| 1 | Isolation and identification of novel casein-derived bioactive peptides and potential functions in fermented casein with Lactobacillus helveticus显示文摘The present study here establishes a complete and effective method for isolating,purifying and identifying extracellular and intracellular peptides,and also describes the characters and bioactivities of peptides from fermented casein with Lactobacillus helveticus.Intracellular peptides are much larger in quantity and more complex in composition than extracellular peptides,between which the correlation reveals proteolytic and metabolic mechanisms.In addition,totally 241 different peptide sequences were identified by Nano LC–MS/MS from casein(212)and Lactobacillus helveticus proteins(29).These casein-derived peptides mostly originated from-casein,followed byS1-casein,-casein,andS2-casein,and came from extracell(69)and intracell(143),in which common peptides have a total of 27.Forty-four of the identified peptides were previously described as bioactive,including angiotensinconverting enzyme(ACE)-inhibitory,antioxidant,immunomodulating,antimicrobial,DPP-IV inhibitory,antiamnesic and anticancer effects and so on.Thirteen peptides with the potential of some biological activities are obtained,which were described in previous studies.A total of 47 novel peptides of 5 to 26 amino acids that were not disclosed were obtained.The new sources of natural bioactive peptides may have the very high application value as potential new peptide drugs for treatment human diseases.The product peptide DELQDKIHPF found in both extracell and intracell was quantitatively analyzed using the MRM mode of UPLC-U3Q,23.1 and 9.76 ng/mL,respectively.The quantitative analysis of the potential bioactive peptide may also advance the production of peptide products in the future. | Mengzhu Fan Tingting Guo Wanru Li Jing Chen Fushuo Li Chao Wang Yi Shi David Xi-an Li Shaohui Zhang | 2019 | Food Science and Human Wellness2019,8,2: | 5 |
| 2 | A botanical medicine dragon’s blood exhibited clinical antithrombosis efficacy similar to low molecular weight heparin显示文摘Deep vein thrombosis(DVT)is a common complication following traumatic fracture with a 0.5%–1%annual incidence.Low molecular weight heparin(LMWH)is the most commonly used anticoagulation drug for DVT prevention,but treatment with LMWH is invasive.Our aim is to compare the antithrombotic effect of dragon’s blood,an oral botanical anticoagulant medicine approved by the Chinese FDA,with LMWH in patients undergoing hip fracture surgery and to explore the molecular mechanisms of anticoagulation treatment.Our study recruited patients and divided them into LMWH and dragon’s blood treatment group.Coagulation index tests,Doppler ultrasound and mRNA sequencing were performed before and after anticoagulation therapy.There was no significant difference in postoperative DVT incidence between the two groups(23.1%versus 15.4%,P=0.694).D-dimer(D-D)and fibrinogen degradation product(FDP)showed significant reductions in both groups after anticoagulation treatments.We identified SLC4A1,PROS1,PRKAR2B and seven other genes as being differentially expressed during anticoagulation therapy in both groups.Genes correlated with coagulation indexes were also identified.Dragon’s blood and LMWH showed similar effects on DVT and produced similar gene expression changes in patients undergoing hip fracture surgery,indicating that dragon’s blood is a more convenient antithrombosis medicine(oral)than LMWH(hypodermic injection). | Jiangbin Liang Song Mei Xiangyu Qiao Wei Pan Yan Zhao Shaohui Shi Yaling Zhai Haizhao Wen Guoping Wu Chengyu Jiang | 2021 | Science China(Life Sciences)2021,64,10: | 4 |
| 3 | Degradation of the antibiotic sulfamonomethoxine sodium in aqueous solution by photo-Fenton oxidation显示文摘In this study, photo-Fenton oxidation was applied to degradation of sulfamonomethoxine sodium (SMMS) in aqueous solution. The operation parameters of pH, temperature, and concentrations of H2O2, Fe2+ and SMMS were investigated. The optimum conditions for the photo-Fenton process were determined as follows: [SMMS]=4.53 mg/L, pH 4.0, [H2O2]=0.49 mmol/L, [Fe2+]= 19.51 μmol/L and T=25°C. Under these conditions 98.5% of the SMMS degraded. The kinetics were also studied, and degradation of SMMS by the photo-Fenton process could be described by first-order