1MiR-146a-5p targeting SMAD4 and TRAF6 inhibits adipogenensis through TGF-β and AKT/mTORC1 signal pathways in porcine intramuscular preadipocytes显示文摘Background: Intramuscular fat(IMF) content is a vital parameter for assessing pork quality. Increasing evidence has shown that microRNAs(miRNAs) play an important role in regulating porcine IMF deposition. Here, a novel miRNA implicated in porcine IMF adipogenesis was found, and its effect and regulatory mechanism were further explored with respect to intramuscular preadipocyte proliferation and differentiation.Results: By porcine adipose tissue miRNA sequencing analysis, we found that miR-146a-5p is a potential regulator of porcine IMF adipogenesis. Further studies showed that miR-146a-5p mimics inhibited porcine intramuscular preadipocyte proliferation and differentiation, while the miR-146a-5p inhibitor promoted cell proliferation and adipogenic differentiation. Mechanistically, miR-146a-5p suppressed cell proliferation by directly targeting SMAD family member 4(SMAD4) to attenuate TGF-β signaling. Moreover, miR-146a-5p inhibited the differentiation of intramuscular preadipocytes by targeting TNF receptor-associated factor 6(TRAF6) to weaken the AKT/mTORC1 signaling downstream of the TRAF6 pathway.Conclusions: MiR-146a-5p targets SMAD4 and TRAF6 to inhibit porcine intramuscular adipogenesis by attenuating TGF-β and AKT/mTORC1 signaling, respectively. 2Preparation and visible-light photocatalytic property of nanostructured Fe-doped TiO_2 from titanium containing electric furnace molten slag显示文摘Nanostructured Fe-doped titanium dioxide was synthesized from titanium containing electric furnace molten slag(TCEFMS)by using an alkali fusion,followed by a hydrolyzation-acidolysis-calcination route.The efects of alkali/slag mass ratio,calcinating temperature,calcinating time,and water/slag mass ratio on the extraction efciency and purity of products were systematically studied in this paper.It is indicated that the best extraction efciency of nanostructured Fedoped titanium dioxide is 99.35%,when the molten slag is calcinated at 700 C for 1 h with the mass ratio of alkali/molten slag of 1.5:1.The influence of alkali/slag mass ratio on the photocatalytic activity of final products was evaluated by the photodegradation of methyl blue under visible light irradiation.A maximum photodegradation efciency of 88.12%over30 min was achieved under the optimum conditions.Yang Li Yi Yue Zai-qing Que Mei Zhang Min Guo 2013International Journal of Minerals,Metallurgy and Materials2013,20,10:10
3LAG-3 expression on tumor-infiltrating T cells in soft tissue sarcoma correlates with poor survival显示文摘Objective: To elucidate the role and prognostic significance of lymphocyte activation-gene-3(LAG-3) in soft tissue sarcoma(STS).Methods: The expression of LAG-3 in patient and matched normal blood samples was analyzed by flow cytometry. The localization and prognostic values of LAG-3^+ cells in 163 STS patients were analyzed by immunohistochemistry. In addition, the expression of tumor-infiltrating CD3^+ T, CD4^+ T, and CD8^+ T cells and their role in the prognosis of STS were evaluated by immunohistochemistry. The effect of LAG-3 blockade was evaluated in an immunocompetent MCA205 fibrosarcoma mouse model.Results: Peripheral CD8^+ and CD4^+ T cells from STS patients expressed higher levels of LAG-3 than those from healthy donors.LAG-3 expression in STS was significantly associated with a poor clinical outcome(P = 0.038) and was correlated with high pathological grade(P < 0.001), advanced tumor stage(P = 0.016). Additionally, LAG-3 expression was highly correlated with CD8^+ T-cell infiltration(r = 0.7034, P < 0.001). 