1Effects of Nogo-neutralizing antibody and neurotrophin-3 on axonal regeneration following spinal cord injury in rats显示文摘BACKGROUND: Recent studies have suggested that regeneration of the central nerve fiber following spinal cord injury occurs under specific conditions. OBJECTIVE: To study the effects of Nogo-neutralizing antibody (IN-1), in combination with neurotrophin-3 (NT-3), on axonal regeneration and motor function following spinal cord injury in the rat. DESIGN, TIME AND SETTING: A randomized, controlled, animal study combining immunohistochemistry was performed at the Laboratory of Neuroanatomy of Xiangya Medical College, and Central Laboratory of Xiangya the Third Hospital, Central South University from January 2006 to December 2007. MATERIALS: Eighteen healthy, Sprague Dawley rats were randomly divided into three groups, with six rats per group: control, IN-1, and IN-1/NT-3. Hemisectioned spinal cord injury models were established by cutting the posterior 2/3 of spinal cord, which is equivalent to the T8 level. METHODS: A polyethylene tubing was inserted through into subarachnoid cavity, equivalent to the superior margin at the T8 level. Saline, IN-1, and IN-1/NT-3 were respectively injected into control, IN-1, and IN-1/NT-3 groups, three times/day for seven consecutive days. MAIN OUTCOME MEASURES: At 2 weeks post-surgery, biotin dextran amine (10%) was injected into the right sensorimotor cortex area. At day 28 post-surgery, spinal cord tissue was prepared for frozen sections. Positive astrocytic expression was observed with glial fibrillary acidic protein (GFAP) immunohistochemical staining whose proliferation level was represented by gray value, i.e. the higher the gray value was, the less the positive cells were, and growth of positive fibers was observed with a biotin dextran amine histological reaction. Motor function was measured according to BBB scores pre-operatively, as well as at days 1, 7, 14, 21, and 28 post-operatively. RESULTS: Three rats died during experimentation. By random supplement, a total of 18 rats were included. GFAP-positive astrocytes were observed in all the three groups. In the control group, astrocytes were characterized according to active function, hyperplasia, proliferation, hypertrophy, and increasing processes as compared to IN-1 group and IN-1/IN-3 group. Astrocyte hyperplasia represented by gray value in the IN-1 group was less than the control group. Gray value of GFAP-positive products in the IN-1/IN-3 group was higher than other two groups (P < 0.05). Biotin dextran amine tracing demonstrated no corticospinal tract fiber outgrowth following spinal cord injury; the fibers were incapable of passing through the glial scar in the control group. Several fibers were distributed in the proximal scar tissue region in the IN-1 group, and the regenerated fibers were disarranged. Many nerve fibers were distributed throughout the scar tissue, and even several biotin dextran amine-positive fibers were observed at the distal end of the injured segment. Post-operative Basso, Beattie, Bresnahan scores were greater than pre-operative ones, while Basso, Beattie, Bresnahan scores in the IN-1/NT-3 group were significantly greater than the other two groups at days 14, 21, and 28 post-surgery (P < 0.05). CONCLUSION: IN-1, in combination with NT-3, promoted axonal regeneration following spinal cord injury, inhibited the colloidal effect, and enhanced the correlation between proximal and distal processes to recover motor function. 2Three Types of Spatial Function Zoning in Key Ecological Function Areas Based on Ecological and Economic Coordinated Development: A Case Study of Tacheng Basin, China显示文摘Three types of spatial function zoning is an effective measure for regional environmental protection and orderly development.For ecological and economic coordinated development, spatial function zones should be divided scientifically to clear its direction of development and protection. Therefore, based on ecological constraints, a beneficial discussion would be about the key ecological function areas adopting the concept of ecological protection restriction and supporting socioeconomic development for spatial function zoning. In this paper, the researchers, taking Tacheng Basin, Xinjiang of China as an example, choose township as basic research unit and set up an evaluation index system from three aspects, namely, ecological protection suitability, agricultural production suitability, and urban development suitability, which are analyzed by using spatial analysis functions and exclusive matrix method. The results showed that: 1) This paper formed a set of multilevel evaluation index systems for three types of spatial function zoning of the key ecological function areas based on a novel perspective by scientifically dividing Tacheng Basin into ecological space, agricultural space, and urban space,which realized the integration and scientific orientation for spatial function at the township scale. 