2罗格列酮和全反式维甲酸对多发性骨髓瘤裸鼠移植瘤生长及血管形成的影响显示文摘目的研究罗格列酮(RGZ)和全反式维甲酸(ATRA)对多发性骨髓瘤裸鼠移植瘤生长和血管形成的影响。方法分别以不同浓度的RGZ和ATRA或RGZ+ATRA处理U266细胞24h,采用半定量RT—PCR法检测各组U266细胞中血管内皮细胞生长因子(VEGF)的表达。将U266细胞接种于4周龄Balb/c裸鼠一侧肩胛部皮下,接种1周后分别以RGZ和RGZ+ATRA腹腔注射干预,连续给药21d。第21天处死裸鼠,剥离皮下肿瘤并称重,将肿瘤组织分别进行HE和CD34、VEGF免疫组化染色。结果未经处理的对照组U266细胞中VEGFmRNA高表达;经RGZ处理后,VEGFmRNA的表达水平下调;经RGZ+ATRA处理后,VEGFmRNA的表达水平较RGZ组更低。U266细胞接种后,在裸鼠皮下均可形成肿瘤。腹腔用药后,RGZ组裸鼠移植瘤的体积和重量分别为(785±262)mm3和(1748±365)mg,均明显低于对照组(均P〈0.01);RGZ+ATRA组移植瘤的体积和再量分别为(154±89)mm3和(626±102)mg3均明显低于RGZ组(均P〈0.05)。各组裸鼠移植瘤中均有VEGF蛋白的表达,其中对照组VEGF蛋白的平均染色强度(IS)为2.20±0.40,明显高于RGZ组(1.48±0.37,P〈0.01);RGZ+ATRA组的Is为0.58±0.26,明显低于RGZ组(P〈0.01)。各组裸鼠移植瘤中均有CD34的表达,其中对照组CD34表达最强,微血管密度(MVD)为(56.4±15.2)个/高倍视野,明显高于RGZ组[(44.6±11.2)个/高倍视野,P〈0.05];RGZ+ATRA组CD34表达最弱,MVD为(21.5±8.6)个/高倍视野,明显低于RGZ组(P〈0.01)。结论RGZ和ATRA存体内环境中也能发挥抑制骨髓瘤细胞增殖和生长的作用。通过抑制骨髓瘤细胞中VEGF的表达从而阻断血管形成,可能也足RGZ和ATRA抗肿瘤作用的重要机制之一。黄海雯 陈萍 李炳宗 傅晋翔 李军 张晓慧 刘睿 范银银 张宏 Howard C.H. 10罗格列酮与全反式维甲酸对骨髓瘤细胞分化的影响及其可能机制显示文摘目的探讨过氧化物酶体增殖物活化受体γ(PPARγ)的配体罗格列酮(RGZ)与全反式维甲酸(ATRA)对骨髓瘤细胞分化的影响及其可能机制。方法以RGZ和ATRA处理人骨髓瘤细胞U266和RPMI-8226,用^3H-掺入法检测细胞增殖变化;碘化丙啶(PI)染色后,以流式细胞仪检测细胞周期变化;瑞氏-姬母萨染色观察细胞形态学变化;流式细胞仪分析细胞表面CD49e的表达变化;Western blot方法检测p27^Kip1和p21^Waf1的表达变化。结果RGZ能显著抑制U266和RPMI-8226细胞的增殖,并且这种抑制作用呈剂量依赖性。经5μmol/LRGZ处理后,U266和RPMI-8226细胞G0/G1期细胞的比例分别为(45.2±6.7)%和(40.3±7.3)%,较对照组明显增加(均P〈0.05);G2/M期和S期细胞的比例分别为(52.2±7.4)%和(57.4±9.5)%,较对照组明显减少(均P〈0.05);CIM9e的表达率分别为(8.7±1.5)%和(7.9±1.6)%,较对照组明显上调(均P〈0.05)。以10μmol/LRGZ处理后,U266和RPMI-8226细胞的G0/G1期、G2/M期和S期细胞比例、CD49e的表达率均较5μmol/LRGZ组的变化更加显著。同时,经RGZ处理后,U266和RPMI-8226细胞的形态学变化也符合骨髓瘤细胞分化成熟的规律,p27^Kip1和p21^Waf1蛋白的表达量也较对照组明显增加。RGZ和ATRA联合对U266和RPMI-8226细胞影响较RGZ单独作用更加明显。结论RGZ可能通过上调p27^Kip1和p21^Waf1蛋白的表达,介导骨髓瘤细胞的周期阻滞,并诱导骨髓瘤细胞分化,抑制骨髓瘤细胞增殖。ATRA能增强RGZ对骨髓瘤细胞的上述作用,两者联合具有协同效应。黄海雯 陈广华 常惠荣 Howard C.H. 13Ⅲ/Ⅴ-on-Si MQW lasers by using a novel photonic integration method of regrowth on a bonding template显示文摘Silicon photonics is becoming a mainstream data-transmission solution for next-generation data centers,highperformance computers,and many emerging applications.The inefficiency of light emission in silicon still requires the integration of a Ⅲ/Ⅴ laser chip or optical gain materials onto a silicon substrate.A number of integration approaches,including flip-chip bonding,molecule or polymer wafer bonding,and monolithic Ⅲ/Ⅴ epitaxy,have been extensively explored in the past decade.Here,we demonstrate a novel photonic integration method of epitaxial regrowth of Ⅲ/Ⅴ on a Ⅲ/Ⅴ-on-SOI bonding template to realize heterogeneous lasers on silicon.This method decouples the correlated root causes,i.e.,lattice,thermal,and domain mismatches,which are all responsible for a large number of detrimental dislocations in the heteroepitaxy process.The grown multi-quantum well vertical p-i-n diode laser structure shows a significantly low dislocation density of 9.5×104 cm^(−2),two orders of magnitude lower than the state-of-the-art conventional monolithic growth on Si.This low dislocation density would eliminate defect-induced laser lifetime concerns for practical applications.The fabricated lasers show room-temperature pulsed and continuous-wave lasing at 1.31μm,with a minimal threshold current density of 813 A/cm^(2).This generic concept can be applied to other material systems to provide higher integration density,more functionalities and lower total cost for photonics as well as microelectronics,MEMS,and many other applications.Yingtao Hu Di Liang Kunal Mukherjee Youli Li Chong Zhang Geza Kurczveil Xue Huang Raymond G.Beausoleil 2019Light(Science & Applications)2019,8,1:1
