| 1 | The combined genotypes effect of ESR and FSHβ genes on litter size traits in five different pig breeds显示文摘Estrogen receptor (ESR) and Follicular-stimula-ting hormone beta subunit (FSHβ) genes were chosen as candidates to determine whether they control litter size and some other reproductive traits in swine. 269 sows from five different pig breeds were genotyped by an established PCR-RFLPs protocol at both ESR and FSHβ loci. The effects of both ESR and FSHβ on pig reproductive traits, including total number born (TNB) and number born alive (NBA), are analyzed by SAS software (version 6.12). These computation results demonstrated that both ESR locus and FSHβ locus are the major genes influencing litter size in pigs. The sows of BBBB combined genotype of ESR and FSHβ loci generally produce 1.85-3.01 TNB and 2.0-3.0 NBA more than those of ABAA combined genotype. The notable effect of ESR locus and FSHβ locus on litter size of pigs have made it possible to improve the pig reproduction by Marker-assisted selection (MAS). | 10 | Epigenome-wide development and validation of a prognostic methylation score in intrahepatic cholangiocarcinoma based on machine learning strategies显示文摘Background:Clinical parameter-based nomograms and staging systems provide limited information for the prediction of survival in intrahepatic cholangiocarcinoma(ICC)patients.In this study,we developed a methylation signature that precisely predicts overall survival(OS)after surgery.Methods:An epigenome-wide study of DNA methylation based on whole-genome bisulfite sequencing(WGBS)was conducted for two independent cohorts(discovery cohort,n=164;validation cohort,n=170)from three hepatobiliary centers in China.By referring to differentially methylated regions(DMRs),we proposed the concept of prognostically methylated regions(PMRs),which were composed of consecutive prognostically methylated CpGs(PMCs).Using machine learning strategies(Random Forest and the least absolute shrinkage and selector regression),a prognostic methylation score(PMS)was constructed based on 14 PMRs in the discovery cohort and confirmed in the validation cohort.Results:The C-indices of the PMS for predicting OS in the discovery and validation cohorts were 0.79 and 0.74,respectively.In the whole cohort,the PMS was an independent predictor of OS[hazard ratio(HR)=8.12;95% confidence interval(CI):5.48-12.04;P<0.001],and the C-index(0.78)of the PMS was significantly higher than that of the Johns Hopkins University School of Medicine(JHUSM)nomogram(0.69,P<0.001),the Eastern Hepatobiliary Surgery Hospital(EHBSH)nomogram(0.67,P<0.001),American Joint Committee on Cancer(AJCC)tumor-node-metastasis(TNM)staging system(0.61,P<0.001),and MEGNA prognostic score(0.60,P<0.001).The patients in quartile 4 of PMS could benefit from adjuvant therapy(AT)(HR=0.54;95%CI:0.32-0.91;log-rank P=0.043),whereas those in the quartiles 1-3 could not.However,other nomograms and staging system failed to do so.Further analyses of potential mechanisms showed that the PMS was associated with tumor biological behaviors,pathway activation,and immune microenvironment.Conclusions:The PMS could improve the prognostic accuracy and identify patients who would benefit from AT for ICC patients,and might facilitate decisions in treatment of ICC patients. | Xing Chen Liangqing Dong Lu Chen Yuan Wang Jinpeng Du Lijie Ma Xiaokai Yan Jiwei Huang Mingheng Liao Xiangzheng Chen Dongming Liu Jin Li Bo Zhang Wen Teng Kefei Yuan Deqiang Sun Qiang Gao Yong Zeng | 2023 | Hepatobiliary Surgery and Nutrition2023,12,4: | 0 |

| 12 | Quantitative Gas-in-place Comparison of Original and Bitumen-free Lacustrine Shale显示文摘Residual bitumen in organic-rich shale of oil windows exists widely, and its effect on the gas storage capacity of shale could be two-fold. Bitumen could occupy and block the nanopores of shale, thereby reducing the gas storage capacity. On the other hand, gas could be dissolved in bitumen in shale gas reservoirs, leading to enhanced gas storage capacity. To quantify the effect of bitumen on the gas-in-place(GIP) estimation of lacustrine organic-rich shale, the micropore characteristics and methane sorption capacity of original and bitumen-free shale from the Triassic Yangchang Formation of the Ordos Basin, combined with the methane dissolution capacity for the isolated bitumen, were analyzed and compared. GIP for the original and bitumen-free shale in the depth range of 500–2500 m was evaluated. The results show that micropores in the shale samples were mainly related to organic matter. Clay mineral-hosted pores contributed slightly to microporosity. Bitumen significantly reduced the micropore surface area and volume of the original shale, with average percentages of 28.09% and 51.26%, respectively. The methane sorption capacity decreased after bitumen removal. When normalized to the original shale mass, the sum of the methane sorption capacity for bitumen-free shale and the methane dissolution capacity for isolated bitumen was similar to the methane sorption capacity of the original shale, indicating that the lack of methane absorbed on bitumen is the main reason for the decrease in methane sorption capacity after bitumen removal. The contribution of absorbed methane on bitumen to sorbed methane in shale could be higher than 36.23%. Dual effects of bitumen on shale GIP were observed. | 13 | Sugarcane bagasse amendment improves the quality of green waste vermicompost and the growth of Eisenia fetida显示文摘Vermicomposting is a feasible method for disposing of lignocellulosic waste while generating a useful product.The current study assessed the potential of vermicomposting green waste mixed with sugarcane bagasse in proportions of 25%,50%,and 75%(v:v,based on dry weight).The suitability was evaluated based on the agrochemical properties,earthworm biomass,and phytotoxicity.The final vermicomposts exhibited near-neutral pH values(7.1-7.6),and lower EC values(0.43-0.72 mS/cm)and C:N ratios(14.1-19.9).The content of available nutrients and CEC for all the vermicomposts exceeded those of the control compost(without earthworms).For vermicomposts,the average values of NO3-N,AP,AK,and CEC were 53,517,1362 mg/kg,and 158 cmol/kg,respectively.The total contents of heavy metals increased in all vermicompost treatments compared to control composts with the following average final percentages:Zn(2.0%),Cr(15.5%),Pb(23.4%),and Cu(44.3%),but these amounts were safe for application in agroforestry.The addition of sugarcane bagasse to green waste significantly increased the content of total humic substance,humic acid and urease activity,acid and alkaline phosphatase activity,and Eisenia fetida reproduction.The addition of 25%sugarcane bagasse to green waste decreased the toxicity to germinating seeds.These results revealed that vermicomposting is a feasible way to degrade green waste into a value-added chemical product. | Linlin Cai Xiangyang Sun Dan Hao Suyan Li Xiaoqiang Gong Hao Ding Kefei Yu | 2020 | Frontiers of Environmental Science & Engineering2020,14,4: | 0 |