2) Under the guidance of three types of spatial pattern, the functional orientation and suggestions of development and protection was clearly defined for ecological protection zones,ecological economic zones, agricultural production zones, and urban development zones. 3) A new idea of space governance is provided to promote the coordinated and sustainable development between ecology and economy, which can break the traditional mode of thinking about regional economic development, and offers a scientific basis and reference for macro decision-making.WANG Guiling YANG Degang XIA Fuqiang ZHONG Ruisen XIONG Chuanhe 2019Chinese Geographical Science2019,29,4:2
3Effects of combined application of Nogo-neutralizing antibody IN-1 and neurotrophin-3 on c-Fos and c-Jun expression in a rat model of hemisection spinal cord injury显示文摘BACKGROUND:Nogo-neutralizing antibody IN-1 accelerates axon growth and enhances recovery of spinal cord function by inhibiting growth inhibitory factors. Neurotrophin-3 (NT-3)contributes to regeneration of nerve fibers in the spinal cord and motor function recovery. The combination of Nogo-neutralizing antibody IN-1 and NT-3 is hypothesized to produce better outcomes and facilitate axonal regeneration by affecting c-Fos and c-Jun protein expression. OBJECTIVE:To investigate the combined effects of Nogo-neutralizing antibody IN-1 and NT-3 on c-Fos and c-Jun protein levels in the injured spinal cord. DESIGN,TIME AND SETTING:A randomized,controlled study was performed at the Laboratory of Neuroanatomy,Xiangya Medical College,Central South University and the Central Laboratory of Third Xiangya Hospital of China from June 2005 to December 2007. MATERIALS:NT-3 (Peprotech,USA) and Nogo-neutralizing antibody IN-1 (Santa Cruz Biotechnology,USA) were used in this study. METHODS:Hemisectioned spinal cord injury models were established by cutting the posterior 2/3 of rat spinal cord,which is equivalent to the T8 level in the human spine. A total of 120 rats were equally and randomly assigned to three groups:model (0.2 μL saline),IN-1 (0.2 μL IN-1),and IN-1/NT-3 (0.2 μL IN-1 + 0.2 μL NT-3). The compounds were separately infused into transection sites on the side of head. MAIN OUTCOME MEASURES:Western blot analysis was employed to measure c-Fos and c-Jun protein expression in the injured spinal cord at 15,30 minutes,1,2,4,6,8,and 12 hours following surgery. RESULTS:Following spinal cord injury,c-Fos and c-Jun protein expression were increased and peaked at 4-6 hours. Following injection of IN-1 or the combination of IN-1 and NT-3,c-Fos protein expression was significantly reduced in the injured spinal cord (P < 0.05 or P < 0.01) (with the exception of the 15 minute time point). However,c-Jun protein expression was significantly increased (P < 0.05 or P < 0.01) (with the exception of the 15 and 30 minute time points). Combined application of IN-1 and NT-3 resulted in significantly altered protein expression compared to IN-1 alone. CONCLUSION:IN-1 increases c-Jun protein levels and protects the injured spinal cord by inhibiting c-Fos protein levels. 4Divergent apparent temperature sensitivity of terrestrial ecosystem respiration显示文摘Aims Recent studies revealed convergent temperature sensitivity of ecosys-tem respiration(Re)within aquatic ecosystems and between terrestrial and aquatic ecosystems.We do not know yet whether various terres-trial ecosystems have consistent or divergent temperature sensitivity.Here,we synthesized 163 eddy covariance flux sites across the world and examined the global variation of the apparent activation energy(Ea),which characterizes the apparent temperature sensitivity of and its interannual variability(IAV)as well as their controlling factors.Methods We used carbon fluxes and meteorological data across FLUXNET sites to calculate mean annual temperature,tempera-ture range,precipitation,global radiation,potential radiation,gross primary productivity and Re by averaging the daily values over the years in each site.Furthermore,we analyzed the sites with>8 years data to examine the IAV of Ea and calculated the standard deviation of Ea across