A novel human mAb (MERS-GD27) provides prophylactic and postexposure efficacy in MERS-CoV susceptible mice显示文摘Dear Editor,Since September 2012, the Middle East respiratory syndrome coronavirus (MERS-CoV) cases have been reported in more than 27 countries, and more than 2,000 cases have been confirmed in the laboratory (http://gffzz36b398b81f8a4a4ch6u0qpxwn0coc6qwv.ffgz.tsg.suse.edu.cn/emergencies/mers-cov/en/). MERS-CoV causes an acute and severe respiratory illness with a high mortality rate(~35%) in humans (Shi et al., 2017, Zaki et al., 2012).Neutralizing antibodies targeting the spike of MERS-CoV have been shown to be a therapeutic option for treatment of lethal disease (Agrawal et al., 2016, Ying et al., 2014).Peihua Niu Guangyu Zhao Yao Deng Shihui Sun Wenling Wang Yusen Zhou Wenjie Tan 2018Science China(Life Sciences)2018,61,10:11
CD147-spike protein is a novel route for SARS-CoV-2 infection to host cells显示文摘In face of the everlasting battle toward COVID-19 and the rapid evolution of SARS-CoV-2,no specific and effective drugs for treating this disease have been reported until today.Angiotensin-converting enzyme 2(ACE2),a receptor of SARS-CoV-2,mediates the virus infection by binding to spike protein.Although ACE2 is expressed in the lung, kidney, and intestine, its expressing levels are rather low, especially in the lung.Considering the great infectivity of COVID-19,we speculate that SARS-CoV-2 may depend on other routes to facilitate its infection. Here,we first discover an interaction between host cell receptor CD147 and SARS-CoV-2 spike protein. The loss of CD147 or blocking CD147 in Vero E6 and BEAS-2B cell lines by anti-CD147 antibody,Meplazumab, inhibits SARSCoV-2 amplification.Expression of human CD147 allows virus entry into non-susceptible BHK-21 cells, which can be neutralized by CD147 extracellular fragment.Viral loads are detectable in the lungs of human CD147(hCD147)mice infected with SARS-CoV-2,but not in those of virus-infected wild type mice.Interestingly, virions are observed in lymphocytes of lung tissue from a COVID-19 patient.Human T cells with a property of ACE2 natural deficiency can be infected with SARS-CoV-2 pseudovirus in a dosedependent manner, which is specifically inhibited by Meplazumab.Furthermore,CD147 mediates virus entering host cells by endocytosis.Together, our study reveals a novel virus entry route,CD147-spike protein, which provides an important target for developing specific and effective drug against COVID-19.Ke Wang Wei Chen Zheng Zhang Yongqiang Deng Jian-Qi Lian Peng Du Ding Wei Yang Zhang Xiu-Xuan Sun Li Gong Xu Yang Lei He Lei Zhang Zhiwei Yang Jie-Jie Geng Ruo Chen Hai Zhang Bin Wang Yu-Meng Zhu Gang Nan Jian-Li Jiang Ling Li Jiao Wu Peng Lin Wan Huang Liangzhi Xie Zhao-Hui Zheng Kui Zhang Jin-Lin Miao Hong-Yong Cui Min Huang Jun Zhang Ling Fu Xiang-Min Yang Zhongpeng Zhao Shihui Sun Hongjing Gu Zhe Wang Chun-Fu Wang Yacheng Lu Ying-Ying Liu Qing-Yi Wang Huijie Bian Ping Zhu Zhi-Nan Chen 2020Signal Transduction and Targeted Therapy2020,5,1:9

Rational development of a human antibody cocktail that deploys multiple functions to confer Pan-SARS-CoVs protection显示文摘Structural principles underlying the composition and synergistic mechanisms of protective monoclonal antibody cocktails are poorly defined. Here,we exploited antibody cooperativity to develop a therapeutic antibody cocktail against SARS-CoV-2.On the basis of our previously identified humanized cross-neutralizing antibody H014,we systematically an a lyzed a fully human n aive antibody library and rationally identified a potent neutralizing antibody partner,P17,which confers effective protection in animal model.Cryo-EM studies dissected the nature of the P17 epitope, which is SARS-CoV-2 specific and distinctly different from that of HOI4.High-resolution structure of the SARS-CoV-2 spike in complex with HOM and P17,together with functional investigations revealed that in a two-antibody cocktail, synergistic neutralization was achieved by S1 shielding and conformational locking, thereby blocking receptor attachment and viral membrane fusion, conferring high potency as well as robustness against viral mutation escape.Furthermore, cluster analysis identified a hypothetical 3rd antibody partner for further reinforcing the cocktail as pan-SARS-CoVs therapeutics.Hangping Yao Yao Sun Yong-Qiang Deng Nan Wang Yongcong Tan Na-Na Zhang Xiao-Feng Li Chao Kong Yan-Peng Xuc Qi Chen Tian-Shu Cao Hui Zhao Xintian Yan Lei Cao Zhe Lv Dandan Zhu Rui Feng Nanping Wu Wenhai Zhang Yuhao Hu Keda Chen Rong-Rong Zhang Qingyu Lv Shihui Sun Yunhua Zhou Run Yan Guan Yang Xinglu Chanjuan Liu Xiangyun Lu Linfang Cheng Hongying Qiu Xing-Yao Huang Tianhao Weng Danrong Shi Weidong Jiang Junbin Shao Lei Wang Jie Zhang Tao Jiang Guojun Lang Cheng-Feng Qinc Lanjuan Li Xiangxi Wang 2021Cell Research2021,31,1:5
