| 1 | Transgenic expression of MYB15 confers enhanced sensitivity to abscisic acid and improved drought tolerance in Arabidopsis thaliana显示文摘Abiotic stresses cause serious crop losses. Knowledge on genes functioning in plant responses to adverse growth conditions is essential for developing stress tolerant crops. Here we report that transgenic expression of MYB15, encoding a R2R3 MYB transcription factor in Arabidopsis thaliana, conferred hypersensitivity to exogenous abscisic acid (ABA) and improved tolerance to drought and salt stresses. The promoter of MYB15 was active in not only vegetative and reproductive organs but also the guard cells of stomata. Its transcript level was substantially upregulated by ABA, drought or salt treatments. Compared with wild type (WT) control, MYB15 overexpression lines were hypersensitive to ABA in germination assays, more susceptible to ABA-elicited inhibition of root elongation, and more sensitive to ABA-induced stomatal closure. In line with the above findings, the transcript levels of ABA biosynthesis (ABA1, ABA2), signaling (ABI3), and responsive genes (AtADH1, RD22, RD29B, AtEM6) were generally higher in MYB15 overexpression seedlings than in WT controls after treatment with ABA. MYB15 overexpression lines displayed improved survival and reduced water loss rates than WT control under water deficiency conditions. These overexpression lines also displayed higher tolerance to NaCl stress. | 2 | Significance of dredging on sediment denitrification in Meiliang Bay,China:A year long simulation study显示文摘An experiment for studying the effects of sediment dredging on denitrification in sediments was carried out through a one-year incubation of undredged (control) and dredged cores in laboratory. Dredging the upper 30 cm of sediment can significantly affect physico-chemical characteristics of sediments. Less degradation of organic matter in the dredged sediments was found during the experiment. Denitrification rates in the sediments were estimated by the acetylene blockage technique, and ranged from 21.6 to 102.7 nmol N2/(g dry weight (dw)·hr) for the undredged sediment and from 6.9 to 26.9 nmol N2/(g dw·hr) for dredged sediments. The denitrification rates in the undredged sediments were markedly higher (p<0.05) than those in the dredged sediments throughout the incubation, with the exception of February 2006. The importance of various environmental factors on denitrification was assessed, which indicated that denitrification was regulated by temperature. Nitrate was probably the key factor limiting denitrification in both undredged and dredged sediments. Organic carbon played some role in determining the denitrification rates in the dredged sediments, but not in the undredged sediments. | 6 | Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. | 8 | Measurements of ^(134)Cs and ^(137)Cs in urine and estimation of the internal dose of an adult exposed to the Chernobyl Accident显示文摘To estimate the internal dose of a Chinese visiting scholar internally contaminated because of the Cherno- byl Accident, the contents of 134Cs and 137Cs in urine were measured using a Ge(Li) γ-spectrometer. The internal doses were calculated based on data from the ICRP Publications. The effective doses from 134Cs and 137Cs were estimated to be 61 μSv and 98 μ Sv, respectively. The sum of 159 μSv was lower than the total effective dose (310 μSv), from the inhalation and ingestion of natural radionuclides. The dose of 131I was also reviewed referring to the UNSCEAR 2000 Report. The equivalent effective dose of 131I was estimated to be 2.9 mSv, 18 times more than the amount of 134Cs and 137Cs. | 9 | Effect of artificially accelerated aging on the vigor of Metasequoia glyptostroboides seeds显示文摘Metasequoia glyptostroboides is an endangered relict plant native to China that has been widely introduced into many countries and areas around the world. However,its seeds germinate at a very low percentage. Consequently,population regeneration by seed is low under natural conditions, which probably contributes to the endangered status of this plant species. The present study aimed to describe the aging mechanism of M. glyptostroboides seeds. Our objective was to elucidate causes of the low germination rate in an effort to enhance potential for conservation of the species. We used germination tests,relative electrical conductivity and malondialdehyde content determination, ultrastructural observation of embryo cells, and analysis of superoxide dismutase, catalase,ascorbate peroxidase, and dehydroascorbate reductase