years at each site to character-ize IAV.Important Findings The results showed a widely global variation of Ea,with significantly lower values in the tropical and subtropical areas than in temperate and boreal areas,and significantly higher values in grasslands and wetlands than that in deciduous broadleaf forests and evergreen for-ests.Globally,spatial variations of Ea were explained by changes in temperature and an index of water availability with differing contribution of each explaining variable among climate zones and biomes.IAV and the corresponding coefficient of variation of Ea decreased with increasing latitude,but increased with radiation and corresponding mean annual temperature.The revealed patterns in the spatial and temporal variations of Ea and its controlling factors indicate divergent temperature sensitivity of Re,which could help to improve our predictive understanding of Re in response to climate change.Bing Song Shuli Niu Ruisen Luo Yiqi Luo Jiquan Chen Guirui Yu Janusz Olejnik Georg Wohlfahrt Gerard Kiely Asko Noormets Leonardo Montagnani Alessandro Cescatti Vincenzo Magliulo Beverly Elizabeth Law Magnus Lund Andrej Varlagin Antonio Raschi Matthias Peichl Mats BNilsson Lutz Merbold 2014Journal of Plant Ecology2014,7,5:2

10Bending wave intensity in a thick beam显示文摘In this paper the expressions are derived for estimating bending wave intensity and its components at arbitrary frequency in one-dimensional structures such as beams. There is a critical frequency fo above which there are two bending waves with different wave numbers propagating along a structure and the estimates of bending wave intensity and its components become quite complicated, and it may not be possible to measure bending wave intensity using the existing intensity techniques. The expressions are also derived for calculating approximate estimate error resulting from using the existing intensity theory. 15THE ESTIMATE AND MEASUREMENT OF TRANSVERSE WAVE INTENSITY显示文摘Transverse waves are a type of structural waves and should be considered in the analy-sis of high frequency vibration because the energy carried by transverse waves increases with the in-crease of frequency and becomes important at high frequencies. This paper studies the estimate theoryand measuring technique of the transverse wave intensity in two dimensional homogeneous structures.In general, the intensity vector is the sum of the effective intensity vector and the intensity variationvector. Each axial intensity component is proportional to two imaginary parts of cross spectral densitiesand its estimate is complicated. For the special case where transverse waves propagate in one direction,the intensity variation is zero and the estimate of the intensity is simplified. The intensity technique isformed based on the finite difference principle. Transverse wave intensity can be measured using a pairof two-transducer arrays lying in the orthogonal direction for the general case or a two-transducer ar-ray lying in the propagating direction for the special case. In order to assess the measurement accuracyof transverse wave intensity, the coupling loss factors from bending to transverse waves in buildingstructures were measured using the intensity technique and compared with the results predicted andmeasured using the conventional method. 17Pangenome of water caltrop reveals structural variations and asymmetric subgenome divergence after allopolyploidization显示文摘Water caltrop(Trapa spp.,Lythraceae)is a traditional but currently underutilized non-cereal crop.Here,we generated chromosome-level genome assemblies for the two diploid progenitors of allotetraploid Trapa.natans(4x,AABB),i.e.,diploid T.natans(2x,AA)and Trapa incisa(2x,BB).In conjunction with four published(sub)genomes of Trapa,we used gene-based and graph-based pangenomic approaches and a pangenomic transposable element(TE)library to develop Trapa genomic resources.The pangenome displayed substantial gene-content variationwith dispensable and private gene clusters occupying a large proportion(51.95%)of the total cluster sets in the six(sub)genomes.Genotyping of presence-absence variation(PAVs)identified 40453 PAVs associated with 2570 genes specific to A-or B-lineages,of which 1428were differentially expressed,andwere enriched in organ development process,organic substancemetabolic process and response to stimulus.Comparative genome analyses showed that the allotetraploid T.natans underwent asymmetric subgenome divergence,with the B-subgenome being more dominant than the A-subgenome.Multiple factors,including PAVs,asymmetrical amplification of TEs,homeologous exchanges(HEs),and homeolog expression divergence,together affected genome evolution after polyploidization.Overall,this study sheds lights on the genome architecture and evolution of Trapa,and facilitates its functional genomic studies and breeding program.Xinyi Zhang Yang Chen Lingyun Wang Ye Yuan Mingya Fang Lin Shi Ruisen Lu Hans Peter Comes Yazhen Ma Yuanyuan Chen Guizhou Huang Yongfeng Zhou Zhaisheng Zheng Yingxiong Qiu 2023Horticulture Research2023,10,11:0