Trusted GPSR protocol without reputation faking in VANET显示文摘In vehicular ad hoc network(VANET), misbehaviors of internal nodes, such as discarding packets, may lead to a rapid decline in packet delivery ratio. To solve this problem, an improvement of greedy perimeter stateless routing(GPSR) protocol is presented. In the new protocol, trustworthiness is considered in the route selection process. The trustworthiness is measured by an objective trust model based on the subjective trust model Dy Trust. And the reputation value which reflects the trustworthiness of each node is calculated and broadcasted by the intersection nodes. Specially, besides resisting the packet-discarding behavior of selfish nodes, this protocol also includes a location detection process to resist the location-faking behavior of malicious nodes. As a result, the selfish nodes and the malicious nodes can be excluded from the network. In addition, compared with improved GPSR protocol, the presented one is able to resist one kind of reputation-faking attack and has better performance in simulation.Xiao Ya Zheng Shihui Sun Bin 2015The Journal of China Universities of Posts and Telecommunications2015,22,5:1
Screening of Brown Planthopper Resistant miRNAs in Rice and Their Roles in Regulation of Brown Planthopper Fecundity显示文摘MicroRNAs(miRNAs)can participate in plant-insect interactions, which regulate plant defense networks. In this study,we analyzed the miRNA expression profiles of six rice varieties before and after brown planthopper(BPH)-feeding.We identified 45 differentially expressed miRNAs between BPH-susceptible and BPH-resistant rice varieties and 144 miRNAs that responded to BPH-feeding.Thus,miRNAs may be involved in multiple pathways regulating rice defense response against BPH.In addition,we found that the genetic history of rice varieties determined the regulation mode of the miRNA and affected the amounts, types, changing trends and response periods of miRNAs in response to BPH-feeding.To conclude,we scanned seven potential cross-kingdom miRNAs,of which miR5795 may target the vitellogenin gene in BPH,causing a 16.07%reduction in BPH oviposition. The results provide new miRNA information of rice-BPH interactions and BPH-resistant rice variety breeding.LÜJun LIU Jinhui CHEN Lin SUN Jiawei SU Qin LI Shihui YANG Jianhua ZHANG Wenqing 2022Rice science2022,29,6:1
Improved genetic distance-based spatial deployment can effectively minimize inbreeding in seed orchard显示文摘Background:Inbreeding in seed orchards is expected to increase with the advancement of breeding cycles, which results in the delivery of crops with suboptimal genetic gain, reduced genetic diversity, and lower seed set. Here,a genetic distance-dependent method for clonal spatial deployment in seed orchards was developed and demonstrated, which reduced the inbreeding levels. The method’s main evaluation parameter of inbreeding is the genetic distance among individuals and the deployment method used an improved adaptive parallel genetic algorithm(IAPGA)based on Python language.Using inbreeding-prone Chinese Mongolian pine breeding population material originating from a single natural population, the proposed method was compared to a traditional orchard design and a distance-based design;namely, complete randomized block(RCB)and optimum neighborhood(ONA)designs, respectively. Results:With the advancement of selective breeding cycles, group separation among orchard related individuals is expected to increase.Based on the genetic distance among individuals, the IAPGA design was superior in significantly reducing the inbreeding level as compared to the two existing designs, confirming its suitability to advanced-generation orchards where relatedness among parents is common. In the 1st,2nd,and mixed generations clonal deployment schemes, the IAPGA design produced lower inbreeding with 87.22%,81.49%,and 87.23%of RCB,and 92.78%,91.30%,and 91.67%of ONA designs, respectively. Conclusions:The IAPGA clonal deployment proposed in this study has the obvious advantage of controlling inbreeding, and it is expected to be used in clonal deployment in seed orchards on a large-scale.Further studies are needed to focus on the actual states of pollen dispersal and mating in seed orchards, and more assumptions should be taken into account for the optimized deployment method.Boning Yang Haihong Sun Jiandong Qi Shihui Niu Yousry AEl-Kassaby Wei Li 2020Forest Ecosystems2020,7,1:1