activities during accelerated aging treatment. | 10 | Electric conductivity and electric convertibility of potassium acetate in water, ethanol, 2,2,2–trifluoroethanol, 2–propanol and their binary blends显示文摘Salinity gradient energy between the concentrated and diluted electrolyte solutions can be converted to electric energy by using reverse electrodialysis(RED) technology. Electrolyte solution is a vital factor that impacts the energy conversion efficiency. Potassium acetate(KAc) was chosen as solute, and water, ethanol, 2,2,2-trifluoroethanol(TFE), 2-propanol(IPA) and several of their binary mixtures were selected as solvents. Electric conductivity of these solutions were measured under varying conditions. KAc was easily ionized in water and possessed the maximum electric conductivity, following by KAc–H_2O–TFE and KAc–H_2O–ethanol, and then KAc in pure TFE, ethanol, and IPA respectively. For electric convertibility of these solutions working in a RED power generation system, it was found that the KAc–H_2O possessed the maximum power density, and the KAc–ethanol–H_2O possessed the larger open circuit voltage than aqueous KAc solution under the same working condition. Besides, it was observed that both the electric conductivity and electric convertibility were significantly influenced by the concentration and temperature of solution. With the increasing of concentration,electric conductivity of these solutions increased firstly and then reached to the peak, but later it decreased.Solution temperature took a positive impact role to the electric conductivity. | 11 | De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. | 12 | Prognostic potential of an immune score based on the density of CD8+ T cells, CD20+ B cells, and CD33+/p-STAT1+ double-positive cells and HMGB1 expression within cancer nests in stage ⅢA gastric cancer patients显示文摘Objective: There is heterogeneity in the prognosis of gastric cancers staged according to the tumornodes-metastasis(TNM) system.This study evaluated the prognostic potential of an immune score system to supplement the TNM staging system.Methods: An immunohistochemical analysis was conducted to assess the density of T cells, B cells, and myeloid-derived suppressor cells(MDSCs) in cancer tissues from 100 stage ⅢA gastric cancer patients; the expression of the high-mobility group protein B1(HMGB1) was also evaluated in cancer cells.The relationship between the overall survival(OS), disease-free survival(DFS), and immunological parameters was analyzed.Results: An immune score system was compiled based on the prognostic role of the density of T cells, B cells, MDSCs, and the expression of HMGB1 in cancer tissues.The median 5-year survival of this group of patient was 32%.However, the 5-year survival rates of 80.0%, 51.7%, 0%, 5.8%, and 0% varied among the patients with an immune score of 4 to those with an immune score of 0 based on the immune score system, respectively.Similarly, differences in DFS rates were observed among the immune score subgroups.Conclusions: An immune score system could effectively identify the prognostic heterogeneity within stage ⅢA gastric cancer patients, implying that this immune score system may potentially supplement the TNM staging system, and help in identifying a more homogeneous group of patients who on the basis of prognosis can undergo adjuvant therapy. | Jun Dong Jiao Li Shiming Liu Xingyu Feng Shi Chen Zhiwei Zhou Yingbo Chen Xiaoshi Zhang | 2016 | Chinese Journal of Cancer Research2016,28,5: | 3 |
| 13 | Evaluation of Snow Depth and Snow Cover Fraction Simulated by Two Versions of the Flexible Global Ocean–Atmosphere–Land System Model显示文摘Based on historical runs, one of the core experiments of the fifth phase of the Coupled Model Intercomparison Project(CMIP5), the snow depth(SD) and snow cover fraction(SCF) simulated by two versions of the Flexible Global Ocean– Atmosphere–Land System(FGOALS) model, Grid-point Version 2(g2) and Spectral Version 2(s2), were validated against observational data. The results revealed that the spatial pattern of SD and SCF over the Northern Hemisphere(NH) are simulated well by both models, except over the Tibetan Plateau, with the average spatial correlation coefficient over all months being around 0.7 and 0.8 for SD and SCF, respectively. Although the onset of snow accumulation is captured well by the two models in terms of the annual cycle of SD and SCF, g2 overestimates SD/SCF over most mid- and high-latitude areas of the NH. Analysis showed that g2 produces lower temperatures than s2 because it considers the