18Molecular mechanisms of seasonal photoperiod effects of the pineal gland on the hippocampus in rats显示文摘Background:Based on the theory of“five Zang-organs corresponding to the seasons”in traditional Chinese medicine(TCM),physiological functions including emotions vary with the seasons.We aimed to investigate the seasonal photoperiod effects of melatonin(MT)released from the pineal gland on the MT receptor(MTR)-Gs/Gi-cyclic adenosine monophosphate(cAMP)-protein kinase A(PKA)-cAMP response element-binding protein(CREB)signaling pathway in the hippocampus.Methods:Rats were divided into three groups:control,operation(surgery with pineal gland removal),and pseudo-operation groups(same surgery as operation group but without removing pineal gland),and fed at specific time across the four seasons.The levels of MTR,adenylate cyclase(AC),cAMP,PKA,and CREB in the hippocampus were analyzed using ELISA.The concentrations of Gs and Gi were analyzed using Western blot.The expression of CREB mRNA was detected by PCR.Results:For intragroup comparisons,compared with spring,the levels of Gs/Gi in the control group were higher in summer,autumn,and winter(P=.009 in summer;P<.001 in autumn and winter);the levels of MTR,cAMP,PKA,and CREB in the control group were significantly higher in autumn and winter than in spring(all P<.001).The levels of MTR,cAMP,PKA,and CREB in each season were significantly lower in the operation group than in the control group(all P<.05).Significant differences were noted in Gs/Gi levels between the operation group and control group in spring,autumn,and winter(all P<.05).Conclusion:Our findings suggest that MTR-Gs/Gi-cAMP-PKA-CREB signaling pathway is involved in the seasonal photoperiod effects of the pineal gland on the hippocampus and may underpin seasonal changes in emotions.It can support the prevention and treatment of the seasonal onset of mental disorders,and enrich the theory of“five Zang-organs corresponding to the seasons”.Qi Han Guowen Du Leilei Liu Lepeng Wang Wenna Li Hairong Zhang Yike Sun Pei Zhu Ruisen Hao Shuran Ma 2021Journal of Traditional Chinese Medical Sciences2021,8,2:0
19The measurement of bending wave intensity in a two-dimensional structure显示文摘In a two-dimensional bending wavefield, the intensity vector is the sum of the effec-tive intensity vector and the intensity variation vector. The effective intensity vector gives the magnitude and direction of bending wave power flow per unit width in structures. The illtensity variation vector characterizes the circulation of local bending wave energy and varies harmon ically with space and wavenumber. This paper studies the relationship between the analysis frequency bandwidth and the measurement acctiracy of effective bending wave intensity, and ex-amines the accuracy of source localization using measured intensity vectors in two-dimensional structures. It is shown that if the analysis frequency bandwidth is wider than the sum of several one-third octave frequency bandwidths, the intensity variation can be suppressed to a negligible value in comparison with the effective intensity Moreover, a structure-borne source can be localized by measuring effective bending wave intensity vectors at a few positions, but the accuracy is dependent on the analysis frequency bandwidth, the property of the wavefield and the distance between source and measurement position. 20Three bias errors in the measurement of structural intensity显示文摘There are many possible bias errors in the measurement of structural intensity and some of them have been theoretically examined. Attempting to analyse all the bias errors at the same time results in a very complicated analysis and makes it difficult to draw clear conclusions.The bias errors are usually analysed individually. In this paper a theoretical study of three bias errors in the measurement of structural intensity is presented by using the twor-accelerometer array technique. It is assumed that the physical and material properties of the test structure are known.