kinetics. The apparent activation energy was calculated as 23.95 kJ/mol. Mineralization of the process was investigated by measuring the chemical oxygen demand (COD), and the COD decreased by 99% after 120 min. | 8 | Pathogenic analysis of coxsackievirus A10 in rhesus macaques显示文摘Coxsackievirus A10(CV-A10)is one of the etiological agents associated with hand,foot and mouth disease(HFMD)and also causes a variety of illnesses in humans,including pneumonia,and myocarditis.Different people,particularly young children,may have different immunological responses to infection.Current CV-A10 infection animal models provide only a rudimentary understanding of the pathogenesis and effects of this virus.The characteristics of CV-A10 infection,replication,and shedding in humans remain unknown.In this study,rhesus macaques were infected by CV-A10 via respiratory or digestive route to mimic the HFMD in humans.The clinical symptoms,viral shedding,inflammatory response and pathologic changes were investigated in acute infection(1–11 day post infection)and recovery period(12–180 day post infection).All infected rhesus macaques during acute infection showed obvious viremia and clinical symptoms which were comparable to those observed in humans.Substantial inflammatory pathological damages were observed in multi-organs,including the lung,heart,liver,and kidney.During the acute period,all rhesus macaques displayed clinical signs,viral shedding,normalization of serum cytokines,and increased serum neutralizing antibodies,whereas inflammatory factors caused some animals to develop severe hyperglycemia during the recovery period.In addition,there were no significant differences between respiratory and digestive tract infected animals.Overall,all data presented suggest that the rhesus macaques provide the first non-human primate animal model for investigating CV-A10 pathophysiology and assessing the development of potential human therapies. | Suqin Duan Fengmei Yang Yanyan Li Yuan Zhao Li Shi Meng Qin Quan Liu Weihua Jin Junbin Wang Lixiong Chen Wei Zhang Yongjie Li Ying Zhang Jingjing Zhang Shaohui Ma Zhanlong He Qihan Li | 2022 | Virologica Sinica2022,37,4: | 1 |
| 9 | Noninvasive fluid bubble detection based on capacitive micromachined ultrasonic transducers显示文摘Ultrasonic fluid bubble detection is important in industrial controls,aerospace systems and clinical medicine because it can prevent fatal mechanical failures and threats to life.However,current ultrasonic technologies for bubble detection are based on conventional bulk PZT-based transducers,which suffer from large size,high power consumption and poor integration with ICs and thus are unable to implement real-time and long-term monitoring in tight physical spaces,such as in extracorporeal membrane oxygenation(ECMO)systems and dialysis machines or hydraulic systems in aircraft.This work highlights the prospect of capacitive micromachined ultrasonic transducers(CMUTs)in the aforementioned application situations based on the mechanism of received voltage variation caused by bubble-induced acoustic energy attenuation.The corresponding theories are established and well validated using finite element simulations.The fluid bubbles inside a pipe with a diameter as small as 8 mm are successfully measured using our fabricated CMUT chips with a resonant frequency of 1.1 MHz.The received voltage variation increases significantly with increasing bubble radii in the range of 0.5–2.5 mm.Further studies show that other factors,such as bubble positions,flow velocities,fluid medium types,pipe thicknesses and diameters,have negligible effects on fluid bubble measurement,demonstrating the feasibility and robustness of the CMUT-based ultrasonic bubble detection technique. | Jiawei Yuan Zhikang Li Qi Ma Jie Li Zixuan Li Yihe Zhao Shaohui Qin Xuan Shi Libo Zhao Ping Yang Guoxi Luo Xiaozhang Wang Kwok Siong Teh Zhuangde Jiang | 2023 | Microsystems & Nanoengineering2023,9,1: | 0 |