4Psychological impact of the COVID-19 pandemic on healthcare workers:a cross-sectional study in China显示文摘Background Healthcare workers fighting against the coronavirus disease 2019(COVID-19)pandemic are under tremendous pressure,which puts them at an increased risk of developing psychological problems.Aims This study aimed to investigate the prevalence of psychological problems in different healthcare workers(ie,physicians,medical residents,nurses,technicians and public health professionals)during the COVID-19 pandemic in China and explore factors that are associated with the onset of psychological problems in this population during this public health crisis.Methods A cross-sectional,web-based survey was conducted in February 2020 among healthcare workers during the COVID-19 pandemic.Psychological problems were assessed using the Generalized Anxiety Disorder Scale,Patient Health Questionnaire and Insomnia Severity Index.Logistic regression analyses were used to explore the factors that were associated with psychological problems.Results The prevalence of symptoms of anxiety,depression,insomnia and the overall psychological problems in healthcare workers during the COVID-19 pandemic in China was 46.04%,44.37%,28.75%and 56.59%,respectively.The prevalence of the overall psychological problems in physicians,medical residents,nurses,technicians and public health professionals was 60.35%,50.82%,62.02%,57.54%and 62.40%,respectively.Compared with healthcare workers who did not participate in front-line work,front-line healthcare workers had a higher risk of anxiety,insomnia and overall psychological problems.In addition,attention to negative or neutral information about the pandemic,receiving negative feedback from families and friends who joined front-line work,and unwillingness to join front-line work if given a free choice were three major factors for these psychological problems.Conclusions Psychological problems are pervasive among healthcare workers during the COVID-19 pandemic.Receiving negative information and participating in front-line work appear to be.important risk factors for psychological problems.The psychological health of different healthcare workers should be protected during the COVID-19 pandemic with timely interventions and proper information feedback.Jianyu Que Le Shi Jiahui Deng Jiajia Liu Li Zhang Suying Wu Yimiao Gong Weizhen Huang Kai Yuan Wei Yan Yankun Sun Maosheng Ran Yanping Bao Lin Lu 2020General Psychiatry2020,33,3:8
5Gangba sheep in the Tibetan plateau:Validating their unique meat quality and grazing factor analysis显示文摘Gangba sheep are known for having typical sensory characteristics attributed to free range conditions and grazing on wild plants.The genuine Gangba mutton was selected as the experimental group,and the commercial Tibetan mutton was selected as the control group,the nutritive composition of basic chemical components,amino acids and fatty acids in mutton were investigated to correlate its unique meat quality and eating satisfaction.The results showed that fatty acids were significantly higher(P<0.05)in Gangba mutton than in commercial mutton,and the higher content of flavoring amino acids(glutamic acid and aspartame)were primarily responsible for the taste attributes umami of meat juices.Moreover,the trace elements analysis in mutton and grazing factors(forage,water source and soil)were conducted,to explain the source of essential trace elements in mutton.The concentrations of essential trace elements show that the Gangba mutton was a valuable source for highly available Cu and Zn in human nutrition,and well managed with few detected of toxicity metal.The concentrations of essential trace elements in mutton are closely related to the trace elements in environmental grazing factors.In conclusion,the congenital grazing conditions(a highly mineralized water resource,natural forages and clean soils)were shown to contribute to the unique meat characteristics of Gangba sheep.Qiangying Zhang Ming Que Wei Li Shuang Gao Xin Tan Duo Bu 2021Journal of Environmental Sciences2021,33,3:8