EDVAM:a 3D eye-tracking dataset for visual attention modeling in a virtual museum显示文摘Predicting visual attention facilitates an adaptive virtual museum environment and provides a context-aware and interactive user experience.Explorations toward development of a visual attention mechanism using eye-tracking data have so far been limited to 2D cases, and researchers are yet to approach this topic in a 3D virtual environment and from a spatiotemporal perspective. We present the first 3D Eye-tracking Dataset for Visual Attention modeling in a virtual Museum, known as the EDVAM.In addition,a deep learning model is devised and tested with the EDVAM to predict a user’s subsequent visual attention from previous eye movements. This work provides a reference for visual attention modeling and context-aware interaction in the context of virtual museums.Yunzhan ZHOU Tian FENG Shihui SHUAI Xiangdong LI Lingyun SUN Henry Been-Lirn DUH 2022Frontiers of Information Technology & Electronic Engineering2022,23,1:0
Screening HLA-A-restricted T cell epitopes of SARS-CoV-2 and the induction of CD8^(+)T cell responses in HLA-A transgenic mice显示文摘Since severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)-specific T cells have been found to play essential roles in host immune protection and pathology in patients with coronavirus disease 2019(COVID-19),this study focused on the functional validation of T cell epitopes and the development of vaccines that induce specific T cell responses.A total of 120 CD8^(+)T cell epitopes from the E,M,N,S,and RdRp proteins were functionally validated.Among these,110,15,6,14,and 12 epitopes were highly homologous with SARS-CoV,OC43,NL63,HKU1,and 229E,respectively;in addition, four epitopes from the S protein displayed one amino acid that was distinct from the current SARS-CoV-2 variants.Then,31 epitopes restricted by the HLA-A2 molecule were used to generate peptide cocktail vaccines in combination with Poly(I:C),R848 or poly(lactic-co-glycolic acid)nanoparticles, and these vaccines elicited robust and specific CD8^(+)T cell responses in HLA-A2/DR1 transgenic mice as well as wild-type mice.In contrast to previous research, this study established a modified DC-peptide-PBL cell coculture system using healthy donor PBMCs to validate the in silico predicted epitopes, provided an epitope library restricted by nine of the most prevalent HLA-A allotypes covering broad Asian populations, and identified the HLA-A restrictions of these validated epitopes using competitive peptide binding experiments with HMy2.CIR cell lines expressing the indicated HLA-A allotype, which initially confirmed the in vivo feasibility of 9-or 10-mer peptide cocktail vaccines against SARS-CoV-2.These data will facilitate the design and development of vaccines that induce antiviral CD8^(+)T cell responses in COVID-19 patients.Xiaoxiao Jin Yan Ding Shihui Sun Xinyi Wang Zining Zhou Xiaotao Liu Miaomiao Li Xian Chen Anran Shen Yandan Wu Bicheng Liu Jianqiong Zhang Jian Li Yi Yang Haibo Qiu Chuanlai Shen Yuxian He Guangyu Zhao 2021Cellular & Molecular Immunology2021,18,12:0
Cryptanalysis on ‘an arbitrated quantum signature protocol based on the chained CNOT operations encryption’显示文摘Arbitrated quantum signature(AQS) is an important branch in quantum cryptography to authenticate quantum information, and cryptanalysis on AQS protocols helps to evaluate and improve security of AQS. Recently, it is discovered that an AQS protocol based on chained controlled-NOT(CNOT) algorithm is vulnerable to a novel attack because a transformation from binary keys into permutations and the chained CNOT algorithm have special properties, which enables a malicious receiver to forge signatures with probability 1/2. Moreover, a malicious signer can also deny his signatures with probability 1/4. Then, two possible improved methods are presented to resist these attacks: one is padding constants to reduce probability of the successful attacks, and the other is a circular chained CNOT algorithm to make the attack strategy invalid. And the security analysis shows that both the two improve methods could well resist these attacks.Wen Kai Zheng Shihui Sun Bin 2019The Journal of China Universities of Posts and Telecommunications2019,26,3:0