| 10 | The SMAD2/miR-4256/HDAC5/p16^(INK4a) signaling axis contributes to gastric cancer progression显示文摘The dysregulation of exosomal microRNAs(miRNAs)plays a crucial role in the development and progression of cancer.This study investigated the role of a newly identified serum exosomal miRNA miR-4256 in gastric cancer(GC)and the underlying mechanisms.The differentially expressed miRNAs were firstly identified in serum exosomes of GC patients and healthy individuals using next-generation sequencing and bioinformatics.Next,the expression of serum exosomal miR-4256 was analyzed in GC cells and GC tissues,and the role of miR-4256 in GC was investigated by in vitro and in vivo experiments.Then,the effect of miR-4256 on its downstream target genes HDAC5/p16^(INK4a) was studied in GC cells,and the underlying mechanisms were evaluated using dual luciferase reporter assay and Chromatin Immunoprecipitation(ChIP).Additionally,the role of the miR-4256/HDAC5/p16^(INK4a) axis in GC was studied using in vitro and in vivo experiments.Finally,the upstream regulators SMAD2/p300 that regulate miR-4256 expression and their role in GC were explored using in vitro experiments.miR-4256 was the most significantly upregulated miRNA and was overexpressed in GC cell lines and GC tissues;in vitro and in vivo results showed that miR-4256 promoted GC growth and progression.Mechanistically,miR-4256 enhanced HDAC5 expression by targeting the promoter of the HDAC5 gene in GC cells,and then restrained the expression of p16^(INK4a) through the epigenetic modulation of HDAC5 at the p16INK4a promoter.Furthermore,miR-4256 overexpression was positively regulated by the SMAD2/p300 complex in GC cells.Our data indicate that miR-4256 functions as an oncogene in GC via the SMAD2/miR-4256/HDAC5/p16^(INK4a) axis,which participates in GC progression and provides novel therapeutic and prognostic biomarkers for GC. | MIN WANG HAILIANG ZHAO WEIWEI CHEN CAIQUN BIE JINYING YANG WENRUI CAI CHUTIAN WU YANFANG CHEN SHUFEN FENG YING SHI YUTING LI HUIJUN TANG LIXIAN ZHONG LILIANGZI GUO SISI CHEN LINJING LONG SHAOHUI TANG | 2023 | Oncology Research2023,31,4: | 0 |

| 13 | Remediation mechanism of“double-resistant”bacteria—Sedum alfredii Hance on Pb-and Cd-contaminated soil显示文摘Background:Concentrations of heavy metals continue to increase in soil environments as a result of both anthro-pogenic activities and natural processes.Cadmium(Cd)and lead(Pb)is one of the most toxic heavy metals and pose health risks to both humans and the ecosystem.Therefore,effectively solving the problem of heavy metal pollution is the concern of soil workers.Among the existing remediation techniques,only the combined use of microorganisms and plants for remediation of heavy metal-contaminated soil is the greenest and most developed one.Consequently,based on this background,this study investigates the remediation mechanism of Pb and Cd heavy metals using the combined action of bacteria and Sedum alfredii Hance.Methods:In order to enrich the research theory of combined plant and microorganism remediation of heavy metal-contaminated soil,we constructed a heavy metal composite pollution remediation system by combining Pb and Cd-tolerant bacteria with the Pb and Cd hyperaccumulator plant—Sedum alfredii Hance to investigate its combined remediation effect on Pb and Cd composite contaminated soil.Results:The results showed that resistant bacteria were able to promote enrichment of Pb and Cd in Sedum alfredii Hance and J2(200 ml of bacterial solution)was significantly(P<0.05)more effective than J1(100 ml of bacterial solution).The resistant bacteria were able to alleviate the toxic effects of Pb and Cd heavy metals on Sedum alfredii Hance and promote growth while reducing rhizosphere soil pH.The resistant bacteria were able to significantly reduce the effective state of Pb and Cd in the rhizosphere soil(P<0.05),with the greatest reduction in the effective state of Pb in treatment A(Cd7Pb100 mg/kg),where J2 was reduced by 9.98%compared to J0,and the greatest reduction in the effective state of Cd in treatment C(Cd28Pb400 mg/kg),where J2 was 43.53%lower than J0.In addition,the resistant bacteria were able to increase the exchangeable state Cd content by 0.97 to 9.85%.The resistant bacteria had a weakly promoting effect and a highly inhibitory effect on the absorption of Pb by Sedum alfredii Hance.Conclusions:The resistant bacteria can change the rhizosphere environment and significantly improve the remedia-tion effect of Sedum alfredii Hance on heavy metal cadmium.The role of“double-resistant”bacteria in promoting the accumulation of Cd was greater than that of Pb. | Tongdong Mo Daihua Jiang Dingding Shi Shaohui Xu Xuejiao Huang Zhigang Huang | 2022 | Ecological Processes2022,11,1: | 0 |