6Delivery room resuscitation and short-term outcomes of extremely preterm and extremely low birth weight infants: a multicenter survey in North China显示文摘Background:Delivery room resuscitation assists preterm infants,especially extremely preterm infants(EPI)and extremely low birth weight infants(ELBWI),in breathing support,while it potentially exerts a negative impact on the lungs and outcomes of preterm infants.This study aimed to assess delivery room resuscitation and discharge outcomes of EPI and ELBWI in China.Methods:The clinical data of EPI(gestational age[GA]<28 weeks)and ELBWI(birth weight[BW]<1000 g),admitted within 72 h of birth in 33 neonatal intensive care units from five provinces and cities in North China between 2017 and 2018,were analyzed.The primary outcomes were delivery room resuscitation and risk factors for delivery room intubation(DRI).The secondary outcomes were survival rates,incidence of bronchopulmonary dysplasia(BPD),and risk factors for BPD.Results:A cohort of 952 preterm infants were enrolled.The incidence of DRI,chest compressions,and administration of epinephrine was 55.9%(532/952),12.5%(119/952),and 7.0%(67/952),respectively.Multivariate analysis revealed that the risk factors for DRI were GA<28 weeks(odds ratio[OR],3.147;95%confidence interval[CI],2.082–4.755),BW<1000 g(OR,2.240;95%CI,1.606–3.125),and antepartum infection(OR,1.429;95%CI,1.044–1.956).The survival rate was 65.9%(627/952)and was dependent on GA.The rate of BPD was 29.3%(181/627).Multivariate analysis showed that the risk factors for BPD were male(OR,1.603;95% CI,1.061–2.424),DRI(OR,2.094;95% CI,1.328–3.303),respiratory distress syndrome exposed to≥2 doses of pulmonary surfactants(PS;OR,2.700;95%CI,1.679–4.343),and mechanical ventilation≥7 days(OR,4.358;95% CI,2.777–6.837).However,a larger BW(OR,0.998;95% CI,0.996–0.999),antenatal steroid(OR,0.577;95%CI,0.379–0.880),and PS use in the delivery room(OR,0.273;95%CI,0.160–0.467)were preventive factors for BPD(all P<0.05).Conclusion:Improving delivery room resuscitation and management of respiratory complications are imperative during early management of the health of EPI and ELBWI.Shuai-Jun Li Qi Feng Xiu-Ying Tian Ying Zhou Yong Ji Yue-Mei Li Shu-Fen Zhai Wei Guo Fang Zhang Rong-Xiu Zheng Hai-Ying He Xia Liu Jun-Yi Wang Hua Mei Hong-Yun Wang Hua Xie Chao-Mei Zeng Li Ma Ping-Ping Zhang Jin-Yu Li Xiao-Ying Wang Li-Hua Li Hong Cui Shu-Lan Yang Lu Chen Xiao-Hong Gu Yan-Ju Hu Sheng-Shun Que Li-Xia Sun Ming Yang Wen-Li Zhao Qiu-Yan Ma Hai-Juan Wang Jiu-Ye Guo 2021Chinese Medical Journal2021,,13:7

During meiosis, the callose wall is formed outside of the meiocyte/microspore to form a tetrad. Then, primexine is deposited between the callose wall and the microspore plasma membrane which will become undulated. The sporopollenin deposits on top of the undulated membrane and develops into the pollen wall pattern, while the callose wall is gradually degraded. In recent years, much progress has been made in the study of pollen wall pattern formation, at both molecular and genetic levels. 9Using Materialise’s interactive medical image control system to reconstruct a model of a patient with rectal cancer and situs inversus totalis: A case report显示文摘BACKGROUND Situs inversus totalis(SIT)is a rare congenital anomaly that refers to a completely reversed location of abdominal and thoracic organs.An extremely small number of patients with this condition,especially those with rectal neoplasms,have been reported.Surgery in these patients is technically challenging.Therefore,we reconstructed a three-dimensional(3D)digital model with the Materialise’s interactive medical image control system(Mimics)as a guide for laparoscopic resection.CASE SUMMARY We report the case of a 68-year-old woman with rectal neoplasms and SIT diagnosed by electronic colonoscopy biopsy and enhanced computed tomography(CT),which showed that there was a soft tissue mass protruding into the lumen in the lower rectal segment,a lesion that involved the serosal layer,multiple enlarged peripheral lymph nodes,and visceral situs abnormalities.Based on the CT images,we reconstructed a 3D model with Mimics to assist with our surgical planning.Then,we performed laparoscopyassisted radical resection of the rectal