indirect effects of aerosols in its atmospheric component, which is the primary driver for the SD/SCF difference between the two models. In addition, both models simulate the significant decreasing trend of SCF well over(30°–70°N) in winter during the period 1971–94. | 16 | Deep Learning in Sheet Metal Bending With a Novel Theory-Guided Deep Neural Network显示文摘Sheet metal forming technologies have been intensively studied for decades to meet the increasing demand for lightweight metal components.To surmount the springback occurring in sheet metal forming processes,numerous studies have been performed to develop compensation methods.However,for most existing methods,the development cycle is still considerably time-consumptive and demands high computational or capital cost.In this paper,a novel theory-guided regularization method for training of deep neural networks(DNNs),implanted in a learning system,is introduced to learn the intrinsic relationship between the workpiece shape after springback and the required process parameter,e.g.,loading stroke,in sheet metal bending processes.By directly bridging the workpiece shape to the process parameter,issues concerning springback in the process design would be circumvented.The novel regularization method utilizes the well-recognized theories in material mechanics,Swift’s law,by penalizing divergence from this law throughout the network training process.The regularization is implemented by a multi-task learning network architecture,with the learning of extra tasks regularized during training.The stress-strain curve describing the material properties and the prior knowledge used to guide learning are stored in the database and the knowledge base,respectively.One can obtain the predicted loading stroke for a new workpiece shape by importing the target geometry through the user interface.In this research,the neural models were found to outperform a traditional machine learning model,support vector regression model,in experiments with different amount of training data.Through a series of studies with varying conditions of training data structure and amount,workpiece material and applied bending processes,the theory-guided DNN has been shown to achieve superior generalization and learning consistency than the data-driven DNNs,especially when only scarce and scattered experiment data are available for training which is often the case in practice.The theory-guided DNN could also be applicable to other sheet metal forming processes.It provides an alternative method for compensating springback with significantly shorter development cycle and less capital cost and computational requirement than traditional compensation methods in sheet metal forming industry. | Shiming Liu Yifan Xia Zhusheng Shi Hui Yu Zhiqiang Li Jianguo Lin | 2021 | IEEE/CAA Journal of Automatica Sinica2021,8,3: | 2 |

Methods Animals were injected with low dose gentamicin(100 mg/kg/day) for14 days, From the 14 th to 28 th days, the mice were maintained free of gentamicin treatment. Immunohistochemistry labeling was employed to trace RIBEYE, a major presynaptic componment of ribbon synapses.RIBEYE/CtBP2 expression levels were assessed and compared with hearing threshold shifts. Auditory function was assessed by auditory brainstem responses. The stereocilia of outer hair cells(OHCs) and IHCs was examined by scanning electron microscopy(SEM). Results Hearing thresholds were elevated with peak hearing loss observed on the 7th day after gentamicin exposure, followed by improvement after the 7th day.RIBEYE/CtBP2 expression directly correlated with observed hearing threshold shifts. Strikingly, we did not see any obvious changes in stereocilia in both OHCs and IHCs until the 28 th day. Mild changes in stereocilia were only observed in OHCs on the 28 th day. Conclusions These findings indicate that presynapse cochlear ribbons, rather than stereocilia, may be sensitive to aminoglycoside ototoxic exposure in mice cochleae. | 18 | Phenotypic similarities in pigs with SOX10^(c.321dupC)and SOX10^(c.325A>T)mutations implied the correlation of SOX10 haploinsufficiency with Waardenburg syndrome显示文摘SOX10 is a causative gene of Waardenburg syndrome(WS)that is a rare genetic disorder characterized by hearing loss and pigment disturbance.More than 100 mutations of SOX10 have been found in patients with Type 2 WS(WS2),Type 4 WS(WS4),and more complex syndromes.However,no mutation hotspot has been detected in SOX10,and most cases are sporadic,making it difficult to establish a correlation between the high phenotypic and genetic variability.In