community after half a year of algal blooms removed by chitosan-modified soils in Taihu Lake was assessed and the rationality of carrying out the process semiannually was evaluated in the present study.Morphological composition and genetic diversity of zooplankton community were investigated by microscope checkup and polymerase chain reaction-denatured gradient gel electrophoresis(PCR-DGGE) .A total of 44 zooplankton taxa(23 protozoa,17 rotifers,3 copepoda and 1 cladocera) were detected by microscope checkup,and a total of 91 bands(28 bands amplified by primers F1427-GC and R1616,63 bands amplified by primers Fung-GC and NS1) were detected by PCR-DGGE.The results of cluster analysis or detrended correspondence analysis indicated that there was no considerable diflerence in morphological composition of zooplankton and DGGE profiles between experimental and control sites,and DGGE profiles could represent the biologic diversity.The study showed that zooplankton community could recover original condition after half year of algal blooms removed by chitosan-modified soils and it was acceptable to apply this process semiannually.In addition,the results revealed that PCR-DGGE could be applied to investigate the impacts of the environmental protection or restoration engineering on zooplankton community diversity.Jiajia Ni Yuhe Yu Weisong Feng Qingyun Yan Gang Pan Bo Yang Xiang Zhang Xuemei Li 2010Journal of Environmental Sciences2010,22,10:8

14The biogenesis and secretion of exosomes and multivesicular bodies(MVBs):Intercellular shuttles and implications in human diseases显示文摘Exosomes carry and transmit signaling molecules used for intercellular communication.The generation and secretion of exosomes is a multistep interlocking process that allows simultaneous control of multiple regulatory sites.Protein molecules,mainly RAB GTPases,cytoskeletal proteins and soluble N-ethylmaleimide-sensitive fusion attachment protein receptor(SNARE),are specifically regulated in response to pathological conditions such as altered cellular microenvironment,stimulation by pathogenic factors,or gene mutation.This interferes with the smooth functioning of endocytosis,translocation,degradation,docking and fusion processes,leading to changes in the secretion of exosomes.Large numbers of secreted exosomes are disseminated by the flow of body fluids and absorbed by the recipient cells.By transmitting characteristic functional proteins and genetic information produced under disease conditions,exosomes can change the physiological state of the recipient cells and their microenvironment.The microenvironment,in turn,affects the occurrence and development of disease.Therefore,this review will discuss the mechanism by which exosome secretion is regulated in cells following the formation of mature secretory multivesicular bodies(MVBs).The overall aim is to find ways to eliminate disease-derived exosomes at their source,thereby providing an important new basis for the clinical treatment of disease.Minxue Xu Jie Ji Dandan Jin Yue Wu Tong Wu Renjie Lin Shengze Zhu Feng Jiang Yifei Ji Baijun Bao Mei Li Weisong Xu Mingbing Xiao 2023Genes & Diseases2023,10,5:0
15An Adaptive Parameter-Free Optimal Number of Market Segments Estimation Algorithm Based on a New Internal Validity Index显示文摘An appropriate optimal number of market segments(ONS)estimation is essential for an enterprise to achieve successful market segmentation,but at present,there is a serious lack of attention to this issue in market segmentation.In our study,an independent adaptive ONS estimation method BWCON-NSDK-means++is proposed by integrating a newinternal validity index(IVI)Between-Within-Connectivity(BWCON)and a newstable clustering algorithmNatural-SDK-means++(NSDK-means++)in a novel way.First,to complete the evaluation dimensions of the existing IVIs,we designed a connectivity formula based on the neighbor relationship and proposed the BWCON by integrating the connectivity with other two commonly considered measures of compactness and separation.Then,considering the stability,number of parameters and clustering performance,we proposed the NSDK-means++to participate in the integrationwhere the natural neighbor was used to optimize the initial cluster centers(ICCs)determination strategy in the SDK-means++.At last,to ensure the objectivity of the estimatedONS,we designed a BWCON-based ONS estimation framework that does not require the user to set any parameters in advance and integrated the NSDK-means++into this framework forming a practical ONS estimation tool BWCON-NSDK-means++.The final experimental results showthat the proposed BWCONand NSDK-means++are significantlymore suitable than their respective existing models to participate in the integration for determining theONS,and the proposed BWCON-NSDK-means++is demonstrably superior to the BWCON-KMA,BWCONMBK,BWCON-KM++,BWCON-RKM++,BWCON-SDKM++,BWCON-Single linkage,BWCON-Complete linkage,BWCON-Average linkage and BWCON-Ward linkage in terms of the ONS estimation.Moreover,as an independentmarket segmentation tool,the BWCON-NSDK-means++also outperforms the existing models with respect to the inter-market differentiation and sub-market size.Jianfang Qi Yue Li Haibin Jin Jianying Feng Dong Tian Weisong Mu 2023Computer Modeling in Engineering & Sciences2023,,10:0