neoplasms and total excision of the lesion.Adjuvant chemotherapy with the XELOX regimen(oxaliplatin 150 mg,D1+Xeloda 1.0 g,Bid,D1-14)was initiated 1 mo after the operation.The patient recovered well after surgery,and her physical condition remained stable.CONCLUSION Preoperative 3D reconstruction of the imaging results could help reduce the unknown risks during surgery caused by anatomical abnormalities and improvethe perioperative safety for patients.Tao Chen Yu-Tao Que Yi-Hao Zhang Fei-Yu Long Yi Li Xin Huang Ya-Nan Wang Yan-Feng Hu Jiang Yu Guo-Xin Li 2020World Journal of Clinical Cases2020,8,4:5

12A Teosinte-derived Allele of a MYB Transcription Repressor Confers Multiple Disease Resistance in Maize.显示文摘Natural alleles controlling multiple disease resistances (MDR) are valuable for crop breeding. However, only one MDR gene have been cloned in maize, and molecular mechanisms of MDR are not clear. By map-based cloning, we have cloned a teosinte-derived allele of a resistance gene, Mexicana lesion mimic 1 (ZmMM1), which has a lesion mimic phenotype and confers resistance to northern leaf blight (NLB), gray leaf spot (GLS) and southern corn rust (SCR). Strong MDR conferred by the teosinte allele is linked with the polymorphisms in the 3' untranslated region of the ZmMM1 gene that cause increased accumulation of ZmMM1 protein. ZmMM1 acts as a transcription repressor and negatively regulates transcription of specific target genes including ZmMM1-target gene 3 (ZmMT3), which functions as a negative regulator of plant immunity and associated cell death. 13Extraction and separation of nickel and cobalt from saprolite laterite ore by microwave-assisted hydrothermal leaching and chemical deposition显示文摘Extraction and separation of nickel and cobalt from saprolite laterite ore were studied by using a method of microwave-assisted hydrothermal leaching and chemical deposition. The effects of leaching temperature and time on the extraction efficiencies of Ni2+ and Co2+ were investigated in detail under microwave conditions. It is shown that the extraction efficiencies of Ni2+ and Co2+ from the ore pre-roasted at 300 C for 5 h were 89.19% and 61.89% when the leaching temperature and time were about 70 C and 60 min, respectively. For the separation process of Ni and Co, the separation of main chemical components was performed by adjusting the pH values of sulfuric leaching solutions using a NaOH solution based on the different pH values of precipitation for metal hydroxides. The final separation efficiencies of Ni and Co were 77.29% and 65.87%, respectively. 14Influence of gangue existing states in iron ores on the formation and flow of liquid phase during sintering显示文摘Gangue existing states largely affect the high-temperature characteristics of iron ores. Using a micro-sintering method and scanning electron microscopy, the effects of gangue content, gangue type, and gangue size on the assimilation characteristics and fluidity of liquid phase of five different iron ores were analyzed in this study. Next, the mechanism based on the reaction between gangues and sintering materials was unraveled. The results show that, as the SiO2 levels increase in the iron ores, the lowest assimilation temperature(LAT) decreases, whereas the index of fluidity of liquid phase(IFL) increases. Below 1.5wt%, Al2O3 benefits the assimilation reaction, but higher concentrations proved detrimental. Larger quartz particles increase the SiO2 levels at the local reaction interface between the iron ore and CaO, thereby reducing the LAT. Quartz-gibbsite is more conductive to assimilation than kaolin. 