this study,a duplication of the 321th cytosine(c.321dupC)was introduced into SOX10 in pigs,which induced premature termination of the translation of SOX10(p.K108QfsX45).The premature stop codon in Exon 3 triggered the degradation of mutant mRNA through nonsense-mediated mRNA decay.However,SOX10^(c.321dupC) induced a highly similar phenotype of WS2 with heterogeneous inner ear malformation compared with its adjacent missense mutation SOX10^(c.325A>T).In addition,a site-saturation mutation analysis of the SOX10 N-terminal nuclear localization signal(n-NLS),where these two mutations located,revealed the correlation between SOX10 haploinsufficiency and WS by an in vitro reporter assay.The analysis combining the in vitro assay with clinical cases may provide a clue to clinical diagnoses. | Tingting Lin Lihua Luo Weiwei Guo Wei Ren Chuanhong Liu Hong Wei Shiming Yang Yong Wang | 2020 | Journal of Genetics and Genomics2020,47,12: | 2 |
| 19 | Olfactory and gustatory dysfunctions of COVID-19 patients in China:A multicenter study显示文摘Introduction:With the spread of the epidemic worldwide,an increasing number of doctors abroad have observed the following atypical symptoms of coronavirus disease 2019(COVID-19):olfactory or taste disorders.Therefore,clarifying the incidence and clinical characteristics of olfactory and taste disorders in Chinese COVID-19 patients is of great significance and urgency.Materials and Methods:A retrospective study was conducted,which included 229 severe acute respiratory syndrome coronavirus 2 confirmed patients,through face-to-face interviews and telephone follow-up.Following the completion of questionnaires,the patients participating in the study,were categorized according to the degree of olfactory and taste disorders experienced,and the proportion of each clinical type of patient with olfactory and taste disorders and the time when symptoms appeared were recorded.Results:Among the 229 patients,31(13.54%)had olfactory dysfunction,and 44(19.21%)had gustatory dysfunction.For the patients with olfactory dysfunction,6(19.35%)developed severe disease and became critically ill.Olfactory dysfunction appeared before the other symptoms in 21.43%of cases.The proportion of females with olfactory and gustatory dysfunction was higher than that of males(P<0.001).Conclusions:The incidence of olfactory and gustatory dysfunction was much lower than that reported abroad;the prognosis of patients with olfactory dysfunction is relatively favorable;olfactory and gustatory dysfunction can be used as a sign for early screening;females are more prone to olfactory and gustatory dysfunction. | Jianhui Li Yi Sun Enqiang Qin Hu Yuan Mingbo Liu Wenqi Yi Zhu Chen Chengcheng Huang Fengjie Zhou Ruiyao Chen Leibo Zhang Ning Yu Qiong Liu Xuejun Zhou Jingjing He Boyu Li Fusheng Wang Changliang Yang Shiming Yang | 2022 | World Journal of Otorhinolaryngology-Head and Neck Surgery2022,8,4: | 2 |
| 20 | Nitrogen addition and mowing alter drought resistance and recovery of grassland communities显示文摘Nitrogen enrichment and land use are known to influence various ecosystems,but how these anthropogenic changes influence community and ecosystem responses to disturbance remains poorly understood.Here we investigated the effects of increased nitrogen input and mowing on the resistance and recovery of temperate semiarid grassland experiencing a three-year drought.Nitrogen addition increased grassland biomass recovery but decreased structural recovery after drought,whereas annual mowing increased grassland biomass recovery and structural recovery but reduced structural resistance to drought.The treatment effects on community biomass/structural resistance and recovery were largely modulated by the stability of the dominant species and asynchronous dynamics among species,and the community biomass resistance and recovery were also greatly driven by the stability of grasses.Community biomass resistance/recovery in response to drought was positively associated with its corresponding structural stability.Our study provides important experimental evidence that both nitrogen addition and mowing could substantially change grassland stability in both functional and structural aspects.Our findings emphasize the need to study changes across levels of ecological organization for a more complete understanding of ecosystem responses to disturbances under widespread environmental changes. | Zhuwen Xu Heyong Liu Yani Meng Jinfei Yin Haiyan Ren Mai-He Li Shan Yang Shiming Tang Yong Jiang Lin Jiang | 2023 | Science China(Life Sciences)2023,66,7: | 2 |