A polygalacturonase gene OsPG1 modulates water homeostasis in rice显示文摘A dynamic plant architecture is the basis of plant adaptation to changing environments.Although many genes regulating leaf rolling have been identified, genes directly associated with water homeostasis are largely unknown. Here,we isolated a rice mutant, dynamic leaf rolling 1(dlr1),characterized by‘leaf unfolding in the morning-leaf rolling at noon-leaf unfolding in the evening’during a sunny day.Water content was decreased in rolled leaves and water sprayed on leaves caused reopening, indicating that in vivo water deficiency induced the leaf rolling.Map-based cloning and expression tests demonstrated that an A1400G single base mutation in Oryza sativa Polygalacturonase 1(OsPG1)/PHOTO-SENSITIVE LEAF ROLLING 1(PSL1)was responsible for the dynamic leaf rolling phenotype in the dlr1 mutant.OsPG1 encodes a polygalacturonase, one of the main enzymes that degrade demethylesterified homogalacturonans in plant cell walls.OsPG1 was constitutively expressed in various tissues and was enriched in stomata.Mutants of the OsPG1 gene exhibited defects in stomatal closure and decreased stomatal density, leading to reduced transpiration and excessive water loss under specific conditions, but had normal root development.Further analysis revealed that mutation of OsPG1 led to reduced pectinase activity in the leaves and increased demethylesterified homogalacturonans in guard cells. Our findings reveal a mechanism by which OsPG1 modulates water homeostasis to control dynamic leaf rolling, providing insights for plants to adapt to environmental variation.Qinwen Zou Ranran Tu Jiajun Wu Tingting Huang Zhihao Sun Zheyan Ruan Hongyu Cao Shihui Yang Xihong Shen Guanghua He Hong Wang 2024The Crop Journal2024,12,1:0

Effects of telomere length on leukemogenesis显示文摘Dear Editor,Most cancer cells maintain the length of their telomeres via telomerase(Günes and Rudolph,2013;Kim et al.,1994).However,in some cancers, telomeres are maintained not by telomerase but by alternative lengthening of telomeres(Bryan et al.,1997).Telomerase activity appears to be reduced in some bone marrow(BM)diseases, including chronic myeloid leukemia, acute myeloid leukemia(AML),and myeloproliferative neoplasms(MPN),leading to shortened telomeres that correlate with leukemogenesis(Bouillon et al.,2018;Engelhardt et al.,2000;Shay et al.,1996).Our current study aimed to determine the definitive roles of telomeres and telomerase in leukemogenesis.Shihui Ma Guohuan Sun Shangda Yang Zhenyu Ju Tao Cheng Hui Cheng 2020Science China(Life Sciences)2020,63,2:0
Advances in regenerative medicine applications of tetrahedral framework nucleic acid-based nanomaterials: an expert consensus recommendation显示文摘With the emergence of DNA nanotechnology in the 1980s, self-assembled DNA nanostructures have attracted considerable attention worldwide due to their inherent biocompatibility, unsurpassed programmability, and versatile functions. Especially promising nanostructures are tetrahedral framework nucleic acids(t FNAs), first proposed by Turberfield with the use of a one-step annealing approach. Benefiting from their various merits, such as simple synthesis, high reproducibility, structural stability, cellular internalization, tissue permeability, and editable functionality, t FNAs have been widely applied in the biomedical field as threedimensional DNA nanomaterials. Surprisingly, t FNAs exhibit positive effects on cellular biological behaviors and tissue regeneration, which may be used to treat inflammatory and degenerative diseases. According to their intended application and carrying capacity,t FNAs could carry functional nucleic acids or therapeutic molecules through extended sequences, sticky-end hybridization, intercalation, and encapsulation based on the Watson and Crick principle. Additionally, dynamic t FNAs also have potential applications in controlled and targeted therapies. This review summarized the latest progress in pure/modified/dynamic t FNAs and demonstrated their regenerative medicine applications. These applications include promoting the regeneration of the bone, cartilage, nerve, skin, vasculature, or muscle and treating diseases such as bone defects, neurological disorders, joint-related inflammatory diseases, periodontitis, and immune diseases.Yunfeng Lin Qian Li Lihua Wang Quanyi Guo Shuyun Liu Shihui Zhu Yu Sun Yujiang Fan Yong Sun Haihang Li Xudong Tian Delun Luo Sirong Shi 2022International Journal of Oral Science2022,14,4:0