16Herbal formula of Bushen Jianpi(补肾健脾方)combined with sorafenib inhibits hepatocellular carcinoma growth by promoting cell apoptosis and blocking the cell cycle显示文摘OBJECTIVE:To investigate the efficacy of an herbal formula of Bushen Jianpi(补肾健脾方,BSJP)combined with sorafenib on hepatocellular carcinoma(HCC)in vitro and in vivo,and to study the underlying mechanisms of action.METHODS:BSJP,a mixture of 12 raw herbs,was extracted in 70%alcohol/30%water and freeze-dried into a powder.The in vitro effects of BSJP alone,sorafenib alone,and their combination on cell survival,apoptosis,and cell cycle distribution were evaluated in HCC cell lines HCCLM3,HepG2,and SMMC-7721.The expression of B-cell lymphoma-2(Bcl-2),caspase-3,and caspase-9 in HCCLM3 cells was measured using Western blots after drug administration.The in vivo effects of BSJP and sorafenib were evaluated in a tumor surgical resection model using 4-week old male athymic BALB/c nude mice injected with HCCLM3 cells.Immunohistochemical analysis of tumor tissues was performed to evaluate the effects of BSJP alone,sorafenib alone,and their combination on the expression of caspase-3,caspase-9,and Bcl-2.RESULTS:BSJP decreased the survival rate of HCC cell lines,and the combination of BSJP and sorafenib further decreased the survival rate.BSJP significantly promoted cell apoptosis and blocked cell-cycle progression in HCCLM3,HepG2,and SMMC-7721 cells in a dose-dependent manner.Furthermore,the administration of BSJP and sorafenib inhibited the growth of HCCLM3 cell xenografts in nude mice,with no reduction in body weight.In vivo and in vitro experiments showed that BSJP combined with sorafenib could significantly decrease the expression of Bcl-2.CONCLUSION:Our findings suggest that the herbal formula of BSJP is a potential HCC antitumor agent.ZHOU Zhangjie LIU Xinhua WU Tingting QUE Zujue WU Zhonghua WU Weizhong FU Shujuan ZHANG Shiqiang YANG Yun JIANG Haiyan XIA Xiaoting LV Junqiang DU Boqian LI Yun LU Tao ZHANG Zhihui ZHONG Yi 2021Journal of Traditional Chinese Medicine2021,41,2:3

19Controllable synthesis of nanorod/nanodisk TiO_2 from titanium-containing electric furnace molten slag显示文摘Nanostructured TiO2 with different morphologies and crystal phases was successfully synthesized from titanium-containing electric furnace molten slag by using a hydrothermal method followed by reflux process in acid solution. The effects of acid concentration, reflux time, and acid type on the formation of TiO2 were systematically investigated by scanning electron microscopy, X-ray diffraction(XRD), transmission electron microscopy(TEM),high-resolution transmission electron microscopy(HRTEM),and X-ray fluorescence(XRF). It is indicated that TiO2 nanorod with rutile phase is achieved in nitric or hydrochloric acid, while anatase TiO2 nanodisk is formed in sulfuric acid.With the increase of the concentration of hydrochloric acid from 0.3 to 1.5 mol L-1, the dispersibility and crystallinity of the final product can be improved. With prolonging the reflux time from 6 to 14 h, the rutile TiO2 nanorod with uniform crystal size and high crystallinity is obtained. 20Tacrolimus reduces scar formation and promotes sciatic nerve regeneration显示文摘A sciatic nerve transection and repair model was established in Sprague-Dawley rats by transecting the tendon of obturator internus muscle in the greater sciatic foramen and suturing with nylon sutures. The models were treated with tacrolimus gavage (4 mg/kg per day) for 0, 2, 4 and 6 weeks. Specimens were harvested at 6 weeks of intragastric administration. Masson staining revealed that the collagen fiber content and scar area in the nerve anastomosis of the sciatic nerve injury rats were significantly reduced after tacrolimus administration. Hematoxylin-eosin staining showed that tacrolimus significantly increased myelinated nerve fiber density, average axon diameter and myelin sheath thickness. Intragastric administration of tacrolimus also led to a significant increase in the recovery rate of gastrocnemius muscle wet weight and the sciatic functional index after sciatic nerve injury. The above indices were most significantly improved at 6 weeks after of tacrolimus gavage. The myelinated nerve fiber density in the nerve anastomosis and the sciatic nerve functions had a significant negative correlation with the scar area, as detected by Spearman's rank correlation analysis.
